Abstract:Ziziphi Spinosae Semen is the dried seeds of Ziziphus jujuba Mill. var. spinosa (Bunge) Hu ex H. F. Chou, and its extract has broad application prospects in the development of sleep-aid functional foods. However, the quality parameters of Ziziphi Spinosae Semen extracts currently available in the market are not uniform and there is a lack of unified standards. Therefore, it is important to establish an accurate and comprehensive method for quality evaluation. In view of the problems that the UV responses of flavonoids and saponins in the Ziziphi Spinosae Semen extracts vary dramatically and the saponin content in Ziziphi Spinosae Semen water extract is very low, high performance liquid chromatography (HPLC) was used to establish the fingerprint and quantify spinosin. The separation was carried out on a Waters XSelect HSS C18 column (250 mm×4.6 mm, 5 μm), and the mobile phase was acetonitrile-0.1% (v/v) phosphoric acid aqueous solution for gradient elution. The eight common peaks in the fingerprint of the Ziziphi Spinosae Semen extracts, identified by HPLC-quadrupole time-of-flight mass spectrometry, were attributed to flavonoids by reference substance identification, literature comparison, and high-resolution mass spectrometry data analysis. Semi-quantitative analysis of seven flavonoids and quantitative analysis of spinosin were conducted using the established HPLC quantitative fingerprint. The contents of jujuboside A and jujuboside B were determined by ultra-high performance liquid chromatography-triple quadrupole mass spectrometry. Chromatographic separation was performed on a Waters ACQUITY UPLC BEH C18 column (50 mm×2.1 mm, 1.7 μm) by gradient elution using a mobile phase of acetonitrile-0.1%(v/v) formic acid aqueous solution. The target compounds were analyzed in multiple reaction monitoring mode with positive electrospray ionization. The semi-quantitative and quantitative data of the above-mentioned 10 components are displayed in the form of radar. Using the above methods, three batches of Ziziphi Spinosae Semen water extracts prepared in the laboratory and 15 batches of extract samples obtained from 15 suppliers were analyzed and compared. The results showed that although the raw materials of three batches of Ziziphi Spinosae Semen water extracts prepared in the laboratory were from different enterprises, the overall difference was not significant. However, the component contents of the samples provided by different manufacturers were greatly different, suggesting that there are some problems associated with the different manufacturers, such as dilution of excipients, adulteration of Ziziphi Mauritianae Semen, alcohol extraction, purification, and enrichment. For example, the representative composition contents in the Ziziphi Spinosae Semen extracts obtained from manufacturers B, C, E, F, G, H, I, and O were low, which were approximately 1/10 of corresponding contents in the normal water extracts prepared in the laboratory. It is speculated that to reduce the unit price of the product, the manufacturer used fewer raw materials or a large number of auxiliary materials to dilute the Ziziphi Spinosae Semen extracts. The contents of some flavonoids in the Ziziphi Spinosae Semen extract from manufacturer N were slightly higher than that in the self-preparation Ziziphi Spinosae Semen water extract, but it did not contain jujuboside A; thus, it was speculated that the Ziziphi Mauritianae Semen might be used for extraction. The contents of 10 components in the Ziziphi Spinosae Semen extract obtained from manufacturer D were all higher than the corresponding ones in the self-preparation Ziziphi Spinosae Semen water extract. Combined with the quality label of total saponin content > 20% and poor water solubility, it was speculated that the product might be prepared by alcohol extraction or purified and enriched by using resin. These results provided the basis for the enterprise to establish internal control quality standards for Ziziphi Spinosae Semen extracts and to select qualified suppliers.

