Abstract:Structural characterization of metalorganic frameworks (MOFs) is crucial, since an understanding of the relationship between the macroscopic properties of these industrially relevant materials and their molecular-level structures allows for the development of new applications and improvements in current performance. In many MOFs, the incorporated metal centers dictate the short- and long-range structure and porosity of the material. Here we demonstrate that solid-state NMR (SSNMR) spectroscopy targeting NMR-active metal centers at natural abundance, in concert with ab initio density functional theory (DFT) calculations and X-ray diffraction (XRD), is a powerful tool for elucidating the molecular-level structure of MOFs. Zr-91 SSNMR experiments on MIL-140A are paired with DFT calculations and geometry optimizations in order to detect inaccuracies in the reported powder XRD crystal structure. In-115 and La-139 SSNMR experiments on sets of related MOFs at two different magnetic fields illustrate the sensitivity of the In-115/La-139 electric field gradient tensors to subtle differences in coordination, bond length distribution, and ligand geometry about the metal center. Ti-47/49 SSNMR experiments reflect the presence or absence of guest solvent in MIL-125(Ti), and when combined with DFT calculations, these SSNMR experiments permit the study of local hydroxyl group configurations within the MOF channels. Zn-67 SSNMR experiments and DFT calculations are also used to explore the geometry near Zn within a set of four MOFs as well as local disordering caused by distributions of different linkers around the metal. SSNMR spectroscopy of metal centers offers an impressive addition to the arsenal of techniques for MOF characterization and is particularly useful in cases where XRD information may be ambiguous, incomplete, or unavailable.
19F: a versatile reporter for non-invasive physiology and pharmacology using magnetic resonance

Abstract:The fluorine atom provides an exciting tool for diverse spectroscopic and imaging applications using Magnetic Resonance. The organic chemistry of fluorine is widely established and it can provide a stable moiety for interrogating many aspects of physiology and pharmacology in vivo. Strong NMR signal, minimal background signal and exquisite sensitivity to changes in the microenvironment have been exploited to design and apply diverse reporter molecules. Classes of agents are presented to investigate gene activity, pH, metal ion concentrations (e.g., Ca(2+), Mg(2+), Na(+)), oxygen tension, hypoxia, vascular flow and vascular volume. In addition to interrogating speciality reporter molecules, (19)F NMR may be used to trace the fate of fluorinated drugs, such as chemotherapeutics (e.g., 5-fluorouracil, gemcitabine), anesthetics (e.g., isoflurane, methoxyflurane) and neuroleptics. NMR can provide useful information through multiple parameters, including chemical shift, scalar coupling, chemical exchange and relaxation processes (R1 and R2). Indeed, the large chemical shift range (approximately 300 ppm) can allow multiple agents to be examined, simultaneously, using NMR spectroscopy or chemical shift selective imaging.

Abstract:The fluorine-19 nucleus was recognized early to harbor exceptional properties for NMR spectroscopy. With 100% natural abundance, a high gyromagnetic ratio (83% sensitivity compared to 1H), a chemical shift that is extremely sensitive to its surroundings and near total absence in biological systems, it was destined to become a favored NMR probe, decorating small and large molecules. However, after early excitement, where uptake of fluorinated aromatic amino acids was explored in a series of animal studies, 19F-NMR lost popularity, especially in large molecular weight systems, due to chemical shift anisotropy (CSA) induced line broadening at high magnetic fields. Recently, two orthogonal approaches, (i) CF3 labeling and (ii) aromatic 19F-13C labeling leveraging the TROSY (Transverse Relaxation Optimized Spectroscopy) effect have been successfully applied to study large biomolecular systems. In this perspective, we will discuss the fascinating early work with fluorinated aromatic amino acids, which reveals the enormous potential of these non-natural amino acids in biological NMR and the potential of 19F-NMR to characterize protein and nucleic acid structure, function and dynamics in the light of recent developments. Finally, we explore how fluorine NMR might be exploited to implement small molecule or fragment screens that resemble physiological conditions and discuss the opportunity to follow the fate of small molecules in living cells.

