Abstract:The emission spectra and photodimerization of fluorinated alkyl 2-naphthoates (NpFCn) and 9-anthrylmethyl perfluoroalkanoates (AnFC(n)) in aqueous-organic binary solvents have been investigated. The fluorescence spectra of NpFCn in dimethyl sulfoxide-water mixture (DMSO-H2O) are dominated by excimer emission. Addition of long chain fluorocarbon compounds to the solution results in a reduction in the excimer emission and an enhancement of monomer emission. Selective excitation of the naphthalene moiety in the mixture solution of NpFCn and AnFC(n) leads to strong emission from AnFC(n). Photoirradiation of NpFCn yields a 'cubane-like' photodimer with a relatively high quantum yield. All these observations suggest that hydrophobic interactions force molecules with long fluorocarbon chains to form aggregates in aqueous-organic binary solvents.

Abstract:In aqueous organic solvents perfluoroalkyl 2-naphthoates (NpFCn) with long fluorocarbon chain show strong excimer fluorescence. Excitation of NpFCn in the mixed solution of NpFCn and 9-anthrylmethyl perfluoroalkanoates (AnFCn) results in strong emission of AnFCn. Abstract:Classical molecular dynamics (MD) simulations were performed to study the association of KF and NaF ion pairs in water in a wide range of temperatures (423–1273 K) and densities (0.10–0.60 g/cm3). The association constants (KA) derived from the potentials of mean force (PMF) exhibit a rising tendency as temperature increases or density decreases. The association of NaF is similar to KF under most conditions but evidently larger at higher temperatures and lower densities. The radial distribution function (RDF) and coordination number (CN) reveal the characteristics of ion-water microstructures. There is significant water clustering near the K+/Na+ at high temperatures or low densities. The results indicate that fluoride ion acts as an important ligand complexing with K+/Na+ and promotes their migration and concentration in hydrothermal fluids.

Abstract:Solutions of alkali metal fluorides in aliphatic diols or carboxylic acids have been investigated by 19F n.m.r. at concentrations of ca. 0.5 mol kg–1 and greater. Heats of association of potassium fluoride with ethane-1,2-diol and methanoic acid are obtained from variable-temperature n.m.r. studies. The nature of the strong hydrogen bond and its influence on the n.m.r. spectra are discussed.

Abstract:Fluorine-containing medicinal compounds frequently allow modulation of physical-chemical properties. Here, we address the effect of fluorine, near the ethylamino side chain, on conformational flexibility and noncovalent interactions (NCIs) of the selected jet-cooled monohydrated cluster of 2-(2-fluoro-phenyl)-ethylamine (2-FPEA) by mass-selected resonance-enhanced two-photon ionization and ionization-loss stimulated Raman spectroscopies. Our results show that Raman spectral signatures of the 2-FPEA-H2O cluster match the scaled harmonic vibrational Raman frequencies, resulting from density functional theory calculations of the most stable 2-FPEA gauche conformer hydrogen-bonded (HB) to water, confirming the three-dimensional cluster structure. This predicted electronic structure, together with NCI analysis, allows visualization and assessment of the attractive and repulsive interactions. The comparison of the NCIs and revealed red (O-H and N-H stretches) and blue shifts (C-H stretches and CH2 out-of-plane bends) of the cluster to other class members confirm O-H···N, N-H···π, C-H···O, and C-H···F HB formation and their contribution to structure stabilization, uncovering the potential of the approach.
Micromolar fluoride contamination arising from glass NMR tubes and a simple solution for biomolecular applications

Abstract:Fluorine ( F) NMR is emerging as an invaluable analytical technique in chemistry, biochemistry, material science, and medicine, especially due to the inherent rarity of naturally occurring fluorine in biological, organic, and inorganic compounds. Thus, we were surprised to identify an unexpected peak in our F NMR spectra, corresponding to free fluoride, which appears to leach out from various types of new and unused glass NMR tubes over the course of several hours. We quantified this contaminant to be at micromolar concentrations for typical NMR sample volumes across multiple glass types and brands. We find that this artefact is undetectable for samples prepared in quartz NMR tubes within the timeframes of our experiments. We also observed that pre-soaking new glass NMR tubes combined with rinsing removes this contamination below micromolar levels. Given the increasing popularity of F NMR across a wide range of fields, the long collection times required for relaxation studies and samples of low concentrations, and the importance of avoiding contamination in all NMR experiments, we anticipate that our simple solution will be useful to biomolecular NMR spectroscopists.

