Abstract:A concise and efficient synthetic approach was developed for the synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones via a one-pot, three-component reaction of 4-aminocoumarin, arylglyoxal monohydrates, and aromatic amines promoted by KHSO4 in toluene under reflux condition. The attractive features of this protocol are operational simplicity, short reaction time, easy purification of products and good to excellent yields.

Abstract:A mild, efficient, and environmentally benign one-pot synthesis of functionalized chromeno[4,3-b]pyrrol-4(1H)-ones by a three-component domino reaction of 4-aminocoumarins, arylglyoxal monohydrates, and 1,3-dicarbonyl compounds in refluxing ethanol without the use of catalyst is reported. Abstract:An efficient tandem process was developed to synthesize diversified benzo[a]xanthones from 2-methyl-3-(1-alkynyl)chromones with electron-deficient chromone-fused dienes. This unusual reaction, involving multiple steps and not requiring the use of transition metal catalysts or an inert atmosphere, results in the formation of three new C-C bonds and one C-O bond.

Abstract:A hexafluoroisopropanol（HFIP）-catalyzed highly diastereoselective formal [4 + 2] cyclization between ortho-hydroxyphenyl para-quinone methides and difluoroenoxysilanes is developed. Abstract:A selective copper(I)-catalyzed benzylic C(sp3)–H geminal difunctionalization reaction of a benzylic-type sp3 carbon was developed. This novel strategy allowed simultaneous introduction of amide and hydroxyl group in a highly selective and efficient way via successive oxidative intramolecular amidation and hydroxylation. This method was also applied to the synthesis of 3-hydroxy-2,3-dihydro-1H-pyrrolo[3,4-b]quinoxalinones from readily available 3-methyl-N-substituted quinoxaline-2-carboxamides in moderate to good yields. The five-membered cyclic hemiaminal moiety of the pyrrolo[3,4-b]quinoxalinones can serve as an intermediates for subsequent transformations into other useful functional groups.

Abstract:A simple arylamine-catalyzed Mannich-cyclization cascade reaction was developed for facile synthesis of substituted 2H-benzo[h]chromenes. The notable feature of the process included the efficient generation of ortho-quinone methides (o-QMs) catalyzed by a simple aniline. The mild reaction conditions allowed for a broad spectrum of 1- and 2-naphthols and trans-cinnamaldehydes to engage in the cascade sequence with high efficiency.

Abstract:A tandem Prins/Friedel-Crafts cascade strategy has been developed for the synthesis of hexahydro-2H-benzo[g]chromene derivatives. Under action of equimolecular amount of BF3 etherate, delta,epsilon-unsaturated alcohols and arylacetaldehydes are coupled to form oxocarbenium ion intermediates, which are captured by the alkene moiety through a favored six-membered transition state. The Prins-cyclization resultant tertiary carbenium ions undergo consecutive intramolecular Friedel-Crafts cyclization to create the tricyclic products. This process enables a tandem formation of three new chemical bonds in one single operation with high yields and stereoselectivity in most cases. When cyclic substituents are involved, a variety of spiro derivatives could also be synthesized successfully, which further proved the robustness of this methodology.

Abstract:An efficient synthesis of novel functionalized 2-aryl-4-(indol-3-yl)-4H-chromenes has been developed, in the presence of a catalytic amount of iodine, from easily available starting materials, 2-hydroxychalcone derivatives. Abstract:A one-pot synthesis of substituted (dihydro)thieno[2,3-c]chromene derivatives was developed through a Lewis base catalyzed domino Michael addition/intramolecular cyclization reaction between 3-nitro-2H -chromenes and thioamides. The desired (dihydro)thieno[2,3-c]chromene derivatives were prepared with a large substrate scope (22 examples) and excellent diastereoselectivity in moderate to excellent yields.

Abstract:A facile and efficient strategy for modular access to furo[3,2- c ]chromen-4-ones using 4-hydroxycoumarin and β-nitroalkenes via Lewis acid-catalyzed formal [3 + 2] annulation protocol is described. This reaction proceeds via cascade Michael addition/nucleophilic addition/elimination in the presence of Yb(OTf) 3 , which involves the formation of two new σ (C–C and C–O) bonds for the construction of a novel furan ring in a single operation. This protocol affords a variety of functional groups, thereby providing a practical and efficient method for the fabrication of a furo[3,2- c ]chromen-4-one framework.

Abstract:Accomplished herein is a rearrangement strategy for the highly efficient assembly of synthetically cumbersome while medicinally significant 2-amino-3-formyl chromones via palladium-catalyzed ring-opening, rearrangement, and cyclization process. Such a sequence enables the formation of one C(sp2)–O bond and one C(sp2)–C(sp2) bond, and reconstruction of the benzo-γ-pyrone moiety in a single operation, thus producing the difunctionalized chromone-incorporated derivatives. The reaction proceeds in a shorter reaction time (30 min for 3-iodochromones in most cases) in a highly atom- and step-economical manner. The synthetic application of the current protocol is further demonstrated by late-stage modification of pharmaceuticals and their intermediates, gram-scale reactions, transformations of functional groups, as well as the synthesis of bioactive molecules and drugs. Mechanistic studies indicate that a nucleophilic ring-opening process of the benzo-γ-pyrone moiety, phenoxy anion intermediate, and an intramolecular palladium-catalyzed cyclization process might be involved in the present reaction system.