Abstract:Pulsed Fourier transform nuclear magnetic resonance (FT-NMR) has reigned supreme in high-resolution, high-field spectroscopy─particularly when targeting complex liquid-state samples involving multiple sharp peaks spread over large spectral bandwidths. It is known, however, that if spectral resolution is not a must, the FT-based approach is not necessarily the optimal route for maximizing NMR sensitivity: if T2 ≈ T1, as often found in solutions, Carr's steady-state free-precession (SSFP) approach can in principle provide a superior signal-to-noise ratio per √(acquisition_time) (SNRt). A rapid train of pulses will then lead to a transverse component that reaches up to 50% of the thermal equilibrium magnetization, provided that pulses are applied at repetition times TR ≪ T2, T1, and that a single suitable offset is involved. It is generally assumed that having to deal with multiple chemical shifts deprives SSFP from its advantages. The present study revisits this assumption by introducing an approach whereby arbitrarily short SSFP-derived free induction decays (FIDs) can deliver high-resolution spectra, without suffering from peak broadenings or phase distortions. To achieve discrimination among nearby frequencies, signals arising from a series of regularly phase-increased excitation pulses are collected. Given SSFP's amplitude and phase sensitivity to the spins' offset, this enables the resolution of sites according to their chemical shift position. In addition, the extreme fold-over associated with SSFP acquisitions is dealt with by a customized discrete FT of the interpulse time-domain signal. Solution-state 13C NMR spectra which compare well with FT-NMR data in terms of sensitivity, bandwidth, and resolution can then be obtained.

Abstract:A simple 1,1′-bi-2-naphthol (1,1′-BINOL) boron complex has been rationally designed and synthesized, the structure of which was confirmed by single crystal X-ray diffraction analysis. Furthermore, the complex exhibited turn-on fluorescence for fluoride ions with high selectivity and sensitivity. The recognition mechanism for promoting was determined by 1H, 19F and 11B NMR titrations, which indicated that both the photo-induced electron-transfer (PET) and the intramolecular charge transfer (ICT) effects were functionalized. In addition, the complex showed strong solid-state fluorescence, and the emission spectrum and quantum efficiency (ΦF) of the solid powders were also measured.

An NMR and relativistic DFT investigation of one-bond nuclear spin-spin coupling in solid triphenyl group-14 chlorides

Abstract:A solid-state nuclear magnetic resonance and zeroth-order regular approximation density functional theory, ZORA-DFT, study of one-bond nuclear spin-spin coupling between group-14 nuclei and quadrupolar 35/37Cl nuclei in triphenyl group-14 chlorides, Ph3XCl (X = C, Si, Ge, Sn and Pb), is presented. This represents the first combined experimental and theoretical systematic study of spin-spin coupling involving spin-pairs containing quadrupolar nuclei. Solid-state NMR spectra have been acquired for all compounds in which X has a spin-1/2 isotope--13C, 29Si, [117/119]Sn and 207Pb-at applied magnetic fields of 4.70, 7.05 and 11.75 T. From simulations of these spectra, values describing the indirect spin-spin coupling tensor-the isotropic indirect spin-spin coupling constant, 1J(X, 35/37Cl)iso and the anisotropy of the J tensor, Delta1J(X, 35/37Cl)--have been determined for all but the lead-chlorine spin-pair. To better compare the indirect spin-spin coupling parameters between spin-pairs, 1J(iso) and Delta1J values were converted to their reduced coupling constants, 1K(iso) and Delta1K. From experiment, the sign of 1K(iso) was found to be negative while the sign of Delta1K is positive for all spin-pairs investigated. The magnitude of both 1K(iso) and Delta1K was found to increase as one moves down group-14. Theoretical values of the magnitude and sign of 1K(iso) and Delta1K were obtained from ZORA-DFT calculations and are in agreement with the available experimental data. From the calculations, the Fermi-contact mechanism was determined to provide the largest contribution to 1K(iso) for all spin-pairs while spin-dipolar and paramagnetic spin-orbit mechanisms make significant contributions to the anisotropy of K. The inclusion of relativistic effects was found to influence K(Sn,Cl) and K(Pb,Cl).
Investigation of zeolitic imidazolate frameworks using 13C and 15N solid-state NMR spectroscopy

Abstract:Zeolitic imidazolate frameworks (ZIFs) are a subclass of metal-organic frameworks (MOFs) with extended three-dimensional networks of transition metal nodes (bridged by rigid imidazolate linkers), with potential applications in gas storage and separation, sensing and controlled delivery of drug molecules. Here, we investigate the use of 13C and 15N solid-state NMR spectroscopy to characterise the local structure and disorder in a variety of single- and dual-linker ZIFs. In most cases, a combination of a basic knowledge of chemical shifts typically observed in solution-state NMR spectroscopy and the use of dipolar dephasing NMR experiments to reveal information about quaternary carbon species are combined to enable spectral assignment. Accurate measurement of the anisotropic components of the chemical shift provided additional information to characterise the local environment and the possibility of trying to understand the relationships between NMR parameters and both local and long-range structure. First-principles calculations on some of the simpler, ordered ZIFs were possible, and provided support for the spectral assignments, while comparison of these model systems to more disordered ZIFs aided interpretation of the more complex spectra obtained. It is shown that 13C and 15N NMR are sufficiently sensitive to detect small changes in the local environment, e.g., functionalisation of the linker, crystallographic inequivalence and changes to the framework topology, while the relative proportion of each linker present can be obtained by comparing relative intensities of resonances corresponding to chemically-similar species in cross polarisation experiments with short contact times. Therefore, multinuclear NMR spectroscopy, and in particular the measurement of both isotropic and anisotropic parameters, offers a useful tool for the structural study of ordered and, in particular, disordered ZIFs.