Abstract:The fluorine-19 nucleus was recognized early to harbor exceptional properties for NMR spectroscopy. With 100% natural abundance, a high gyromagnetic ratio (83% sensitivity compared to 1H), a chemical shift that is extremely sensitive to its surroundings and near total absence in biological systems, it was destined to become a favored NMR probe, decorating small and large molecules. However, after early excitement, where uptake of fluorinated aromatic amino acids was explored in a series of animal studies, 19F-NMR lost popularity, especially in large molecular weight systems, due to chemical shift anisotropy (CSA) induced line broadening at high magnetic fields. Recently, two orthogonal approaches, (i) CF3 labeling and (ii) aromatic 19F-13C labeling leveraging the TROSY (Transverse Relaxation Optimized Spectroscopy) effect have been successfully applied to study large biomolecular systems. In this perspective, we will discuss the fascinating early work with fluorinated aromatic amino acids, which reveals the enormous potential of these non-natural amino acids in biological NMR and the potential of 19F-NMR to characterize protein and nucleic acid structure, function and dynamics in the light of recent developments. Finally, we explore how fluorine NMR might be exploited to implement small molecule or fragment screens that resemble physiological conditions and discuss the opportunity to follow the fate of small molecules in living cells.

Abstract:13C and 15N chemical shifts of the intact farnesyl (31R)-bacteriochlorophyll (BChl) c have been measured in methanol and carbon tetrachloride solutions. Two sets of resonances have been observed in carbon tetrachloride for all carbon and nitrogen atoms, indicating a formation of highly stable dimeric species with asymmetric configurations. Complete assignments have been made based on a combination of homonuclear and heteronuclear correlation experiments using the 13C- and 15N-labeled BChl c samples. Changes of the 13C chemical shift in the two solvents can be interpreted in terms of mixed effects arising from (a) ring current due to the overlap of the macrocycles, (b) coordination state of the central magnesium, (c) excitation state of the π-electron system, (d) polarity of the solvents used, and (e) hydrogen bonding. Substantial ring current effect is observed on the 13C chemical shifts for the carbon atoms around pyrrolic ring I upon the dimer formation. Remarkable differences in the line widths observed for all propionic carbons and some carbons of the farnesyl group suggest that the propionic-farnesyl side chains may adopt a “return” structure over the region from 171 to f2 carbons with much different conformation and mobility in the dimer. No clear evidence is obtained for a hydrogen bond formed with the C131 carbonyl group in CCl4 solution, nor for ring overlap over the ring V. Comparison between the 15N chemical shifts in both solvents indicates that the paramagnetic shielding effect is predominant and NIV nitrogen is most sensitive to the dimer formation, followed by NII, NI, and NIII, respectively. The result reveals a high sensitivity of 15N chemical shift to the electronic state and N−Mg bond length for each nitrogen atom in the dimer.

Abstract:C-13 and H-1 MAS NMR spectra of solid 1,8-bis(dimethylamino)naphthalene (DMAN) and several of its complexes have been measured. The presence of four resonances from quaternary carbons, six from protonated carbons, and four from methyl carbons demonstrates that the DMAN molecule is asymmetric. This is in agreement with the actual results from single-crystal X-ray diffraction but contrary to the accepted interpretation. In reality, the molecule is asymmetric as a result of interactions in the crystal. The estimated energies of intramolecular interactions permit the assignment of resonances from all 14 carbon atoms in the molecule. We discuss the role of protonation of DMAN in the redistribution of electron density.
Strong Hydrogen-Bonding in Aliphatic Diol Solutions of Alkali-Metal Fluorides

