The structure of this new arabinoside was elucidated on the basis of extensive NMR and HRMS spectroscopy. Abstract:Three new bisdesmosidic saponins named conyzasaponins A, B, and C (1-3) and one new monodesmosidic saponin, conyzasaponin G (4), were isolated from the aerial parts of Conyza blinii. Their structures were elucidated on the basis of extensive NMR (DEPT, DQF-COSY, HOHAHA, HMQC, HMBC, and NOESY) and MS studies. Compounds 1-3 share a common prosapogenin, bayogenin 3-O-beta-D-xylopyranosyl-(1-->3)-beta-D-glucopyranoside, which is identical with conyzasaponin G (4), and differ in the structures of the ester-linked sugar moieties at C-28. Conyzasaponin A (1) is the 28-O-beta-D-apiofuranosyl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester, conyzasaponin B (2), the 28-O-beta-D-apiofurano- syl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-[alpha-L-arabinopyranosyl-(1-->3)]-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester, and conyzasaponin C (3), the 28-O-alpha-L-rhamnopyranosyl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-[beta-D-apiofuranosyl-(1-->3)]-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester of the prosapogenin, respectively.

Abstract:Four new apiose-containing triterpenoid saponins, conyzasaponins D, E, F, and H, were isolated from the aerial parts of Conyza blinii. They are the first examples of triterpenoid saponins containing one apiose unit whose C-2 is substituted by a galactose. Conyzasaponins D and F have two apiose units, a sugar that has been rarely observed in natural products research. Their structures were established on the basis of extensive NMR studies and chemical degradation. Conyzasaponins D and F exhibited moderate cytotoxicity against HL-60 cell lines.

Abstract:Nine new triterpenoid saponins, conyzasaponins I-Q (1-9), were isolated from the aerial parts of Conyza blinii. Their structures were established on the basis of extensive 1D and 2D NMR spectra as well as by chemical degradations. Among these compounds, conyzasaponins M-O (5-7) share a common pentasaccharide unit attached to C-28 of the aglycon, 28-O-beta-D-apiofuranosyl-(1-->3)-beta-D-xylopyranosyl-(1-->4)-[beta-D-apiofuranosyl-(1-->3)]-alpha-L-rhamnopyranosyl-(1-->2)-alpha-L-arabinopyranosyl ester, which contains two apiofuranosyl residues. To the best of our knowledge, they are among the few examples of natural products possessing two apiofuranose units in a single sugar chain.

Abstract:Objective: To study the chemical constituents in the air-dried part of Conyza blinii. Method: The constituent isolation was done by solvent-extraction together with column chromatography. Several methods, such as IR, 1HNMR, 13CNMR and MS, were used to determine the structures of the isolated constituents. Result: Three compounds were isolated from the air-dried part of Conyza blinii and identified as friedelinol, n-triacontanol and daucosterol on the basis of physical and chemical properties and spectroscopic analysis. Conclusion: These compounds have not been discovered previously in this plant.

Abstract:Five new dibenzocyclooctadiene lignans, schisphenlignans A-E (1-5), together with eight known ones, were isolated from the stems of Schisandra sphenanthera. The structures of 1-5 were elucidated based on the analysis of their NMR, MS and circular dichroism (CD) spectra. Some isolates were tested for their acute activities on insulin sensitivity in 3T3-L1 differentiated adipocytes, but none of them showed significant bioactivity with 10 mu M administration of the tested compounds.

Abstract:Fifteen new labdane-type diterpenoids, sublyratins A-O (1-15), along with four known analogues (16-19) were isolated from the aerial parts of Croton sublyratus. Their structural assignments were challenging due to the stereoisomeric features evident and were achieved by analyzing comprehensively the spectroscopic data and electronic circular dichroism spectra and using X-ray crystallographic analysis. Compounds 9 and 16-18 displayed cytotoxic activity against the HL-60 cell line with IC50 values of 1.5-2.8 μM.

Abstract:Conyza blinii Le'vl is a medicinal herb used for the treatment of inflammation in Chinese folk medicine. Its major bioactive constituents are triterpene saponins, most of which contain 6-8 sugar residues. In this report, electrospray ionization tandem mass spectrometry fragmentation behaviors of bisdesmosidic triterpene saponins (conyzasaponin A, B, and C) were studied in both positive and negative ion modes with an ion-trap mass spectrometer. In full scan mass spectrometry, these saponins gave predominant [M-H](-) and [M+Na](+) ions, which determined the molecular weights. In tandem mass spectrometry (MSn, n = 2-4), the [M-H](-) and [M-Na](+) ions yielded fragments [Y-0 alpha-H](-) and [B-alpha+Na](+), which were diagnostic for the structures of the triterpene skeleton and sugar chains. The structural elucidation was approved by accurate mass data using IT-TOF-MS. An interpretation guideline based on MSn (n = 2-4) diagnostic ions was proposed in order to elucidate the chemical structures of unknown triterpene saponins in C. blinii extract. The saponins in C. blinii were separated by liquid chromatography with a methanol/acetonitrile/water solvent system, and then analyzed by ion-trap and IT-TOF mass spectrometers. Based on the interpretation guideline, a total of 35 triterpenoid saponins were tentatively identified. Among them, 15 saponins had been previously reported, and the other 20 saponins were reported from Conyza species for the first time. This study indicates that LC/MS is a powerful technology for the rapid characterization of complicated saponins in herbal extracts. Copyright (C) 2010 John Wiley & Sons, Ltd.