Abstract:Reliable, reproducible and valid fingerprint analysis methods using high-performance liquid chromatography-photodiode array detection (HPLC-DAD) for characteristic bioactive flavonoids and saponins of Semen Ziziphi spinosae (SZS) were developed and validated. HPLC separation of the chemical constituents of SZS was performed on an YMC-PACK ODS-A RP-18 column and detected at 270 and 204nm for flavonoids and saponins, respectively. A mobile phase consisted of acetonitrile and 0.1% phosphoric acid aqueous solution was used with linear gradient elution. Using spinosin and jujuboside B as the reference markers of flavonoids and saponins respectively, 9 common fingerprint peaks of flavonoids and 10 common fingerprint peaks of saponins were specified based on the fingerprint analysis of 10 batches of SZS from different regions in China. The fingerprint analysis methods developed are reliable, reproducible and valid, and might be used as a more convenient approach for the species identification and quality monitoring and assessment of SZS.
Determination of Syringin in Bushen Zhuanggu Capsule by HPLC-DAD

Abstract:RP-HPLC was used to determine the content of syringin in Bushen Zhuanggu capsule. The analytical column was Symmetry C_(18) column. The mobile phase consisted of methanol-water with 0.2% glacial acetic acid (55∶45), the flow rate was 1 mL/min,column temperature was (30 ℃),DAD (diode array detector) at 265 nm was used for detection,the content of syringin in Bushen Zhuanggu capsule is 1.46 mg/g, the average recovery of syringin was 98.25%. The results showed that the method is simple,accurate and repeatable and it is suitable for quality control of Bushen Zhuanggu capsule.

Abstract:A rapid, sensitive and selective high-performance liquid chromatography–tandem mass spectrometric method (HPLC–MS–MS) has been developed and validated for the determination of soyasaponins Ba and Bb in human serum using glycyrrhizin as internal standard (I.S.). Soyasaponins Ba and Bb were extracted from human serum by liquid–liquid extraction and cleaned up by C18 solid-phase extraction (SPE), followed by separation on a C18 reversed-phase column using acetonitrile/water containing 0.025% acetic acid as a mobile phase for gradient elution. Soyasaponins Ba and Bb, and I.S. were ionized by negative ion pneumatically assisted electrospray and detected by HPLC–MS–MS in the multiple-reaction monitoring (MRM) mode using precursor → product ion combinations at m/z 958 → 940, 942 → 924 and 822 → 351, respectively. The calibration curves were linear (r2 > 0.991) in the concentration range of 0.5–100.0 ng/mL, with lower limits of quantification of 0.5 and 0.3 ng/mL for soyasaponins Ba and Bb, respectively, in human serum. Intra-day and inter-day relative standard deviations (R.S.D.) were less than 7.9 and 11.3%, respectively. The mean recoveries of soyasaponins Ba and Bb ranged from 92 to 101% and from 85 to 94%, respectively.

Abstract:A simple,reproducible and reliable HPLC fingerprint analysis method of Ziziphi Spinosae Semen (ZSS) and Ziziphi Mauritianae Semen (ZMS) was established,and then it was applied to analyze samples collected from different locations.A total of 12 common fingerprint peaks were designated in ZSS and ZMS,respectively,and five of which were definitely identified.Of these common peaks,10 peaks were presented in both ZSS and ZMS,whereas two were characteristic for ZSS and ZMS,respectively.Furthermore,the two characteristic peaks of ZMS were definitely assigned as 6'"-sinapoylspinosin and frangufoline by HPLC guided purification and structural elucidation.The HPLC fingerprint analysis was proved to be useful in identifying and discriminating ZSS and ZMS,which was beneficial for quality control of ZSS and its products.

Abstract:In this study, high‐performance liquid chromatography‐evaporative light scattering detection (HPLC‐ELSD) and HPLC‐mass spectrometry (HPLC‐MS) were employed for the quantitative analysis of steroidal saponins of Semen Allii Fistulosi. The HPLC‐ELSD method was simple, accurate, and repeatable, and the recoveries were 97.91%∼99.77%. Similarly, the HPLC‐MS method was rapid, sensitive, accurate, and reproducible, and the recoveries ranging from 95.61% to 99.70% in the control samples. The sensitivity of the HPLC‐MS method was several times higher than that of the HPLC‐ELSD method. These results collectively demonstrated that these methods are suitable for the content determination of steroidal saponins of Semen Allii Fistulosi and the identification of its plant source.

