Abstract:A 200-fold inhibition in the titer of infectious vesicular stomatitis virus (VSV) was produced in cultures of Ly cells treated with 30 reference units of interferon per milliliter. Virus particle production, as measured by VSV particle-associated transcriptase, or nucleocapsid protein was inhibited by a maximum of tenfold. The glycoprotein and membrane protein content was reduced in VSV derived from interferon-treated cells. Thus interferon-treated cells may have produced VSV particles with low infectivity, which may be related to the reduced amount of glycoprotein incorporated into such particles. These findings resemble those reported in interferon-treated cells infected with murine leukemia viruses.

Abstract:Silicosis is a particular form of lung fibrosis attributable to occupational exposure to crystalline silica. The occupational exposure to crystalline silica also increases the risk of chronic obstructive pulmonary disease (COPD), cancer and lung infections, especially pulmonary tuberculosis. Silicosis is currently diagnosed in previously exposed workers by standard chest X-ray, when lesions are visible and irreversible. Therefore, it would be necessary to find specific and non-invasive markers that could detect silicosis in earlier stages, before the occurrence of X-ray opacities. In this narrative review, we present several diagnostic, monitoring and predictive biomarkers with high potential in the management of silicosis, such as: pro- and anti-inflammatory cytokines (TNF (Tumour necrosis factor-α), IL-1 (Interleukin-1), IL-6, IL-10), CC16 (Clara cell 16, an indirect marker of epithelial cell destruction), KL-6 (Krebs von den Lungen 6, an indirect marker of alveolar epithelial damage), neopterin (indicator of cellular immunity) and MUC5B gene (Mucin 5B, a gel-forming mucin in mucus). Studies have shown that all the aforementioned markers have a high potential for early diagnosis or evaluation of progression in silicosis and represent promising alternatives to radiology. We consider that a multicentric study is needed to evaluate these biomarkers in correlation with occupational history, histopathological examination, imaging signs and pulmonary functions tests on large groups of subjects to better evaluate the accuracy of the presented biomarkers.
Diagnostic performance of distinct metabolic features in the plasma of patients with silicosis

Abstract:Abstract Background Silicosis is a progressive pneumoconiosis characterized by interstitial fibrosis following exposure to silica dust. This study aimed to identify potential noninvasive metabolic biomarkers for the diagnosis and monitoring of this condition by pilot and validation analyses of patients with silicosis in metabolomics studies. Methods Patients with silicosis, dust-exposed workers (DEWs) without silicosis and age-matched healthy controls were recruited in a case-control study. Plasma samples were collected, and metabolomics analyses by ultra-high-performance liquid chromatography-mass spectrometry (UHPLC-MS) were conducted. Distinct metabolic features (DMFs) among the groups were identified in the pilot study and were validated in the validation study. The enriched signalling pathways of these DMFs were determined. The ability of DMFs to discriminate among the groups in the validation study was analysed through receiver operating characteristic (ROC) curves. The correlations between DMFs and clinical features were also explored. Results Twenty-nine DMFs and 9 DMFs were detected in the plasma of the DEW and silicosis groups, respectively, compared with the control group; these features showed the same trend in the pilot study and the validation study. Sphingolipid metabolism was the major metabolic pathway in the DEWs, and arginine and proline metabolism was associated with silicosis. Twenty DMFs in the DEWs and 3 DMFs in the patients with silicosis showed a discriminatory ability with ROC curve analysis. The abundance of kynurenine was higher in Stage III silicosis than in Stage I or Stage II silicosis. L-arginine and kynurenine were both negatively correlated with the percentage of forced vital capacity predicted in silicosis. Conclusions Distinct metabolic features of plasma samples related to sphingolipid metabolism and arginine and proline metabolism were identified in the DEW and silicosis groups, respectively. L-arginine and kynurenine may have a predictive role in the diagnosis and severity of silicosis.

