Abstract:Cysteine (Cys) plays important roles in many physiological processes of eukaryotic cells and its detection in cells is of fundamental significance. However, glutathione (GSH), homocysteine, N-acetyl-L-cysteine and other thiols greatly hamper the detection of Cys. In particular, GSH strongly interferes with the detection of cellular Cys (30-200 mu M) due to its high intracellular concentration (1-10 mM). In this work, an off on fluorescent probe (HOTA) for the detection of Cys is presented. This probe possesses both excellent sensitivity and satisfactory selectivity for cellular Cys detection: with the addition of 200 mu M Cys, the fluorescence intensity of the probe (10 mu M) enhanced 117-fold and the detection limit was calculated to be 13.47 mu M, which is lower than the cellular Cys concentration; the probe also selectively detected 30-200 mu M cysteine over 1-10 mM glutathione. Consequently, cell imaging experiments were performed with probe HOTA. Furthermore, the results of the thiol-blocking and GSH synthesis inhibiting experiments confirmed that the intracellular emission mainly originates from the interaction between Cys and HOTA.

Abstract:A new fluorescent probe based on fluorescein fluorophore displays an excellent selectivity and sensitivity for GSH in buffer solution containing CTAB. The fluorescence intensity of sensor is linearly proportional to GSH concentrations ranging from 0 to 10 μM, and as low as 22 nM concentration of GSH was evaluated with 3.3 σ/s. The optical changes are attributed to the release of free fluorescein after the electron withdrawing dinitrophenyl ether reacted efficiently with GSH in CTAB micelles. The enhanced selectivity toward GSH over Cys and Hcy considered to be related to the carboxyl groups in thiols. The confocal microscopy experiments implied that this probe is a promising candidate for imaging of GSH in living cells.

Abstract:Cysteine is associated with various diseases in the organism. Monitoring Cys levels is important for the study of related diseases. Here, a ratiometric fluorescent probe based on a novel fluorophore was designed for detecting Cys. The fluorophore exhibits a large Stokes shift and good photostability. The probe QL‐A shows high specificity and has been used to visualize Cys in living cells. It is a potential tool for exploring the role of Cys in biological processes and diseases. As a biothiol, cysteine (Cys) is essential to both physiological and pathological processes and has been associated with many diseases, including neurological disorders, rheumatoid arthritis, and renal dysfunction. Therefore, the development of a high‐performance probe for detecting Cys levels can help prevent and diagnose disease. In this study, a ratiometric fluorescent probe based on a novel fluorophore was developed for detecting Cys, and it showed high specificity and a rapid response time toward Cys. This probe demonstrates excellent biocompatibility and has been utilized effectively for the imaging of Cys in living cells.

Abstract:Cysteine (Cys) and homocysteine (Hcy) are involved in maintaining homeostasis in the body and are relevant to various diseases. While the level of Cys and Hcy is much lower than GSH in the living system, which leads to a major challenge to selectively identify Cys/Hcy in the presence of large amounts of GSH. In this paper, an AIE fluorescent probe SQM-NBD was obtained by connecting NBD to the hydroxyl group of the fluorophore SQM-OH for selective detection of Cys/Hcy. Probe SQM-NBD had no fluorescence itself. But, under the disturbance of GSH, the fluorescence signal of probe SQM-NBD could be turned on by Cys/Hcy. The study of the response mechanism showed that probe SQM-NBD could release both SQM-OH and Cys/Hcy-NBD after reacting with Cys/Hcy. While Cys/Hcy continued to quench the fluorescence of SQM-OH through the combination of Michael addition and the ion rich environment, resulting in only the fluorescence signal of Cys/Hcy-NBD being observed. SQM-NBD appeared superior sensitivity to Cys/Hcy, with LOD of 54 nM and 72 nM, respectively. Significantly, probe SQM-NBD realized the application of fluorescence imaging of Cys/Hcy in HeLa cells, indicating that probe SQM-NBD has the potential for Cys/Hcy identification under physiological and pathological conditions.

Abstract:In this paper, by modifying the thioxanthene-benzothiozolium fluorophore, BCy-Cys, a lysosome-targeted near-infrared (NIR) fluorescent probe was synthesized for the detection of cysteine (Cys) from homocysteine (Hcy)/glutathione (GSH). As expected, BCy-Cys exhibited high selectivity and high sensitivity for detection of Cys over Hcy/GSH, with an extremely low limit of detection at 0.31 μM, marked by obvious color changes. HRMS was conducted to confirm that the fluorescence intensity at 795 nm was significantly enhanced by the enhancement of intramolecular charge transfer (ICT). Importantly, BCy-Cys could be used to visualize both exogenous and endogenous lysosomal Cys, signifying its potential application in complex organismal systems.