Content and Antioxidant Activity of Components in Different Polar Parts of Semen Ziziphi Spinosae and Their Correlation Analysis

Abstract:Objective: Using Semen Ziziphi Spinosae as the research subject, the antioxidant activity and chemical composition in various solvent extraction sections were investigated, and their correlation was examined. Methods: Semen Ziziphi Spinosae was extracted by the water decoction method,the extracts were successively extracted with petroleum ether, chloroform, ethyl acetate, and n-butanol, and five polar parts were obtained, including water extract (S1), petroleum ether extract (S2), chloroform extract (S3), ethyl acetate extract (S4), and n-butanol extract (S5).The contents of total phenolic acids, total flavonoids, total polysaccharides, total saponins, jujuboside A, and spinosa in different polar parts were determined by ultraviolet spectrophotometry and high-performance liquid chromatography. The antioxidant activities of different polar parts were studied by classical in vitro antioxidant and H2O2 cell oxidative damage methods. The correlation between six chemical components and antioxidant activities was analyzed by partial least squares regression. Results: Compared with other components in one part, the contents of total polysaccharides in S1, S2, S3 extraction parts were the highest, which were (17.40±0.47), (5.38±0.05) and (5.43±0.01) mg/g, respectively. The content of total flavonoids in S4 and S5 extraction parts was the highest, which were (17.49±0.47) and (23.18±0.20) mg / g, respectively. Each extraction site had good antioxidant activity, and the n-butanol extraction site (S5) had the strongest antioxidant capacity, showing a significant dose-effect relationship.Correlation analysis showed that total saponins (X2), total polysaccharides (X3), and spinosa content (X5) had a great influence on the antioxidant capacity of ZSS. Conclusion: The n-butanol extract of Semen Ziziphi Spinosae had the strongest antioxidant capacity. The active ingredients with an antioxidant effect were total saponins, total flavonoids, and spinosa. This study provides a theoretical basis for the in-depth research and development of wild Semen Ziziphi Spinosae as a natural antioxidant and antioxidant food.
Comparison between HPLC and HPTLC densitometry for the determination of spinosin from Ziziphus jujuba Mill. fruit extracts

Abstract:Two validated, rapid and precise methods based on HPTLC and RP-HPLC with UV detection for quantitative determination of spinosin in edible extracts of Ziziphus jujuba fruit, is described. The HPTLC chromatograms were developed using a mobile phase of ethyl acetate –dichloromethane – methanol – water (18:10:15:5, v/v/v/v) on a pre-coated plate of silica gel 60 F254 and quantified by densitometry absorbance/remission mode at 334 nm in the range of 10–120 ng/mL. A gradient elution scheme with methanol-water at a flow rate of 1 mL/min and C18-Hypersil (250 × 4.6 mm, 5 µm) analytical column in the range of 32–400 ng/mL were used at 334 nm in the HPLC method. The limit of detection and limit of quantification were 11.59 and 35.12 ng/mL for HPTLC and 33.78 and 102.36 ng/mL for HPLC method, respectively. The relative standard deviation ranged between 0.94–1.27 (intra-day) and 0.39–1.69 (inter-day) for the HPTLC method and 1.02–1.56 (intra-day) and 0.89–1.71 (inter-day) for HPLC method. The results indicated that the developed methods were accurate and precise. The spinosin contents from different extracts were determined and compared by HPLC and HPTLC densitometry. The quantitative results of both analytical methods did not show any statistically significant differences between them.

Abstract:A rapid and sensitive UPLC-MS/MS method was established for the simultaneous quantification of 6 '''-feruloylspinosin, spinosin, jujuboside A, and jujuboside B in rat plasma after the oral administration of traditional and dispensing granules (DG) decoction of Ziziphi Spinosae Semen (ZSS). The four components were separated using 0.1% formic acid and acetonitrile as a mobile phase by gradient elution at a flow rate of 0.3 mL/min equipped with a C-18 column (2.1 x 50 mm, 1.7 mu m particle size, Acquity BEH C-18). The mass spectrometer was operated under multiple reaction monitoring mode. An aliquot of 100 mu L rat plasma was deproteinized by 300 mu L methanol. The supernatant was injected into the UPLC-MS/MS system for analysis. The calibration curves displayed good linearity. The intra-day and inter-day precisions (RSD) were less than 7.3%. The accuracies ranged from -1.3 to 6.1%. The extraction recoveries ranged from 95.8 to 101.9%, and the matrix effects were satisfactory. For DG, half-life values (t(1/2)) of 6 '''-feruloylspinosin and C-max of jujuboside A were elevated remarkably. MRT0-t of jujuboside B was significantly increased. No significant variation was observed for the pharmacokinetic parameters of spinosin. The results could provide a scientific basis for the clinical application of traditional and DG decoction of ZSS.

