Abstract:We present a microfluidic synthesizer made entirely of Teflon material for solid phase peptide synthesis (SPPS). Solvent-resistant perfluoroalkoxy (PFA) was used to construct chip-sized devices featuring multiple tri-layer pneumatic microvalves. Using these devices, model peptides were automatically synthesized and cleaved in situ in a continuous-flow manner. The total coupling and cleavage time was significantly reduced compared to conventional bulk reactors. The synthesis of a decapeptide, for instance, took less than 6 h using our device while it usually takes more than three days using conventional reactors.
Porous pDMS Microreactor Array Chip for De Novo DNA Synthesis

Abstract:This paper reports a facile fabrication method of polymeric chip with about five-fold increasement in reaction area by in-situ constructing porous microreactor array, which is therefore beneficial for low-cost, high throughput and high capacity de novo DNA synthesis. Microwell arrays were laser-engraved on polytetrafluoroethylene (PTFE) substrate, filled with sugar and then casted with polydimethylsiloxane (PDMS). After sugar removed, porous PDMS (pPDMS) arrays would be incorporated easily. A 40-mer oligonucleotides were successfully synthesized on the pPDMS array and validated by fluorescent hybridization experiments. With the high reaction sites provided by pPDMS, the yield of oligo is about four times higher than the pristine planar PDMS with the same footprint. In addition, the porosity of pPDMS can be easily adjusted to meet different yield requirements.

Abstract:Polydimethylsiloxane (PDMS) is a popular choice for the microfluidic chip fabrication; however, it still meets several significant challenges, including poor solvent compatibility, non-reprocessability, and non-recyclability. The innovation of new elastomers that overcomes these drawbacks pushes forward the application of microfluidics in organic synthesis and avoids potential environmental threats from chip waste. Herein, we reported a solvent-resistant perfluoropolyether (PFPE)-based thermoset elastomer that can be reprocessed like thermoplastics and degraded back to its raw material. The solvent-resistant elastomer was synthesized by crosslinking PFPE-diol into a dynamic covalent network. Since the PFPE chain is repellent to most solvents, this PFPE-based elastomer exhibited much lower swelling ratios in comparison with PDMS. Dye diffusion experiments on microfluidic chips convinced about the success in preventing swelling-induced dye leakage or diffusion. To present the thermoplasticity and reprocessability, one specific spiral microfluidic chip was cut into small pieces and reshaped into another chip that allowed laminar flow of two types of solutions. Moreover, the PFPE-based elastomer was chemically degraded and over 90% PFPE-diol was recycled. This solvent-resistant and almost fully recyclable elastomer provides a new material choice for microfluidics and is bound to play an important role in microfluidic studies with respect to organic chemistry.

Abstract:Reverse DNA synthesis (5′-3′) plays diverse functional roles in cellular biology, biotechnology, and nanotechnology. However, current microfluidic systems for synthesizing single-stranded DNAs at a laboratory scale are limited. In this work, we develop a simple and efficient polydimethylsiloxane- (PDMS-) based microfluidic system for the reverse chemical synthesis of short-chain oligonucleotides (in the 5′-3′ direction) under ambient conditions. The use of a microfluidics device and anhydrous conditions effectively surpass the problem of moisture sensitivity during oligonucleotide synthesis. With optimized microfluidic synthesis conditions, the system is able to synthesize up to 21 bases-long oligonucleotides in air atmosphere. The as-synthesized oligonucleotides, without further purification, are characterized using matrix-assisted laser desorption ionization–time of flight (MALDI-TOF/TOF) mass spectroscopy (MS) supported by the denatured polyacrylamide gel electrophoresis (PAGE) analysis. This developed system is highly promising for producing the desired sequence at the nanomolar scale on-chip and on-demand in the near future.

Abstract:An easy method is introduced allowing fast polydimethylsiloxane (PDMS) replication of nanofluidic lab-on-chip devices using accurately fabricated molds featuring cross-sections down to 60 nm. A high quality master is obtained through proton beam writing and UV lithography. This master can be used more than 200 times to replicate nanofluidic devices capable of handling single DNA molecules. This method allows to fabricate nanofluidic devices through simple PDMS casting. The extensions of YOYO-1 stained bacteriophage T4 and λ−DNA inside these nanochannels have been investigated using fluorescence microscopy and follow the scaling prediction of a large, locally coiled polymer chain confined in nanochannels.