Abstract:A novel fluorescent probe was developed by integrating chlorinated coumarin and benzothiazolylacetonitrile and exploited for simultaneous detection of cysteine (Cys), homocysteine (Hcy), and glutathione (GSH). Featuring four binding sites and different reaction mechanisms for different biothiols, this probe exhibited rapid fluorescence turn-on for distinguishing Cys, Hcy, and GSH with 108-, 128-, 30-fold fluorescence increases at 457, 559, 529 nm, respectively, across different excitation wavelengths. Furthermore, the probe was successfully applied to the fluorescence imaging of endogenous Cys and GSH and exogenous Cys, Hcy, and GSH in living cells.

Abstract:Small-molecule biothiols, including cysteine (Cys),homocysteine (Hcy), and glutathione (GSH), participate invarious pathological and physiological processes. It is still a challenge to simultaneously distinguish Cys and Hcy because of their similar structures and reactivities, as well as the interference from the high intramolecular concentration of GSH. Herein, anovelfluorescent probe,CySI, based on cyanine and thioester was developed to differentiate Cys and Hcy through a single-wavelength excitation and two distinctly separated emissionchannels. The probe exhibited a turn-onfluorescence responseto Cys at both 625 nm (the red channel) and 740 nm (the near-infrared channel) but only showedfluorescence turn-on to Hcy at740 nm (the near-infrared channel) and nofluorescent response toGSH. With the aid of built-in self-calibration of single excitation and dual emissions, simultaneous discriminative determinations ofCys and Hcy were realized through red and near-infrared channels.CySIexhibited excellent selectivity toward Cys and Hcy with afast response. This probe was further exploited to visualize exogenous Cys and Hcy in cells through dual emission channels underone excitation. Moreover, it could efficiently target mitochondria and was applied to monitor the endogenous Cys fluctuations independently in mitochondria through the red emission channel.

Abstract:A new selective probe with a quinoxalinone structure, QP-1, has been developed for detection of Cys from biothiols. QP-1 features superb selectivity to Cys and a wide pH range. QP-1 has selectivity to Cys over Hcy, GSH, other amino acids and ions. HRMS spectra confirmed that the detection process was a conjugate addition addition-elimination reaction. Moreover, QP-1 has been successfully applied in the imaging of Cys in living cells. Finally, QP-1 has been used to detect Cys in rat urine samples.

Abstract:A benzothiazole-based near-infrared (NIR) ratiometric fluorescent probe (HBT-Cys) was developed for discriminating cysteine (Cys) from homocysteine (Hcy) and glutathione (GSH). The probe was designed by masking phenol group in the conjugated benzothiazole derivative with methacrylate group that could be selectively removed by Cys, and therefore an intramolecular charge transfer (ICT) fluorescence was switched on in the NIR region. In the absence of Cys, the probe exhibited a strong blue fluorescence emission at 431 nm, whereas a NIR fluorescence emission at 710 nm was significantly enhanced accompanied by a decrease of emission at 431 nm in the presence of Cys, allowing a ratiometric fluorescence detection of Cys. The fluorescence intensity ratio (I710nm/I431nm) showed a good linear relationship with Cys concentration of 1-40 μM with the detection limit of 0.5 μM. The sensing mechanism was explored based on MS experimental analysis and DFT theoretical calculation. Moreover, the fluorescent probe was successfully used for fluorescence bioimaging of Cys in living cells.

Abstract:We demonstrate a concept-proof work of using fluorescence (FL) "turn-on" probes for the discriminatory detection of cysteine (Cys) over homocysteine (Hcy). The fluorogens are provided with aggregation-induced emission (AIE) characteristic and functionalized with two aldehyde-groups (DMTPS-ALD and TPE-ALD). All the detections were carried out in a biocompatible medium (10 mm HEPES buffer and DMSO, pH 7.4). In principle, the formation of thiazinane/thiazolidine through the chemical reaction of aldehydes on the probe molecules and the residue of Cys/Hcy determines the selective recognition of Cys and Hcy over other amino acids and glucose. The FL responses originate from the AIE property of thiazinane/thiazolidine resultants, which have low solubility and precipitate (aggregate) in the detection medium. The discrimination between Cys and Hcy comes from the difference in reaction kinetics of TPE-ALD/DMTPS-ALD with Cys and Hcy, thereby the FL responses show different time courses and intensity enhancement. It is worth noting that TPE-ALD outshined the other two probes in performance with fast response, a high FL enhancement up to 16-fold, high sensitivity, and good specificity and selectivity. Moreover, its FL response threshold at 250 mu m is very close to the lower limit of the normal level of Cys in human plasma, which implies that TPE-ALD could be applied as a potential indicator of Cys deficiency.