Abstract:Beck (1972) pointed out that grouping tasks, where one set of elements is distinguished from another, do not imply associative links within either set of elements but could rely on processes of segregation. Although many physiological theories of grouping are purely associative there are not many methods of isolating associative processes psychophysically. One method has been to measure common resolutions of elements when they are presented under conditions of depth and motion ambiguity. In the present paper a number of the issues surrounding grouping are considered, particularly in relation to associative processes. These include whether grouping is post-constancy and post-completion, grouping with and without emergent features, the role of top-down processing and attention and the explanatory role of general principles such as simplicity and likelihood.
Exosomal proteomics and cytokine analysis distinguish silicosis cases from controls

Abstract:Occupational silica exposure caused a serious disease burden of silicosis. There is currently a lack of sensitive and effective biomarkers for silicosis, and the pathogenesis of silicosis is unclear. Exosomes were significant in the pathogenesis of silicosis, and our study was carried out from exosomal proteomics and cytokine analysis. Firstly, the plasma levels of cytokines were detected using a Luminex multiplex assay, and the results indicated that the plasma levels of TNF-α, IL-6, CCL2, CXCL10, and PDGF-AB were significantly higher in silicosis patients than in silica-exposed workers and controls (p < 0.05). After correlation analysis, the plasma levels of cytokines were positively correlated with exosomal protein concentration. Secondly, data-independent acquisition (DIA) was performed on plasma-derived exosomes in the screening population, which identified 88, 151, 293, and 53 differentially expressed proteins (DEPs) in exposure/control, silicosis/control, silicosis/exposure, and silicosis stage Ⅲ/silicosis stage Ⅰ groups respectively. After parallel reaction monitoring (PRM) in an independent verification population, the results indicated that the changing trend of 15 DEPs was coincident in screening and verification results. The result of correlation analysis indicated that the plasma level of TNF-α was negatively correlated with the expression of exosomal DSP, KRT78, SERPINB12, and CALML5. The AUC of combined determination of TNF-α and CALML5 reached 0.900, with a sensitivity of 0.714 and a specificity of 0.933. Overall, our study revealed the exosomal proteomic profiling of silicosis patients, silica-exposed workers, and controls, indicating that exosomes were significant in the pathogenesis of silicosis. It also revealed that the combined of the plasma levels of cytokines and the expression of exosomal DEPs could increase determination efficiency. This study provided directions for the development of silicosis biomarkers and a scientific basis for the pathogenesis research of silicosis in the future.
Exploration study on serum metabolic profiles of Chinese male patients with artificial stone silicosis, silicosis, and coal worker's pneumoconiosis

Abstract:Long-term exposure to inhaled silica dust induces pneumoconiosis, which remains a heavy burden in developing countries. Modern industry provides new resources of occupational SiO<sub>2</sub> leading to artificial stone silicosis especially in developed countries. This study aimed to characterize the serum metabolic profile of pneumoconiosis and artificial stone silicosis patients. Our case-control study recruited 46 pairs of pneumoconiosis patients and dust-exposed workers. Nontargeted metabolomics and lipidomics by ultra-high-performance liquid chromatography-tandem mass spectrometry platform were conducted to characterize serum metabolic profile in propensity score-matched (PSM) pilot study. 54 differential metabolites were screened, 24 of which showed good screening efficiency through receiver operating characteristics (ROC) in pilot study and validation study (both AUC > 0.75). 4 of the 24 metabolites can predict pneumoconiosis stages, which are 1,2-dioctanoylthiophosphatidylcholine, phosphatidylcholine(O-18:1/20:1), indole-3-acetamide and l-homoarginine. Kynurenine, N-tetradecanoylsphingosine 1-phosphate, 5-methoxytryptophol and phosphatidylethanolamine(22:6/18:1) displayed the potential as specific biomarkers for artificial stone silicosis. Taken together, our results confirmed that tryptophan metabolism is closely related to pneumoconiosis and may be related to disease progression. Hopefully, our results could supplement the biomarkers of pneumoconiosis and provide evidence for the discovery of artificial stone silicosis-specific biomarkers.

Abstract:Long-term exposure to respirable dust containing silica leads to pneumoconiosis/silicosis. The disease is irreversible and incurable, and only preventive steps such as job rotation, use of personal protective equipment, etc., remain solutions to the problem. Under such a situation, early diagnosis or prediction may become very useful to control the disease. Biomarkers are biological parameters in blood serum that change their values with deposition of dust in the lung and onset of lung fibrosis. Since these biomarkers can help us to diagnose and in the prognosis of the disease before it is actually diagnosed by the conventional X-ray technique and lung function test used for diagnosis of silicosis. The present paper describes the various types of available biomarkers, their application and usefulness. The paper also suggests that further study of the behavior/level of these biomarkers on a specific subject with time may provide more useful information of a confirmatory nature for prevention of dust-linked diseases.