Abstract:Two new 18-norschiartane-type schinortriterpenoids, namely wuwezidilactones Q (1) and R (2) were isolated from the stems of Schisandra lancifolia. Their structures were determined on the basis of extensive spectroscopic analysis. The absolute configurations of the new compounds were further determined by ROESY and an empirical comparison of their experimental ECD spectra with literature.

Their chemical structures were elucidated on the basis of MS, NMR spectroscopic analyses coupled with chemical degradation. The cytotoxic activities of two isolated compounds were evaluated against two human gastric cancer cell lines BGC-823 and AGS using MTT assay.

Abstract:Two new bislabdane-type diterpenoids lanceolatin (1), lancelatol (2) and three new diterpenoids 15-nor-14-oxo-8(17),12-labdanien-14,18-dionic acid (3), 8(17),12,14-labdantrien-18-oic acid (4) and 8(17),12,14-labdantrien-18-ol (5), together with 7 known diterpenoids were isolated from the roots of Cunninghamia lanceolata. Their structures were determined by spectroscopic and chemical methods. All of these compounds were isolated from the species for the first time. One of the isolated compounds showed a significant inhibition effect on mouse hind-paw edema induced by carrageenan.

Abstract:Three unusual sesterterpenoids featuring unprecedented rearranged colquhounane (C25) and tetranorcolquhounane (C21) frameworks, colquhounoids E (1) and F (3) and norcolquhounoid F (2), were isolated from a Lamiaceae medicinal plant Colquhounia coccinea var. mollis. Their structures were elucidated by spectroscopic analysis and quantum chemical calculations. A biomimetic inspired regioselective cyclopropane cleavage was achieved under acidic conditions. The immunosuppressive activities of these new sesterterpenoids were also evaluated.

Abstract:Two new lignans, named as 6-hydroxyhinokinin and 6-hydroxyhinokinin-6- O-β-D-glucopyranoside, together with five known compounds: elemol, oplodiol, catechin, β-sitosterol and daucosterol were isolated from Kadsura heteroclita. The structures of these compounds were characterized by extensive 1D and 2D NMR spectral experiments, chemical analysis and comparison with literature data. The absolute structures of the novel lignans were elucidated as 8 S, 8′ S, based on their optical rotations.

Abstract:Objective To study the chemical constituents in the whole herb of Conyza blinii and seek its antiulcerogenic principles. MethodsCompounds were separated and purified by solvent extraction, re-crystallization, and chromatography on silica gel and polyamide column. Their structures were elucidated on the basis of 1H-NMR, 13C-NMR, and MS spectral data and co-TLC with authentic samples. Pylorus-ligated rat model was used for the evaluation of antiulcerogenic activity. ResultsEight compounds were identified: 5, 8, 3′, 4′-tetrahydroxy-7-methoxy flavone (Ⅰ), 5, 8, 4′-trihydroxy-7, 3′-dimethoxy flavone (Ⅱ), β-sitosterol (Ⅲ), spinasterol (Ⅳ)、n-hexacosanic acid (Ⅴ), 5, 7-dihydroxy-3, 3′, 4′-trimethoxy flavone (Ⅵ), 5, 7-dihydroxy-3, 8, 4′-trimethoxy flavone (Ⅶ), blinin (Ⅷ). Blinin could significantly narrow the ulcer area and lower the level of gastric mucous MDA. ConclusionCompounds Ⅰ-Ⅲ and Ⅴ-Ⅶ are obtained from the title plant for the first time. Blinin is found to possess significant antiulcer property.

Abstract:Two new lappaconitine‐type C18‐diterpenoid alkaloids, named as leucostosines C (1) and D (2), together with six known compounds (3‐8), were isolated from the roots of Aconitum leucostomum Worosch. Their structures were elucidated by various spectroscopic analyses, including IR, HR‐ESI‐MS, NMR spectra and X‐ray experiments. Leucostosine C is the first diterpenoid alkaloid bearing the 7‐amino group. The isolated compounds were tested for the acetylcholinesterase (AChE) inhibitory effect and neuroprotective activity, none of them showed significant activities.
