Abstract:This paper introduces the application of a calling and queuing system for blood sample collection in a large hospital in China. Besides the basic function, it has following functions. (a) A real name system: get the number according to the laboratory application form to prevent the phenomena of buying a number and an empty number. (b) Two times waiting: the patient should wait at the main hall, then at the blood sampling window so as to improve the work efficiency. (c) The flowchart for an outpatient blood testing is as following: getting the number --> waiting --> blood sampling --> getting the test information report. This system is capable of not only optimizing the work flow, but also improving the clinical environment. It shortens the patient's waiting time and raises the laboratory quality as well.
An Automatic Lab-on-Disc System for Blood Typing

Abstract:A blood-typing assay is a critical test to ensure the serological compatibility of a donor and an intended recipient prior to a blood transfusion. This article presents a lab-on-disc blood-typing system to conduct a total of eight assays for a patient, including forward-typing tests, reverse-typing tests, and irregular-antibody tests. These assays are carried out in a microfluidic disc simultaneously. A blood-typing apparatus was designed to automatically manipulate the disc. The blood type can be determined by integrating the results of red blood cell (RBC) agglutination in the microchannels. The experimental results of our current 40 blood samples show that the results agree with those examined in the hospital. The accuracy reaches 97.5%.

Abstract:Background: The ABO blood group is closely related to clinical blood transfusion, transplantation, and neonatal hemolytic disease. It is also the most clinically significant blood group system in clinical blood transfusion. Objective: The purpose of this paper is to review and analyze the clinical application of the ABO blood group. Methods: The most common ABO blood group typing methods in clinical laboratories are hemagglutination test and microcolumn gel test, while genotype detection is mainly adopted in clinical identification of suspicious blood types. However, in some cases, the expression variation or absence of blood type antigens or antibodies, experimental techniques, physiology, disease, and other factors affect the accurate determination of blood types, which may lead to serious transfusion reactions. Results: The mistakes could be reduced or even eliminated by strengthening training, selecting reasonable identification methods, and optimizing processes, thereby improving the overall identification level of the ABO blood group. ABO blood groups are also correlated with many diseases, such as COVID-19 and malignant tumors. Rh blood groups are determined by the RHD and RHCE homologous genes on chromosome 1 and are classified as Rh negative or positive according to the D antigen., the agglutination method is often used in clinical settings, while genetic and sequencing methods are often used in scientific research. Conclusion: Accurate ABO blood typing is a critical requirement for the safety and effectiveness of blood transfusion in clinical practice. Most studies were designed for investigating rare Rh blood group family, and there is a lack of research on the relationship between Rh blood groups and common diseases.

Abstract:Objective: To analyze the cause of ABO typing discrepancy and to explore the strategy of blood transfusion. Methods: ABO blood group was identified by serological method, and the cases with ABO typing discrepancy were confirmed by molecular biological method. Results: Seventy-four cases of ABO typing discrepancy were detected in84 487 cases, the discrepancy rate was 0.09%. Removing repeated cases, there were 67 cases: red blood cell antigen weakened 43 cases, blood group antibody weakened 9 cases, both antigen and antibody weakened 2 cases, subtypes 12 cases, others 1 case. In 36 hospitalized patients of ABO typing discrepancy, 25 cases received blood transfusion, and 22 were patients with hematological disorder. Excluding subtype cases, analysis of the effect of blood transfusion between 15 cases of same ABO genotype RBC and 5 cases of O blood type washed RBC showed no significant difference(P=0.225>0.05). Conclusions: The largest number of ABO typing discrepancy are patients with hematological disorders. Compatible ABO or same gene type RBC should be used in clinical blood transfusion. Both serological and molecular biological methods should be taken into account.

Abstract:The 15-channel Technicon blood typing machine is shown to be equally useful for patient and donor blood typing. It provides increased accuracy as well as increased speed and economy. Test capabilities are broadened by permitting additional simultaneous determinations such as the automated reagin test for syphilis or tests for additional erythrocyte antigens. Automated antibody screening, performed on the blood typing machine in conjunction with patient typing, was found to be of limited use because it failed to detect as much as 60 per cent of some clinically significant antibodies.

Abstract:Objective To evaluation Techno TwinStation automatic blood type instrument for blood cross matching accuracy.Methods A total of 4,500 patients'samples were tested using Techno TwinStation automatic for blood cross matching,and it was compared with manual polybrene test and anti-human globulin method.Results(1)The test results were all the same in 200 specimens with a negative antibody in screening in all the three cross matching methods,and in 4,280 specimens with a negative antibody in both the automatic instrument method and manual polybrene test.(2)As for the 20 specimens with a positive antibody in screening,the test results were as follow: the incompatibility occurred in 20 specimens in automatic blood type instrument,in 16 specimens in manual polybrene test method,and in 18 specimens in anti-human globulin test method,among which there were no statistically significant differences(P>0.05).Conclusion The results of Techno TwinStation blood type instrument used for cross matching blood testing are reliable.Its experimental operation is normalized and standardized with an advantage of low incidence of human error.Moreover,the experimental results can be permanently preserved,which provides a convenience to search for medical proof.

