Abstract:Hochsubstituierte α,β-ungesättigte Cyclohexenone, ein zentrales Strukturmotiv in vielen Naturstoffen, sind mit der Titelreaktion zugänglich (siehe Schema). Die Diastereoselektivität bezüglich der beiden stereogenen Zentren (4- und 6-Position) ist hoch. Mit optisch aktiven Ausgangsmaterialien wurden die erwünschten Produkte ohne nachweisbare Racemisierung gebildet. Eine mögliche Erklärung der Stereoselektivität wird präsentiert.

Abstract:The addition reaction of sulfonyl chlorides with acetylenes in the presence of a copper salt is described. The reaction proceeds in toluene in the presence of CuCl with Me2S as the additive to afford only (Z)-beta-chlorovinyl sulfones in moderate to excellent yields.

Abstract:Oxazolones are structural subunits in numerous natural products and designed molecules with substantial pharmacological properties. Here, a palladium-catalyzed chemodivergent regio-and enantioselective allylic alkylation of 4-or 5 -substituted oxazol-2(3H)-ones with Morita-Baylis-Hillman (MBH) adducts has been developed using a spiroketal-based diphosphine as the ligand (50 examples). Interestingly, 4-substituted oxazol-2(3H)-ones acted as a C-nucleophiles in the reaction to afford a range of chiral 4,5-substituted oxazol-2(3H)-ones in high yields (72-99%) with good to excellent chemo-, regio-, and enantioselectivities (C/N 95:5->99:1, b/l 91:9->99:1, 85-98% ee). When a N-nucleophile was used under otherwise identical conditions, 5 -substituted oxazol-2(3H)-ones delivered a range of chiral 3,5-substituted oxazol-2(3H)-ones in high yields (68- 98%) with good regio-and enantioselectivities (b/l 71:29-91:9, 66-94% ee). The synthesis can be readily performed on gram scale under fairly low catalyst loadings, and the utility of the protocol was showcased in the facile transformation of the products into more elaborate chiral molecules.

Abstract:Novel C-2-symmetrical bissulfonamide ligands were easily prepared in three simple steps and applied in the enantioselective addition of alkynylzinc reagents to aldehydes. Compound 7a was found to be an outstanding ligand for this reaction. When the catalyst loading, 4 mol % of 7a, was used, high enantioselectivity with an ee value up to 97% was achieved under very mild conditions. When the amount of ligand 7a was lowered to 2 mol %, excellent levels of enantiocontrol up to 95% ee were still achieved. So far the most economic catalyst system was developed for the addition of terminal alkynes to aromatic aldehydes in terms of ligand loading and enantioselectivity. Moreover, ligand 7a in combination with Ti(O-i-Pr)(4) was found to be effective in promoting the addition reaction of an alkynylzinc reagent to unactivated simple ketones under very mild conditions. The corresponding chiral tertiary propargylic alcohols were obtained with enantiomeric excesses of up to 81%.

Abstract:We describe a new method for the stereoselective synthesis of (Z)-1,2-dithio-1-alkenes via copper-catalyzed cross-coupling of (Z)-2-bromovinyl sulfides and thiols. The desired products are obtained in good to excellent yields. The double bond geometry of the starting (Z)-2-bromovinyl sulfides is retained under these reaction conditions.

Abstract:Catalysed by the nitrile hydratase/amidase-containing Rhodococcus sp. AJ270 cells, a number of beta-aryl- and beta-alkyl-beta-hydroxy-alpha-methylenepropiononitriles (the Baylis-Hillman nitriles) 1 underwent hydrolysis under mild conditions to produce the corresponding enantiomerically enriched Baylis-Hillman amides 2 and acids 3. The enantioselectivity of the biotransformations was strongly determined by the steric effect of the substituents at the beta-position of the substrates. The protection of the free hydroxy of beta-phenyl-beta-hydroxy-alpha-methylenepropiononitrile 1a by methylation led to the enhancement of enantiocontrol of the biohydrolysis.

Abstract:A Ag-CH3SO3H co-catalyzed direct Z-selective synthesis of 1,2-cis-dibromoalkenes from alkynes has been developed. Mechanistic studies showed that the reaction underwent the formation of a silver allene cation intermediate followed by sequential bromine free radical addition and substitution. This discovery might open up a new arena for silver catalysis and important allene chemistry. Additionally, an efficient PPh3-promoted E-selective synthesis of 1,2-trans-dibromoalkenes from alkynes is also disclosed. Both protocols utilized easy-to-handle N-bromosuccinimide as a sole brominating reagent and reacted under much mild reaction conditions.

Abstract:Cyclization–oxidation of Baylis–Hillman adducts provides a convenient method to stereoselectively synthesize variety of multi-substituted bicyclo[3.1.0] ring systems via Pd(II)/Pd(IV)-catalyzed reactions. We also disclose that C–Pd(IV) intermediate can undergo reductive elimination through SN2-type attack by the latent nucleophile of vinyl acetate to afford Csp3–Csp3 bond formation with inversion of configuration at the Pd(IV)-bounded carbon.