Abstract:Ziziphi Spinosae Semen (ZSS), the seeds of Ziziphus jujuba var. spinosa, is widely used in China or other Asian countries for the treatment of insomnia and palpitation. In our previous work, chemical constituents in ZSS were profiled by ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (UHPLC/Q-TOF MS). Notably, characterization of substances in vivo was of great importance to reveal the therapy basis or mechanism in further work. Till now, there were few reports about in vivo substances' investigation of ZSS. In the present study, an integrated strategy contained represented compounds and diagnostic ions extraction was applied to characterize metabolism feature of ZSS in rats based on UHPLC/Q-TOF MS method. First, the metabolic information of four compounds (spinosin, isovitexin, jujuboside B, betulinic acid) featuring three representative chemical structures (flavonoids, saponins, terpenes) in ZSS was conducted, and their metabolism features were summarized, especially for flavonoid C-glycosides. Second, the absorbed compounds and representative compounds-related metabolites were quickly screened out; during this time, the diagnostic ions were sorted out. Last, with the help of diagnostic ions and summarized metabolic reactions, other metabolites were characterized. As a result, a total of 151 xenobiotics (58 prototypes and 93 metabolites) were identified or tentatively characterized in rats after ingestion of ZSS. Among them, 16 substances were presented in plasma, 114 in urine, 51 in bile, and 120 in feces, respectively. Hydrogenation, hydrolysis, and glucuronidation were the major metabolic reactions of ZSS in rats. The present study provided meaningful data for further pharmacological mechanism research or pharmacokinetics evaluation of ZSS.

Characterization of Fatty Oil of Zizyphi spinosi semen Obtained by Supercritical Fluid Extraction

Abstract:Zizyphi spinosi semen (ZSS) has been widely used for treatment of insomnia in oriental countries. The aim of this study is to characterize the fatty oil of ZSS obtained by supercritical fluid extraction in terms of chemical composition and physicochemical properties. The chemical composition, including fatty acids and unsaponifiable constituents, was analyzed by gas chromatography–mass spectrometer (GC–MS). The results revealed that 9-octadecenoic acid (43.38 ± 0.03%) and 9,12-octadecadienoic acid (40.58 ± 0.03%) were the main fatty acids, and β-sitosterol (37.39 ± 0.02%) and squalene (30.79 ± 0.01%) were the key unsaponifiables. Furthermore, four indexes were assayed according to Chinese Pharmacopeia (2005) to reflect the physicochemical properties of ZSS oil, their values being determined as follows: acid value (10.3 ± 0.1 mg KOH/g), peroxide value (0.05 ± 0.01 g/100 g), saponification value (194.4 ± 0.5 mg KOH/g) and iodine value (109.7 ± 0.8 g I/100 g). The basic information obtained provides data support for quality evaluation and efficacy research of ZSS oil, and suggests its prospects for development in pharmaceutical and food industries.
Simultaneous qualitative and quantitative analysis of triterpenic acids, saponins and flavonoids in the leaves of two Ziziphus species by HPLC-PDA-MS/ELSD