Abstract:Elastomeric materials, most prominently polydimethylsiloxane, are of great importance for microfluidic systems. Unfortunately, most elastomeric materials used in these systems so far show poor chemical resistance. Fluorinated elastomers are promising for chemistry-on-chip applications performed on these microfluidic systems, but challenging to shape on the microscale and difficult to bond which is a required step for the fabrication of embedded chips. In this work, a custom-synthesized fluorinated photocurable resin which can be structured using vat photopolymerization 3D printers, and shows a high elasticity combined with a remarkable chemical resistance was demonstrated. The material provides excellent stretchability, with a linear deformation capability of 523% and remains stretchable even after being soaked in tetrahydrofuran for 24 hours. By using multi-material printing microfluidic chips with bonded hard and soft segments of different fluorinated resins were fabricated. Also, a variety of chemically resistant microfluidic components including pneumatic valves and peristaltic pumps were proven feasible. This novel fluorine-containing resin is expected to initiate further advancements of chemistry-on-chip application.

Abstract:We report the design and synthesis of a new perfluoropolyether-based material, which has liquid-like viscosity and can be cured into a tough, highly durable elastomer when “clicked” with selected tri-pod organic small molecules. This highly fluorinated elastomer exhibits remarkable resistance to a variety of organic solvents, water, heat and even harsh acidic and basic conditions. Whereas PDMS-based microfluidic devices are commonly used for aqueous based applications, their limited chemical resistance and high swellability in many common organic solvents make it unfeasible for microfluidic applications involving organic solvents and/or harsh conditions. With excellent chemical resistance and low swellability, our newly synthesized fluoro-elastomers will hopefully provide an alternative material for organic based microfluidic devices. Furthermore, the alkyne–azide “click” chemistry employed in curing not only provides high efficiency of synthesis and ease of device fabrication, but, more importantly, produces 1,2,3-triazole linkages that are very stable against harsh acidic or basic conditions. This work has great potential to expand microfluidics to a series of novel applications especially in organic and medicinal chemistry.
In Situ Synthesis of Oligonucleotides on Plasma-Modified PTFE for Fabricating DNA Chips

Abstract:Oligonucleotide probe arrays were in-situ synthesized on the H-2/N-2 plasma modified poly(terafluoroethylene) (PTFE) surface via micro-fluidic channels connected with an automated synthesizer. A contact angle measurement of water droplets was used to ascertain the hydrophilicity of the surface. X-ray photoelectron spectroscopy (XPS) analysis confirmed the presence of polar amino groups on the surface. Ultra-violet (UV) spectrum analysis indicated that the surface showed a coupling efficiency higher than 98% for in situ synthesis of oligonucleotides arrays. The probe array specificity was discriminated by hybridizing with fluorescent target sequences. Oligonucleotide probe arrays on modified PTFE surface showed high stability and durability after repetitive denaturing and hybridizing. The results implied the plasma modified PTFE surface was extremely stable, performed well in DNA hybridization assays and could service as a good substrate for high-density oligonucleotide array synthesis.

Abstract:We have developed a method for rapid prototyping of hard polymer microfluidic systems using solvent imprinting and bonding. We investigated the applicability of patterned SU-8 photoresist on glass as an easily fabricated template for solvent imprinting. Poly(methyl methacrylate) (PMMA) exposed to acetonitrile for 2min then had an SU-8 template pressed into the surface for 10min, which provided appropriately imprinted channels and a suitable surface for bonding. After a PMMA cover plate had also been exposed to acetonitrile for 2min, the imprinted and top PMMA pieces could be bonded together at room temperature with appropriate pressure. The total fabrication time was less than 15min. Under the optimized fabrication conditions, nearly 30 PMMA chips could be replicated using a single patterned SU-8 master with high chip-to-chip reproducibility. Relative standard deviations were 2.3% and 5.4% for the widths and depths of the replicated channels, respectively. Fluorescently labeled amino acid and peptide mixtures were baseline separated using these PMMA microchips in <15s. Theoretical plate numbers in excess of 5000 were obtained for a ∼3cm separation distance, and the migration time relative standard deviation for an amino acid peak was 1.5% for intra-day and 2.2% for inter-day analysis. This new solvent imprinting and bonding approach significantly simplifies the process for fabricating microfluidic structures in hard polymers such as PMMA.

