Abstract:Reliable, reproducible and valid fingerprint analysis methods using high-performance liquid chromatography-photodiode array detection (HPLC-DAD) for characteristic bioactive flavonoids and saponins of Semen Ziziphi spinosae (SZS) were developed and validated. HPLC separation of the chemical constituents of SZS was performed on an YMC-PACK ODS-A RP-18 column and detected at 270 and 204nm for flavonoids and saponins, respectively. A mobile phase consisted of acetonitrile and 0.1% phosphoric acid aqueous solution was used with linear gradient elution. Using spinosin and jujuboside B as the reference markers of flavonoids and saponins respectively, 9 common fingerprint peaks of flavonoids and 10 common fingerprint peaks of saponins were specified based on the fingerprint analysis of 10 batches of SZS from different regions in China. The fingerprint analysis methods developed are reliable, reproducible and valid, and might be used as a more convenient approach for the species identification and quality monitoring and assessment of SZS.

Abstract:Ziziphi spinosae semen has been widely used to treat insomnia and anxiety. To profile its chemical components, an online comprehensive two‐dimensional LC‐MS was developed. In this two‐dimensional LC system, a novel phthalic anhydride bonded stationary phase column was combined with a C18 column. As a result, this new stationary phase exhibited remarkable differences in separation selectivity from C18, achieving a good orthogonality of 83.3%. Moreover, this new stationary phase with weaker hydrophobicity than C18 realized solvent compatibility in the online configuration. Coupled with tandem MS, 154 compounds were identified, including 51 unreported compounds. Compared with one‐dimensional LC‐MS, this online two‐dimensional LC‐MS system exhibited a much higher resolving power in isomer separation. This work provided an effective separation and characterization method for the material basis of Ziziphi spinosae semen. Abstract:RATIONALE: The fragmentation behavior of seven secoiridoid glycosides (SIGs) isolated from Gentiana triflora Pall., a traditional Chinese medicine, was investigated by electrospray ionization ion trap mass spectrometry (ESI-IT-MS). METHODS: Multistage electrospray ionization mass spectrometry (ESI-MSn) experiments both in positive-and negative-ion mode were used to elucidate the main fragmentation pathways of these compounds. RESULTS: In full scan mass spectra, different deprotonated or protonated molecules were observed for compounds with different basic structure. Ring cleavages of the aglycon moiety were observed in MS2 spectra and the characteristic ions provided valuable information on the basic structural skeletons. Their fragmentation pathways and fragment ion structures were proposed. CONCLUSIONS: The established fragmentation patterns have been successfully used to identify eleven SIGs in the extract of Gentiana triflora Pall. Copyright (C) 2014 John Wiley & Sons, Ltd.

Abstract:Abstract Semen Ziziphi Spinosae (SZS) is used to treat neurasthenia, insomnia, and anxiety in China. The extract from SZS contains many bioactive components, such as flavonoids, alkaloids, and terpenoids. Among these, the flavonoids are the main bioactive compounds that have been proven to be responsible for the sedative effects. In this study, a total of 13232 differentially expressed genes and 83 flavonoid metabolites were identified at three different growth and development stages (T1, T2, and T3) of SZS using transcriptomic and metabolomic. The main flavonoid metabolic components of SZS were catechin, L-epicatechin, (-)-epigallocatechin, (+)-gallocatechin, spinosin and its derivatives. A total of 53 unigenes encoding 15 enzymes were identified as candidate genes involved in flavonoid biosynthesis in SZS. Among, PAL, C4H, 4CL, CHS, CHI, FLS, FNS, ANS, ANR, LAR, and UFGT genes were all downregulated in T3 than in T1. The expression levels of these genes influence the accumulation of flavonoids in SZS. Our results should provide valuable information for flavonoid metabolites and candidate genes involved in flavonoid biosynthesis on SZS.