Abstract:In this study, by applying 19F, 23Na and 7Li high-resolution NMR methods, the evolution of the [Zr x F y ]4x-y local ionic structures in FLiNaK-ZrF4 salt mixtures were elucidated. K3ZrF7, Na3ZrF7 and Na7Zr6F31 crystal phases were identified when the melt salts were being solidified. The distribution of these [Zr x F y ]4x-y species was dependent on the content of ZrF4 in FLiNaK eutectic salts. Moreover, K3ZrF7 phase transition from an orthorhombic lattice into a disordered cubic lattice was clarified, thereby causing dynamics of the coordinated F- ions to be reduced and the well-ordered crystal lattices to be destroyed. These mentioned results provide a further insight into the Zr-F based ionic structure and the formation of the disordered Zr-F structure in ZrF4-based eutectic salts.

Abstract:High‐resolution nuclear magnetic resonance (NMR) spectroscopy represents a key methodology for studying biomolecules and their interplay with other molecules. Recent developments in labeling strategies have made it possible to incorporate fluorine into proteins and peptides reliably, with manageable efforts and, importantly, in a highly site‐specific manner. Paired with its excellent NMR spectroscopic properties and absence in most biological systems, fluorine has enabled scientists to investigate a rather wide range of scientific objectives, including protein folding, protein dynamics and drug discovery. Furthermore, NMR spectroscopic experiments can be conducted in complex environments, such as cell lysate or directly inside living cells. This review presents selected studies demonstrating how 19F NMR spectroscopic approaches enable to contribute to the understanding of biomolecular processes. Thereby the focus has been set to labeling strategies available and specific NMR experiments performed to answer the underlying scientific objective.

Abstract:Quantifying low-level components in solid-state analysis presents a significant challenge for most thermal, diffractometric, vibrational, and spectroscopic techniques. In pharmaceutical analysis, identifying and quantifying the physical form of the drug substance in solid dosages is a critical task to ensure the quality of drug products. For example, recrystallization of active pharmaceutical ingredients in amorphous solid dispersions can compromise the stability and bioavailability of drug products. Herein, we have developed and demonstrated fluorine-19 solid-state nuclear magnetic resonance (19F ssNMR) methods and pushed the boundary to quantify minor crystalline contents in amorphous pharmaceuticals. Calibration curves suggest that 19F direct polarization and 1H-19F cross-polarization ssNMR can readily quantify 0.1% w/w crystalline compound I, a commercial fluorinated drug molecule developed by Merck & Co., Inc., Rahway, NJ, U.S.A., in its amorphous formulation. 1H-19F multiple cross-polarization (MultiCP) has been implemented, for the first time, and compared with conventional cross-polarization methods. Most importantly, a relaxation-filtered 19F ssNMR method was utilized to unambiguously identify and quantify as low as 0.04% w/w crystalline components, that is, 6 μmol in a 100 mg tablet at 25% drug loading, by suppressing the signal from the amorphous counterpart. Such a low level of detection offers high confidence and sensitivity to quantify trace amounts of phase change in pharmaceutical amorphous materials in the solid state, which can facilitate formulation development as well as quality control.

Abstract:Solid-state nuclear magnetic resonance (NMR) spectroscopy is used to study heavily Yb3+ and Er3+ doped, fluorescent NaY1–x–yYbxEryF4 nanoparticles. An understanding of the 19F wide line NMR response suggests that the 0 ppm portion of the 19F NMR spectrum can be used as a probe of trivalent lanthanide content via spin–lattice relaxation time changes. A Yb3+ and Er3+ magnetic interaction is manifest as a cooperative contribution to the 19F spin–lattice relaxation rate in heavily co-doped nanoparticle samples. The results from this study will aid in the understanding of the mechanism of enhanced optical up-conversion among these well-known nanostructures.