Abstract:Abstract Background Silicosis and asbestosis are progressive pneumoconiosis characterized by interstitial fibrosis exposure to silica dust or asbestos fibres. This study aimed to explore the potential biomarkers for diagnosis of silicosis or asbestosis. Methods Plasma concentrations of CC-chemokine ligand 18 (CCL18), CXC motif chemokine 13 (CXCL13), osteopontin (OPN), periostin, and fibulin-3 were compared between silicosis, asbestosis, and healthy controls (HCs). The receiver operating characteristic (ROC) curve was used to monitor the diagnostic sensitivity, and their correlation to disease severity was examined. Results Plasma CCL18, CXCL13, and OPN concentrations in both patient groups were high. For silicosis patients, these plasma concentrations were significantly correlated with predicted forced vital capacity (FVC%) and lung diffusing capacity for carbon monoxide (DL CO SB%). The order of diagnostic accuracy was CCL18, OPN, and CXCL13 in silicosis or asbestosis. Combining CCL18, OPN, and CXCL13 can improve diagnostic accuracy. Conclusions CCL18, CXCL13, and OPN are available biomarkers for the diagnosis of silicosis and asbestosis.
Significance of blood levels of silicon in early diagnosis of silicosis

Abstract:Objective To explore the significance of blood silicon levels in early diagnosis of silicosis.Methods Silicon exposed rats were randomly divided into eleven groups according to the body weight,detecting the serum silicon regularly by inductively coupled plasma mass spectrometry(ICP-MS)after exposure,the expression of transforming growth factor-β1,and tumor necrosis factor-α in lung tissue were also observed by immunohistochemistry-SP(streptavidin-perosidase)method.Results Comparing the controls,the serum silicon levels were raised at the first day after exposure(the peak value was at the 16th hour),the subsequent 7 d,14 d,21 d and 28 d were also significantly higher than that of control group(P<0.05),meanwhile,the expressions of TGF-β1 and TNF-α in the lung tissue of exposed rats were all obviously increased at 7 d,14 d,21 d and 28 d after exposure compared with control group(P<0.05).Conclusions The results showed that the rise of serum silicon level was even earlier than the expression of TGF-β1 and TNF-α in lung tissues,suggesting that this index may have some useful value in the early diagnosis of silicosis.

Abstract:Background Silicosis shows an increasing trend with the development of new industries. However, the potential biomarkers for predicting the disease severity are lacking. A novel inflammatory marker, the systemic immune-inflammation Index (SII), has not been studied in silicosis. Methods In this retrospective study, we used data from a big database platform of a tertiary general hospital in Beijing, which was established based on the electronic medical records of the hospital. The clinical data of adult patients diagnosed with silicosis at the Department of Occupational Medicine and Toxicology from 2013 to 2022 were collected. The data extracted from the database were in de-identified form. Only patients with a first diagnosis of silicosis and without conditions that might affect the parameters of routine blood tests were included in the analysis. Analyses were performed to assess the relationship between SII and the advanced stage of silicosis. Results A total of 246 participants were included in the study. Most of the patients were exposed to silica particles during excavation and digging ( n = 149, 60.6%). SII level was significantly higher in patients with advanced stages of silicosis. A multivariate logistic regression analysis revealed that a higher SII level was associated with the advanced stage of silicosis [odds ratio (OR) = 1.002; 95% confidence interval (CI): 1.000–1.003, p < 0.001] after adjusting for all covariates. The best cutoff value of SII was 444.1. The results of the subgroup analysis also showed a significant correlation between SII level over 444.1 and the advanced stage of silicosis in groups stratified by gender, history of smoking, and duration of silica exposure. Moreover, our results showed a significant but weak negative correlation between the level of SII and some lung function parameters in silicosis. Conclusion Higher SII is associated with the advanced stage of silicosis and impaired lung function. More long-term, large-scale studies are needed to confirm these findings.