Abstract:Heavy alkali-metal fluorides are soluble in low-molecular-weight liquid aliphatic diols. Low-field shifts in the proton magnetic resonances of selected fluoride–diol solutions may be interpreted as being due largely to strong hydrogen bonding between the fluoride anion and the diol hydroxyls, although cation effects are also evident. Variable-temperature 1H n.m.r. studies on KF–1,2-ethanediol, CH3CN–1,2-ethanediol, and neat 1,2-ethanediol have been carried out and values of –(50 ± 10) kJ mol–1 for ΔH⊖ and –(130 ± 25) J K–1 mol–1 for ΔS⊖ have been determined for the (F ⋯ HO)– hydrogen bond.
Ion spatial distributions at the liquid-vapor interface of aqueous potassium fluoride solutions

Abstract:X-Ray photoemission spectroscopy operating under ambient pressure conditions is used to probe ion distributions throughout the interfacial region of a free-flowing aqueous liquid micro-jet of 6 M potassium fluoride. Varying the energy of the ejected photoelectrons by carrying out experiments as a function of X-ray wavelength measures the composition of the aqueous-vapor interfacial region at various depths. The F(-) to K(+) atomic ratio is equal to unity throughout the interfacial region to a depth of 2 nm. The experimental ion profiles are compared with the results of a classical molecular dynamics simulation of a 6 M aqueous KF solution employing polarizable potentials. The experimental results are in qualitative agreement with the simulations when integrated over an exponentially decaying probe depth characteristic of an APPES experiment. First principles molecular dynamics simulations have been used to calculate the potential of mean force for moving a fluoride anion across the air-water interface. The results show that the fluoride anion is repelled from the interface, consistent with the depletion of F(-) at the interface revealed by the APPES experiment and polarizable force field-based molecular dynamics simulation. Together, the APPES and MD simulation data provide a detailed description of the aqueous-vapor interface of alkali fluoride systems. This work offers the first direct observation of the ion distribution at an aqueous potassium fluoride solution interface. The current experimental results are compared to those previously obtained for saturated solutions of KBr and KI to underscore the strong difference in surface propensity between soft/large and hard/small halide ions in aqueous solution.
19F: a versatile reporter for non-invasive physiology and pharmacology using magnetic resonance

Abstract:The fluorine atom provides an exciting tool for diverse spectroscopic and imaging applications using Magnetic Resonance. The organic chemistry of fluorine is widely established and it can provide a stable moiety for interrogating many aspects of physiology and pharmacology in vivo. Strong NMR signal, minimal background signal and exquisite sensitivity to changes in the microenvironment have been exploited to design and apply diverse reporter molecules. Classes of agents are presented to investigate gene activity, pH, metal ion concentrations (e.g., Ca(2+), Mg(2+), Na(+)), oxygen tension, hypoxia, vascular flow and vascular volume. In addition to interrogating speciality reporter molecules, (19)F NMR may be used to trace the fate of fluorinated drugs, such as chemotherapeutics (e.g., 5-fluorouracil, gemcitabine), anesthetics (e.g., isoflurane, methoxyflurane) and neuroleptics. NMR can provide useful information through multiple parameters, including chemical shift, scalar coupling, chemical exchange and relaxation processes (R1 and R2). Indeed, the large chemical shift range (approximately 300 ppm) can allow multiple agents to be examined, simultaneously, using NMR spectroscopy or chemical shift selective imaging.
XAFS Studies of Solution-Phase Complexes of Cesium with Dibenzo-18-crown-6 Ethers

