Abstract:Objective To explore the significance of blood level of silicon in the pathogenesis of silicosis. Methods Serum silicon was detected by inductively coupled plasma mass spectrometry (ICP-MS) and the expression of transforming growth factor-β1 (TGF-β1),tumor necrosis factor-α(TNF-α) in lung tissue of rats were observed by immunohistochemistry-SP.Results The serum level of silicon was significantly increased at the 16th hour of the first day after being exposed to silica and markedly higher than that of the control rats[(0.666±0.167) mg/L versus (0.055±0.040) mg/L,P0.01].It was also higher in the group on the subsequent 7,14,21 and 28 days compared with control. The expression of TGF-β1 in the lung tissue of model rats reached its peak value at 7th day and was significantly higher than that of the control group at 7,14,21 and 28 days (P0.05).The expression of TNF-α in the lung tissue of the model rats was significantly higher than that of the control group at 7,14,21 and 28 days (P0.05).Conclusion The blood level of silicon in silicosis rats was rapidly increased after exposed to silica and this change was prior to the expression of TGF-β1 and TNF-α,which indicated that this change might be meaningful for the study of silicosis pathogenesis and used as a biomarker for the early diagnosis of silicosis.
