Abstract:An unexpected process from tetrahydrodiazirino[3,1-a]isoquinolines and diversified sulfonyl chlorides RSO2Cl (R=aryl, alkyl) to deliver highly yields 4,5-dihydro-3H-benzo[d][1,2]diazepine skeleton compounds was disclosed via N-insertion ring-opening reaction. The structural framework of 2,3-benzodiazepines is unambiguously confirmed by single-crystal X-ray crystallographic analysis. The present protocol features mild reaction conditions, high yields and feasibility of large-scale synthesis and catalyst-free access to benzo[2,3]diazepines skeleton. In contrast, azomethine imines were produced by employing tetrahydrodiazirino[3,1-a]isoquinoline reacts with aroyl chlorides under the same conditions.

Abstract:New methods have been developed for the synthesis of the substituted quinolines and quinazolinones derivatives by utilizing N-(2-aminobenzoyl)benzotriazoles under mild reaction conditions. 3-Acyl-2-alkyl(aryl)-4-hydroxyquinolines were obtained in modarete yields by the reaction of N-(2-aminobenzoyl)benzotriazoles and diketones in the presence of tert-BuOK. 3-Acylamino-4(3H) quinazolinones were obtained in good yields via N-(2-aminobenzoyl)benzotriazoles, orthoester and hyrazides in one-pot.

Abstract:It was found that 4-hydroxy-2-butenoic ester (11) could not react with 3,4-dihydro-isoquinoline (4a). Individual addition reactions of γ-mercapto-α,β-unsaturated esters (18) and -unsaturated amide (19) with 3,4-dihydroisoquinolines (4) were carried out under appropriate conditions to provide the corresponding thiazolo[2,3-α]isoquinoline derivatives with good yields (up to 87%) and significant diastereomeric selectivity. The mechanism of the crucial reaction was discussed.

Abstract:A facile and novel approach to the synthesis of 2-phenylquinazolines was developed via a tandem reaction following sp(3) C-H functionalization. Twenty-five examples of 2-phenylquinazolines were obtained from easily available 2-aminobenzophenones and benzylic amines with good to excellent yields.

Abstract:A convenient method for preparing 3-aryl isoquinolines via a base-promoted tandem reaction is presented. Simply combining commercially available 2-methyl-arylaldehydes, benzonitriles, NaN(SiMe(3))(2), and Cs(2)CO(3) enabled the synthesis of a variety of isoquinolines (23 examples, ≤90% yield). Among the syntheses of isoquinolines, the transition metal-free method described here is straightforward, practical, and operationally simple.

Abstract:A facile and efficient method for the synthesis of novel 2-substituted 4-tosyl-4,5-dihydrooxazolo[5,4-c]isoquinolines from 4-diazoisoquinolin-3-ones and nitriles is reported. The reaction proceeded through a TfOH-promoted formal [3 + 2] cycloaddition and the products could be conveniently converted to 2-aryloxazolo[5,4-c]isoquinolines and the subsequent 2-(oxazolo[5,4-c]isoquinolin-2-yl)phenol which emitted bright green light in dilute dichloromethane solution and in solid form as well. Simple operation, metal-free and mild reaction conditions, short reaction time and broad substrate scope are the prominent features of this methodology.

Abstract:A novel, practical, highly efficient, and transition metal free nitrogen insertion reaction for the synthesis of 2,3-disubstituted quinazolinone derivatives was developed. Diverse functionalized 3-indolinone-2-carboxylates and nitrosoarenes with a wide range of substituted nitrosobenzenes, nitrosopyridines, dibenzofuranyl, or dibenzothienyl nitroso compounds worked smoothly to give 2,3-disubstituted quinazolinone derivatives in good to excellent yields (69-98%). A gram-scale reaction was achieved, and an afloqualone analogue was synthesized under the mild reaction conditions.

Abstract:A broad scope of quinazolin-4( 3H )-ones or benzothiadiazine 1,1-dioxides was obtained via a transition-metal free and convenient method. This approach involving KHSO 4 promoted cyclization and dehydration between diverse functionalized 2-aminobenzamides or 2-aminobenzenesulfonamides and N, N -dimethylformamide derivatives. The protocol offers moderate to excellent yields (45%-93%) under mild condition in a short of reaction time of 6 hours. The use of an environment-friendly and less expensive catalyst KHSO 4 also makes this protocol more attractive.

Abstract:The synthesis of 2-aminoquinazoline derivatives is achieved by using hydrochloric acid as a mediator in the [4+2] annulation reaction between N-benzyl cyanamides and 2-amino aryl ketones. In addition, 2-amino-4-iminoquinazolines are synthesized by the reaction of 2-aminobenzonitriles, instead of 2-amino aryl ketones, with N-benzyl cyanamides. A wide range of substrates can be used and high yields are obtained, demonstrating the practicality of this method for the synthesis of 2-aminoquinazoline derivatives.