Abstract:AbstractSynthetic oligonucleotides (oligos) are important tools in the fields of molecular biology and genetic engineering. For applications requiring a large number of oligos with high concentration, it is critical to perform high throughput oligo synthesis and achieve high yield of each oligo. This study reports a microreactor chip for oligo synthesis. By incorporating silica beads in the microreactors, the surface area of the solid substrate for oligo synthesis increases significantly in each microreactor. These beads are fixed in the microreactors to withstand the flushing step in oligo synthesis. Compared to conventional synthesis methods, this design is able to avoid protocols to hold the beads and integrate more microreactors on a chip. An inkjet printer is utilized to deliver chemical reagents in the microreactors. To evaluate the feasibility of oligo synthesis using this proof-of-concept synthesizer, an oligo with six nucleotide units is successfully synthesized.

Abstract:We demonstrate a highly integrated microfluidic chip with the function of DNA amplification. The integrated chip combines giant electrorheological-fluid actuated micromixer and micropump with a microheater array, all formed using soft lithography. Internal functional components are based on polydimethylsiloxane (PDMS) and silvercarbon black-PDMS composites. The system has the advantages of small size with a high degree of integration, high polymerase chain reaction efficiency, digital control and simple fabrication at low cost. This integration approach shows promise for a broad range of applications in chemical synthesis and biological sensinganalysis, as different components can be combined to target desired functionalities, with flexible designs of different microchips easily realizable through soft lithography.

Abstract:A microfluidic polymer chip for the self-assembly of DNA conjugates through DNA-directed immobilization is developed. The chip is fabricated from two parts, one of which contains a microfluidic channel produced from poly(dimethylsiloxane) (PDMS) by replica-casting technique using a mold prepared by photolithographic techniques. The microfluidic part is sealed by covalent bonding with a chemically activated glass slide containing a DNA oligonucleotide microarray. The dimension of the PDMS-glass microfluidic chip is equivalent to standard microscope slides (76 x 26 mm(2)). The DNA microarray surface inside the microfluidic channels is configured through conventional spotting, and the resulting DNA patches can be conveniently addressed with compounds containing complementary DNA tags. To demonstrate the utility of the addressable surface within the microfluidic channel, DNA-directed immobilization (DDI) of DNA-modified gold nanoparticles (AuNPs) and DNA-conjugates of the enzymes glucose oxidase (GOx) and horseradish peroxidase (HRP) are carried out. DDI of AuNPs is used to demonstrate site selectivity and reversibility of the surface-modification process. In the case of the DNA-enzyme conjugates, the patterned assembly of the two enzymes allows the establishment and investigation of the coupled reaction of GOx and HRP, with particular emphasis on surface coverage and lateral flow rates. The results demonstrate that this addressable chip is well suited for the generation of fluidically coupled multi-enzyme microreactors.

Abstract:Fused deposition modeling (FDM) is well suited for microfluidic prototyping due to its low investment cost and a wide range of accessible materials. Nevertheless, most commercial FDM materials exhibit low chemical and thermal stability. This reduces the scope of applications and limits their use in research and development, especially for on-chip chemical synthesis. In this paper, we present FDM fabrication of microfluidic chips with polyvinylidene fluoride (PVDF) for applications that require high thermal or chemical resistance. Embedded microchannels with a minimum channel width and heights of ~200 μm × 200 μm were fabricated, and the resistance against common solvents was analyzed. A procedure was developed to increase the optical transmission to result in translucent components by printing on glass. Chips for fluid mixing were printed, as well as microreactors that were packed with a catalytically active phase and used for acetal deprotection with a conversion of more than 99%. By expanding the use of fluorinated polymers to FDM printing, previously challenging microfluidic applications will be conducted with ease at the lab scale.