Abstract:Objective To explore Auto Vue Innova automatic instrument for identification of blood groups and unexpected antibody screening.Methods Forward and reverse ABO/Rh typing and unexpected antibody screening were performed on 9 652 samples using the Auto Vue Innova automatic instrument and manual method,then a comparison was made.Positive samples would be confirmed by conventional indirect anti-globulin technique.Results The rates of first accurate ABO typing by automatic instrument and tube method were 99.71%(9 624/9 652) and 99.75%(9 628/9 652) respectively.There was no significant difference between the two methods.The rates of first accurate RhD typing by the two methods were all 100.00%.The rates of unexpected antibody detection and the number of false positive were 0.47%(45/9 652),7 and 0.41%(40/9 652),2 respectively.The throughput per hour using Auto Vue Innova automatic instrument and manual method were 96,145 and 21,48 respectively.Conclusion The automatic instrument is accurate,safe and has higher sensitivity.It is easy to inquiry and quote and is suited for batch operations.

Abstract:The scientific, reasonable, safe and effective blood transfusion is the principle in clinical practice. Most of the medical institutions currently use information management system in blood transfusion(IMSBT). Due to lack of the corresponding national standards, there are various problems in practical application of IMSBT system with respect to blood transfusion and inventory management without characteristics of institutions, lack of unique patient identifier, deficiency in closed-loop real-time records of blood usage in institutions and absence of“urgent”blood issued module for emergency. This paper discusses the problems to which attention should be paid in the development of IMSBT system. We hope that through constant improvement, the IMSBT system will become a good assistant of clinical and blood transfusion, thus promotes the development of clinical blood transfusion.

Abstract:Objective To explore the feasibility and safety of electronic crossmatching technology in clinical blood transfusion in our country.Methods Blood group identification and antibody screening in patients and blood donors were carried out by using the automatic blood group instrument and test tube method.The results from the identification of two blood groups in patients and blood donors were consistent.The antibody screening test results were negative for both patients and blood donors,and were consistent with the traditional serological cross matching approaches.On these samples,a simulated electronic crossmatching was conducted.Results On 4 435 suspended erythrocyte samples that were consistent with both electronic crossmatching and traditional serological crossmatch,2 330 patients were infused in a simulation.No adverse transfusion reactions,such as ABO/RhD incompatibility,were observed.65 samples showed inconsistent results in electronic crossmatch with the traditional serological crossmatch.The main reason was that patients were detected with anti-Mur antibody from irregular antibody screening detection.Patients with positive C3 from direct antiglobulin test were infused with washed red blood cells.Positive IgG in patients determined by direct antiglobulin test,were also found in donors.Conclusion On the basis of improving reagent antigen spectrum in blood group irregular antibody screening and strengthening the foundation of direct antihuman globulin test in patients and blood donors,the electronic cross matching technology is feasible and safe in clinical blood transfusion.For the safety of clinical blood transfusion,serological crossmatch of blood should be used as a supplement if patients do not qualify for electronic crossmatch.

Conclusion Can be blood group identification right according to select the appropriate method of ABO blood group identification of abnormal results,guarantee the safety of clinical blood transfusion patients.

Abstract:Blood transfusion requires a mandatory cross-match test to examine the compatibility between donor and recipient blood groups. Generally, in all cross-match tests, a specific chemical reaction of antibodies with erythrocyte antigens is carried out to monitor agglutination. Since the visual inspection is no longer useful for obtaining precise quantitative information, therefore there is a wide variety of different technologies reported in the literature to recognize the agglutination reactions. Despite the classical methods, modern biosensors and molecular blood typing strategies have also been considered for straightforward, accurate and precise analysis. The interfacial part of a typical sensor device could range from natural antibodies to synthetic receptor materials, as designed by molecular imprinting and which is suitably integrated with the transducer surface. Herein, we present a comprehensive overview of some selected strategies extending from traditional practices to modern procedures in blood group typing, thus to highlight the most promising approach among emerging technologies.

Abstract:A closed-loop transfusion management system is constructed that covers blood preservation, transportation, transfer, distribution of blood, distribution, clinical blood specimen collection and blood transfusion process, which can monitor the implementation of doctor's advice, view the transport process of blood and blood samples, and record blood transfusion and adverse reaction information. These measurements can play a good effect in reduction of manual records and handover links in blood transfusion management, enhance the blood bank management, guarantee safely using blood, and realize the goal of real-time monitoring and closed-loop management.

Abstract:Objective: Analyze the reasons for the mismatch between forward and reverse typing of ABO blood types and the mismatch between cross matching. Methods: When the forward and reverse typing do not match, use physiological saline method, polyamine method, anti human globulin method, and anti screening positive samples are used for antibody identification. Results: The factors contributing to discrepancies in blood typing between forward and reverse typing include weakened serum typing, condensation, monoclonal immunoglobulin influence, bone marrow transplantation, and blood type subtypes. The causes of cross matching incompatibility include homologous antibody, warm Autoantibody, cold Autoantibody and daretozumab. Conclusion: Regular red blood cell homologous antibody screening should be conducted based on disease type, blood transfusion history, and medication history. Antigen matched red blood cells should be selected for cross matching, and different experimental methods should be used for testing to ensure the safety of clinical blood transfusion.