Abstract:X-ray absorption fine structure (XAFS) measurements have been made at the Cs LIII absorption edge on (1) 0.04 M acetonitrile solutions with respect to both CsBr and dibenzo-18-crown-6 ether (Cs−D18C6), (2) solids produced by drying the aforementioned Cs−D18C6 solution complex, and (3) crystalline CsBr, CsCl, and CsF powders that were used as standards. XAFS measurements have also been made at the Br K absorption edge on the same 0.04 M solution mentioned above. Due to the many difficulties associated with obtaining high-quality XAFS data on these systems, a custom-manufactured 5-mil-thick Sc foil was used in conjunction with a Soller slit assembly to improve the XAFS signal-to-noise ratio by almost a factor of 6. XAFS analyses of the Cs−D18C6 solution show the presence of a 1:1 Cs−D18C6 complex with a Br contact ion, consistent with previous references derived from 133Cs nuclear magnetic resonance investigations. For interpretation of the Cs LIII edge data, the choice of a Br counterion reduces the error in determining the presence of a single counterion among the lighter backscattering O and C atoms of the crown ether complex. Observation of the normalized XAFS data for the dried Cs−crown ether solution shows that the Cs atom's local environment changes from a 1:1 Cs−D18C6 complex to a predominantly crystalline CsBr solid phase. This recrystallization during drying illustrates the importance of in situ methods to characterize chemical speciation in solution. The ability of XAFS to directly probe the ternary Cs−D18C6−Br contact ion pair opens many exciting avenues for improving extraction methodologies.

Abstract:We measured the rotationally resolved infrared spectra of helium solvated methyl fluoride at 3 μm and 10 μm, wherein lies C‐H and C‐F stretching bands, respectively. The linewidths (FWHM) were found to increase with increasing vibrational energy and range from 0.002 cm‐1 in the v3 band (C‐F stretch) at ~1047 cm‐1, to 0.65 cm‐1 in the v4 band (asymmetric C‐H stretch) at ~2997 cm‐1. In between these two bands we observed the lower and upper components of the Fermi triad bands (ν1/2ν2/2ν5) at ~2859 and ~2961 cm‐1. We carried out Stark spectroscopy on the lower band on account of its narrower linewidths (0.04 vs. 0.14 cm‐1, respectively). The objective of performing Stark spectroscopy was to see if there is any evidence for a rotational linewidth dependence on the external field strength, due to a reduced difference in between methyl fluorides rotational energy gap and the roton‐gap of superfluid helium. We did not find any evidence for such an effect, which we largely attribute to the rotational energy gap not increasing significantly enough by the external field. We point to another molecule (formaldehyde) whose energy levels are predicted to show a more promising response to application of an external field.

Abstract:Low-temperature 13C and 1H MAS NMR and 14N NQR spectra of solid 1,8-bis(dimethylamino)naphthalene (DMAN) have been measured. X-Ray diffraction shows that the molecule is more symmetric than at room temperature. The low-temperature geometry of DMAN is very similar to the ab initio optimized geometry. Solid-state NMR and nuclear quadrupole resonance (NQR) confirm the symmetry of the molecule at low temperature despite the asymmetry of intermolecular interactions in the crystal. Experimental values of the principal components of electric field gradients at 14N, which characterize the properties of the lone pair electrons of the nitrogen atoms, are in good agreement with the ab initio calculated values.

Abstract:A new chemical/spectroscopic couple that differentiates aromatic from aliphatic carboxylic acid polymers was developed. The method is complementary to more traditional methods of identification (IR) and is applicable to the analysis of complex mixtures where IR determinations are complicated by extensive vibrational band overlap. The method entails (1) conversion of carboxylic acids into acid fluorides and (2) specific detection of the resonances of the acid fluoride carbon and of the carbon directly attached to the carbonyl carbon by solid state (19)F-(13)C cross polarization (CP)/MAS (13)C NMR. The assignment of the chemical shift of the latter resonance to either the sp(2) or sp(3) carbon resonance manifold specifies the nature of the acid functionality. The preparation of the acid fluoride derivatives of several polymers containing aliphatic and aromatic carboxylic acid functionality was evaluated using sulfur tetrafluoride (SF(4)), diethylaminosulfur trifluoride (DAST), cyanuric fluoride, and thionyl fluoride. Room temperature reactions using DAST in methylene chloride or neat SF(4) gave the acid fluorides in yields ≥90% for the acids studied. Aromatic acid fluoride yields were essentially quantitative.
