Establishment of a Rat Chronic Elevated Intraocular Pressure Model by Episcleral Veins Ligation and Subconjunctival 5-Fu Injection

Abstract:Objective In order to investigate the mechanism of glaucomatous optic nerve injury and protection,the aim of the present study is to establish a stable chronic elevated intraocular pressure glaucoma model in rat.Methodes Sixty-five male SD rats weighing 300±20 g were included in this study.The rats were divided into two groups: improved experiment group(60 rats) and control experiment group(5 rats).Ligation of 2 episcleral veins with subconjunctival 5-Fu injection was performed in the improved experiment group and only ligation of 2 episcleral veins was conducted in the control experiment group,to induce elevated intraocular pressure.Intraocular pressure(IOP) was measured by a tonometer in rats under ketamine-xylazine anesthesia.The degree of retinal ganglion cells(RGC) loss was assessed by cresyl violet staining at 1,4,6,10 weeks after operation in the experiment group.Results Elevated IOP was consistently maintained for up to 10 weeks in the improved experiment group.The survival rate of RGCs in operation eyes was 90.16%,83.50%,75.01%,62.37% at 1,4,6,10 weeks after operation,respectively,in comparison with the values in control eyes.However,elevated IOP only maintained 2 weeks in control group.Conclusion An ideal rat chronic elevated intraocular pressure glaucoma model has been successfully established by ligation of 2 episcleral veins with subconjunctival 5-Fu injection in rat eyes.This method can provide an excellent animal model maintaining long-term elevated intraocular pressure for glaucoma research and the preparation procedure of this model is rather simple,reliable and reproducible.
