Study on Levels of Silicon and CC16 in Serum of Rats Exposed to Silica Dust

Abstract:Objective To explore the change of serum silicon levels in rats with different exposure doses,and compare to the changes of serum Clara cell protein(CC16)which is an early biomarker of inflammation in lung,and clarify their roles in pathogenesis of silicosis.Methods Male rats were randomly divided into 28 groups according to their exposure doses to silica(10,30,50 or 70 mg/ml)and their exposure time(1 d,3 d,5 d,7 d,14 d,21 d and 28 d).The serum levels of silicon were determined by inductively coupled plasma mass spectrometry(ICP-MS),the serum levels of CC16 were examined by ELISA method.Meantime,the pathological changes in lung were also observed using hematoxylin-eosin staining.Results The silicon level in serum was gradually increased in a dose-and time-dependent manner,however,the CC16 level in serum was dose-dependently decreased but later than that of silicon level.Conclusions The results showed that the changes of serum levels of silicon and CC16 in rats appear quite early after exposure to silica dust,especially serum silicon level,and the changes were dose-and time-dependent.It is suggested that these indices mentioned above might provide some earlier evidence of silicosis,therefore should have certain clinical significance.
