Abstract:The development of simple‐to‐operate methods for the preparation of indolizidine derivatives are of great importance. A mild one‐pot protocol for the [3+2] cycloaddition of isoquinolinium/pyridinium ylides and electron‐deficient alkenes using NaBH3CN as reductant has been developed. The protocol has a broad substrate scope (more than 40 examples) and provides various novel hexahydroindolizidine derivatives in high yields and excellent diastereoselectivities (up to >20/1).

Abstract:An efficient synthesis of diversified indolizine derivatives was developed via CuBr-catalyzed reaction of pyridines, methyl ketones and alkenoic acids under solvent free conditions in oxygen atmosphere. This synthesis involves cascade processes of copper-catalyzed bromination of the methyl ketone, 1,3-dipolar cycloaddition of the pyridinium ylide with the alkenoic acid, followed by oxidative decarboxylation and dehydrogenative aromatization of the primary cycloadduct. By this protocol, a wide range of indoliznes with different substitution patterns were selectively prepared in one pot from simple substrates in good to excellent yields.

Abstract:The copper-catalyzed azide-alkyne cycloaddition (CuAAC) reaction is regarded as a prime example of "click chemistry", but the asymmetric click cycloaddition of internal alkynes still remains challenging. A new asymmetric Rh-catalyzed click cycloaddition of N-alkynylindoles with azides was developed, providing atroposelective access to C-N axially chiral triazolyl indoles, a new type of heterobiaryl, with excellent yields and enantioselectivity. This asymmetric approach is efficient, mild, robust and atom-economic, and features very broad substrate scope with easily available Tol-BINAP ligands.

Abstract:Palladium-catalyzed [4+2] annulation of aryl and alkenyl carboxamides with 1,3-dienes via C–H functionalization is developed using air as the terminal oxidant. Abstract:The development and utilization of new dienes and dienophiles for the controlled synthesis of isoquinuclidines is highly appealing. Herein, we describe a novel strategy for diastereoselective synthesis of indoline-fused isoquinuclidines via copper-catalyzed dearomative Diels-Alder reaction of cyclic amidines with indoles. This protocol avoids the use of unstable DHPs and activated alkenes, offering a more efficient and selective approach to synthesize isoquinuclidines.

Abstract:A general protocol for regioselective electrochemical [2+2] cyclization of N-acyl indoles with alkynes was described. This novel method was carried out under mild, chemical oxidant-free, and transition-metal-free conditions, with a broad substrate scope and good functional group tolerance to provide cyclobutene-fused indolines with high regioisomeric ratios and was readily scalable to the gram scale. Mechanistic studies have suggested that the selective reduction of radical cations at the cathode to form a double radical intermediate is the key step in the reaction.

Abstract:Herein, a straightforward and efficient approach to iodinated indolo[1,2-c]quinazoline amines via I2/CHP-promoted cascade annulation of diarylalkynes and isocyanides has been described. This method features excellent regio- and chemoselectivities, good functional group compatibility, as well as mild reaction conditions. Mechanistic studies suggest that CHP is essential to the catalytic cycle of the reaction. The newly formed indolo[1,2-c]quinazoline scaffolds can be further modified, showing potential applications in medicinal chemistry and materials science.

Abstract:A novel and practical procedure for the preparation of 3-unsubstituted indolizines by 1,3-dipolar cycloaddition was developed. The requisite pyridinium N-methylides were generated simply from the corresponding N-(carboxymethyl)pyridinium halides. In the presence of MnO2, electron-deficient alkenes, instead of alkynes or vinyl bromides, were used successfully as dipolarophiles. This general method features cheap reagents, simple workup procedure and gives the products in moderate to high yields (57-92%).

Abstract:A facile and general one-step method is presented for the synthesis of aromatic indolizine compounds 3a–n in moderate to high yields (53–99%) by reaction of the pyridinium N-ylides 1a–e, the quinolinium N-ylide 1f and the isoquinolinium N-ylide 1g with various olefinic dipolarophiles, such as acrylonitrile 2a, methyl acrylate 2b, acrylamide 2c, diethyl maleate 2d and methyl crotonate 2e, respectively, in the presence of a new oxidant TPCD [Copy4(HCrO4)2, tetrapyridinecobalt(II) dichromate] at 90 °C for 2 h in DMF.

Abstract:An unprecedented iridium-catalyzed asymmetric [4 + 3] cycloaddition of racemic 4-indolyl allylic alcohols with azomethine ylides is reported. The ability of acid promoter zinc triflate to perform multiple roles is the key factor for the success of this strategy. This method provides scalable and efficient access to biologically important azepino[3,4,5-cd] indoles in good yields with generally excellent diastereo- and enantioselectivities (up to >20:1 dr and >99% ee). Mild reaction conditions, easily accessible substrates and chiral catalyst, and broad substrate scope highlight the practicality of this methodology.

Abstract:We have developed cobalt-catalyzed, bidentate 2-(1-methylhydrazinyl)pyridine (MHP)-directed C(sp(2))-H alkylation/ annulation of benzoic hydrazides with various alkenes. Notably, diverse cyclopenta[c]isoquinolinones and dihydroisoquinolinones were obtained via this functional group-tolerant protocol. The reaction can be performed on a gram scale while maintaining an excellent yield, and the directing group can be removed efficiently under mild conditions. Furthermore, density-functional theory (DFT) calculations provide an incisive understanding of the observed regioselectivities for different olefins.
