Abstract:The reaction of o-aminoacetophenones and arylboronic acids catalyzed by copper(II) salts in the presence of pyridine under an O2 atmosphere provides a general and efficient one-pot preparation of biologically interesting acridones. This relay reaction comprises an intermolecular Suzuki cross-coupling, intramolecular oxidative C(sp3)-H amination, and C(sp2)-H activation with simultaneous rearrangement of the generated isatin intermediates. This strategy tolerates both electron-donating and -withdrawing functionalities to afford various acridones in good to excellent yields.

Abstract:A Lewis-acid-promoted approach to the synthesis of highly functionalized dihydrobenzofuran derivatives was developed. A diverse range of functional groups are tolerated in this type of reaction. The reaction mechanism investigation indicates that the highly reactive phenyl cation intermediate is probably involved in this process. The chirality of substrate is retained under the reaction conditions.

Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:A facile preparation of 3-aminofuran derivatives via multicomponent reactions of thiazole carbenes, aldehydes, and dimethyl acetylenedicarboxylate (DMAD) is reported. In this process, the thiazole carbenes, generated in situ from thiazolium salts, reacted with aldehydes and DMAD at -78 to 0 degrees C in CH2Cl2 to afford the substituted furans in moderate to good yields. Eight substituted thiazolium salts were employed as carbene precursors in the reaction. Besides aryl aldehydes, alpha beta-unsaturated aldehydes, aliphatic aldehydes, and arenedial were also investigated and found to be applicable to this reaction.

Abstract:Asymmetric Wittig-type olefination of 4-substituted cyclohexanones with chiral ligand-modified stable arsonium ylides has been examined. The 8-phenylmenthol-derived chiral arsonium ylide of 4 reacted with prochiral ketones 9a-d at -15 degrees C to give the 4-substituted cyclohexylideneacetates 11a-d in 58-69% yield and in up to 80% diastereomeric excess (de). Abstract:An efficient and practical method for the synthesis of medicinally important acridines from readily available o-aminoaryl ketones and arylboronic acids was developed using copper(II)-mediated relay reactions that involve intermolecular Chan-Lam cross-coupling and subsequent intramolecular Friedel-Crafts-type reactions. A sole promoter, i.e., Cu(OTf)2, was used; therefore, strongly acidic and basic conditions, nonreadily available or expensive substrates, additives, and noble-metal catalysts were not needed.

Abstract:Amine compounds are widely found in natural products, pharmaceuticals, and pesticides. We propose a base‐facilitated synthesis method of polysubstituted aromatic amines. A simple, rapid, plateau‐utilization, and highly regioselective non‐in‐situ amination reaction was achieved by using aryl thianthrenium salt as the aryne precursor, and the arylynyl distortion induced by the substituents. This method provides new possibilities for organic synthesis.

Abstract:A novel and simple synthesis of 3-aroyl-2,5-diaryl-2,4-cyclopentadiene-1,1-dicarbonitriles is described. The reactive 1:1 zwitterionic intermediate, formed by the addition of triphenylphosphine to diaroylacetylenes, was trapped by arylidenemalononitriles to produce the title compounds under mild reaction conditions in fairly good yields.

Abstract:Arynes undergo a wide range of diverse chemical transformations making them highly versatile reactive in-termediates for organic synthesis. Access to arynes has long been dominated by pre-functionalized reagents such as the venerable o-trimethylsilylaryl triflate class of substrates. However, a move toward the development of methods to access arynes that are both mild and efficient has prompted research into aryl “onium” aryne precursors. We have leveraged aryl “onium” species as intermediates in an overall net dehydrogenation of simple arenes as an efficient way to generate and trap arynes. Herein, we describe two strategies to achieve mild and efficient access to arynes. 1) The development of a one-pot method that converts simple arenes into aryl thianthrenium salts and uses them in situ to generate arynes; the net transformation of arene to trapped aryne is achieved within two hours. 2) The development of a two-step process to convert arenes into aryl(Mes)iodonium salts and ultimately trapped arynes to expand the scope of arenes compatible; the net transformation is complete within four hours. Mechanistic analysis through competition experiments, deuterium kinetic isotope effects (DKIE) and Density Functional Theory (DFT) provide key insight into the similarity and differences of the two approaches described in this work and yield a user’s guide for selecting the appropriate strategy based on the arene.

Abstract:Catalysts for direct arylations have been extensively studied for the efficient synthesis of (hetero)biaryl compounds. Phosphine ligands are pivotal in this field, and electron-deficient phosphine ligands are advantageous for direct arylation. Due to its relatively weak σ-donation ability, arsenic is considered an ideal candidate for designing electron-deficient ligands. However, no studies have explored the relationship between the structure of the arsine ligands and their catalytic activity for direct arylation. In this study, we synthesized 27 arsine ligands with varying electron-donating and steric properties. Tris(o-trifluorotolyl)arsine, an electron-deficient and bulky ligand, exhibited the highest efficiency in a model reaction between 2-propylthiophene and 4-bromobenzotrifluoride. Both experimental and computational studies revealed the mechanism of direct arylation using arsine ligands. This is the first comprehensive investigation of an arsine-ligated catalytic direct arylation.

Abstract:Sulfonated tetrahydropyridine derivatives are accessed through a radical reaction of 1,6-enynes, sulfur dioxide, and aryldiazonium tetrafluoroborates under mild conditions. Sulfonated pyrrolidine can be generated when terminal alkyne is used as the substrate. This reaction proceeds efficiently in dichloroethane without the addition of any catalysts or additives, providing sulfonated tetrahydropyridine derivatives in moderate to good yields. During this transformation, aryldiazonium tetrafluoroborates cooperate with DABCO·(SO2)2 leading to sulfonyl radicals, which initiate the radical cyclization process. Two molecules of aryldiazonium tetrafluoroborates are involved in the reaction.