Polymeric Microfluidic Devices Fabricated Using Epoxy Resin for Chemically Demanding and Day-Long Experiments

Abstract:Polydimethylsiloxane (PDMS) is a widely used material in laboratories for fabricating microfluidic devices with a rapid and reproducible prototypingability, owing to its inherent properties (e.g., flexibility, air permeability, and transparency). However, the PDMS channel is easily deformed under pressures applied to generate flows because of its elasticity, which can affect the robustness of experiments. In addition, air permeability of PDMS causes the pervaporation of water, and its porous structure absorbs oil and even small hydrophobic molecules, rendering it inappropriate for chemically demanding or day-long experiments. In this study, we develop a rapid and reproducible fabrication method for polymer-based rigid microfluidic devices, using epoxy resin that can overcome the limitations of PDMS channels, which are structurally and chemically robust. We first optimize a high-resolution fabrication protocol to achieve convenient and repeatable prototyping of polymeric devices via epoxy casting using PDMS soft molds. In addition, we compare the velocity changes in PDMS microchannels by tracking fluorescent particles in various flows (~133 μL/min) to demonstrate the structural robustness of the polymeric device. Furthermore, by comparing the adsorption of fluorescent hydrophobic chemicals and the pervaporation through channel walls, we demonstrate the excellent chemical resistance of the polymeric device and its suitability for day-long experiments. The rigid polymeric device can facilitate lab-on-chip research and enable various applications, such as high-performance liquid chromatography, anaerobic bacterial culture, and polymerase chain reaction, which require chemically or physically demanding experiments.
Composite Elastomer-Enabled Rapid Photofabrication of Microfluidic Devices

Abstract:Microfluidics, as an emerging technology, is highly dependent on the evolution of device materials and fabrication techniques. While replica molding of polydimethylsiloxane and hot embossing/injection molding of thermoplastics are most popular, they are either hard to scale up or inappropriate for laboratory-scale prototyping. Recently, photocurable resins, as a huge class of materials, have attracted extensive interest. However, very few of them can now be used in device fabrication due to the challenge in machining these materials. In response, we herein propose a novel concept of composite elastomers, which can covalently link with and consequently offer a flexible support to photocured thin films. This effect would allow most photocurable resins to be used in microfluidic device fabrication, greatly enriching the material choices for diverse applications. Moreover, the whole fabrication process becomes very simple and rapid, with an impressive throughput of at least hundreds of replicas per day. With these features, it is reasonably expected that the composite elastomer-enabled rapid photofabrication method will be very competent for laboratory prototyping, providing not only the ease of fabrication but also a possibility to select the materials specifically for ultimate applications and promising potential for volume production without the redevelopment process. Abstract:A microfluidic system combining tempera tu re-controlled reactor, analyte delivery, chip electrophoresis (CE) separation, and fluorescence detection was developed, in which the heaters, resistance temperature detectors, enzymatic reactors, CE channels, and pneumatic valves/pumps were integrated onto a single glass-PDMS chip. The microdevice was used to perform the digestion reaction, followed by on-line electrophoresis separation and detection of the resulting fragments with endonuclease BamHI and FokI as models. Pneumatic valves/pumps served not only for isolating the reaction region from the separation medium to prevent contamination, but also for delivering and quantitatively diluting the fluid from the reaction chamber to the CE section. Thus enzymatic reaction and electrophoresis separation could be insulated and connected as needed. A dynamic coating procedure with the use of PVP and mannitol was firstly adopted for glass-PDMS hybrid chip-based DNA separations, leading to an improved separation efficiency with reproducible migration time and theoretical plates. The expected 263- and 287-bp digestion products of BamHI and FokI were definitely verified by the size-based electrophoretic separation and detection. The whole integrated reaction-CE system can be manipulated in a simple manner with good reproducibility, which is expected to be applied in other on-line analysis of various biochemical reactions.