Abstract:The very close structural similarities between cysteine and homocysteine present a great challenge to achieve their selective detection using regular fluorescent probes, limiting the biological and pathological studies of these two amino thiols. A coumarin-based fluorescent probe was designed featuring pH-promoted distinct turn-on followed by ratiometric fluorescence responses for Cys and turn-on fluorescence response for Hcy through two different reaction paths. Abstract:Cysteine, as a vital endogenous small molecule mercaptan, plays a crucial role in various physiological processes. The high sensitivity and selectivity of fluorescent probes provide a method to monitor cysteine, which is helpful to understand the mechanism of cysteine in physiological processes more comprehensively. However, the detection of cysteine can be interfered by other small molecule biothiols. Therefore, the design of fluorescent probe based on the structural characteristics and reactivity of cysteine has become research focus currently. Given the biological compatibilities, biological targets, the metabolic pathway of 3-hydoxythalidomide, and its unique fluorescent properties, herein, we have designed a chemodosimeter, 2-(2,6-dioxopiperidin-3-yl)-1,3-dioxoisoindolin-4-yl acrylate, for the detection of cysteine based on a tandem reaction of thiol-ene click chemistry and aminolysis involving 3-hydroxythalidomide as a parent compound. Experimental data exhibited that the probe showed unique selectivity and sensitivity for cysteine over other amino acid and biothiols. In addition, the fluorescent intensity at 511 nm increased linearly as a function of cysteine concentration in the range of 0-6 × 10-7 M (regression factor, R2 = 0.999), with a limit of detection of 6.1 nM. The sensing mechanism was confirmed through 1H NMR titration and density functional theory calculations. Additionally, the probe was also successfully utilized for the detection of cysteine in sewage and for bioimaging in HeLa cells.

Abstract:Cysteine (Cys), an essential biological amino acid, participates several crucial functions in various physiological and pathological processes. The sensitive and specific detection of Cys is of great significance for understanding its biological function to disease diagnosis. Herein, we designed and synthesized a simple fluorescence sensor 2-(benzothiophen-2-yl)-4-oxo-4H-chromen-3-yl acrylate (BTCA) composed of a flavonol skeleton as the fluorophore and acrylic ester group as the recognition receptor. Probe BTCA displayed high selectivity and extremely fast response toward Cys in phosphate buffer solution in the presence of other competitive species even Homocysteine (Hcy) and Glutathione (GSH) owing to a specific conjugate addition-cyclization reaction between the acrylate moiety and Cys. The photoluminescence mechanism of probe BTCA toward Cys was modulated by excited state intramolecular proton transfer (ESIPT) process. The sensing property for Cys was studied by UV-Visible, fluorescence spectrophotometric analyses and time-dependent density functional theory (TD-DFT) calculations, those results indicated that probe BTCA possessed excellent sensitivity, higher specificity, dramatically "naked-eye" fluorescence enhancement (30-fold), high anti-interference ability, especially immediate response speed (within 40 s). Additionally, the practicability of sensor BTCA in exogenous and endogenous Cys imaging in living cells and zebrafish was elucidated as well, suggesting that it has remarkedly diagnostic significance in physiological and pathological process.

Abstract:Based on Excited-State Intramolecular Proton Transfer (ESIPT) fluorophore and an acrylate moiety as Michael addition and leaving site, a simple but colorimetric turn-on fluorescent probe FVCA was designed and synthesized. Upon addition of Cys to the solution of FVCA, the absorption spectra changed from 375 nm to 460 nm (Δ85 nm) and afforded visible color change from colorless to yellow. Meanwhile, the fluorescence emission spectra emerged at 554 nm with remarkable changes, accompanying with an obvious fluorescence color change from colorless to bright green. In addition, FVCA exhibited rapid response (∼ 60 s), high sensitivity (detection limit: 8.37 nM), large strokes shift (∼ 94 nm) and excellent selectively to Cys over other biological mercaptans. Moreover, FVCA had strong anti-interference ability for Cys without any interference from other biologically-relevant thiols even Hcy and GSH, even obvious visible and fluorescence color changes. More, the sensing mechanism was proved to be the ESIPT-based addition-cyclization reaction process by 1H NMR, HRMS and TD-DFT. Importantly, FVCA can be used to image exogenous and endogenous Cys in living A549 cells with low toxicity.

Abstract:Biothiols, associated with multiple physiological and pathological processes, have structural similarities. Monitoring Biothiols selectively in organisms is of great significance. Burdened by the aggregation-caused quenching (ACQ) effect, the applications of conventional biothiols fluorescent probes are extremely limited. Herein, we developed a "turn-on" type aggregation-induced emission (AIE) fluorescent probe BQM-NBD, which was composed of a BQM-OH fluorophore molecule with AIE effect and the recognition group 7-nitro-1,2,3-benzoxadiazole (NBD). Non-fluorescent BQM-NBD produces strong fluorescence after the addition of cysteine (Cys) or homocysteine (Hcy). BQM-NBD exhibited excellent linearity for selective detection of Cys (0-100 Μm) and Hcy (0-50 μM) with detection limits of 6.0 × 10-8 M and 8.4 × 10-8 M, respectively. Simultaneously, after treatment with glutathione (GSH), it appeared no fluorescence. The results demonstrated BQM-NBD exhibited good selectivity to Cys/Hcy. Furthermore, BQM-NBD was successfully performed in the imaging of Cys in living cells with low cytotoxicity, which provides a feasible strategy for the selective detection of Cys in the living system.