Abstract:Ziziphi Spinosae Semen (ZSS), the seeds of Ziziphus jujuba var. spinosa, is widely used in China or other Asian countries for the treatment of insomnia and palpitation. In our previous work, chemical constituents in ZSS were profiled by ultra-high-performance liquid chromatography coupled with quadrupole time-of-flight tandem mass spectrometry (UHPLC/Q-TOF MS). Notably, characterization of substances in vivo was of great importance to reveal the therapy basis or mechanism in further work. Till now, there were few reports about in vivo substances' investigation of ZSS. In the present study, an integrated strategy contained represented compounds and diagnostic ions extraction was applied to characterize metabolism feature of ZSS in rats based on UHPLC/Q-TOF MS method. First, the metabolic information of four compounds (spinosin, isovitexin, jujuboside B, betulinic acid) featuring three representative chemical structures (flavonoids, saponins, terpenes) in ZSS was conducted, and their metabolism features were summarized, especially for flavonoid C-glycosides. Second, the absorbed compounds and representative compounds-related metabolites were quickly screened out; during this time, the diagnostic ions were sorted out. Last, with the help of diagnostic ions and summarized metabolic reactions, other metabolites were characterized. As a result, a total of 151 xenobiotics (58 prototypes and 93 metabolites) were identified or tentatively characterized in rats after ingestion of ZSS. Among them, 16 substances were presented in plasma, 114 in urine, 51 in bile, and 120 in feces, respectively. Hydrogenation, hydrolysis, and glucuronidation were the major metabolic reactions of ZSS in rats. The present study provided meaningful data for further pharmacological mechanism research or pharmacokinetics evaluation of ZSS.

Characterization of Fatty Oil of Zizyphi spinosi semen Obtained by Supercritical Fluid Extraction

Abstract:Zizyphi spinosi semen (ZSS) has been widely used for treatment of insomnia in oriental countries. The aim of this study is to characterize the fatty oil of ZSS obtained by supercritical fluid extraction in terms of chemical composition and physicochemical properties. The chemical composition, including fatty acids and unsaponifiable constituents, was analyzed by gas chromatography–mass spectrometer (GC–MS). The results revealed that 9-octadecenoic acid (43.38 ± 0.03%) and 9,12-octadecadienoic acid (40.58 ± 0.03%) were the main fatty acids, and β-sitosterol (37.39 ± 0.02%) and squalene (30.79 ± 0.01%) were the key unsaponifiables. Furthermore, four indexes were assayed according to Chinese Pharmacopeia (2005) to reflect the physicochemical properties of ZSS oil, their values being determined as follows: acid value (10.3 ± 0.1 mg KOH/g), peroxide value (0.05 ± 0.01 g/100 g), saponification value (194.4 ± 0.5 mg KOH/g) and iodine value (109.7 ± 0.8 g I/100 g). The basic information obtained provides data support for quality evaluation and efficacy research of ZSS oil, and suggests its prospects for development in pharmaceutical and food industries.

Abstract:A simple,reproducible and reliable HPLC fingerprint analysis method of Ziziphi Spinosae Semen (ZSS) and Ziziphi Mauritianae Semen (ZMS) was established,and then it was applied to analyze samples collected from different locations.A total of 12 common fingerprint peaks were designated in ZSS and ZMS,respectively,and five of which were definitely identified.Of these common peaks,10 peaks were presented in both ZSS and ZMS,whereas two were characteristic for ZSS and ZMS,respectively.Furthermore,the two characteristic peaks of ZMS were definitely assigned as 6'"-sinapoylspinosin and frangufoline by HPLC guided purification and structural elucidation.The HPLC fingerprint analysis was proved to be useful in identifying and discriminating ZSS and ZMS,which was beneficial for quality control of ZSS and its products.
Aryl hydrocarbon hydroxylase activity in human placenta and threatened preterm delivery.