Abstract:The structural and transport features of the behavior of the fluorine ion sublattice of Pb 0.78 Sr 0.19 K 0.03 F 1.97 solid solution have been studied by the ab initio molecular dynamics method. It is shown that the local diffusion of fluorine anions changes with the nature of dopant atom, which is consistent with the experimental transport characteristics.

Abstract:In this contribution, the structural and electronic effects of fluoride doping in both crystalline and amorphous indium oxides are investigated by both experimental and theoretical techniques. Pristine crystalline and amorphous fluoride-doped indium oxide (F:In–O) phases were prepared by solution-based combustion synthesis and sol–gel techniques, respectively. The chemical composition, environment, and solid-state microstructure of these materials were extensively studied with a wide array of state-of-the-art techniques such as UV–vis, X-ray photoelectron spectroscopy, grazing incidence X-ray diffraction, 19F and 115In solid-state NMR, high-resolution transmission electron microscopy (HR-TEM), and extended X-ray absorption fine structure (EXAFS) as well as by density functional theory (DFT) computation combined with MD simulations. Interestingly, the UV–vis data reveal that while the band gap increases upon F–-doping in the crystalline phase, it decreases in the amorphous phase. The 19F solid-state NMR data indicate that upon fluorination, the InO3F3 environment predominates in the crystalline oxide phase, whereas the InO4F2 environment is predominant in the amorphous oxide phase. The HR-TEM data indicate that fluoride doping inhibits crystallization in both crystalline and amorphous In–O phases, a result supported by the 115In solid-state NMR, EXAFS, and DFT-MD simulation data. Abstract:Natrium super ionic conductor (NASICON) compounds form a rich and highly chemically tunable family of crystalline materials that are of widespread interest because they include exemplars with high ionic conductivity, low thermal expansion, and redox tunability. This makes them suitable candidates for applications ranging from solid-state batteries to nuclear waste storage materials. The key to an understanding of these properties, including the origins of effective cation transport and low, anisotropic (and sometimes negative) thermal expansion, lies in the lattice dynamics associated with specific details of the crystal structure. Here we closely examine the prototypical NASICON compound, NaZr2(PO4)3, and obtain detailed insights into such behavior via variable-temperature neutron diffraction and 23Na and 31P solid-state NMR studies, coupled with comprehensive density functional theory-based calculations of NMR parameters. Temperature-dependent NMR studies yield some surprising trends in the chemical shifts and the quadrupolar coupling constants that are not captured by computation unless the underlying vibrational modes of the crystal are explicitly taken into account. Furthermore, the trajectories of the sodium, zirconium, and oxygen atoms in our dynamical simulations show good qualitative agreement with the anisotropic thermal parameters obtained at higher temperatures by neutron diffraction. The work presented here widens the utility of NMR crystallography to include thermal effects as a unique probe of interesting lattice dynamics in functional materials.

Abstract:1H, 19F and 15N NMR spectroscopy has been carried out on the soluble hydrogen bonded complex (CH3CH2)4N+F− (uracil). The studies suggest that deprotonation of the uracil does not occur and that in a medium dielectric constant solvent, hydrogen bonding occurs at the N(1)H site. Spectroscopic monitoring of the controlled thermal decomposition of the complex shows that complex breakdown occurs via the uracil anion, HF−2 and neutral uracil.

Abstract:A new chemical/spectroscopic couple that differentiates aromatic from aliphatic carboxylic acid polymers was developed. The method is complementary to more traditional methods of identification (IR) and is applicable to the analysis of complex mixtures where IR determinations are complicated by extensive vibrational band overlap. The method entails (1) conversion of carboxylic acids into acid fluorides and (2) specific detection of the resonances of the acid fluoride carbon and of the carbon directly attached to the carbonyl carbon by solid state (19)F-(13)C cross polarization (CP)/MAS (13)C NMR. The assignment of the chemical shift of the latter resonance to either the sp(2) or sp(3) carbon resonance manifold specifies the nature of the acid functionality. The preparation of the acid fluoride derivatives of several polymers containing aliphatic and aromatic carboxylic acid functionality was evaluated using sulfur tetrafluoride (SF(4)), diethylaminosulfur trifluoride (DAST), cyanuric fluoride, and thionyl fluoride. Room temperature reactions using DAST in methylene chloride or neat SF(4) gave the acid fluorides in yields ≥90% for the acids studied. Aromatic acid fluoride yields were essentially quantitative. Aliphatic acid fluorides were contaminated with the anhydride of the acid. The acid fluoride polymers were characterized by solid state (13)C and (19)F MAS/NMR.
