Abstract:Propolis is a kind of bee products with various biological activities and health functions.The antihyperglycemic effect is one of biological activities of propolis.In this article,the research progress on the antihyperglycemic effect of propolis as well as its main flavonoids and phenolic acid were summarized.The molecular mechanisms,including IRSPI3K pathway,PPARs transcription factors,AMPK pathway and antioxidant,were also summarized aiming to provide references for the future research of biological activities and further applications of propolis.

Abstract:Propolis is an adhesive substance mixed with plant resins collected by honeybees (Apis mellifera) and secretions from their mandibular gland and wax gland, with wide pharmacological activity and healthcare functions. Its antioxidant activity has long been regarded as one of the most important biological activities of propolis. This article summarizes studies on the antioxidant activity of propolis extracts from different geographic origins and with different extraction methods, as well as several important monomer active ingredients in propolis, and concludes the potential molecular mechanism of antioxidant activity of propolis and its monomer ingredients, with the aim of providing ideas for further studies on pharmacological activity of propolis, as well as reference for in-depth development of propolis products.
Biological Activities of Chinese Propolis and Brazilian Propolis on Streptozotocin-Induced Type 1 Diabetes Mellitus in Rats

Abstract:Propolis is a bee-collected natural product and has been proven to have various bioactivities. This study tested the effects of Chinese propolis and Brazilian propolis on streptozotocin-induced type 1 diabetes mellitus in Sprague-Dawley rats. The results showed that Chinese propolis and Brazilian propolis significantly inhibited body weight loss and blood glucose increase in diabetic rats. In addition, Chinese propolis-treated rats showed an 8.4% reduction of glycated hemoglobin levels compared with untreated diabetic rats. Measurement of blood lipid metabolism showed dyslipidemia in diabetic rats and Chinese propolis helped to reduce total cholesterol level by 16.6%. Moreover, oxidative stress in blood, liver and kidney was improved to various degrees by both Chinese propolis and Brazilian propolis. An apparent reduction in levels of alanine transaminase, aspartate transaminase, blood urea nitrogen and urine microalbuminuria-excretion rate demonstrated the beneficial effects of propolis in hepatorenal function. All these results suggested that Chinese propolis and Brazilian propolis can alleviate symptoms of diabetes mellitus in rats and these effects may partially be due to their antioxidant ability.

Propolis modulates the gut microbiota and improves the intestinal mucosal barrier function in diabetic rats

Abstract:Objective: Diabetes mellitus is associated with gut microbiota disturbance and intestinal mucosal injuries. This study investigated the influence of propolis on the gut microbiota and intestinal mucosa in rats with diabetes. Methods: Sprague-Dawley (SD) rats were randomly assigned to the control group, model group, and three propolis groups (supplemented with 80, 160, and 240 mg/kg·bw propolis, respectively). A high-fat diet combined with a streptozotocin (STZ) abdominal injection were used to induce diabetes in the rats. After 4 weeks, the intestinal histopathological analysis of the ileum was observed by transmission electron microscopy. The fasting blood glucose (FBG), plasma insulin, glucose tolerance (OGTT) and glycosylated hemoglobin (HbA1c) levels were measured. The expression of tight junction (TJ) proteins in the ileum was measured using western blotting. The molecular ecology of the fecal gut microbiota was analyzed by 16S rDNA high-throughput sequencing. The contents of the short-chain fatty acids (SCFAs) in feces were measured using high-performance liquid chromatography (HPLC). Results: After propolis treatment, compared to the model group, FBG and HbA1c levels declined, while the glucose tolerance and insulin sensitivity index (ISI) increased. The levels of TJ proteins in the ileum increased in the propolis groups. The tight junctions and gap junctions of the intestinal epithelium were also improved in the propolis groups. The contents of the feces acetic acid, propionic acid and butyrate were increased in the propolis groups. 16S rDNA high-throughput sequencing revealed that the composition of the gut microbiota of rats in the propolis supplement group was significantly improved. Conclusions: Compared to the model group, propolis exerted hypoglycemic effects in diabetic rats, and it repaired intestinal mucosal damage, benefited the communities of the gut microbiota and increased SCFA levels in diabetic rats.

Abstract:Propolis is a strong anti-inflammatory agent and indicated improvement of various inflammations.In this article we reviewed the effect of propolis on inflammation and proinflammatory mediators.The possible action mechanism and active composition of propolis were analyzed.In addition,the theory about combination of anti-inflammatory drug and propolis showed synergic effect on inflammation gives out the suggestion to exploitation of anti-inflammatory drugs.