Abstract:The leaves of Ziziphus jujuba and Z. jujuba var. spinosa have been utilized as crude drugs for their health benefits in China for thousands of years. To control their quality, a reliable method based on high-performance liquid chromatography coupled with photo diode array and electrospray ionization tandem mass spectrometry detection (HPLC-PDA-ESI-MS/MS) was developed for exploration of the chemical profiles of these jujube leaves. As the results, fourteen constituents including three flavonoids, two saponins and nine triterpenic acids were identified or tentatively characterized. Then, twelve of them such as quercetin-3-O-rutinoside, zizyphus saponins I and II, ceanothic acid, alphitolic acid, maslinic acid, 2α-hydroxyursolic acid, zizyberanalic acid, epiceanothic acid, ceanothenic acid, betulinic acid, and oleanolic acid were selected as the chemical markers and were determined using an HPLC coupled with evaporative light scattering detection (ELSD) method. The separation was carried out on a Waters Sunfire C₁₈ column with 0.2 % acetic acid and acetonitrile as the mobile phase under gradient elution. The operating conditions of ELSD were set as 80°C for drift tube temperature and 2.7 l/min for nitrogen flow rate. The developed method was fully validated in terms of linearity, sensitivity, precision, repeatability as well as recovery, and subsequently applied to evaluate the quality of eight batches of Z. jujuba and Z. jujuba var. spinosa leaves from different collections.
Determination of tenacissosides J by HPLC and its application

Abstract:Objective To analysis the contents of tenacissosides J by HPLC method.Methods Applied to the Hypersil ODS column(5μm,250mm×4.5mm),the separation was achieved in acetonitrile-water-triethylamine(8∶92∶0.2,v/v/v) as the mobile phase and a flow rate of 0.5ml/min.20μl sample of tenacissosides J were introduced into column of and wave length monitored at 223 nm and sensibility was 0.5 AUFS.Results The determination could be completed within 15 min and the analytical results of samples demonstrated a good linear relationship(r=0.9995,n=6) within the 43.8~1810μg/ml.The relative standard deviation(RSD) was 3.45% and that of intra-day was 1.66%,inter-day was 2.53%.The average recoveries were 97.9%.The contents of tenacissosides J in different producting place and of extractive materials on different position of Marsdeniae tenacissosides all had significant difference(P<0.05).Conclusion The method is accurate,simple,sensitive and reliable,which can be applied to the determination of tenacissosides J content.

Abstract:As a well-known traditional Chinese medicine, Ziziphi Spinosae Semen has been used for treating anxiety and insomnia for a long time. Spinosin, the main active C-glycoside flavonoid in Ziziphi Spinosae Semen, has attracted much attention because of its many pharmacological activities, including strong hypnotic effects, anxiolytic-like effects, and so on. In the present work, high-purity spinosin was separated from Ziziphi Spinosae Semen using the HPD-300 resin followed by preparative high-performance liquid chromatography. The adsorption kinetics curve of spinosin on the HPD-300 resin was studied and fitted well by the pseudo-second-order equation. The adsorption isotherms were also constructed, and low temperature favored the adsorption reaction. The separation parameters were optimized using dynamic adsorption and desorption tests. After a one-run treatment with HPD-300 resin, the concentration of spinosin increased 11.8-fold from 0.99% to 11.7% with a recovery yield of 80.4%. Furthermore, the purity of spinosin could surpass above 98% after separation by preparative high-performance liquid chromatography and recrystallization with a recovery yield of 72.6%. The developed method was effective and suitable for the large-scale preparation of spinosin. Moreover, it was confirmed that HPD-300 resin could enable good selection for the enrichment of flavonoids from different plants. This article is protected by copyright. All rights reserved.