Silicon, an important exposure marker in vivo in silicosis research

Abstract:Purpose: The degree of silicosis exposure is closely related to the progress of silicosis. At present, we use animal and human studies to explore whether silicon can be an important exposure marker in the development of silicosis. Methods: Rats were randomly divided into 2 groups: (1) controls; and (2) silicosis. Rats in the silicosis group were killed at 4, 8, 12, 16, 24 h, 3, 7, 14, 21, and 28 days. Hematoxylin-eosin (HE) and immunohistochemistry (IHC) were performed to observe the histomorphology of lung tissue. The expression levels of CC16 and SP-D were detected using ELISA kits. In addition, we conducted a population study. Workers who have been selected to work in an iron mine for more than 1 year as research objects. The population was divided into four groups: silicosis exposure group (workers exposed to silica dust for more than 1 year in an iron mine were selected); patients group (silicosis patients); observation group (evidence of disease not meeting formal diagnostic criteria) and control group. Both the levels of trace silicon in the urine and blood of rats and human subjects were measured with ICP-MS. Results: Serum levels of silicon were immediately increased in rats exposed to silicon dust. Similarly, our population study revealed that the silicon level in the silica exposure group and the observing group (exposed but no obvious symptoms) were significantly increased over that of the control group (P < 0.05). In subjects with extended exposure to silica, the serum and urine silicon level in exposed workers appeared to rapidly increase, reaching its peak in 1-5 years, followed by a gradual decline thereafter. Workers exposed to dust for less than 10 years were divided into subgroups by 2-year limit. The levels of serum silicon, urine silicon, TGF-β1, and TNF-α were significantly higher than that of control group. Conclusion: Changes of the serum levels of silicon occurred earlier than the expression of cytokines such as TNF-α, TGF-β1, CC16, and SP-D. The level of silicon in workers rapidly increased after exposure to silica, and the change occurred before the expression of TGF-β1 and TNF-α. As a whole, the findings suggest that determining the level of silicon in vivo might be an effective exposure marker in the diagnosis and pathogenesis of silicosis.
Integrating routine blood biomarkers and artificial intelligence for supporting diagnosis of silicosis in engineered stone workers

Abstract:Engineered stone silicosis (ESS), primarily caused by inhaling respirable crystalline silica, poses a significant occupational health risk globally. ESS has no effective treatment and presents a rapid progression from simple silicosis (SS) to progressive massive fibrosis (PMF), with respiratory failure and death. Despite the use of diagnostic methods like chest x-rays and high-resolution computed tomography, early detection of silicosis remains challenging. Since routine blood tests have shown promise in detecting inflammatory markers associated with the disease, this study aims to assess whether routine blood biomarkers, coupled with machine learning techniques, can effectively differentiate between healthy individuals, subjects with SS, and PMF. To this end, 107 men diagnosed with silicosis, ex-workers in the engineered stone (ES) sector, and 22 healthy male volunteers as controls not exposed to ES dust were recruited. Twenty-one primary biochemical markers derived from peripheral blood extraction were obtained retrospectively from clinical hospital records. Relief-F features selection technique was applied, and the resulting subset of 11 biomarkers was used to build five machine learning models, demonstrating high performance with sensitivities and specificities in the best case greater than 82% and 89%, respectively. The percentage of lymphocytes, the angiotensin-converting enzyme, and lactate dehydrogenase indexes were revealed, among others, as blood biomarkers with significant cumulative importance for the machine learning models. Our study reveals that these biomarkers could detect a chronic inflammatory status and potentially serve as a supportive tool for the diagnosis, monitoring, and early detection of the progression of silicosis.
Engineering solutions to breath tests based on an e-nose system for silicosis screening and early detection in miners

Abstract:Abstract This study aims to develop an engineering solution to breath tests using an electronic nose (e-nose), and evaluate its diagnosis accuracy for silicosis. Influencing factors of this technique were explored. 398 non-silicosis miners and 221 silicosis miners were enrolled in this cross-sectional study. Exhaled breath was analyzed by an array of 16 organic nanofiber sensors along with a customized sample processing system. Principal component analysis was used to visualize the breath data, and classifiers were trained by two improved cost-sensitive ensemble algorithms (random forest and extreme gradient boosting) and two classical algorithms (K-nearest neighbor and support vector machine). All subjects were included to train the screening model, and an early detection model was run with silicosis cases in stage I. Both 5-fold cross-validation and external validation were adopted. Difference in classifiers caused by algorithms and subjects was quantified using a two-factor analysis of variance. The association between personal smoking habits and classification was investigated by the chi-square test. Classifiers of ensemble learning performed well in both screening and early detection model, with an accuracy range of 0.817–0.987. Classical classifiers showed relatively worse performance. Besides, the ensemble algorithm type and silicosis cases inclusion had no significant effect on classification ( p > 0.05). There was no connection between personal smoking habits and classification accuracy. Breath tests based on an e-nose consisted of 16× sensor array performed well in silicosis screening and early detection. Raw data input showed a more significant effect on classification compared with the algorithm. Personal smoking habits had little impact on models, supporting the applicability of models in large-scale silicosis screening. The e-nose technique and the breath analysis methods reported are expected to provide a quick and accurate screening for silicosis, and extensible for other diseases.

