Abstract:The process of preparing 4-hydroxy-3-methoxyphenylacetonitrile,as one of the important intermediates for synthesis was studied.Beginning from the raw material—vanillin,the title compound was prepared,via protection of hydroxyl,reduction,chloridization and nitrilation.The influences of various reaction conditions on the yield were investigated,and the optimal reaction conditions(reagent,temperature,time and yield) were as follows:acetic anhydride as acetylation reagent,reacting at room temperature for 3 h and yield was 99%;Raney Ni as catalyst,reaction time was 8 h at room temperature,and yield was 96.8%;dropping thionyldichloride at 0℃,then reacting at room temperature for 2 h,and the yield was 78%;refluxing with sodium cyanide for 9 h,and the yield was 78.5%.The products were characterized with MS,1H NMR and elemental analysis.The results showed that the process proposed overcame the defects of traditional synthesis route.

Abstract:Aromatic aldehydes are very important organic intermediates,which are widely used in pharmaceuticals,pesticides and other areas.In this paper,an improved synthetic method of aromatic aldehydes containing electrowithdrawing groups in ortho or para(or both) position of the aromatic ring was introduced.4-choloro-2-nitrotoluene,4-nitrotoluene and 4-cyanotoluene were used as raw materials,respectively,via enamination with N,N-dimethylformamide dimethyl acetal(DMFDMA) in the present of N,N-dimethylformamide(DMF) as solvent and under reflux,the corresponding styrylamine compounds were produced;then the intermediates were further oxidized by H2O2 under different reaction conditions.The effects of solvent species,reaction temperature and the used amount of H2O2 on the product yield were studied,and the optimal reaction conditions were determined.Under those optimal conditions,the best yields of different aldehydes are 75.6%(4-chloro-2-nitrobenzaldehyde),58.3%(4-nitrobenzaldehyde) and 68.2%(4-cyanobenzaldehyde),respectively with the advantages of simple process,low cost and being environment-friendly..The main intermediates and objective products were confirmed by HPLC and 1H-NMR.

Abstract:2-Bromo-5-hydroxy-4-methoxybenzaldehyde(I)and 5-hydroxy-4-methoxy-2-nitrobenzaldehyde(II)were synthesized.The intermediates 2-bromo-4,5-dimethoxybenzaldehyde and 4,5-dimethoxy-2-nitrobenzaldehyde were synthesized firstly from veratraldehyde through bromination and nitration respectively.In the presence of molar quantity methionine,the 5th-position methoxys of the intermediates were in turn hydrolysed to hydoxy by concentrated sulfuric acid to give I and II.The chemical structures of the titled compounds were confirmed by 1H-NMR,IR and MS spectrum.The yields of compounds I and II are 85.0% and 65.0% respectively.The orientations of the bromination and nitration steps were suggested and discussed.The mechanism of methionine in the hydrolysis step may involve in the formation of the multiple hydrogen bonded association complex,and the calculated single point energy(E=-1532.5 hartree)of the multiple hydrogen bonded association complex seems to show it is possibly existed at the RHF/6-31G level.

Abstract:As an intermediate for COX-2 inhibiters,4-(methylthio)benzeneacetic acid(Ⅳ) was prepared via Friedel-Crafts and Willgerodt-Kindler reaction from thioanisole(Ⅰ).When the molar ratio of Ⅰ to acetyl chloride and AlCl_3 was 1∶1.1∶1.3 and CS_2 was used as solvent,after reacting at(0°C) for 6 h the yield of 4-(methylthio) acetophenone(Ⅱ) was 75.9%.Ⅱ was refluxed 5 h with morpholine and sulfur to give thioacetmorpholide(Ⅲ),then Ⅲ was refluxed in 10% NaOH for 10 h to give Ⅳ with an overall yield of 63.5% and purity of 99.0%(by HPLC).The structure was confirmed by IR and()~1H NMR.

