Abstract:The dehydrogenation of vapor phase methyl isobutyl carbinol to produce methyl isobutyl ketone over a CuO-ZnO-Al2O3 catalyst prepared by co-precipitation method under the atmospheric pressure was studied. Using a single-tube reactor, the influence of the reaction temperature and the space velocity of methyl isobutyl carbinol on the dehydrogenation activities was investigated experimentally. The results show that under the temperature of 483~513 K and space velocity of 1.0~2.0 h-1, the maximum conversion rate of methyl isobutyl carbinol per pass is 91.11%, and the selectivity for methyl isobutyl ketone is over 99.0%. Based on these experimental results, a rate equation for the reaction was obtained by a least square method as -r=k(p(1.975/MIBC))-(1/k)P(1.980/MIBC), in which k = 9.542×107exp(-E / RT), E = 58.08 kJ·mol-1. The correlation index and variance error calculations indicate that, using the rate equation proposed, the calculated conversion data is in good agreement with the experimental data.

Abstract:The dehydrogenation of methyl isohutyl carbinol(MIBC)to produce methyl isobutyl ketone(MIBK)was studied with a self-prepared CuO/ZnO/Al_2O_3 catalyst in a fixed fed reactor,at ambient pressure.The experimental results showed that the conversion of MIBC was 82.2％ with a selectivity of MIBK 99.0％ under the appropriate reaction conditions of the space velocity of MIBC 1.0-1.5 h~(-1)and the reaction temperature 240-250℃.And the experimental results indicated a good stability of the catalyst.

Abstract:A stepwise isopropylation reaction of acetone is developed for preparation of methyl isobutyl ketone (MIBK) through acetone condensation and a recoverable Pt/C-catalyzed transfer hydrogenation of mesityl oxide (MO) with isopropanol (IPA). Almost quantitative MO conversion (up to 99.9%) and excellent MIBK selectivity (up to 99.3%) could be achieved by the method. Since IPA can be obtained from propane, a cheap petrochemical product, the by-product acetone (AT) generated in the reduction of MO could be easily recovered and reused in the preparation of MO, and the Pt/C catalyst could be recycled and reused, this H-2-and waste-free reaction may be a green and practical method for preparation of the useful fine chemical MIBK. Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:The production capacity of acetone is excess in China at present. Conversion of acetone into high-value products with huge market demands is of practical value. Methyl isobutyl ketone (MIBK) is an important industrial intermediate with very broad applications and large market requirement. Thus, there are tremendous prospects for industrial applications for the synthesis of MIBK from acetone. This article reports a convenient access to MIBK from acetone through condensation and reduction procedures. Compared with conventional procedures, the method employed isopropanol as an alternative reductant to the dangerous H-2 to reduce mesityl oxide (MO) into MIBK. The by-product from the reductant isopropanol was acetone and could be reused as the starting material to produce MO. After conditional optimizations, the MO conversion rate was enhanced to 95.8%, while the selectivity of MIBK was 98.8%. The two steps of reaction could be performed in one-pot.

Abstract:Methyl 3-oxo-2,6,6-trimethylcyclohex-1-ene-1-carboxylate was synthesised over eight steps from the starting material of citral instead of α-ionone. KMnO4, HC(OEt)3 and O2/TEMPO/CuCl were substituted for NaIO4, CH3I and pyridinium chlorochromate, respectively. Every step was simplified and gave a 48% overall yield. The procedure is suitable for large scale production.

Abstract:With wide applications, isoprene faces a growing global market. The increasing concerns about the environmental problems brought by petrochemical industry propel the research on bioproduction of isoprene. To date, numerous efforts have been made in the characterization and engineering of the isoprene biosynthesis pathway, leading to successful production of isoprene by bacteria and yeast. This chapter summarizes the research progresses in isoprene biosynthesis and introduces the isoprene biosynthesis in nature, the key enzyme isoprene synthase, and the development of isoprene-producing microorganisms. In particular, the efforts made toward industrial biosynthesis of isoprene using bacteria and yeast are discussed in detail, with special attention given to the metabolic engineering strategies adopted to achieve isoprene-producing strains with improved performance indices. The persisting issues and future research needs are presented as well.

