However, phosphodiesterase type 5 inhibitor therapy often has reduced efficacy in DMED patients compared to the general population. This study was designed to determine whether chronic Sildenafil administration can modify the impaired vascular endothelial growth factor (VEGF) system and improve the erectile function in rats with diabetic erectile dysfunction. METHODS Diabetes was induced in a group of Sprague Dawley rats (n=30) by intraperitoneal injection of streptozotocin (40mg/kg). These rats were screened for erectile dysfunction by subcutaneous injection of apomorphine (100μg/kg). Rats with DMED were given either vehicle or sildenafil (5mg/kg or 10mg/kg) for 10 weeks. A non-diabetic and age-matched control group (n=10) was also allocated and fed a normal diet for the same period. Assessments were performed in all groups 36 h after the last dose of sildenafil. Penile intracavernous pressure (ICP), mean arterial pressure (MAP), and the area under the curve (AUC) were measured. In addition, immunohistological analysis of penile tissue and Western blotting for VEGF, vascular endothelial growth factor receptor 1(VEGFR1), and endothelial nitric oxide synthase (eNOS) were performed. RESULTS Significant increases in ICP, ICP/MAP ratio, and AUC were observed in both sildenafil-treated groups compared to DMED rats that did not receive Sildenafil. Immunohistochemical staining of the penile tissue from these rats showed a decrease in VEGF, VEGFR1, and eNOS staining in penile tissue DMED rats; however, chronic Sildenafil administration led to an increase in staining for all three factors. Western blot analysis confirmed these results. CONCLUSIONS We demonstrated that daily Sildenafil administration led to a progressive improvement in both endothelial function and erectile function in DMED rats, and our data suggests that this may be the result of improved signaling through the VEGF/eNOS signaling cascade. © 2011 by American Urological Association Education and Research, Inc.FiguresReferencesRelatedDetails Volume 185Issue 4SApril 2011Page: e675-e676 Advertisement Copyright & Permissions© 2011 by American Urological Association Education and Research, Inc.MetricsAuthor Information XiangZhou Sun Guangzhou, China, People's Republic of More articles by this author GuiHua Liu Winston-Salem, NC More articles by this author Yuanyuan Zhang Winston-Salem, NC More articles by this author ChunHua Deng Guangzhou, China, People's Republic of More articles by this author Expand All Advertisement Advertisement PDF downloadLoading ...
