Abstract:A facile and direct intramolecular oxidative C(aryl)-H amidation reaction was developed for the synthesis of quinolino[3,4-b]quinoxalin-6(5H)-ones in moderate to excellent yields starting from readily available materials by tethering the adjacent N-methoxyamide and aryl portions in the presence of phenyliodine(III) bis(trifluoroacetate) at room temperature. This metal-free approach is a valuable addition to the traditional methods already available for the preparation of these molecules.

Abstract:A reduction of quinolines to synthetically versatile tetrahydroquinoline molecules with I2 and HBpin is described. In the presence of iodine (20 mol%) as a catalyst, reduction of quinolines and other N-heteroarenes proceeded readily with hydroboranes as the reducing reagents. The broad functional-group tolerance, good yields and mild reaction conditions imply high practical utility.

Abstract:An efficient ring-closing reaction was developed for the synthesis of spirocyclopropane quinolinediones from 2,2-disubstituted 2-benzoylacetamides in the presence of [bis-(trifluoroacetoxy)iodo]benzene (PIFA). This reaction proceeds at room temperature in the absence of a metal catalyst to provide spirocyclopropane quinolinediones in good to excellent yields. Furthermore, these compounds were readily converted to the corresponding pyrrolo[3,2-c] quinolinones to provide tricyclic ring structures similar to those found in the natural products martinellic acid and martinelline via an intermolecular amine ringopening cyclization reaction.

Abstract:The Winterfeldt oxidation (NaOH, DMF, air, rt) of substituted 1,2,3,4-tetrahydro-gamma-carbolines has been developed, which provides a convenient and efficient method for the synthesis of the corresponding dihydropyrrolo[3,2-b]quinolones in moderate to excellent yields (38-94%). The generality and substrate scope of this reaction are explored and a possible mechanism is proposed. The results imply that electron-withdrawing groups on N-2 of tetrahydro-gamma-carbolines and N-5-H are necessary. The synthesis of 5 or 7-substituted pyrrolo[3,2-b]quinolones in near quantitative yields was also achieved through deprotection and aromatization of N-1-Boc-dihydropyrrolo[3,2-b]quinolones.

Abstract:2-Perfluoroalkylated quinolines were efficiently synthesized via the Michael addition of perfluoroalk-2-ynoates with isatins, followed by the reaction with alcohols through an intramolecular rearrangement cyclization in the presence of Na2CO3. Under the mild and metal-free reaction conditions, a broad scope of quinolines was synthesized and the mechanism was proposed.

Abstract:An efficient iodine-catalyzed [3+3+1] annulation for the construction of dihydrooxepine scaffolds with quinoline units was developed. This strategy involves a seven-membered dihydrooxepine with a broad substrate scope through a formal three-component tandem reaction. Further derivation of the target product produced a trioxabicycle scaffold, which formed the basic core of natural products and pharmaceutical molecules.

Abstract:Herein, a straightforward and efficient approach to iodinated indolo[1,2-c]quinazoline amines via I2/CHP-promoted cascade annulation of diarylalkynes and isocyanides has been described. This method features excellent regio- and chemoselectivities, good functional group compatibility, as well as mild reaction conditions. Mechanistic studies suggest that CHP is essential to the catalytic cycle of the reaction. The newly formed indolo[1,2-c]quinazoline scaffolds can be further modified, showing potential applications in medicinal chemistry and materials science.

Abstract:A highly efficient method for the direct construction of amide bonds via a selective cleavage of C–H and CC bonds in indole structures using an iodine-promoted approach was developed. Mechanistic studies indicated the formation of superoxide radicals obtained from molecular oxygen activation as a key intermediate step, which provided a precursor for subsequent oxidative ring-opening and intermolecular cyclization. A broad range of quinazolin-4(3 H )-ones and tryptanthrins were synthesized in moderate to good yields under mild and environmentally benign conditions.

Abstract:Herein, an eco‐friendly and atom‐economical electrochemical methodology with isatins and 1,2,3,4‐tetrahydroisoquinolines through NHPI/O2‐mediated intermolecular oxidation/annulation is disclosed, leading to a variety of polycyclic quinazolinones in 46‐82% yields. This oxidative cyclization proceeded in transition metal‐ and strong oxidant‐free conditions and generated H2 and CO2 as byproducts. Additionally, late‐stage functionalization and broad substrate scope demonstrated the synthetic usefulness of this protocol.
