Abstract:The Baylis-Hillman reaction of cyclic enones was greatly accelerated in basic water solution with imidazoles as catalysts, which resulted in short reaction time, high yields, and expanding substrate scopes. Bicarbonate solution was shown to be the optimal reaction medium for the reaction in this study. The apparent "enhanced basicity" of imidazoles accounted for the rate increase in alkaline solution.

Abstract:Catalytic asymmetric 1,3-dipolar cycloaddition reactions of activated isocyanides with various 2 pi dipolarophiles have been intensively studied, affording a wide range of enantioenriched five-membered N-heterocycles. In sharp contrast, the catalytic enantioselective higher-order cycloaddition of activated isocyanides has not been achieved yet. We present here our recent work on the development of an unprecedented silver-catalyzed highly diastereo- and enantioselective [3+3] cycloaddition of activated isocyanides with azomethine imines. This method features high efficiency, good to excellent stereocontrol, wide substrate scope, as well as operational simplicity. It is also noteworthy that the same catalytic system was proved to be suitable for not only the late-stage functionalization of complex bioactive molecules but also the kinetic resolution of racemic azomethine imines.

Abstract:4-(Alkoxycarbonyl)-pent-4-enimidates were regioselectively synthesized via a copper-catalyzed four-component reaction of Baylis-Hillman adducts with terminal alkynes, sulfonyl azides and alcohols. The procedure is concise, general and efficient. The resulting pentenimidates could be further transformed to 3-methylene-2,3-dihydroindene-1-imidates.

Abstract:Hydroxyl ionic liquid (HIL) has been explored as a novel support for Baylis–Hillman catalyst. The HIL-supported catalyst showed a better catalytic activity compared to other IL-immobilized catalyst that has no hydroxyl group attached to the IL scaffold. The hydroxyl group linked on IL played an important role in facilitating efficient catalysis under solvent-free conditions. The corresponding Baylis–Hillman and aza-Baylis–Hillman adducts were obtained in good to excellent yields in all cases examined. The HIL-supported quinuclidine can be readily recovered and reused for six times without significant loss of catalytic activity.

Abstract:The development of simple‐to‐operate methods for the preparation of indolizidine derivatives are of great importance. A mild one‐pot protocol for the [3+2] cycloaddition of isoquinolinium/pyridinium ylides and electron‐deficient alkenes using NaBH3CN as reductant has been developed. The protocol has a broad substrate scope (more than 40 examples) and provides various novel hexahydroindolizidine derivatives in high yields and excellent diastereoselectivities (up to >20/1).

Abstract:A novel phosphine-catalyzed asymmetric aza-Morita–Baylis–Hillman reaction of endocyclic ketimines with vinyl ketone or acrolein has been presented. This approach resulted in densely functionalized adducts with moderate to high yields and enantioselectivities (up to 97% yield, up to 97% ee), with the feature of a C2 tetra -substituted chiral center on a 3-oxindole scaffold. A wide variety of substrates were compatible with this methodology, and diverse enantiomeric products could be obtained through a catalyst switching strategy. A series of transformations further demonstrated the utility of this methodology.

Abstract:Regarding the green chemistry's goals, ionic liquids (ILs) open a way to introduce amazing and efficient media for different reactions. Therefore, in the present study, a feasible protocol for the preparation of β-cyclodextrin/imidazolium based dicationic ionic liquid and its supported on silica gel, [βCD/Im](OTs)<sub>2</sub>-Silica are presented. Ability of this eco-friendly microvessel and host ionic liquid system in the one-pot three-components Hantzsch condensation reaction of arylaldehydes, ethylacetoacetate or dimedone and ammonium acetate are also described. The mild and easy reaction conditions, utilization of a catalyst with high catalytic activity and good reusability, and simple work-up procedure, makes this method as an interesting option for the facile and efficient synthesis of 1,4-dihydropyridine and polyhydroquinoline derivatives.

Abstract:A novel type of highly efficient chiral C2-symmetric bipyridine-N,N’-dioxides catalysts to catalyze Michael addition/Cyclization of 5-aminopyrazoles with α,β-unsaturated 2-acyl imidazoles has been developed, affording the corresponding adducts in 85-97% yield with up to 99% enantioselectivity. Remarkably, this protocol exhibits extraordinary advantages in terms of reactivity and enantioselectivity, given the fact that as low as 2.2 mol % of L1 and 2.0 mol % of Ni(OTf)2 can promote the title reaction on gram scale to afford desired product with excellent enantioselectivity.