Abstract:Isoprene is facing a growing global market due to its wide industrial applications. Current industrial production of isoprene is almost entirely petroleum-based, which is influenced by the shrinking C5 supply, while the natural emission of isoprene is predominantly contributed by plants. To bridge the need gap, a highly efficient fermentation-based process for isoprene production might be a suitable and sustainable solution, and extensive research works have been performed to achieve this goal. Here we review the accomplishments in this field by summarizing the history and prospects of microbial isoprene production. The natural producers and biosynthesis pathways of isoprene, the key enzyme isoprene synthase and the metabolic engineering strategies adopted for developing isoprene-producing microorganisms are introduced. In particular, strategies employed for achieving engineered strains with improved performance indices are discussed based on the published papers and patents. Abstract:Pentanone has three types of isomers: 3-methyl-2-butanone (methyl isopropyl ketone, MIPK), 3-pentanone (diethyl ketone, DEK) and 2-pentanone (methyl propyl ketone, MPK). Since MIPK is the smallest branched ketone and a promising biofuel, it is important to fully understand its combustion kinetics. In this work, the ignition delay times were measured in the temperature range from 1150 to 1600 K at pressures from 1 to 5 bar and for the equivalence ratios of 0.5, 1.0 and 1.5 (Mixes 1-9). A high-temperature detailed combustion kinetic model for MIPK was developed via a hierarchical method. Previous theoretical computations of the H-atom abstraction by H(O)over dot(2) and (O)over dotH K.from the tertiary carbon of MIP were adopted, and the reaction rate coefficients of H-abstraction by (H)over dot from the MIPK tertiary carbon were calculated via quantum chemical techniques. The rate coefficients of the reactions that are related to the acetyl group and the isopropyl group of MIPK were estimated by analogy with MPK and DIPK, respectively. Eleven elementary reactions in the submodel that are related to MIPK were selected for the optimization due to the poor prediction performance of the raw model. The self-adaptive differential evolution algorithm was applied to optimize the raw kinetic model based on the ignition delay times of Mixes 1-6. The optimized MIPK model was validated against the ignition delay times of Mixes 7-9. A brute-force sensitivity analysis, in combination with a flux analysis, demonstrates that the amount of unsaturated fuel-related stable intermediate products is the key factor that determines the reactivity of MIPK and DEK. Abstract:Depleting oil reserves and growing environmental concerns have necessitated the development of sustainable processes to fuels and chemicals. Here we have developed a general metabolic platform in E. coli to biosynthesize carboxylic acids. By engineering selectivity of 2-ketoacid decarboxylases and screening for promiscuous aldehyde dehydrogenases, synthetic pathways were constructed to produce both C5 and C6 acids. In particular, the production of isovaleric acid reached 32 g/L (0.22 g/g glucose yield), which is 58% of the theoretical yield. Furthermore, we have developed solid base catalysts to efficiently ketonize the bio-derived carboxylic acids such as isovaleric acid and isocaproic acid into high volume industrial ketones: methyl isobutyl ketone (MIBK, yield 84%), diisobutyl ketone (DIBK, yield 66%) and methyl isoamyl ketone (MIAK, yield 81%). This hybrid "Bio-Catalytic conversion" approach provides a general strategy to manufacture aliphatic ketones, and represents an alternate route to expanding the repertoire of renewable chemicals.

Abstract:Polyolefins attract considerable attention due to their vast consumption and wide application. In this study, cross-metathesis polymerization (CMP) is demonstrated to be an efficient and feasible method for the synthesis of aliphatic polyketones. The synthetic route features good yield, mild condition and easy operation. The success of CMP relies on the high cross-metathesis tendency between a-olefin and vinyl ketone units of monomer 2. Optimization of polymerization condition was conducted to generate unsaturated polyketone P0 (M-n, GPC = 14 kDa), and the results of kinetic study verified high efficiency and structural integrity of this system. Furthermore, post-polymerization modification of P0 by using thiol-Michael chemistry furnished functional polyketones P1-P6, and full conversion of the internal vinyl ketone groups of P0 was observed in each case. The pristine and functional polyketones P0-P6 were virtually soluble in most common organic solvents. In particular, hydrophobic and fluorescent polyketones (P5 and P6) were also obtained by this means. Therefore, this synthetic strategy shows promising future and exemplifies huge potential for the synthesis of a diverse range of functional polyketones.