The Indirect Efficacy Comparison of DNA Methylation in Sputum for Early Screening and Auxiliary Detection of Lung Cancer: A Meta-Analysis

Abstract:Background: DNA methylation in sputum has been an attractive candidate biomarker for the non-invasive screening and detection of lung cancer. Materials and Methods: Databases including PubMed, Ovid, Cochrane library, Web of Science databases, Chinese Biological Medicine (CBM), Chinese National Knowledge Infrastructure (CNKI), Wanfang, Vip Databases and Google Scholar were searched to collect the diagnostic trials on aberrant DNA methylation in the screening and detection of lung cancer published until 1 December 2016. Indirect comparison meta-analysis was used to evaluate the diagnostic value of the included candidate genes. Results: The systematic literature search yielded a total of 33 studies including a total of 4801 subjects (2238 patients with lung cancer and 2563 controls) and covering 32 genes. We identified that methylated genes in sputum samples for the early screening and auxiliary detection of lung cancer yielded an overall sensitivity of 0.46 (0.41–0.50) and specificity of 0.83 (0.80–0.86). Combined indirect comparisons identified the superior gene of SOX17 (sensitivity: 0.84, specificity: 0.88), CDO1 (sensitivity: 0.78, specificity: 0.67), ZFP42 (sensitivity: 0.87, specificity: 0.63) and TAC1 (sensitivity: 0.86, specificity: 0.75). Conclusions: The present meta-analysis demonstrates that methylated SOX17, CDO1, ZFP42, TAC1, FAM19A4, FHIT, MGMT, p16, and RASSF1A are potential superior biomarkers for the screening and auxiliary detection of lung cancer.
Biomarkers related to silicosis and pulmonary function in individuals exposed to silica

Abstract:Background: The identification of markers that can facilitate the early diagnosis of silicosis has remained challenging. We evaluated the association of inflammatory markers with the presence of silicosis and lung function impairment in individuals exposed to silica. Methods: Individuals exposed and not exposed to silica were assessed by occupational history, clinical findings, lung function, chest imaging findings, and inflammatory markers. Results: Among 297 men evaluated, 51 were unexposed controls (G1), 149 were exposed to silica without silicosis (G2), and 97 were exposed to silica with silicosis (G3). Inflammatory marker levels were higher in G3 than in G2 and G1. Platelet/lymphocyte ratio (PLR), lactate dehydrogenase (LDH), soluble tumor necrosis factor II (sTNFRII), and macrophage inflammatory protein-4 (MIP-4) were associated with silicosis, and LDH, neutrophil/lymphocyte ratio (NLR), sTNFRII, monocyte chemoattractant protein-1 (MCP-1), soluble vascular cell adhesion molecule-1 (sVCAM-1), and fibrinogen were negatively associated with lung function. Conclusion: Blood inflammatory markers are associated with silicosis and impaired lung function.

Abstract:Objective To explore the significance of blood level of silicon in the pathogenesis of silicosis.Methods Serum silicon was detected by inductively coupled plasma mass spectrometry (ICP-MS) and the expression of transforming growth factor-β1 (TGF-β1),tumor necrosis factor-α(TNF-α) in lung tissue of rats were observed by immunohistochemistry-SP.Results The serum level of silicon was significantly increased at the 16th hour of the first day after being exposed to silica and markedly higher than that of the control rats[(0.666±0.167) mg/L versus (0.055±0.040) mg/L,P0.01].It was also higher in the group on the subsequent 7,14,21 and 28 days compared with control.The expression of TGF-β1 in the lung tissue of model rats reached its peak value at 7th day and was significantly higher than that of the control group at 7,14,21 and 28 days (P0.05).The expression of TNF-α in the lung tissue of the model rats was significantly higher than that of the control group at 7,14,21 and 28 days (P0.05).Conclusion The blood level of silicon in silicosis rats was rapidly increased after exposed to silica and this change was prior to the expression of TGF-β1 and TNF-α,which indicated that this change might be meaningful for the study of silicosis pathogenesis and used as a biomarker for the early diagnosis of silicosis.

