The structure of this new arabinoside was elucidated on the basis of extensive NMR and HRMS spectroscopy. Abstract:Objective: To study the chemical constituents in the air-dried part of Conyza blinii. Method: The constituent isolation was done by solvent-extraction together with column chromatography. Several methods, such as IR, 1HNMR, 13CNMR and MS, were used to determine the structures of the isolated constituents. Result: Three compounds were isolated from the air-dried part of Conyza blinii and identified as friedelinol, n-triacontanol and daucosterol on the basis of physical and chemical properties and spectroscopic analysis. Conclusion: These compounds have not been discovered previously in this plant.

Abstract:A new coniferol derivative, named as tripolinolate A (1), and 11 known compounds (2–12) were isolated from whole plants of Tripolium vulgare Nees. The structure of this new compound was determined as 4-(2S-methylbutyryl)-9-acetyl-coniferol based on its NMR and HRESIMS spectral analyses. A simple and efficient method was designed to prepare tripolinolate A and its 19 analogs including nine new chemical entities for bioactive assay. Tripolinolate A and its analog 4,9-diacetyl-coniferol were found to be the two most active compounds that significantly inhibited the proliferation of different cancer cell lines with IC50 values ranging from 0.36 to 12.9μM and induced apoptosis in tumor cells. Structure–activity relationship analysis suggested that the molecular size of acyl moieties at C-4 and C-9 position might have an effect on the activity of this type of coniferol derivatives.

Abstract:Three new bisdesmosidic saponins named conyzasaponins A, B, and C (1-3) and one new monodesmosidic saponin, conyzasaponin G (4), were isolated from the aerial parts of Conyza blinii. Their structures were elucidated on the basis of extensive NMR (DEPT, DQF-COSY, HOHAHA, HMQC, HMBC, and NOESY) and MS studies. Compounds 1-3 share a common prosapogenin, bayogenin 3-O-beta-D-xylopyranosyl-(1-->3)-beta-D-glucopyranoside, which is identical with conyzasaponin G (4), and differ in the structures of the ester-linked sugar moieties at C-28. Conyzasaponin A (1) is the 28-O-beta-D-apiofuranosyl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester, conyzasaponin B (2), the 28-O-beta-D-apiofurano- syl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-[alpha-L-arabinopyranosyl-(1-->3)]-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester, and conyzasaponin C (3), the 28-O-alpha-L-rhamnopyranosyl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-[beta-D-apiofuranosyl-(1-->3)]-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester of the prosapogenin, respectively.

Abstract:Nine new triterpenoid saponins, conyzasaponins I-Q (1-9), were isolated from the aerial parts of Conyza blinii. Their structures were established on the basis of extensive 1D and 2D NMR spectra as well as by chemical degradations. Among these compounds, conyzasaponins M-O (5-7) share a common pentasaccharide unit attached to C-28 of the aglycon, 28-O-beta-D-apiofuranosyl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-[beta-D-apiofuranosyl-(1-->3)]-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester, which contains two apiofuranosyl residues. To the best of our knowledge, they are among the few examples of natural products possessing two apiofuranose units in a single sugar chain.

Abstract:Four new apiose-containing triterpenoid saponins, conyzasaponins D, E, F, and H, were isolated from the aerial parts of Conyza blinii. They are the first examples of triterpenoid saponins containing one apiose unit whose C-2 is substituted by a galactose. Conyzasaponins D and F have two apiose units, a sugar that has been rarely observed in natural products research. Their structures were established on the basis of extensive NMR studies and chemical degradation. Conyzasaponins D and F exhibited moderate cytotoxicity against HL-60 cell lines.

Abstract:Three unusual sesterterpenoids featuring unprecedented rearranged colquhounane (C25) and tetranorcolquhounane (C21) frameworks, colquhounoids E (1) and F (3) and norcolquhounoid F (2), were isolated from a Lamiaceae medicinal plant Colquhounia coccinea var. mollis. Their structures were elucidated by spectroscopic analysis and quantum chemical calculations. A biomimetic inspired regioselective cyclopropane cleavage was achieved under acidic conditions. The immunosuppressive activities of these new sesterterpenoids were also evaluated.

Abstract:Five new dibenzocyclooctadiene lignans, schisphenlignans A-E (1-5), together with eight known ones, were isolated from the stems of Schisandra sphenanthera. The structures of 1-5 were elucidated based on the analysis of their NMR, MS and circular dichroism (CD) spectra. Some isolates were tested for their acute activities on insulin sensitivity in 3T3-L1 differentiated adipocytes, but none of them showed significant bioactivity with 10 mu M administration of the tested compounds.