Abstract:A series of basic and acidic ionic liquids, 1-butyl-3-methylimidazolium hydroxide (BMIMOH), 1-acetyl-3-methylimidazolium chloride (AcMIMCl) and AcMIMCl-FeCl3, or analogues of AcMIMCl, namely 1-potassium acetate-3-methylimidazolium chloride (KAcMIMCl), 1-potassium (sodium, ammonium) acetate-3-methylimidazolium hydroxides (KAcMIMOH, NaAcMIMOH and NH4AcMIMOH), were prepared and used as catalysts for catalytic synthesis of propylene glycol ether via reaction of propylene oxide (PO) with methanol under mild reaction conditions. KAcMIMOH exhibited outstanding catalytic performance with 94.2% of conversion of PO and 99.1% of selectivity to 1-methoxy-2-propanol (MP-2) at 60°C and ambient pressure for 4 h. However, AcMIMCl-FeCl3 showed a good catalysis performance with high selectivity to 2-methoxy-1-propanol (MP-1). The tunable synthesis of MP-2 or MP-1 catalyzed by basic compound KAcMIMOH or acidic ionic liquid AcMIMCl-FeCl3 was realized.

Abstract:Methyl isobutyl ketone (MIBK) is a highly valuable product in the chemical industry. It is widely used as an extracting agent for heavy metals, antibiotics, and lubricating oils. Generally, MIBK can be produced by three-step and one-step liquid-phase methods. These methods are expensive and energy-demanding due to the high pressure and low conversion of acetone. A novel nano-Pd/nano-ZnCr2O4 catalyst was developed to produce MIBK with high conversion and selectivity of 77.3% and 72.1%, respectively, at 350 °C and ambient pressure, eliminating the need for high pressure in conventional MIBK processes. This study is the first that proposes a newly developed process of methyl isobutyl ketone (MIBK) production using the low-pressure one-step gas-phase selective dehydrogenation of acetone. In this work, a novel process flow diagram has been developed for the production of MIBK using the developed nano-catalyst. The process was heat integrated, resulting in a 26% and a 19.5% reduction in the heating and cooling utilities, respectively, leading to a 12.6% reduction in the total energy demand. An economic analysis was performed to determine the economic feasibility of the developed process, which shows that the process is highly profitable, in which it reduced both the capital and operating costs of MIBK synthesis and showed a return on investment (ROI) of 29.6% with a payback period of 2.2 years. It was found that the ROI could be increased by 18% when the reactor temperature is increased to 350 °C. In addition, the economic sensitivity analysis showed that the process is highly sensitive to product prices and least sensitive to utility prices, which is due to the versatility of the process that requires only a low amount of energy.

Abstract:Microbial production of isoprene from renewable feedstock is a promising alternative to traditional petroleum-based processes. Currently, efforts to improve isoprenoid production in Saccharomyces cerevisiae mainly focus on cytoplasmic engineering, whereas comprehensive engineering of multiple subcellular compartments is rarely reported. Here, we propose dual metabolic engineering of cytoplasmic and mitochondrial acetyl-CoA utilization to boost isoprene synthesis in S. cerevisiae . This strategy increases isoprene production by 2.1-fold and 1.6-fold relative to the recombinant strains with solely mitochondrial or cytoplasmic engineering, respectively. By combining a modified reiterative recombination system for rapid pathway assembly, a two-phase culture process for dynamic metabolic regulation, and aerobic fed-batch fermentation for sufficient supply of acetyl-coA and carbon, we achieve 2527, mg l −1 of isoprene, which is the highest ever reported in engineered eukaryotes. We propose this strategy as an efficient approach to enhancing isoprene production in yeast, which might open new possibilities for bioproduction of other value-added chemicals.