Abstract:Silicosis is a global problem, and it has brought about great burdens to society and patients’ families. The etiology of silicosis is clear, preventable, and controllable, but the onset is hidden and the duration is long. Thus, it is difficult to diagnose it early and treat it effectively, leaving workers unaware of the consequences of dust exposure. As such, a lack of details in the work history and a slow progression of lung disease contribute to the deterioration of patients until silicosis has advanced to fibrosis. These issues are the key factors impeding the diagnosis and the treatment of silicosis. This article reviews the literature on the early identification, diagnosis, and treatment of silicosis as well as analyzes the difficulties in the diagnosis and the treatment of silicosis and discusses its direction of future development.

[Analysis of systemic cytokine levels in silicosis complicated with pulmonary infection]

Abstract:Objective: To investigate the changes of cytokine profile in elderly patients with silicosis and lower respiratory tract infection and its clinical significance. Methods: In Oclober 2019, a retrospective study was conducted to select 167 elderly patients with silicosis from Xuzhou Mining Group General Hospital as subjects, including 115 patients with silicosis and lower respiratory tract infection as infected group, including 41 patients with stage I silicosis, 38 patients with stage II silicosis and 36 patients with stage III silicosis. There were 52 cases of silicosis without lower respiratory tract infection as non-infected group, and 48 cases of healthy examination in our hospital were selected as control group. All the participants were tested for cytokines (IL-6, IL-10, IL-17A, TNF-α, IFN-γ) and statistically analyzed the relevant experimental data. Results: The levels of IL-6, IL-10, IL-17A, TNF-α and IFN-γ in the infected group after treatment were significantly lower than those before treatment (P<0.05) . The levels of IL-6, IL-10, IL-17A, TNF-α and IFN-γ before and after treatment in the infected group were higher than those in the non-infected group and the control group (P<0.05) . The levels of IL-10, IL-17A, TNF-α and IFN-γ in the infected group were higher than those in the control group, and the difference were statistically significant (P<0.05) . The levels of IL-6, IL-10, IL-17A and IFN-γ in patients with stage III silicosis were higher than those in stage I silicosis in infected group (P<0.05) . The levels of IL-6 and IL-17A in the patients with stage Ⅲ silicosis were higher than those in the patients with stage II silicosis in infected group (P<0.05) . The level of IL-10 in patients with stage II silicosis was higher than that in stage I in infected group (P<0.05) . ROC curve analysis of the indicators for diagnosis of senile silicosis with lower respiratory tract infection and silicosis without lower respiratory tract infection found that IL-6 had the largest AUC (0.910) . And its specifuity and sensithcity were 85.2% and 98.1% respectivehy. Conclusion: The detection of cytokines such as IL-6 and IL-10 has better specificity and higher diagnostic efficiency in the early diagnosis and treatment of elderly patients with silicosis complicated with lower respiratory tract infection. It has good clinical application value and can provide important laboratory evidence for early treatment for clinicians.

Abstract:Objectives: In this study, by comparing the difference in protein expression in bronchoalveolar lavage fluid between silicosis patients in different stages and healthy controls, the pathogenesis of pneumoconiosis was discussed, and a new idea for the prevention and treatment of pneumoconiosis was provided. Methods: The lung lavage fluid was pretreated by 10 K ultrafiltration tube, Agilent 1100 conventional liquid phase separation, strong cation exchange column (SCX) HPLC pre-separation, and C18 reverse phase chromatography desalting purification, and protein was labeled with isotope. GO, KEGG pathway, and PPI analysis of differential proteins were conducted by bioinformatics, and protein types and corresponding signal pathways were obtained. Results: Thermo Q-Exactive mass spectrometry identified 943 proteins. T-test analysis was used to evaluate the different significance of the results, and the different protein of each group was obtained by screening with the Ratio≥1.2 or Ratio≤0.83 and P<0.05. We found that there are 16 kinds of protein throughout the process of silicosis. There are different expressions of protein in stages III/control, stages II/control, stage I/control, stages III/ stages II, stages III/ stage I and stages II/ stage I groups. The results of ontology enrichment analysis of total differential protein genes show that KEGG pathway enrichment analysis of differential protein suggested that there were nine pathways related to silicosis. Conclusion: The main biological changes in the early stage of silicosis are glycolysis or gluconeogenesis, autoimmunity, carbon metabolism, phagocytosis, etc., and microfibril-associated glycoprotein 4 may be involved in the early stage of silicosis. The main biological changes in the late stage of silicosis are autoimmunity, intercellular adhesion, etc. Calcium hippocampus binding protein may participate in the biological changes in the late stage of silicosis. It provides a new idea to understand the pathogenesis of silicosis and also raises new questions for follow-up research.