Abstract:Inspired by the high affinity of copper with DNA and RNA, a uracil-copper catalytic system was developed to promote ring-opening allylation of cyclopropanols with allylic alcohols under water-tolerant conditions. A new C-OH bond-breaking model can well resolve the trade-off between the need for acidic activators for C(allyl)-OH bond cleavage and the demand for strong basic conditions for generating homoenolates. Therefore, Morita-Baylis-Hillman alcohols, rather than pre-activated versions, could be incorporated directly into dehydrative cross-coupling with cyclopropanols delivering water as the only by-product. A variety of functionalized δ,ε-unsaturated ketones were obtained in good to high yield with high E-selectivity.

Abstract:A highly α-regioselective N-nucleophilic allylic substitution of cyclic MBH alcohols and acetates with imidazole or benzimidazole, in toluene at reflux with an azeotropic distillation, was successfully carried out with no catalysts or additives, affording the corresponding N-substituted imidazole derivatives in good yields. On the other hand, in refluxing toluene or methanol, the aza-Michael addition of imidazole onto acyclic MBH alcohols was performed using DABCO as an additive, leading to the corresponding 1,4-adducts in 70-84% yields.

Abstract:CuI-catalyzed aerobic oxidative synthesis of imidazoheterocycles has been achieved. Four hydrogen atoms were removed in one step. This protocol was compatible with a broad range of functional groups, and it has been also successfully extended to unsaturated ketones, bringing about the formation of alkenyl-substituted imidazoheterocycles, which were difficult to prepare by previous means. Preliminary mechanistic studies indicated that this reaction was most likely to proceed through a catalytic Ortoleva-King reaction. By using this method, the marketed drug Zolimidine could be prepared with 90% yield on a gram scale from commercially available materials.

Abstract:The first catalytic cycloamidination of aminoalkenes with nitriles has been achieved by using rare-earth complexes. This reaction is equivalent to the desired intramolecular hydroamination of alkenylamidines, and allows a new direct access to substituted 2-imidazolines and tetrahydropyrimidines in high yields under operationally simple reaction conditions. Moreover, the methodology is also efficient for synthesis of symmetric and unsymmetric bridged diimidazolines. Compared with the traditional stepwise-mediated synthetic approaches, the present method avoids the use of additives and harsh reaction conditions, and thus leads to a completely different product distribution. Mechanistic data suggest that the reaction involves the initial NH activation by lanthanide complex followed by nitrile insertion into a Ln-N bond to form an amidinate lanthanide intermediate which undergoes the cyclization.

Abstract:A highly efficient and stereoselective synthesis of enantioenriched imidazolidines by rhodium-catalyzed intermolecular [3 + 2] cycloaddition reaction of chiral vinyl aziridines and oxime ethers has been successfully developed. Notably, both aldoximes and ketoximes are suitable substrates to afford the corresponding chiral imidazolidines in high yields with good stereoselectivity. This transformation represents an unprecedented example that utilizes ketimine derivatives as an aza-[2C]-component in cycloadditions of vinyl aziridines.

Abstract:CuI/bis(oxazoline)-catalyzed addition of propiolates and terminal ynones to 1-acylpyridinium salt (generated in situ from reaction of pyridine and methyl chloroformate) affords highly functionalized dihydropyridines with excellent enantioselectivity. It is found that the carbonyl group adjacent to the alkyne moiety is essential for the enantioselectivity of the addition. Short synthesis of indolizidines 167B and 223AB is achieved by employing two addition products.

Abstract:We report herein a general and effective system achieving cyclization of β-trifluoromethyl enones with amidines in the presence of 1,3-diiodo-5,5-dimethylhydantoin (DIH), which affords a range of trifluoromethylated 2-imidazolines in synthetically useful yields with good diastereoselectivities (up to 95% yield, up to 98:2 dr) and good functional group tolerance. Furthermore, the one-pot synthesis of trifluoromethylated imidazoles via sequential cyclization and oxidation is demonstrated. More significantly, the reaction mechanism was verified by ESI-MS studies of possible intermediates, and a reasonable reaction mechanism was proposed.