Abstract:This study aims to identification of phytochemical profile of Apis mellifera propolis and explore potential of its anti‐diabetic activity through inhibition on α‐amylase (α‐AE), α‐glucosidase(α‐GE) and finally identified the novel antidiabetic compounds from propolis. Apis mellifera propolis extract (AMPE) exhibited elevated polyphenol 33.26 ± 0.17 (mg GAE/g) and flavonoid (15.45 ± 0.13 mg RE/g), while its also indicated moderate strong antioxidant activity (EC50 793.09 ±1.94 μg/ml). This study found that AMPE displayed promising α‐AE and α‐GE inhibition through in vitro study. Based on LC‐MS/MS screening, 18 unique AMPE compounds were identified, majorly belonging to anthraquinone and flavonoid compounds. In silico study determined that 8 compounds of AMPE compounds exhibited strong binding to α‐AE, interacting to catalytic residue of ASP197. Moreover, 2 compounds exhibit potential inhibition of α‐AG, by interacting to crucial amino acids of ARG315, ASP352, and ASP69. Finally, we suggested 2,7‐Dihydroxy‐1‐(p‐hydroxybenzyl)‐4‐methoxy‐9,10‐dihydrophenanthrene and 3(3‐(3,4‐Dihydroxybenzyl)‐7‐hydroxychroman‐4‐one as novel inhibitors of α‐AE and α‐GE. Notably, these compounds were initially discovered in Apis mellifera propolis, and molecular dynamic analysis confirmed their stable binding with both enzymes over 100 ns simulations. In vivo acute toxicity test reveals AMPE as a practically non‐toxic product with LD50 value of 16050mg/kg.

Abstract:The present study investigates the encapsulated propolis on blood glycemic control, lipid metabolism, and insulin resistance in type 2 diabetes mellitus (T2DM) rats. The animal characteristics and biological assays of body weight, fasting blood glucose (FBG), fasting serum insulin (FINS), insulin act index (IAI), triglycerides (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were measured and euglycemic hyperinsulinemic glucose clamp technique were used to determine these effects. Our findings show that oral administration of encapsulated propolis can significantly inhibit the increasing of FBG and TG in T2DM rats and can improve IAI and M value in euglycemic hyperinsulinemic clamp experiment. There was no significant effects on body weight, TC, HDL-C, and LDL-C in T2DM rats treated with encapsulated propolis. In conclusion, the results indicate that encapsulated propolis can control blood glucose, modulate lipid metabolism, and improve the insulin sensitivity in T2DM rats.
Effects of Propolis on Diabetes Mellitus in SD Rats

Abstract:The effects of ethanol extracted propolis (EEP) and water soluble derivative of propolis (WSP) on diabetes mellitus in SD rats were determined and compared. The results showed that both WSP and EEP decreased levels of fasting blood glucose (FBG), glycosylated hemoglobin (HBAlc), fructosamine (FRU), triglyceride (TG), total cholesterol (TC), creatinine (CREA), blood urea nitrogen (BUN), uric acid (UA), malodialdehyde (MDA), total protein (TP) and albumin (ALB), and decreased the ratio of kidney to body weight in rats with diabetes mellitus. The results indicate that both EEP and WSP improved metabolism of glucose, fat and protein and afforded renal protection for rats with diabetes mellitus.

Abstract:Being abundant in many bioactive substances,propolis has extensive pharmacological activities and health-care functions. Among them, the immunomodulatory activity is the foundation of its multiple biological functions, and more and more people are interested in it.This article compiles data from the related research at home and abroad, focusing on the immunomodulatory activity of propolis and its two important monomer composition-CAPE and ArtepillinC; mainly discussed the immunomodulatory activity on lymphocyte of Brazilian green propolis, and put forward the problems existing in the research, in order to provide a basis for illuminating the mechanism of the immunomodulatory activity and establishing the chemical standardization of propolis.

Abstract:Propolis, a resinous substance produced by honeybees from various plant sources, has been used for thousands of years in traditional medicine for several purposes all over the world. The precise composition of propolis varies according to plant source, seasons harvesting, geography, type of bee flora, climate changes, and honeybee species at the site of collection. This apiary product has broad clinical applications such as antioxidant, anti-inflammatory, antimicrobial, anticancer, analgesic, antidepressant, and anxiolytic as well asimmunomodulatory effects. It is also well known from traditional uses in treating purulent disorders, improving the wound healing, and alleviating many of the related discomforts. Even if its use was already widespread since ancient times, after the First and Second World War, it has grown even more as well as the studies to identify its chemical and pharmacological features, allowing to discriminate the qualities of propolis in terms of the chemical profile and relative biological activity based on the geographic place of origin. Recently, several in vitro and in vivo studies have been carried out and new insights into the pharmaceutical prospects of this bee product in the management of different disorders, have been highlighted. Specifically, the available literature confirms the efficacy of propolis and its bioactive compounds in the reduction of cancer progression, inhibition of bacterial and viral infections as well as mitigation of parasitic-related symptoms, paving the way to the use of propolis as an alternative approach to improve the human health. However, a more conscious use of propolis in terms of standardized extracts as well as new clinical studies are needed to substantiate these health claims.

