Abstract:An improved process was reported to solve the problems such as low yield and high costs in the synthesis of 3-n-propyl-2, 4-dihydroxyacetophenone, which is a key intermediate of Leukotriene receptor antagonist. The target compound was synthesized starting from m-dihydroxybenzene, via acylation, allylation, Claisen rearrangement and hydrogenation reduction. The optimal reaction conditions were as follows. The acylation reaction which used acetic acid as acylation agent and ZnCl 2 as catalyst was carried out under reflux for 5 h, and 77.9% yield was obtained. The allylation reaction using allyl chloride as allylation reagent and NaBr, KI as catalyst was carried out at 40°C for 2 h and continued at 70°C for 14 h, to give 98.6% yield. In Claisen rearrangement carried out at 205°C for 4.5 h, diphenyl ether was used as reaction solvent and NaCl was used as catalyst, and the yield was improved to 67.4%. In the final step, the hydrogenated reduction with Pd/C (w(Pd) is 5%) as catalyst was carried out at 45°C for 6 h, and 88.6% yield was obtained. The process is much of industrial value because of high yield, cheap and available materials, and convenient operations. The structure of the main compounds was confirmed by 1H-NMR, GC-MS and IR.

Abstract:A parallel process for the new synthesis of 2,7-dimethyl-2,4,6-octatriene-1,8-dial was investigated.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-butenal was obtained by a process involving aldehyde protection,ester exchange and catalytic oxidation from 4-acetoxy-2-methyl-2-butenal.A new catalytic oxidation process was proposed to get 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal by oxidating 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol with copper(Ⅰ) chloride、2,2,6,6-tetramethyl-1-piperidinyloxy and oxygen.3-(5,5-Dimethyl-1,3-dioxan-2-yl)-2-buten-1-ol was chlorizated with SOCl_2 in DMF to get 2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane.2,7-Dimethyl-2,4,6-octatriene-1,8-dial was acquired in an overall yield of 48% by one-pot synthesis method from 3-(5,5-dimethyl-1,3-dioxan-2-yl)-2-butenal,2-(3-chloro-1-methyl-1-propenyl)-5,5-dimethyl-1,3-dioxane and triethylphosphite.

Abstract:Cilastatin is an important inhibitor of dehydropeptidase-I.One of its intermediate,7-chloro-2-oxohepanoic acid,was synthesized via two steps: through addition of mono-Grignard reagent of 1-bromo-5-chloro-pentane to diethyl oxalate,the ethyl 7-chloro-2-oxohepanoate was synthesized with 57% yield;then the ester was hydrolyzed to produce 7-chloro-2-oxohepanoic acid with 81% yield.The chemical structure of ethyl 7-chloro-2-oxohepanoate was approved by MS.

Abstract:2,2-Diethoxypropane was prepared by acetone and anhydrous alcohol under triethyl orthoformate as dehydration agent and ammonium chloride as catalyst. Effects of reaction condition on the condensation were investigated, according to the method of single factor test. The preferably condition was that acetone 26 g (0.45 mol), triethyl orthoformate 74 g (0.50 mol), alcohol 75 mL and ammonium chloride 0.72 g were reacted under reflux for 3 h. The yield was 77.5%.

Abstract:A new, practical and cost-effective route for scalable synthesis of 2-[(4-dodecyloxyphenyl)sulfonyl] butanoic acid, a key intermediate of a new cyan dye-forming coupler containing a sulfone group, was developed by adopting phenol as the starting material. The synthesis was accomplished in five steps with etherification, chlorosulfonation, reduction, nucleophilic reaction by C-S coupling and hydrolyzation. An important objective of the new synthetic route was the synthesis of 2-[4-(dodecyloxyphenyl)sulfonyl]butanoate. Overall yield obtained at optimized conditions increased to 66 %. The synthetic strategy was proven to be a process enabling rapid delivery of the target product with high purity.

Abstract:The marine peptides have been a fertile source for new bioactive molecules, and many possess potent, activity, across a broad spectrum of targets. Their diversity in both biological activity and in chemical complexity has made these secondary metabolites being the focus of much work in recent years. As a fragment, (R)-2-methyloctanoic acid-7-yne was always found in those nature materials. In this article, (R)-2-methyl-7-octynoic acid was synthesized with pentane-1, 5-diol as the start material. Stereogenic carbon(C-2) was obtained through alkylation induced with Evans' chiral auxiliary, and alkyne 13 was got by use of Ohira-Bestmann reagent. Through 13-step reactions the target molecule was prepared with over yield of 17. 1%, and e. e. beyond 99%.

Abstract:OBJECTIVE To study the synthetic method of 2-aminoxy-1- aminoethane dihydrochloride. METHOD The objective compound was synthesized by steps of ring-opening, condensation, hydrazinolysis and hydrolysis, starting from phthalic anhydride.RESULTS The chemical structure of synthetic product was confirmed by IR and 1HNMR, the total yield was amount to 39.4%. CONCLUSION This synthetic route is practical.