Abstract:2-(4-Chlorophenyl)ethanol(1) was synthesized by esterification and reduction from 2-(4-chlorophenyl)acetic acid(3).The structure was characterized by 1H NMR and MS.The appropriate reaction conditions were as follows: 3 was 30 mmol,ethanol was 120 mmol,n(3)∶n(SOCl2)∶n(NaBH4) was 1.0 ∶0.5 ∶1.6.The yield of 1 was over 95% by esterification at room temperature for 3 h and reduction at 75 ℃ for 5 h.

Abstract:1-Chloro-2,5-dimethoxybenzene was prepared from p-benzoquinone through chlorination and methylation with total yield of 78%. p-Benzoquinone was firstly converted catalytically to 1-chloro-2,5-dihydroxybenzene in the binary system of dichloroethane and hydrochloric acid at 35℃ with yield of 89%, then the final product was synthesized by methylation of obtained 1-chloro-2,5-dihydroxybenzene with dimethyl sulfate in the presence of sodium hydroxide. The intermediate 1-chloro-2,5-dihydroxybenzene and the obtained title compound 1-chloro-2,5-dimethoxybenzene were characterized by IR and GC-MS respectively. The results showed that their chemical structures were correct and the purity of title compound was over 98%.

Abstract:2, 4-Dimethoxybenzaldehyde was prepared using 1, 3-dimethoxybenzene as raw material through Vilsmeier formylation with DMF directly. The compound was characterized by IR and NMR as a qualified product. Under the optimal conditions the product obtained from the orthogonal experiments could be given in 94.2% yield with purity of 98.64%, after purification the purity of final compound could be reached 99.6%.

Abstract:A procedure for synthesis of 2-methoxyphenothiazine (1) has been developed, starting from resorcinol and aniline by condensation, following methylation and cyclization. p-Toluenesulfonic acid and polysubstituted aromatics were employed as the catalyst of condensation and the solvent of cyclization, respectively, to improve the yield. The use of parallel experiments, statistical experimental design, and multivariate modeling made the total yield of the procedure as high as 74.2%. J. Heterocyclic Chem., (2012).

Abstract:Trifluoronitrobenzene was hydrolyzed in alkali solution to the corresponding potassium phenolate(Ⅰ) .In the presence of potassium iodide,treatment of Ⅰ with monochloroacetone gave 2 acetonyloxy 3,4 difluoronitrobenzene(Ⅱ).At molar ratio of 2,3,4 tyifluoronitrobenzene:monochloroacetone: potassium iodide=1 00∶1 25∶0 06 and temperature of 55 ℃,the yield of Ⅱwas 70 2% and the purity was 97 5%.The drawback of the old method to use hydrochloric acid and potassium carbonate was avoided .This improved method is suitable for industrial production for its low cost .

Abstract:The 1,3-bis(4-nitrophenoxy)benzene was synthesized from the condensation reaction of resorcinol with 4-nitrochlorobenzene.The 1,3-bis(4-aminophenoxy) benzene was prepared from the reduction reaction of 1,3-bis(4-nitrophenoxy) benzene in ethanol solvent with FeCl3.6H2O and C as catalyst,the hydrazine hydrate as reducing agent,and the reaction progress was tracked and studied by thin-layer chromatography(TLC) method,and the structure of target produce was characterized by melting point apparatus,DSC,FT-IR,1H-NMR spectra and elemental analysis etc.ways.The testing results showed when the mass of 1,3-bis(4-nitrophenoxy) benzene was 14.1 g(40.1 mmol),and the volume fraction of hydrazine hydrate was 20 mL,selected the catalyst FeCl3.6H2O’s mass was 1.00 g,and active carbon’s mass was 2.50 g,and control reaction temperature of added the hydrazine hydrate was 70 ℃,after added finish,the reaction mixture was agitated and maintained at the refluence temperature,the yield of 1,3-bis(4-aminophenoxy) benzene was about 60%(according to the 1,3-bis(4-nitrophenoxy) benzene).