Abstract:Creating new functional building blocks that expand the versatility of nanostructures depends on bottom-up self assembly of amphiphilic biomolecules. Inspired by the unique physicochemical properties of hydrophobic perfluorocarbons, coupled with the powerful functions of nucleic acids, we herein report the synthesis of a series of diperfluorodecyl-DNA conjugates (PF-DNA) which can efficiently self-assemble into micelles in aqueous solution. On the basis of the micelle structure, both target binding affinity and enzymatic resistance of the DNA probe can be enhanced. In addition, based on the hydrophobic effect, the PF-DNA micelles (PFDM) can actively anchor onto the cell membrane, offering a promising tool for cell-surface engineering. Finally, the PFDM can enter cells, which is significant for designing carriers for intracellular delivery. The combined advantages of the DNA micelle structure and the unique physicochemical properties of perfluorocarbons make these PFDM promising for applications in bioimaging and biomedicine.
Bead-Droplet Reactor for High-Fidelity Solid-Phase Enzymatic DNA Synthesis

Abstract:Solid-phase synthesis techniques underpin the synthesis of DNA, oligopeptides, oligosaccharides, and combinatorial libraries for drug discovery. State-of-the-art solid-phase synthesizers can produce oligonucleotides up to 200-300 nucleotides while using excess reagents. Accumulated errors over multiple reaction cycles prevent the synthesis of longer oligonucleotides for the genome scale engineering of synthetic biological systems. The sources of these errors in synthesis columns remains poorly understood. Here we show that bead-bead stacking significantly contributes to reaction errors in columns by analyzing enzymatic coupling of fluorescently labelled nucleotides onto the initiated beads along with porosity, particle tracking and diffusion calculations. To circumvent stacking, we introduce dielectrophoretic bead-droplet reactor (DBDR); a novel approach to synthesize on individual microbeads within microdroplets. Dielectrophoretic force overcomes the droplet-medium interfacial tension to encapsulate and eject individual beads from microdroplets in a droplet microfluidic device. Faster reagent diffusion in droplets, and non-uniform electric field induced enhancement in reagent concentration at its surface can improve reaction fidelities in DBDR. Fluorescence comparisons suggest around 3-fold enhancement of reaction fidelity compared to columns. DBDR can potentially enable the high-purity synthesis of arbitrarily long strands of DNA to meet the emerging demands in healthcare, environment, agriculture, materials, and computing.

A microfluidic platform for the synthesis of polymer and polymer-protein-based protocells

Abstract:In this study, we demonstrate the fabrication of polymersomes, protein-blended polymersomes, and polymeric microcapsules using droplet microfluidics. Polymersomes with uniform, single bilayers and controlled diameters are assembled from water-in-oil-in-water double-emulsion droplets. This technique relies on adjusting the interfacial energies of the droplet to completely separate the polymer-stabilized inner core from the oil shell. Protein-blended polymersomes are prepared by dissolving protein in the inner and outer phases of polymer-stabilized droplets. Cell-sized polymeric microcapsules are assembled by size reduction in the inner core through osmosis followed by evaporation of the middle phase. All methods are developed and validated using the same glass-capillary microfluidic apparatus. This integrative approach not only demonstrates the versatility of our setup, but also holds significant promise for standardizing and customizing the production of polymer-based artificial cells.

Abstract:This study develops a new solvent-compatible microfluidic chip based on phenol formaldehyde resin (PFR). In addition to its solvent-resistant characteristics, this microfluidic platform also features easy fabrication, organization, decomposition for cleaning, and reusability compared with conventional chips. Both solvent-dependent (e.g., polycaprolactone) and nonsolvent-dependent (e.g., chitosan) microparticles were successfully prepared. The size of emulsion droplets could be easily adjusted by tuning the flow rates of the dispersed/continuous phases. After evaporation, polycaprolactone microparticles ranging from 29.3 to 62.7 μm and chitosan microparticles ranging from 215.5 to 566.3 μm were obtained with a 10% relative standard deviation in size. The proposed PFR microfluidic platform has the advantages of active control of the particle size with a narrow size distribution as well as a simple and low cost process with a high throughput.
