Abstract:A one-pot two-step reaction of 4-aminocoumarin with arylglyoxal monohydrates and p-toluenesulfonates has been developed, which offers 3-alkoxy-substituted chromeno[4,3-b]pyrrol-4(1H)-ones in moderate to good yields. At first, arylglyoxal monohydrates and 4-aminocoumarin were converted into 3-hydroxychromeno[4,3-b]pyrrol-4(1H)-one intermediates in AcOH under reflux. Then the title compounds in an in situ reaction of the intermediates with p-toluenesulfonates catalyzed by DBU in refluxing toluene were obtained.

Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:Utilizing simple precursors for the exquisite construction of molecules with diversity and complexity is urgent. Herein, a novel A3-X type reaction for functionalized 3-haloquinolines has been developed by PhI (OAc)2-mediated four-component tandem addition/cyclization/oxidation/aromatization process with simple aromatic amine, aldehyde, alkyne and halide salt. Broad functional group tolerance, very mild condition, easy for further transformation illustrate the concision, high efficiency and practicability of this A3-X type reaction.

Abstract:A novel iron-catalyzed one-pot multi-component tandem reaction of acetoacetamide, urea, and two molecules of aryl aldehydes has been developed. This reaction provides a general, straightforward, practical and useful method for the preparation of potential bioactive (E)-6-arylvinyl-3,4-dihydropyrimidin-2(1H)-ones. They are also versatile precursors leading to diverse drug-like dihydropyrimidin-2(1H)-one derivative. The characters of avoiding protonic acid and using a single inexpensive and environmentally benign catalyst make this method more valuable.

Abstract:An efficient tandem intermolecular one-pot aldol condensation/aza-addition reaction of 2-methyl-3-carbamoylpyrroles and aldehydes was developed for the synthesis of 2,3,6,7-tetrahydro-1H-pyrrolo[3,2-c]pyridin-4(5H)-ones. The reaction proceeded using only 3.0 equiv of ammonium acetate promoter in green solvent poly(ethylene glycol)-400 at 100 °C to afford a series of pyrrolo[3,2-c]pyridinone derivatives in good to excellent yields.

Abstract:A one-pot three-component synthesis of dihydropyrimidinones via a molecular iodine-catalyzed tandem reaction of simple readily available mono-substituted urea, alkylaldehyde, and arylaldehyde has been developed. The reaction proceeds with high chemo-and regioselectivity to give highly diverse dihydropyrimidinones in reasonable yields under mild reaction conditions. Moreover, the first catalytic enantioselective version of this reaction was also realized by using chiral spirocyclic SPINOL-phosphoric acids.

Abstract:A facile and efficient one-pot three-component reaction method for the synthesis of thiazine-dicarboxylates is reported. Reaction of an isocyanide and dialkyl acetylenedicarboxylate with 2-amino-4H-1,3-thiazin-4-one derivatives containing both an acidic proton and an internal nucleophile gave the products in good yields of 76–85%. The reactivity of dialkyl acetylenedicarboxylates was further tested in the synthesis of thiazole-pyrimidines where a two-component reaction of 2-aminothiazole with dialkyl acetylenedicarboxylates was successfully converted to a more efficient three-component reaction of a thiourea, α-haloketone and dialkyl acetylenedicarboxylate (DMAD/DEtAD) to give thiazole-pyrimidines in good yields of 70–91%.

Abstract:One-pot chemoselective condensation of 1,3-cyclohexanedione, urea or thiourea with aldehydes has been performed by using TMSCl as a Lewis acid in DMF/MeCN. This extended Biginelli-type reaction afforded two families of fused heterobicyclic and spiro-fused heterobicyclic compounds in excellent yields. para-Substituted benzaldehydes exhibited different chemical behaviors to ortho- or meta-substituted benzaldehydes and aliphatic aldehydes. The possible mechanism was discussed to account for experimental results.
