Ocular toxicity of subconjunctival rapamycin in Lewis rats

Abstract:Objective To investigate the ocular toxicity of subconjunctival administered rapamycin in Lewis rats.Methods Twenty Lewis rats were devided into five groups randomly,four in each group.Rapamycin was subconjunctivally administered in the right eyes of Lewis rats at concentrations of 4 μg·kg-1,20 μg·kg-1,40 μg·kg-1,100 μg·kg-1 and 200 μg·kg-1,respectively.Left eyes received 5 μL balanced salt solution as control group.Slit-lamp biomicroscopy,direct ophthalmoscopy,electroretinography test and histological examination were performed in all eyes before and after the injections.Results The lens was clear;Vitreous body was without cloudiness;No coarctation was found in retinal vessels.There was no statistical difference in eyes with electroretinogram(P>0.05).With pathological examination,corneal structure of all rats were clear;No abnormality was seen in iris stroma and epithelium;Each layer of retina was with clear structure,regular shape,but not with cellular edema.No significant difference was found in all groups with slit-lamp biomicroscopy,direct ophthalmoscopy,electroretinogram and histopathological examination before and after subconjunctival injection.No evidence of toxic reactions,such as conjectival congestion,chemosis and necrosis,was seen in the eyes receiving 4 μg·kg-1,20 μg·kg-1,40 μg·kg-1,100 μg·kg-1 and 200 μg·kg-1 of rapamycin.Conclusion It is safe to subconjunctival inject rapamycin with less than the dose of 200 μg·kg-1 in Lewis rats.No abnormality was found in eyes structure and retinal function.