Abstract:Conyza blinii Le'vl is a medicinal herb used for the treatment of inflammation in Chinese folk medicine. Its major bioactive constituents are triterpene saponins, most of which contain 6-8 sugar residues. In this report, electrospray ionization tandem mass spectrometry fragmentation behaviors of bisdesmosidic triterpene saponins (conyzasaponin A, B, and C) were studied in both positive and negative ion modes with an ion-trap mass spectrometer. In full scan mass spectrometry, these saponins gave predominant [M-H](-) and [M+Na](+) ions, which determined the molecular weights. In tandem mass spectrometry (MSn, n = 2-4), the [M-H](-) and [M-Na](+) ions yielded fragments [Y-0 alpha-H](-) and [B-alpha+Na](+), which were diagnostic for the structures of the triterpene skeleton and sugar chains. The structural elucidation was approved by accurate mass data using IT-TOF-MS. An interpretation guideline based on MSn (n = 2-4) diagnostic ions was proposed in order to elucidate the chemical structures of unknown triterpene saponins in C. blinii extract. The saponins in C. blinii were separated by liquid chromatography with a methanol/acetonitrile/water solvent system, and then analyzed by ion-trap and IT-TOF mass spectrometers. Based on the interpretation guideline, a total of 35 triterpenoid saponins were tentatively identified. Among them, 15 saponins had been previously reported, and the other 20 saponins were reported from Conyza species for the first time. This study indicates that LC/MS is a powerful technology for the rapid characterization of complicated saponins in herbal extracts. Copyright (C) 2010 John Wiley & Sons, Ltd.

Abstract:Objective To study the chemical constituents in the whole herb of Conyza blinii and seek its antiulcerogenic principles. MethodsCompounds were separated and purified by solvent extraction, re-crystallization, and chromatography on silica gel and polyamide column. Their structures were elucidated on the basis of 1H-NMR, 13C-NMR, and MS spectral data and co-TLC with authentic samples. Pylorus-ligated rat model was used for the evaluation of antiulcerogenic activity. ResultsEight compounds were identified: 5, 8, 3′, 4′-tetrahydroxy-7-methoxy flavone (Ⅰ), 5, 8, 4′-trihydroxy-7, 3′-dimethoxy flavone (Ⅱ), β-sitosterol (Ⅲ), spinasterol (Ⅳ)、n-hexacosanic acid (Ⅴ), 5, 7-dihydroxy-3, 3′, 4′-trimethoxy flavone (Ⅵ), 5, 7-dihydroxy-3, 8, 4′-trimethoxy flavone (Ⅶ), blinin (Ⅷ). Blinin could significantly narrow the ulcer area and lower the level of gastric mucous MDA. ConclusionCompounds Ⅰ-Ⅲ and Ⅴ-Ⅶ are obtained from the title plant for the first time. Blinin is found to possess significant antiulcer property.

Abstract:The total synthesis of (+)-cleomeolide (1) has been accomplished. The key construction elements of this cembranoid lactone were (i) improved conversion of optically pure Wieland-Miescher ketone into dienol ether 12 and oxidative cleavage of the latter to aldehydo ester 13; (ii) avoidance of complications arising from steric blockade of C-15 for introduction of the methylene group at that site; (iii) exploitation of an intramolecular Wadsworth-Emmons cyclization for macrocyclic ring construction; (iv) modulation of the conformation adopted by the medium ring by diastereofacial control of epoxidation of the C-3/C-4 double bond; and (v) intramolecular cyclization of the epoxy acid derived from 27 by nucleophilic capture at the more substituted oxiranyl carbon to deliver the target molecule. The deep-seated topographical change that accompanies the formation of 29 projects the macrocyclic ring quasi-axially from the methylenecyclohexane subunit in the manner known by X-ray crystallography to be adopted by the target compound. The intramolecular cyclization to set the bridgehead double bond was thereby facilitated.

Abstract:Conosiligins A–D (1–4), four ring-rearranged sesquiterpenoids, were isolated from cultures of the basidiomycete Conocybe siliginea. Their structures and absolute configurations were determined by detailed spectroscopic analyses and equivalent circulating density (ECD) calculations. Compounds 1 and 2 possess a 5/8-fused ring system, while 3 has a 5/6-fused backbone conjugated with a γ-lactone. Compound 4 is a 5,6-seco tremulane derivative with the loss of a skeletal carbon, featuring a tetracyclic system involving a pyranone moiety. Abstract:Two new clerodane diterpenoids ( 1 and 2 ), a new pyran-2-one derivative ( 3 ), along with five known compounds ( 4 ‒ 8 ), were isolated from Croton crassifolius . Notably, crassifolin X ( 1 ) is a novel clerodane diterpenoid, characterized with a peculiar δ -lactone core being formed between C-1 and C-4. Their structures, including absolute configurations, were established on the basis of spectroscopic methods (UV, IR, HRESIMS and NMR), and circular dichroism experiments. In addition, all compounds were evaluated for their anti-neuroinflammatory activities based on the expression of TNF- α and IL-6 levels on LPS-induced BV2 cells, and compounds 1 ‒ 3 and 5 showed potential anti-neuroinflammatory activity.

Abstract:From the whole plant of Senecio tsoongianus, two pairs of epimers, tsoongianolides A (1) and B (2), and tsoongianolides C (3) and D (4), and two new sesquiterpene lactones, tsoongianolides E (5) and F (6), all of which possess the eremophilane skeleton, were isolated. Two known eremophilane dimers (7) and (8) were also obtained from this species. Their absolute structures were elucidated by 1D- and 2D-NMR techniques and by X-ray diffraction studies, along with chemical evidence.
