Abstract:Several new 6-(1-aryl-5-methyl-1, 2, 3-triazol-4-yl)-3-(4-pyridyl)-s-triazolo[3,4-b]-1, 3, 4-thiadiazoles 3a-j have been synthesized by the condensation of 4-amino-5-mercapto-3-(4-pyridyl)-1, 2, 4-triazole 2 with 1-aryl-4-carboxy-5-methyl-1, 2, 3-triazoles 1a-j in the presence of phosphorus oxychloride. The structures of the products obtained are characterized by elemental analyses and spectral data and the fragmentation pattern of the mass spectra of 3a-j is discussed. 3a-j have been screened for their antibacterial activity against B. subtilis and E. coli.

Abstract:A series of 1,3-bis[3-aryl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazole-6-yl]propanes (3a similar to 3o) were synthesized in good yields by condensation of pentanedioic acid (1) with 3-aryl-4-amino-5-mercapto-1,2,4-triazoles (2a similar to 2o) in the presence of phase transfer catalyst tetrabutylammonium iodide and phosphorus oxychloride. The structures of 3a similar to 3o were confirmed by IR, H-1 NMR, MS spectra and elementary analyses.

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:Condensation of 3-(2′-phenyl-1′,2′,3′-triazole-4′-yl)-4-amino-5- mercapto-1,2,4-triazole(1) with alkyl or aryl acids(2a-r) in the presence of phosphorus oxychloride gave 18 novel 3-(2′-phenyl-1′,2′,3′-triazole-4′-yl)-6-alkyl/ aryl-s-triazolo[3,4-b]-1,3,4-thiadiazoles(3a-r). Their structures were confirmed by elemental and spectral analyses.

Abstract:New fourteen compounds of bis(3-alkyl/aryl)-7H-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazines (3a similar to 3n) have been synthesized by the reaction of 1,4-dibromo-2,3-butanedione (1) with corresponding 3-alkyl/aryl-4-amino-5-mercapto-1,2,4-triazoles (2a similar to 2n). Their structures were characterized by EA, IR, H-1 NMR and MS spectra.

Abstract:A new class of bicyclic silaheterocycles, 6,6-dimethyl-2-aryl-6,7-dihydro- 5H-[1,2,4] triazolo[5,1-b][1,3,5] thiazasilines, which could be developed as potential sila-drugs. The reactions of several 3-aryl- 5-mercapto-1,2,4-triazoles with bis(chloromethyl) dimethylsilane under basic conditions gave high yields of bicyclic products. Reaction, product workup and isolation were streamlined without requiring an anhydrous solvent, inert gas or chromatographic purification. The products were structurally characterized by single-crystal X-ray diffraction.

Abstract:A novel series of pyrazolyl[1,2,4]triazoles has been synthesized by the reaction of polyfunctionalized-triazole 1 with symmetric and asymmetric β -dicarbonyl compounds in ortho -phosphoric acid. Also, we found that the reaction between triazole 1 and arylidene malononitriles in DMF afforded Schiff base compounds 9a,b instead of the target pyrazolyl[1,2,4]triazole 8a,b . The same products 9a,b were synthesized via the reaction of triazole 1 with the corresponding aromatic aldehydes. Moreover, a new series of 6-methyl-6-aryl-5,6-dihydro[1,2,4]triazolo[4,3- b ][1,2,4,5]tetrazine-3-thiol 11a,b and 9-amino-5-methyl-3,5-diphenyl-8-sulfanyl-2,4,5,9-tetrahydro-3 H -[1,2,4]triazolo[5,1- c ][1,2,4]triazepin-3-ol 12a has been successfully synthesized by the reaction of triazoles 1 with acetophenone derivatives.

Abstract:1-(3,3-Dimethyl-2,4-dioxolaneyl-methyl)-4-aryl-1,2,3-triazole(5) was synthesized by 1,3-dipolar cycloaddition of 3,3-dimethyl-2,4-dioxolaneyl-methyl azide and terminal alkyne under ultrasound irradiation for 20 min.The hydroxyl protection of 5 was removed in acidic solution to obtain 1-(2,3-dihydroxylpropyl)-4-aryl-1,2,3-triazole.The structures were characterized by 1H NMR,13C NMR,MS and elemental analysis.

Abstract:Many [1, 2, 4] triazolobenzoheteroazepine derivatives show interesting biological activities. In this paper, the geminal arylazo isocyanato compounds 3 were prepared from chroman-4-ones in three steps, i.e. condensation with arylhydrazine, cycloaddition of the resulting hydrazone with HNCO, and oxidative ring cleavage by KMnO4. Compounds 3 were allowed to react with HBF4, producing the respective 3-spiro substituted 1-aryl-4, 5-dihydro-5-oxo-3H-1, 2, 4-triazolium salts 4. The concurrent rearrangement of compound 4 with insertion of the nitrogen atom into the carbon skeleton provided, after the basic work-up, 2-oxo-[1, 2, 4] triazolo [3, 2-d] [1, 5] benzoxazepines 5a-5h. An unambigous structutal proof was obtained from the X-ray diffraction analysis for compound 5h.

Abstract:2-Trifluoromethyl and 2-difluoromethyl substituted benzimidazole, benzoxazole and benzothiazole derivatives were efficiently prepared through a one-pot reaction of trifluoroacetic acid and difluoroacetic acid, respectively, with commercially available o-phenylenediamines, 2-aminophenols, and 2-aminobenzenethiols in good to excellent yields. Subsequential bromination of 2-difluoromethyl groups by photolysis with NBS led to the formation of bromodifluoromethyl benzo-1,3-diazoles which may be utilized to prepare the new generation of gem-difluoromethylene linked identical or non-identical twin molecules for drug synthesis.

Abstract:A convenient synthetic pathway to 2-aryl-5,6-dihydro-4H-benzo[f][1,2,4]triazolo[1,5-a]azepine derivatives 7 was developed. The synthesis was based on the cycloaddition of the 1,2,3,4-tetrahydronaphthalene α-acetoxy azo compounds 3 with Ar-CN in the presence of AlCl3 and the consecutive ring enlargement.