Abstract:Induction of aryl hydrocarbon hydroxylase (AHH) activity in the placenta has been well documented. This enzyme may be induced by a variety of Polycyclic Aromatic Hydrocarbons (PAHs) and the AHH inducibility is associated with harmful effects of environmental chemicals. Toxic effects of PAHs in tissues such as placenta have been demonstrated to be due to their metabolites, epoxides, which interact with DNA. Thus, environmental PAHs may be related to its alterations in fetal development. Founded on these findings the PAH metabolites could interfere with the normal course of the pregnancy and may be an aborticide, a teratogen or a carcinogen. We hypothesize that low increased activity of placental Aryl Hydrocarbon Hydroxylase (AHH) may be an important determinant of human fetotoxicity. The present investigation was designed to examine the possible implications of PAH exposure at environmental exposure levels on the normal course of the pregnancy using AHH induction as an indicator of PAH exposure. Threatened Preterm Delivery (TPD) was used as an index of problems in the normal course of pregnancy. A group of forty pregnancies at term with TPD was compared with eighty controls for placental AHH induction. Macroscopic placental examination was also performed. A significant increase in prevalence of placental AHH induction with TPD was shown (Odds-Ratio = 2.8; 95% confidence bounds [1.3-6.2]; chi 2 = 6.7 p < 0.01). No such increases were found associated with placental pathology. When taking into account the group of placenta without basal plate calcifications, the significant increase in prevalence of placental AHH induction with TPD above mentioned was greatly increased (Odds-Ratio = 8.9; 95% confidence bounds [2.4-32.9]; chi 2 = 11.1 p < 0.001) controlling for gestational age. The increase in prevalence of placental AHH induction with TPD disappeared when taking into account the subgroup with basal plate or parenchyma calcifications. It is hypothesized that the high estrogen and progesterone at term may explain these associations.

Abstract:In the study, high‐speed counter‐current chromatography was used for separation and purification of magnoflorine, spinosin, and 6‴‐feruloyspinosin from Ziziphi Spinosae Semen. With n‐butanol‐ethyl acetate‐water (2:3:5, v/v) as the optimum solvent system, about 75 mg of magnoflorine, 110 mg of spinosin, and 40 mg of 6‴‐feruloyspinosin were isolated from 0.5 g of crude extract of Z. Spinosae Semen, with the purity of 95.7, 97.2, and 96.4%, respectively. The chemical structures were identified by MS and NMR spectroscopy. In addition, the antidepressant activity of the isolated components was evaluated by PC12 cells injury model and chronic unpredictable mild stress depression mouse model. The results showed that high‐speed counter‐current chromatography could be used to realize the one‐time rapid preparation and separation of magnoflorine, spinosin, and 6‴‐feruloyspinosin from Z. Spinosae Semen and compatibility of these isolated components has certain antidepressant activity.

Abstract:Ziziphi Spinosae Semen (ZSS) is a valued seed renowned for its sedative and sleep-enhancing properties. However, the price increase has been accompanied by adulteration. In this study, chromaticity analysis and Fourier transform near-infrared (FT-NIR) combined with multivariate algorithms were employed to identify the adulteration and quantitatively predict the adulteration ratio. The findings suggested that the utilization of chromaticity extractor was insufficient for identification of adulteration ratio. The raw spectrum of ZMS and HAS adulterants extracted by FT-NIR was processed by SNV + CARS and 1d + SG + ICO respectively, the average accuracy of machine learning classification model was improved from 77.06 % to 97.58 %. Furthermore, the R2 values of the calibration and prediction set of the two quantitative prediction regression models of adulteration ratio are greater than 0.99, demonstrating excellent linearity and predictive accuracy. Overall, this study demonstrated that FT-NIR combined with multivariate algorithms provided a significant approach to addressing the growing issue of ZSS adulteration.

Abstract:As a well-known traditional Chinese medicine, Ziziphi Spinosae Semen has been used for treating anxiety and insomnia for a long time. Spinosin, the main active C-glycoside flavonoid in Ziziphi Spinosae Semen, has attracted much attention because of its many pharmacological activities, including strong hypnotic effects, anxiolytic-like effects, and so on. In the present work, high-purity spinosin was separated from Ziziphi Spinosae Semen using the HPD-300 resin followed by preparative high-performance liquid chromatography. The adsorption kinetics curve of spinosin on the HPD-300 resin was studied and fitted well by the pseudo-second-order equation. The adsorption isotherms were also constructed, and low temperature favored the adsorption reaction. The separation parameters were optimized using dynamic adsorption and desorption tests. After a one-run treatment with HPD-300 resin, the concentration of spinosin increased 11.8-fold from 0.99% to 11.7% with a recovery yield of 80.4%. Furthermore, the purity of spinosin could surpass above 98% after separation by preparative high-performance liquid chromatography and recrystallization with a recovery yield of 72.6%. The developed method was effective and suitable for the large-scale preparation of spinosin. Moreover, it was confirmed that HPD-300 resin could enable good selection for the enrichment of flavonoids from different plants. This article is protected by copyright. All rights reserved.

