Abstract:A convenient approach to N-Boc amino ketones 3a similar to 3l has been developed, which features a SmI2 prompted one-pot radical addition-ring opening process of 2-piperidinone with alpha,beta-unsaturated esters. Moreover, the indolizidine skeleton (6) has been successfully synthesized from the key methyl (S)-8-((tert-butoxycarbonyl)amino)-5-((tert-butyldimethylsilyl)oxy)-4-oxooctanoate (3e), which undergoes one-pot deprotection-condensation-reduction-amination process.

Abstract:Reaction of a—haloketones with aldehydes in the presence of either SmI3 or Sm/I2 formed the carboncarbon double bond to give α,β—unsaturated ketones in good to excellent yields under mild conditions. Abstract:A novel and highly diastereoselective samarium-catalyzed tandem rearrangement/reduction of secondary alpha-hydroxy epoxides, which involves a C1 to C3 carbon migration rearrangement and a very interesting hetero-Tishchenko reduction of the intermediate aldehyde and the reductant aldehyde, has been reported. This reaction could be developed to provide a facile and stereoselective construction of 2-quarternary 1,3-diol units with an hydroxymethyl moiety attached to the diastereogenic quaternary carbon center. Detailed investigations have been carried out concerning the screening of the aldehydes as a reductant, the optimization of reaction conditions, and the substrate scope of this tandem reaction. A catalytic cycle for this reaction, the electronic and steric effects of the reductant aldehydes, and the mechanism for the acyl migration of 1,3-diol monoesters are proposed.

Abstract:This study analyzes the samarium diiodide‐promoted cyclizations of 5‐arylpentan‐2‐ones to dearomatized bicyclic products utilizing density functional theory. The reaction involves a single electron transfer to the carbonyl group, which occurs synchronously with the rate determining cyclization event, and a second subsequent proton‐coupled electron transfer. These redox reactions are accurately computed employing small core pseudo potentials explicitly involving all f‐electrons of samarium. Comparison of the energies of the possible final products rules out thermodynamic control of the observed regio‐ and diastereoselectivities. Kinetic control via appropriate transition states is correctly predicted, but to obtain reasonable energy levels the influence of the co‐solvent hexamethylphosphortriamide has to be estimated by using a correction term. The steric effect of the bulky samarium ligands is decisive for the observed stereoselectivity. Carbonyl groups in para‐position of the aryl group change the regioselectivity of the cyclization and lead to spiro compounds. The computations suggest again kinetic control of this deviating outcome. However, the standard mechanism has to be modified and the involvement of a complex activated by two SmI2 moieties is proposed in which two electrons are transferred simultaneously to form the new C–C bond. Computation of model intermediates show the feasibility of this alternative mechanism.

Abstract:A direct method to construct 3,4-dihydropyridin-2-ones by enzymatic condensation of aldehyde with cyanoacetamide and 1,3-dicarbonyl compounds was developed. One ring and four new bonds (two C–C, one C–N, one CC) were formed in one pot. And reaction conditions involving hydrolases, solvents, substrate molar ratios, and hydrolase loading were optimized. A series of new compounds based on the 3,4-dihydropyridin-2-one core were synthesized by the unprecedented three-component domino reaction.

Abstract:Phthalimide and N-phenylphthalimide smoothly condense with di-p-tolyl ether in triflic acid (CF(3)SO(3)H, TfOH) to obtain the corresponding spiro[isoindoline-1,9'-xanthen]-3-ones. Abstract:An efficient and practical method for highly diastereoselective synthesis of 2-quaternary anti-1,3-diol units with three adjacent stereogenic carbon centers has been developed. The reactions proceed regiospecifically at room temperature via samarium diiodide-promoted tandem Aldol condensation and Evans-Tishchenko reduction. The relative stereochemistry of the resulting 1,3-diol monoesters is identified by X-ray crystallography.

Abstract:A facile one-pot cascade synthesis involving simultaneous in situ pyrazole formation followed by iodine/DMSO-mediated oxidation has been established to afford 1-aryl-2-(3-aryl)-1H-pyrazol-5-yl-ethane-1,2-diones. Primarily, a two-pot approach has been established which includes the reaction of 3-methylthio-1,5-diaryl-2-pentene-1,5-diones with hydrazine in the first step to afford pyrazole, which was eventually oxidized in the next steps in the presence of iodine in DMSO. Further, we performed both steps in the same pot to afford the 1,2-dicarbonyl compounds in good yield. The structure of one of the compounds was confirmed by a single crystal X-ray. DMSO has posed as a solvent as well as an oxidant. Moreover, N-substituted hydrazines provided 1-(1-substituted-3-aryl-1H-pyrazol-5-yl)-2-arylethane-1,2-diones regioselectively. Further, as the synthetic application of this compound, 1-aryl-2-(3-aryl)-1H-pyrazol-5-yl-ethane-1,2-diones was treated with o-phenylenediamine to afford pyrazole functionalized quinoxaline in good yield. The control reaction was carried out to understand the mechanism of product formation.

Abstract:An efficient and practical electrophilic amination method that allows the preparation of useful quaternary carbon-containing α-aminoketones was developed. The reaction proceeds regiospecifically via a samarium enolate intermediate at room temperature in the presence of mild reducing agent SmI2. Unlike the most reported lithium enolate cases, this new amination method does not require the use of strong base such as BuLi or LDA.

Abstract:Abstract 3H-Benzo[1,2]-dithiole-3-thiones were prepared from potassium sulfide and 2-halobenzaldehydes in moderate-to-good yields, and a plausible mechanism for this catalyst-free intramolecular heteroannulation reaction has been proposed. The Knoevenagel condensation reactions of 3H-benzo[1,2]dithiole-3-thiones with active methylene compounds such as ethyl 2-cyanoacetate and diethyl malonate, and the three-component one-pot reactions of potassium sulfide, 2-halobenzaldehydes, and ethyl 2-cyanoacetate, affording the corresponding products, have been developed. Abstract:1,2-Dihydroxanthones (DHXs) are core structures of natural products and useful building blocks in organic synthesis. So far, they have been less studied. In this report, a mild, efficient and green method for the synthesis of 1,2-dihydroxanthones has been developed in one pot through Claisen condensation and O-cyclization under waste-induced relay catalysis with minimum organic solvents. The by-product (HMDS or NH3·H2O) of the first step turned out to be the promoter for the second step, which could efficiently proceed in aqueous media without the addition of other catalysts. The reactions using trifluoroethyl salicylates could be performed under mild conditions to ensure the generation of vulnerable DHXs in high yields. The substrate scope is very broad regardless of the substituent type and its position on the structure. Specifically, the versatility of DHXs was demonstrated by their conversion to xanthones and other complex structures.

Abstract:Anew Sm(III)-catalyzed tandem reaction of a-hydroxy epoxide has been discovered, where two catalytic reactions occur sequentially in one-pot and afford three contiguous stereocenters in a highly diastereoselective manner. It could be developed to a general procedure for diastereoselective construction of 2-quaternary-1,3-diol units. Extensive investigations of the reaction conditions and mechanism are also described.