Chow has been widely used in treating insomnia and anxiety.Aim of the study: In this study, we investigated the hypnotic effect of jujubosides, one of the major components (saponin) of Semen Ziziphi Spinosae, in both day and night period.Materials and methods: After the administration of jujubosides (9 mg/kg once per day for 3 days), sleep parameters were examined by EEG and EMC analysis in normal rats and the action of jujubosides on pentobarbital-induced sleep assessed by the loss-of righting reflex.Results: During daytime (9:00-15:00), jujubosides significantly increased the total sleep and rapid eye movement (REM) sleep without significant influence on non-REM (NREM) sleep. During nighttime (21:00-3:00), jujubosides significantly increased the total sleep and NREM sleep especially the light sleep while showed no significant effect on REM sleep and slow wave sleep (SWS). In pentobarbital-treated mice, jujubosides significantly augmented the hypnotic effect of pentobarbital (45 mg/kg, i.p.), proved by increasing sleep time and this augmentative effect was potentiated by 5-hydroxytryptophan (2 mg/kg, i.p.). Furthermore, jujubosides inhibited the para-chlorophenylalanine-induced suppression of pentobarbital-induced hypnosis.Conclusions: These results suggested that the hypnotic effect of jujubosides on normal rats may be influenced by circadian rhythm and the serotonergic system may involve in the hypnotic effect of jujubosides. jujubosides may be good source of lead compounds for novel hypnotics. Abstract:Objective: To establish a quantitative analysis method for sennoside A, sennoside B and physcion by ultra-high performance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS). Methods: The sample was extracted by methanol-2 mmol/L ammonium formate(9∶1) at 40 ℃ for 1 h. The separation was performed using Agilent Eclipse Plus C_(18 )(2. 1 mm × 50 mm, 1. 8 μm) column with gradient elution. The mobile phase was consisted of 0. 1% formic acid and methanol. Qualitative and quantitative analysis was conducted with an electrospray ionization source operated in the negative ionization(ESI~-) mode and multiple reaction monitoring(MRM) mode. Results: The linear range of three compounds were from 0. 1 to 10 μg/mL with the correlation coefficients(r) above 0. 995. The spiked recoveries were in the range of 81. 9% to 114. 5% at the concentrations of 0. 02, 0. 15 and 1. 60 mg/g with relative standard devisions(RSDs) ranged from 0. 30% to 3. 43%(n=6). The detection limits of sennoside A and sennoside B were 1. 2 μg/g. The detection limit of physcion was 2. 4 μg/g. Sennoside A, sennoside B or physcion were detected in 19 out of 40 batches of samples. The content of sennoside A ranged from 0. 184 to 6. 33 mg/g and the content of sennoside B ranged from 0. 202 to 7. 23 mg/g. The content of physcion ranged from 0. 042 to 0. 79 mg/g. Conclusion: The method is simple, accurate and suitable for the determination of sennoside A, sennoside B and physcion.

Abstract:Ethnopharmacological relevance Semen Ziziphi Spinosae (SZS), the seed of Ziziphus jujuba var. spinosa (Bunge) Hu ex H.F. Chow (Chinese name Suan-Zao-Ren), is widely distributed in China, Laos, Myanmar, and Iran. It is a classic traditional Chinese medicine with sedative and sleeping effects. In clinical practice, there are more than 155 proprietary Chinese medicines containing SZS. However, many commercial SZS products are difficult to qualify using current methods. Moreover, there is a scarcity of quality standards for SZS in proprietary Chinese medicines. Aim of the study The purpose of this study was to clearly reveal the quality indicators during the entire production process of SZS and its products. Materials and methods This study reviewed more than 230 articles and related books on the quality control of SZS and its proprietary Chinese medicines published over the last 40 years (from January 1979 to October 2022). Moreover, where available, information on the quality of was also collected from websites for comparison, including online publications ( e.g . PubMed, CNKI, Google Scholar, and Web of Science), the information at Yaozhi website and China Medical Information Platform, along with some classic books on Chinese herbal medicine. The literature and information search were conducted using keywords such as "Suan-Zao-Ren", " Ziziphus jujuba" and "quality control", and the latest results from various databases were combined to obtain valid information. The active components, which in vivo exposure, and Q-markers were also summarized. Results The jujuboside A, jujuboside B, and spinosin were revealed as the key Q-markers for SZS. Moreover, the advancements and prospects of the quality control for SZS and its extract, proprietary Chinese medicines, health foods, and adulterants were comprehensively summarized. The high-performance liquid chromatography-UV/evaporative light scattering detection and fingerprint analysis were found to be the mainstream methods for the SZS quality control. In particular, the novel quality evaluation method based on the unit content was applied for SZS and its proprietary Chinese medicines. Significant fluctuations were found in the contents of Q-markers. Moreover, the mass transfer rule of Q-markers was comprehensively clarified based on the entire production process, including production origins, ripening time, primary process, processing, compatibility decoction/extract, and storage. Ultimately, the crushing and compatibility of SZS were found to be the key steps affecting the active components. Conclusions In short, this study provides solid evidences to reveal quality indicators for the entire production process of developing rational quality standards for SZS and its products. Moreover, this study also provides a template quality control overview, which could be extended to other traditional Chinese medicines.