Abstract:Ethnopharmacological relevance: Propolis is a traditional natural medicine with various activities such as antioxidant and anti-inflammatory, immunomodulatory, anti-tumour, gastroenteritis treatment and prevention, anti-microbial and parasitic, as well as glucose regulation and anti-diabetes, and is expected to be an anti-diabetic candidate with few side effects, but the mechanism of action of propolis on type 2 diabetes mellitus (T2DM) has not been fully elucidated. Aim of the study: The purpose of this study was to investigate the mechanism of the effect of ethanol extract of propolis (EEP) on the regulation of blood glucose in T2DM mice. Materials and methods: We studied the possible mechanism of EEP on T2DM using an animal model of T2DM induced by a combination of a high-fat diet and intraperitoneal injection of streptozotocin (STZ). The experiment was divided into four groups, namely, the normal group (HC), model group (T2DM), EEP and metformin group (MET). Biochemical indexes and cytokines were measured, and the differences of metabolites in the serum were compared by 1H-NMR. In addition, the diversity of intestinal flora in feces was studied by 16S rDNA amplicon sequencing. Results: The results showed that following treatment with EEP and MET, the weight-loss trend of mice was alleviated, and the fasting blood glucose, insulin secretion level, insulin resistance index, C peptide level and oral glucose tolerance level decreased, whereas the insulin sensitivity index increased, thereby EEP effectively alleviated the occurrence of T2DM and insulin resistance. Compared with the T2DM group, the concentrations of pro-inflammatory cytokines interleukin-1 beta (IL-1β), interleukin-6 (IL-6) and tumor necrosis factor-alpha (TNF-α) decreased significantly in EEP and MET groups, whereas the concentrations of anti-inflammatory cytokine interleukin-10 (IL-10) increased significantly. Metabolomics results revealed that EEP and MET regulate carbohydrate metabolism and restore amino acid and lipid metabolism. Correlation analysis of intestinal flora in mouse feces showed that compared with the HC group, harmful bacteria such as Bilophila, Eubacterium_ventriosum_group, Mucispirillum and Desulfovibrio were found in the T2DM group, whereas the abundance of beneficial bacteria such as Lactobacillus was significantly reduced. Parabacteroides, Akkermansia, Leuconostoc, and Alloprevotella were abundantly present in the EEP group; however, the MET group showed an increase in the genus Parasutterella, which could regulate energy metabolism and insulin sensitivity. Conclusions: The results showed that EEP and MET reduce fasting blood glucose in T2DM mice, followed by alleviating insulin resistance, improving the inflammatory reaction of mice, regulating the metabolism of mice, and affecting the steady state of gut microbiota. However, the overall therapeutic effect of EEP is better than that of MET.

Abstract:The antioxidant activities, polyphenol acid and flavonoid contents were discussed in this article, and studied the relationship between polyphenol acid and an- tioxidant activity. Furthermore, measured the main chemi- cal constituents of propolis collected in various geo- graphic origins. The results showed that antioxidant activ- ities of propolis from different areas were different, and the content of polyphenol acid and flavonoid were also different, and there existed some positive correlation be- tween polyphenol and antioxidant activities. The main chemical constituents of propolis of various areas had great significant different, which indicated that propolis had complex chemical constituents, and antioxidant activi- ties of propolis of various areas were the results of syn- ergistic effect of multiple constituents.
Inhibitory properties of aqueous ethanol extracts of propolis on alpha-glucosidase.

Abstract:The objective of the present study was to evaluate the inhibitory properties of various extracts of propolis on alpha-glucosidase from baker's yeast and mammalian intestine. Inhibitory activities of aqueous ethanol extracts of propolis were determined by using 4-nitrophenyl-D-glucopyranoside, sucrose and maltose as substrates, and acarbose as a positive reference. All extracts were significantly effective in inhibiting α-glucosidase from baker's yeast and rat intestinal sucrase in comparison with acarbose (P < 0.05). The 75% ethanol extracts of propolis (75% EEP) showed the highest inhibitory effect on α-glucosidase and sucrase and were a noncompetitive inhibition mode. 50% EEP, 95%, EEP and 100% EEP exhibited a mixed inhibition mode, while water extracts of propolis (WEP) and 25% EEP demonstrated a competitive inhibition mode. Furthermore, WEP presented the highest inhibitory activity against maltase. These results suggest that aqueous ethanol extracts of propolis may be used as nutraceuticals for the regulation of postprandial hyperglycemia.