Abstract:Background Ziziphi spinosae semen (ZSS), dry mature seeds of Ziziphus jujuba Mill. var. spinosa (Bunge) Hu ex H.F. Chow., are widely used in the treatment of insomnia, fright palpitations, and profuse dreaming in medical treatments. The counterfeit species of ZSS, Ziziphi mauritianae semen (ZMS), and Hovenia dulcis Thunb (HDT) are seeds from Ziziphus incurva Roxb. and Hovenia acerba Lindl., respectively. However, the pharmacopeia is not recorded for ZMS and HDT. In this article, we reviewed the identification methods of ZSS, ZMS, and HDT. Materials and Methods The random amplified polymorphic DNA (RAPD) technique was used to identify the ZSS and its copies. Plant genome DNA was extracted from Z. jujuba Mill., Z. incurva Roxb., and H. acerba Lindl. Random primers were designed to amplify the genome. The amplified products were detected by agarose gel electrophoresis. Results By analyzing the polymorphism of the amplified product DNA fragment, the DNA fingerprint maps of Z. jujuba Mill., Z. incurva Roxb., and H. acerba Lindl. were successfully constructed.

Abstract:Ziziphi Spinosae Semen (ZSS) is the first choice for the treatment of insomnia. This research aimed to reveal the spatial distribution of identifying quality markers of ZSS and to illustrate the metabolite quality characteristics of this herbal medicine. Here, we performed a matrix-assisted laser desorption/ionization-mass spectrometry imaging (MALDI-MSI) in situ to detect and image 33 metabolites in ZSS, including three saponins, six flavonoids, four alkaloids, eight fatty acids, and 12 amino acids. The MALDI images of the metabolites clearly showed the heterogeneous spatial distribution in different regions of ZSS tissues, such as the cotyledon, endosperm, and radicle. The distribution area of two saponins, six flavonoids, and three alkaloids increased significantly after the fried processing of ZSS. Based on the ion images, samples with different processing technologies were distinguished unambiguously by the pattern recognition method of orthogonal partial least squares discrimination analysis (OPLS-DA). Simultaneously, 23 major influencing components exerting higher ion intensities were identified as the potential quality markers of ZSS. Results obtained in the current research demonstrate that the processing of ZSS changes its content and distribution of the medicinal components. The analysis of MALDI-MSI provides a novel MS-based molecular imaging approach to investigate and monitor traditional medicinal plants.

Abstract:A sensitive and reliable ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry method was established to separate and identify the chemical constituents of Zhi-Zi-Da-Huang decoction, a classic traditional Chinese medicine formula. The chromatographic separation was achieved on a Shim-pack XR-ODS C18 column (75 × 3.0 mm, 2.2 μm) using a gradient elution program. The detection was performed on a Waters Xevo G2 Q-TOF mass spectrometer equipped with electrospray ionization source in both positive and negative modes. With the optimized conditions, a total of 82 compounds were identified or tentatively characterized. Of the 82 compounds, 21 compounds were identified by comparing the retention time and MS data with reference standards, the rest were characterized by analyzing MS data and retrieving the reference literature. In addition, 31 compounds were identified from Gardenia jasminoides Ellis, ten compounds were identified from Rheum palmatum L., 33 compounds were identified from Citrus aurantium L., and eight compounds were identified from Sojae Semen Praeparatum. Results indicated that iridoids, anthraquinones, flavonoids, isoflavonoids, coumarins, glycosides of crocetin, monoterpenoids, and organic acids were major constituents in Zhi-Zi-Da-Huang decoction. It is concluded that the developed ultra high performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry method with high sensitivity and resolution is suitable for identifying and characterizing the chemical constituents of Zhi-Zi-Da-Huang decoction, and the analysis provides a helpful chemical basis for further research on Zhi-Zi-Da-Huang decoction.