Abstract:A computerized system was reported capable of automatically doing quantitative analysis of morphological, structural and optical modifications of retinal vessels. This system included hardware for image acuquisition and display and software for performing image archiving, image analysis, retinal vascular networks recognition and artificial intelligence. By clinical experiment, the effectiveness of this system was proved and some numerical index for classification the degree of retinal arteriosclerosis was put forward. The accuracy and reliability of the results, justified by their low variability, made feasible the standardization of quantitative parameters to be used both in the diagnosis and in the prognosis of systemic diseases, and also allowed prospective studies based upon them.

Abstract:Objective To investigate the effect of blood group serology and polymerase chain reaction with sequence-specific primers (PCR-SSP) on identification and genotyping of ambiguous ABO blood group. Methods Eighty suspicious ABO blood group samples were identified by serology and polymerase chain reaction with sequence-specific primers (PCR-SSP). The final blood group type and the strategy of the transfusion of each case were determined according to the results of serology and PCR-SSP. Results 40 cases were confirmed to be subtypes, and the remaining 40 cases were normal types with weakened antigens or missing antibodies due to other reasons. The results of molecular genetic blood group typing based on PCR-SSP were 41 cases of subtypes (There were 3 discrepancies between two methods: one was Ael identified by serological methods, while its gene type was O2O2; one was common type O, while its gene type was BO1; one was type A, while its gene type was AB.) and 39 cases of normal ones. Conclusion Genotyping technology combined with serological typing has an important significance in identification of ABO blood groups.

Abstract:Objective:To evaluate the application of Galileo Echo automatic blood analyzer on blood grouping and irregular antibody screening tests. Method:ABO,Rh(D) blood grouping and the irregular antibodies were studied in 9082 samples using Galileo Echo automatic blood analyzer,at the same time, comparing with manual method(test tube method).Result:The accuracy of Galileo Echo automatic blood analyzer in ABO and Rh(D) blood grouping identification were 99.75%(9059/9082) and 99.88%(9071/9082),there was no significant difference in the manual method.The positive rate of the irregular antibodies was 0.54%(49/9082).Conclusion:The results of Galileo Echo automatic blood analyzer in ABO,Rh(D) blood grouping and the irregular antibodies tests are reliable, its experimental operation is automatically,quickly and convenient.Moreover,the experimental results can be permanently preserved,which is suitable for clinical blood grouping tests.

Abstract:BACKGROUND An accurate interpretation of the ABO blood group of an individual is of utmost importance to ensure patient safety and good transfusion practices. The aim of the study was to determine the incidence and causes of ABO typing discrepancies among patients and analysis of the clinical characteristics as well. METHODS A retrospective observational study was carried out in the Department of Laboratory Medicine in the General Hospital of Chinese PLA from March 2018 to December 2020. Records of patients were collected and analyzed for frequency, clinical characteristics, and influencing factors of ABO typing discrepancies. RESULTS There were 132 ABO typing discrepancies patients (57 females, 75 males), aged from 3 to 84 (50.3 ± 19.3) years, with history of operation 89 cases (67.42%) and blood transfusion 23 cases (17.42%). In the control group, there were 142 cases (63 females and 79 males), aged 10 - 78 years (50.5 ± 15.55), with operation history 68 cases (47.88%) and blood transfusion history 2 cases (1.40%). Among the inconsistent blood types group, there were 59 cases (44.7%) type B, 38 cases (28.8%) type A, 28 cases (21.2%) type AB, and 7 cases (5.3%) type O. There were 21 cases (15.9%) in the department of hematology, 15 cases (11.36%) in the department of orthopedics, 14 cases (10.6%) in the department of hepatobiliary surgery, 11 cases (8.33%) in the department of general surgery, 11 cases (8.33%) in the department of vascular surgery, and less than 5% in other departments. The common cause of ABO typing discrepancies was due to low/weak affinity antibody (3.79%), low/weak affinity antibody B (18.18%), weak A antigen (3.03%), weak B antigen (12.12%), hematopoietic stem cell transplantation (10.60%), irregular antibody (46.70%), subtype (3.79%), and cold agglutination (1.51%). The diseases in the ABO discrepancy group mainly included hematological disorders, malignant tumors, osteoarthritis and so on. Binary logistic regression showed that hematological disorders, malignant tumors, history of operation (p = 0.01), and history of blood transfusion (p = 0.017) were the influencing factors of ABO typing discrepancies. CONCLUSIONS It is possible that the antibody of ABO blood group system could not be detected. The independent influence factors were hematological disorders, malignant tumor, blood transfusion history and operation history for ABO typing discrepancies. It was necessary to analyze the causes correctly and judge the correct blood group by serological methods.