Abstract:6-Hydroxy-8-chlorooctanoate ethyl ester, an intermediate of α-lipoic acid, was synthesized via a chemoenzymatic method. High-throughput screening revealed that keto reductase HGD-1 could effectively catalyze the preparation of 6-hydroxy-8-chlorooctanoate ethyl ester, achieving a substantial conversion rate. The reaction conditions were optimized subsequently, the parameters for the complexation reaction were established as follows: a weight of AlCl 3 1.8 times that of the substrate; a reaction time of 4 h; a temperature range of 20–25 °C. The parameters for the enzyme-catalyzed reaction were as follows: a temperature range of 25–30 °C; a reaction time of 4 h; a solution pH of 6.5–7.5; a substrate concentration of 50 g/L; concentrations of keto reductase HGD-1, coenzyme glucose dehydrogenase, and nicotinamide adenine dinucleotide phosphate of 3 g/L, 4 g/L, and 0.05 g/L, respectively. Under these optimal conditions, the substrate conversion rate exceeded 95%, and 92% yield of 6-hydroxy-8-chlorooctanoate ethyl ester is obtained, suggesting a viable, eco-friendly method for synthesizing α-lipoic acid, and it provides a green process route for the industrial production of α-lipoic acid. Abstract:The novel efficient procedure has been developed for the oxathioacetalization of carbonyl compounds and 2-mercaptoethanol at room temperature under solvent-free condition. The results showed that the novel multi-SO 3 H functionalized ionic liquid was very efficient for the reactions with the good to excellent yields in short time. Operational simplicity, without need of any solvent, room temperature, low cost of the catalyst used, high yields, applicability to large-scale reactions, reusability and chemoselectivity are the key features of this methodology.

Abstract:A convenient, scalable, and highly enantioselective total synthesis of (R)-5,6-octadecadienoic acid was developed by using the recently established ATA (allenylation of terminal alkynes) reaction of TBS-protected (TBS = tert-butyldimethylsilyl) propargyl alcohol with n-dodecanal in the pres-ence of (R)-alpha,alpha-diphenylprolinol as the chiral source. The axial chirality of the allene unit withstood the many common organic transformations that were needed to achieve the total synthesis with a high ee (enantiomeric excess) value.

Abstract:A new synthetic method for ((4R,6S)-6-hydroxymethyl-2,2-dimethyl-1,3-dioxinan-4-yl)-acetic acid tert-butyl ester was accomplished by a chemoenzymatic approach. Racemic 4-(4-methylbenzyloxy)-3-hydroxybutyronitrile (3) was firstly synthesized via condensation and epoxy opening reactions. (S)-4-(4-Methylbenzyloxy)-3-hydroxybutyronitrile (4) was obtained from resolution of racemic 3 by the lipase from Artgribacter sp. with a high enantiomeric excess (98% ee) in the mixture solvent of acetonitrile/hexane (V : V= 1 : 4). Dihydroxy ester 7 was synthesized by a syn-selective triethylborane mediated reduction with a de value of 97%. After acid-catalyzed protection of 7 and hydrogenolysis of the benzyl protecting group, the objective 9 was obtained with a high optical purity.

Abstract:(E)-4-bromo-3-methyl-2-butenoic acid was conveniently synthesized from 3-methyl-2-butenoic acid in 29% overall yield.In this procedure,the title compound was easily obtained by hydrolysis of (E)-trimethylsilyl 4-bromo-3-methyl-2-butenoate,which could be isolated from its mixture of Z-/E-isomer by distillation in vacuo.The isomeric mixture was prepared via the esterification of 3-methyl-2-butenoic acid with chlorotrimethylsilane,followed by bromination with NBS.

Abstract:Z(E)-ethyl 3-oxo-2-(4-phenyl-1,3-dithiolan-2-ylidene)butanoate (1) was prepared in moderate (40.3%) yield by the reaction of ethyl 3-oxobutanoate with carbon disulfide and 1-(1,2-dibromoethyl)benzene with potassium carbonate as the base in DMF.Subsequently,the condensation reaction of 1 with arylalde- hydes was performed in the presence of C_2H_5ONa in C_2H_5OH,in which the product of condensation and ester hydrolysis:(2Z(E),4E)-3-oxo-5-phenyl-2-(4-phenyl-1,3-dithiolan-2-ylidene)pent-4-enoic acid (2) were isolated in good yield.

Abstract:3,4-Hexanedione was synthesized by using cheap propanal as starting material.Propionoin was synthesized from propanal by self-condensation under the catalysis of catalytic amount thiazoliumion and sodium carbonate,and the yield of propionoin was 90%.In the presence of concentrated sulfuric acid,propionoin was oxidized by 30%H2O2/FeSO4·7H2O,3,4-hexanedione was obtained with the yield of 85%.The product was determined by contrasting standard sample via GC.

Abstract:A new route for the synthesis of( 3S,4S)-4-amino-3-hydroxy-5-phenylpentanoic acid derivatives was reported. The target compounds with overall yields of 5. 8% ~ 6. 7% were synthesized by condensation with Weinreb amide,reduction,aldol condensation reaction and bromine imitation reaction,using amino-protected L-phenylalanine as starting material. The structures were confirmed by1 H NMR,13 C NMR and ESI-MS. The exploration of reaction conditions showed that the loading amount of D-proline had no evident effect on the yield,but on stereoselectivity. Larger size protecting groups in substrate can enhanced the stereo-selectivity.
