Abstract:Cysteine (Cys) plays important roles in many physiological processes of eukaryotic cells and its detection in cells is of fundamental significance. However, glutathione (GSH), homocysteine, N-acetyl-L-cysteine and other thiols greatly hamper the detection of Cys. In particular, GSH strongly interferes with the detection of cellular Cys (30-200 mu M) due to its high intracellular concentration (1-10 mM). In this work, an off on fluorescent probe (HOTA) for the detection of Cys is presented. This probe possesses both excellent sensitivity and satisfactory selectivity for cellular Cys detection: with the addition of 200 mu M Cys, the fluorescence intensity of the probe (10 mu M) enhanced 117-fold and the detection limit was calculated to be 13.47 mu M, which is lower than the cellular Cys concentration; the probe also selectively detected 30-200 mu M cysteine over 1-10 mM glutathione. Consequently, cell imaging experiments were performed with probe HOTA. Furthermore, the results of the thiol-blocking and GSH synthesis inhibiting experiments confirmed that the intracellular emission mainly originates from the interaction between Cys and HOTA.

Abstract:A novel one- and two-photon fluorescent probe CB1 has been developed for discriminating Cys and Hcy in a successive manner with high selectivity. The discrete time-dependent fluorescent responses enable us to sequentially detect Cys and Hcy in different time windows. Two-step reaction and kinetic modes were used to explain the sensing mechanism. As a promising biosensor for cell imaging, CB1 has been confirmed to exhibit membrane permeability to intact cells, low cytotoxicity to viable cells and photostability to ultraviolet light excitation. Furthermore, the results from the control assay have shown that the one- and two-photon fluorescence of CB1 within cells is associated with intracellular mercapto biomolecules but yet there is little interference with physiological pH value, viscosity and common bioanalytes. Finally one- and two-photon fluorescent images of CB1 within living SiHa cells have been presented.

Abstract:Cysteine is associated with various diseases in the organism. Monitoring Cys levels is important for the study of related diseases. Here, a ratiometric fluorescent probe based on a novel fluorophore was designed for detecting Cys. The fluorophore exhibits a large Stokes shift and good photostability. The probe QL‐A shows high specificity and has been used to visualize Cys in living cells. It is a potential tool for exploring the role of Cys in biological processes and diseases. As a biothiol, cysteine (Cys) is essential to both physiological and pathological processes and has been associated with many diseases, including neurological disorders, rheumatoid arthritis, and renal dysfunction. Therefore, the development of a high‐performance probe for detecting Cys levels can help prevent and diagnose disease. In this study, a ratiometric fluorescent probe based on a novel fluorophore was developed for detecting Cys, and it showed high specificity and a rapid response time toward Cys. This probe demonstrates excellent biocompatibility and has been utilized effectively for the imaging of Cys in living cells.

Abstract:Cysteine (Cys), one of the biological thiols, which plays critical roles in biological system regulating the balance of redox homeostasis. In order to monitor the level of Cys in the living cells and organisms, a chromogenic fluorescence probe Rhocl-Cys based on Rhodamine chloride exhibiting the preferable performance of fluorescence turn-on response reacting with Cys was presented. Rhocl-Cys responded rapidly to Cys within 20 min, and had stable fluorescence intensity within pH 6.0-10.0, high selectivity towards Cys and the anti-inference capability with a low detection limit of 0.80 mu M. In particular, Rhocl-Cys could qualitatively and quantitatively monitor the level of endogenous and exogenous Cys in living cells and successfully apply to zebrafish detecting Cys. Therefore, these results might further provide the basis exploring the role of Cys in biological system and facilitate as clinical diagnostic molecular tools.

Abstract:As an important sulfur-containing amino acid, aberrant levels of cysteine are closely related with an array of dieases. Although many Cys-specific fluorescent probes have been designed, most of them wouldn't be able to detect Cys in buffer solution, which limits the application of the probe in biological system. In this work, a novel near-infrared fluorescent probe Cys-WR with a large Stokes shift (about 110 nm) was developed for the detection of Cys by conjugating the fluorophore WR-4 with crotonate moiety. The probe exhibited off-on response to Cys with wide linear range of 0-100 mu M, as well as the color of the probe solution changes from deep pink to brown observed by naked eyes. Moreover, the probe showed little cytotoxicity. Imaging of Cys using this probe in living A549 cells and zebrafish had also been well demonstrated.