Abstract:"Zhu She Yong Xue Shuan Tong" lyophilized powder (ZSYXST), consists of a series of saponins extracted from Panax notoginseng, which has been widely used in China for the treatment of strokes. In this study, an ultra-high performance liquid chromatography with quadrupole time-of-flight mass spectrometry (UHPLC-Q-TOF/MS) combined with preparative high performance liquid chromatography (PHPLC) method was developed to rapidly identify both major and minor saponins in ZSYXST. Some high content components were removed through PHPLC in order to increase the sensitivity of the trace saponins. Then, specific characteristic fragment ions in both positive and negative mode were utilized to determine the types of aglycone, saccharide, as well as the saccharide chain linkages. As a result, 94 saponins, including 20 pairs of isomers and ten new compounds, which could represent higher than 98% components in ZSYXST, were identified or tentatively identified in commercial ZSYXST samples.

A hyphenated electrospray ionization and Q-TOF analyzer was used for the determination of accurate mass of the protonated or deprotonated molecule and fragment ion in both negative and positive modes. The chromatographic separation was achieved by UPLC, which used a column with 1.7 μm particle packing which enabled higher speed of analysis, peak capacity, greater resolution and increased sensitivity. The constituents of SZRD were identied and confirmed according to the mass spectrometric fragmentation mechanisms, MS/MS fragment ions, relevant literature and the establishment of an in-house molecular formula database. With this method, a total of 22 compounds of SZTD were tentatively identied based on MS and MS/MS data and comparison with available databases. It is concluded that a rapid and robust platform based on UPLC-ESI-Q-TOF-MS was established, which is useful for identifying multiple-constituent of traditional Chinese medicine (TCM) prescriptions. Our present results proved that the established method could provide helpful chemical information for further pharmacological mechanism research of SZRD.
Identification of Multiple Constituents in Chinese Medicinal Prescription Shensong Yangxin Capsule by Ultra-Fast Liquid Chromatography Combined with Quadrupole Time-Of-Flight Mass Spectrometry

Abstract:A practical method using ultra-fast liquid chromatography in tandem with quadrupole time-of-flight mass spectrometry combined with dynamic background subtraction technology was developed for the rapid separation and identification of the complicated constituents in the Shensong Yangxin capsule (SSYX). The chromatographic separation was performed on a C18 column (2.1 x 100 mm, 2.6 mu m) with a gradient elution program using methanol and 0.1% formic acid aqueous solution as the mobile phase at a flow rate of 0.4 mL min(-1). Accurate mass measurements of the molecular ions in the full scan and the characteristic fragment ions triggered by information-dependent acquisition provided reliable identification criteria. Thus, 99 compounds, including saponins, phenolic acids, tanshinones, lignans, terpenoids, alkaloids and flavonoids, were unambiguously or tentatively identified in 40 min by comparing their retention times and accurate mass measurements for each molecular ion and its subsequent fragment ions with those of authentic standards or literature data. Simultaneously, all the compounds were further assigned to the individual raw materials. In conclusion, these results will provide a basis for quality control and further study of SSYX, and the proposed technique based on high-resolution mass spectrometry would be expected to be adaptable to the analysis of complicated constituents in various complex matrices.

Abstract:The present research deals with a psychometric evaluation of depressive features in a group of laid-off compared with a control group of full-time workers. Sex differences has been taken into account in the comparison. (The laid-off workers belong to a group under an unemployment fund, Cassa Integrazione Guadagni, which continues paying about 80% of the ex-employee's last wage for an undetermined period of time). The psychometric elements considered include the D and Ma scales of MMPI and their difference as proposed by Diamond, and the Beck Depression Rating Scale. The results, still preliminary, show that work deprivation, even with the financial subsidy from the unemployment fund, can be considered a life event able to provoke a depressive symptomatology.