Abstract:(3,4-Dimethoxyphenyl) acetic acid was synthesized by using 3,4-Dimethoxybenzaldehyde as the starting material.Steps included Knoevenagel reaction,potassium borohydride reduction and oxidation under acidic environment.The optimum conditions of the synthesis were got by discussing the reaction mechanism.The purity of the compound was analyzed to be 62.6 % and its structure was confirmed by IR and NMR method.There were many advantages of the synthesis,for the starting material can be easily gained and operation of the reactions was simple and the yield was quite good and avoidance of extremely poisonous reagents.There for,the synthesis could be more fit for industrial production.

Abstract:3-(4-t-Butylphenyl)-1-(4-methoxyphenyl)-2-propen-1-one(1) in yield of 91% was synthesized by condensation reaction of 4-t-butylbenzaldehyde with 4-methoxyacetophenone using sodium methoxide as a catalyst.4-t-Butyl-4′-methoxydibenzoylmethane(2) in overall yield of 81% was prepared by addition reaction of 1 with bromine,and then debromination in presence of sodium methoxide.The structure of 2 was confirmed by UV and 1H NMR.
Two Efficient Methods for the Preparation of 2-Chloro-6-methylbenzoic Acid

Abstract:Two efficient methods for the preparation of 2-chloro-6-methylbenzoic acid were developed: one based on nucleophilic aromatic substitution and the other based on carbonylation. In the first approach, 2-chloro-6-fluorobenzaldehyde was converted to its n-butylimine, then treated with 2 equiv of methylmagnesium chloride in THF to give, after hydrolysis, 2-chloro-6-methylbenzaldehyde. Subsequent oxidation of this compound gave the title compound in 85% overall yield. In the second approach, 3-chloro-2-iodotoluene was efficiently carbonylated in methanol to give methyl 2-chloro-6-methylbenzoate, which after hydrolysis afforded the title compound in 94% yield (84% yield after recrystallization). The carbomethoxylation proceeded smoothly even at a high substrate-to-Pd ratio of 10 000. Both methods do not require isolation of intermediates and are suitable for the preparation of kilogram quantities of 2-chloro-6-methylbenzoic acid.

Abstract:To get an optimal condition for the synthesis of 3-chloro-N-( 4-cyanophenyl) benzamide,3-chloro-N-( 4-cyanophenyl) benzamide was synthesized from 4-nitrobenzonitrile and 3-chlorobenzoic acid by reduction, chloride, amide.The effect of different reaction conditions on product yield was investigated.The target compound could be obtained in 89% by the best condition: 3-chloro-benzoic acid with 4-aminobenzonitrile material ratio of 1∶1.15, dropping temperature 0 ~5 ℃, acid binding agent: N,N-Diisopropylethylamine, in an amount of 2-times amount of amine,reaction for two hours.A synthesis route was established for the synthesis of 3-chloro-N-(4-cyanophenyl) benzamide, the synthetic route is in mild reaction conditions, the process is simple to operate, easy to control.

Abstract:CuI-catalyzed O-arylation of phenols with electron-rich 4-bromoanisole and electron-deficient 3-bromoanisole was described. Electron-deficient 3-bromoanisole showed high reactivity, and electron-rich 4-bromoanisole had low reactivity. O-Arylation of 4-bromoanisole occurred to afford the product by increasing reaction temperature and time. This reaction was applied to the synthesis of 1-chloro-4-(4-methoxyphenoxy)benzene, which was further synthesized to herbicide Clofop.

Abstract:(E)-4-bromo-3-methyl-2-butenoic acid was conveniently synthesized from 3-methyl-2-butenoic acid in 29% overall yield.In this procedure,the title compound was easily obtained by hydrolysis of (E)-trimethylsilyl 4-bromo-3-methyl-2-butenoate,which could be isolated from its mixture of Z-/E-isomer by distillation in vacuo.The isomeric mixture was prepared via the esterification of 3-methyl-2-butenoic acid with chlorotrimethylsilane,followed by bromination with NBS.