Abstract:Based on Excited-State Intramolecular Proton Transfer (ESIPT) fluorophore and an acrylate moiety as Michael addition and leaving site, a simple but colorimetric turn-on fluorescent probe FVCA was designed and synthesized. Upon addition of Cys to the solution of FVCA, the absorption spectra changed from 375 nm to 460 nm (Δ85 nm) and afforded visible color change from colorless to yellow. Meanwhile, the fluorescence emission spectra emerged at 554 nm with remarkable changes, accompanying with an obvious fluorescence color change from colorless to bright green. In addition, FVCA exhibited rapid response (∼ 60 s), high sensitivity (detection limit: 8.37 nM), large strokes shift (∼ 94 nm) and excellent selectively to Cys over other biological mercaptans. Moreover, FVCA had strong anti-interference ability for Cys without any interference from other biologically-relevant thiols even Hcy and GSH, even obvious visible and fluorescence color changes. More, the sensing mechanism was proved to be the ESIPT-based addition-cyclization reaction process by 1H NMR, HRMS and TD-DFT. Importantly, FVCA can be used to image exogenous and endogenous Cys in living A549 cells with low toxicity.

Abstract:We reported here a turn-on fluorescent probe (1) for the detection of cysteine (Cys) by incorporating the recognition unit of 2,4-dinitrobenzenesulfonyl ester (DNBS) to a coumarin derivative. The structure of the obtained probe was confirmed by NMR and HRMS techniques. The probe shows a remarkable fluorescence off-on response (∼52-fold) by the reaction with Cys in 100% aqueous buffer. The sensing mechanism was verified by the HPLC test. Probe 1 also displays high selectivity towards Cys. The detection limit was calculated to be 23 nM. Moreover, cellular experiments demonstrated that the probe is highly biocompatible and can be used for monitoring intracellular Cys.

Abstract:Cysteine (Cys) distribute widely in organisms as the crucial components of proteins, and play important roles in pathophysiological processes of human body. Low level of Cys might induce hepatic injury, edema and growth retardation, while superfluous level of Cys is found to be closely relevant to Alzheimer's and Parkinson's diseases. In this work, a novel near-infrared (NIR) fluorescent probe PFQ-C was developed for highly selective detection of Cys in living cells and mice by utilizing the cyclization removal reaction between acrylate group and Cys. The superior sensitivity (limit of detection, 0.036 μM), NIR emission (655 nm), large Stokes shift (135 nm) and low cytotoxicity of the probe highlight its broad potential for future clinical applications. The response mechanism of the probe towards Cys was clarified by spectroscopy, chromatography and theoretical calculation. In addition, results of fluorescence imaging of cells and mice revealed the good performance of the probe for monitoring the distributions and variations of Cys activity in vivo, which is very useful for the researches on diseases associated with Cys.

Abstract:Cysteine is an essential amino acid for humans and can be generated from the conversion of methionine or the interconversions with cystine. The detection of cysteine in the living system is of great importance for the diagnosis of related diseases. Based on the classical cysteine response mechanism, the sulfhydryl group of cysteine undergoes Michael addition with the acrylate to rapidly respond to form a seven-membered ring that specifically recognizes cysteine. In the present work, we designed a novel ratiometric fluorescent probe N-1, which contained the benzo[cd]indol fluorophore and the response group acrylate. As the concentration of cysteine increased, the fluorescence of I485 was significantly enhanced while the fluorescence intensity at 635 nm was weakened. Moreover, probe N-1 showed excellent sensitivity and selectivity to Cys with the calculated LOD of 4.0 × 10-9 M. Probe N-1 was successfully used for imaging cysteine in the living Hela cells with low cytotoxicity, indicating that this ratiometric fluorescent probe would be a novel and useful tool to monitor cysteine in the physiological or pathological processes.

Abstract:Cysteine (Cys) plays a crucial role in numerous physiological and pathological processes. Therefore, it is imperative to design a highly selective and sensitive near-infrared (NIR) fluorescent probe to monitor Cys. In this study, we have developed a novel NIR fluorescent probe XA based on Xanthene hybrid tetrahydro-acridine salt dye for specifically tracking of Cys, where a chlorine-substituted tetrahydro-acridine acts as a high Cys-reactive site and water-soluble group. Probe XA exhibits a remarkable "turn-on" NIR emission (830 nm) with an extra-large Stokes shift (305 nm) for monitoring Cys. It also has a high selectivity, rapid response time (6 min) and high sensitivity (LOD as 0.5 μM). We fully characterized and discussed the sensing mechanism of XA toward Cys using HPLC and MS spectrums, as well as quantum theory calculations. Furthermore, the excellent properties of NIR fluorescent detection allow this novel probe to successfully monitor fluctuations of exogenous and endogenous Cys concentration levels in living cells and in vivo.

Abstract:The fluorescent probes with good water-solubility, long-wavelength emission and large Stokes shift are greatly desirable for in vivo detection. Herein, we designed a novel 1,8-naphthalimide-based near-infrared (NIR) optical and fluorescent probe (NTC) for sensing cysteine (Cys). Using acrylate as the recognition site, the probe demonstrated high selectivity and sensitivity for Cys with a low detection limit (0.093 μM) in aqueous buffer solution due to the excellent water-solubility. Upon the reaction with Cys, the recovery of intramolecular charge transfer (ICT) in the probe led to about 40-fold fluorescence enhancement. Furthermore, the reaction result was investigated by 1H NMR, and HRMS analyses, and the sensing mechanism was validated by quantum calculations. Finally, NTC was applied to image exogenous and endogenous Cys in HeLa cells and zebrafish selectively, implying that the probe possessed great potential application in biological fluorescence sensing.

Abstract:A benzothiazole-based near-infrared (NIR) ratiometric fluorescent probe (HBT-Cys) was developed for discriminating cysteine (Cys) from homocysteine (Hcy) and glutathione (GSH). The probe was designed by masking phenol group in the conjugated benzothiazole derivative with methacrylate group that could be selectively removed by Cys, and therefore an intramolecular charge transfer (ICT) fluorescence was switched on in the NIR region. In the absence of Cys, the probe exhibited a strong blue fluorescence emission at 431 nm, whereas a NIR fluorescence emission at 710 nm was significantly enhanced accompanied by a decrease of emission at 431 nm in the presence of Cys, allowing a ratiometric fluorescence detection of Cys. The fluorescence intensity ratio (I710nm/I431nm) showed a good linear relationship with Cys concentration of 1-40 μM with the detection limit of 0.5 μM. The sensing mechanism was explored based on MS experimental analysis and DFT theoretical calculation. Moreover, the fluorescent probe was successfully used for fluorescence bioimaging of Cys in living cells.

Abstract:On March 11, 1997, Senator Pete Domenici introduced U.S. Senate Bill 422, the Genetic Confidentiality and Non-Discrimination Act of 1997. The bill specifies that “existing legal protections for genetic information are inadequate to ensure genetic privacy and to prevent genetic discrimination.” The first stated purpose of the bill is “to define the circumstances under which DNA samples may be collected, stored, and analyzed and genetic information may be collected, stored, analyzed, and disclosed.” The bill reinforces statutes already passed in 19 states that guarantee patients certain rights of control over their own personal genetic information (Congressional Record, S2141). This bill fundamentally changes the mechanism whereby molecular biologists acquire tissue, and defines new obligations between researchers and human subjects. Even if this bill is not passed in its present form, it appears inevitable that legislation with a similar intent will soon emerge. Any such legislation will carry the full weight and penalties of law. Because many current research practices may violate these proposed new regulations, researchers should understand the provisions of S. 422. Furthermore, it is crucial that researchers understand the bill now, before it becomes law, because this may be their last opportunity to lobby for any modifications of the bill.

Abstract:A novel class of curcumin-derived fluorescent probes was designed. This kind of probe introduces easy leaving groups methylsulfonyl and phenylsulfonyl respectively to achieve the detection effect through the nucleophilic attack of amino acids. At the same time, BF2 group is introduced to increase the emission wavelength of the probe. Probes 4 and 5 can respond quickly with amino acids, but can specifically recognize Cys. In UV detection, the maximum absorption wavelength of the probes can be blue-shifted by 81 nm with the addition of Cys and still show a strong fluorescence signal. The detection limits for compounds 4 and 5 were determined to be 0.40 μM and 0.87 μM, respectively, with a goodness-of-fit of 0.99. In addition, a rapid response of the probe to Cys could be observed with the naked eye within 1 min. These results provide a new method for rapid detection of Cys; And this kind of probe has the drug structure of curcumin, which can provide ideas for the design of drug-probe.

Abstract:As an indispensable sulfur-containing amino acid, cysteine plays a vital role in various physiological processes of animals and plants. Its abnormal levels have been closely associated with the development and progression of serveral diseases. As a result, there is a surge in research focusing on the detection and measurement of cysteine within plants, which has captured significant attention in recent years. While several fluorescent probes have been designed to target cysteine, the majority of these probes face inherent challenges when applied in complicated biological systems. In this study, we have developed a novel fluorescent probe, MN-Cys, based on rhodamine derivative dyes, which can selectively detect Cys against GSH with a NIR-fluorescence turn-on response mode. Notably, MN-Cys shows low cytotoxicity, and possesses the capability to visualize and assess Cys via bioimaging, including NIR fluorescence imaging in both living cells and zebrafish models and two-photon imaging in Arabidopsis thaliana. The findings in this work underline the potential of MN-Cys as a promising NIR fluorescent probe which incorporates unique reaction sites and demonstrates exceptional sensing properties. Overall, this probe holds significant promise for the rapid and accurate detection of Cys both in vitro and in vivo.

Abstract:A novel fluorescent probe based on azamonardine (Aza) fluorophore was designed and synthesized for the highly selective detection of cysteine (Cys) in vivo and in vitro. After reacting with acryloyl chloride, the fluorescence of Aza is effectively quenched, resulting in the formation of the Aza-acryl probe. Upon the addition of Cys, the ester bond of Aza-acryl is cleaved, releasing a new compound (Compound 1) with strong fluorescence, thereby achieving fluorescence turn-on detection of Cys. The structure of Aza-acryl was characterized using X-ray crystallography and NMR spectroscopy. Additionally, density functional theory was employed to elucidate the quenching mechanism of the acyl group on the Aza. Aza-acryl exhibits high selectivity towards Cys and distinguishes it from other biothiols such as homocysteine (Hcy) and glutathione (GSH). The mechanism of Aza-acryl for detecting Cys was investigated through HPLC, NMR spectroscopy, high-resolution mass spectrometry, and reaction kinetics experiments. Aza-acryl demonstrates excellent imaging capabilities for Cys in cells and zebrafish, providing a reliable and selectable tool for the detection and imaging of Cys in biological systems.

Abstract:Background Cys, as an essential amino acid that can be ingested from daily food, plays an important role in maintaining the oxidative balance in cells. Abnormal Cys levels in organisms will lead to various diseases. Therefore, it is of great significance to construct a fluorescent probe that can detect Cys levels in food and biological systems. Results Here, a turn-off type probe TA had been successfully synthesized, which attached diethylamine as the strong electron donor, acrylate as the weak electron donor, and xanthene as the π-bridge. TA showed wonderful selectivity, low detection limit, good photostability and well live-cell compatibility for Cys by reducing acrylate group to hydroxyl group of TAOH . The reaction mechanism was demonstrated by 1 H NMR, ESI-MS spectra, pH-dependent response experiments, and DFT calculations. Importantly, the reason why TAOH exhibited no fluorescence was the disappearance of the ICT effect in the molecule due to the dominant existence of spirocyclic state of TAOH . In addition, the probe can be used not only for the imaging detection of Cys in A549 cells and zebrafish, but also for the detection of Cys levels in food samples. Significance This work provides a new idea for the design of Cys fluorescent probe, which may be beneficial to the comprehension of the potential mechanism of novel fluorescent probe.

Development of a NIR fluorescent probe for highly selective and sensitive detection of cysteine in living cells and in vivo

Abstract:Cysteine (Cys) plays important physiological roles in the human body, and abnormal Cys concentrations can cause a variety of diseases. Thus, detecting Cys with high selectivity and sensitivity in vivo is important. Near-infrared (NIR) fluorescent probes are widely employed in biological detection because of their excellent optical properties such as minimal damage to biological samples, low background interference and high signal-to-noise ratio. However, few NIR fluorescent probes that can detect Cys over homocysteine (Hcy) and glutathione (GSH) have been reported because of their similar reactivity and structure. In this work, a highly water-soluble NIR probe (CYNA) for detecting Cys whose structure is similar to that of indocyanine green and is based on cyanine skeleton was synthesized and via aromatic nucleophilic substitution–rearrangement (SNAr-rearrangement) to specific recognize the cysteine. The probe showed high selectivity toward Cys and superior biosecurity, excellent biocompatibility and prolonged dynamic imaging. It also has long fluorescence emission wavelength (820 nm), low detection limit (14 nM) and was successfully applied for visualizing Cys in living cells and mice, which has great promise for applications in noninvasive vivo biological imaging and detection.
