Abstract:A new steroidal glycoside (1), (25R)-14α, 17α-hydroxyspirost-5-en-3β-yl 3-O-α-L-rhamnpyranosyl-(1 → 2)-β-D-glucopyranosyl-(1 → 3)-β-D-glucopyranoside, together with three known steroidal glycosides, (25R)-3β-hydroxyspirost-5-en-1β-yl-3-O-α-L-rhamnopyranosyl-(1 → 2)-O-β-D-xylopyranosyl-(1 → 3)-α-L-arabinopyranoside (2), Cixi-ophiopogon B (3) and Cixi-ophiopogon A (4), were obtained from the tuberous roots of Ophiopogon japonicus (Liliaceae). Compound 2 was isolated from the Ophiopogon genus for the first time. Their structures were identified on the basis of extensive mass and nuclear magnetic resonance spectroscopic analysis.

Abstract:Three new triterpenoid derivatives, named (15 alpha)-15-hydroxysoyasapogenol B (1), (7 beta,15 alpha)-7,15-dihydroxysoyasapogenol B (2), and (7 beta)-7,29-dihydroxysoyasapogenol B (3), were isolated from cultures of the plant endophytic fungus Pestalotiopsis clavispora. Their structures and relative configurations were elucidated by extensive spectroscopic analysis and X-ray crystallography.

Abstract:Abstract: A new selaginellin derivative named as selaginellin S (1) was isolated from the whole plants of Selaginella pulvinata (Hook. et Grev.) Maxim. (Selaginellaceae), together with a known one (selaginellin M, 2). Compounds 1 and 2 were separated and purified by silica gel and Sephadex LH-20 column chromatography. Their structures were determined on the basis of extensive spectroscopic analysis including IR, MS, 1D and 2D NMR experiments, as well as ECD calculations. Compound 1 is a key intermidiant in the biosynthesis pathway of selaginellins. Compound 2 is first reported in this plant.

Abstract:A new neolignan, (–)-(7 R ,8 S ,7′ R ,8′ S ,8′′ S )-4′-hydroxy-3′,3′′,5′′-trimethoxy-4′′-O- β -D-glucopyranosyl-7,9,7′,9′-diepoxy-3,4,8′′,9′′-bisoxy-8,8′-sesquineolignan ( 1 ), and five known neolignan derivatives were isolated from an ethanolic extract of Selaginella trichoclada Alston. The chemical structure of these compounds was identified by extensive spectroscopic analyses (UV-vis, 1D/2D NMR, CD, and MS). Furthermore, the molecular docking results indicate that compound 1 may act as an anti-inflammatory inhibitor.

Abstract:One new 18-oxygenated polyhydroxy steroid, (24S)-ergostan-3β,5α,6β,18,25-pentaol 18,25-diacetate (1), along with five known analogues (2-6), was isolated from a soft coral Sarcophyton sp. collected from the South China Sea. The structure of the new compound (1) was elucidated by 1D NMR, 2D NMR and mass spectrometry. The absolute configuration of 1 was determined by the application of the modified Mosher's method. The isolated compounds were assayed for their cytotoxic and antimicrobial activities.

Abstract:Five selaginellin derivatives (1 and 3-6) including a new one, selaginellin T (1), and a new triarylbenzophenone analog, selagibenzophenone A (2), were isolated from the whole plants of Selaginella pulvinata. Their structures were determined by 1D- and 2D-NMR and HR-ESI-MS data. Selagibenzophenone A (2) is the first example of naturally occurring triarylbenzophenone. The results of the phosphodiesterase-4 (PDE4) inhibitory screening assays showed that compounds 1-6 exhibited potent activities with the IC50 values in the range of 1.04-9.35 mu M. [GRAPHICS] .

Abstract:By activity-guided fractionation, two new sterols, 3beta,28xi-dihydroxy-24-ethylcholesta-5,23Z-dien (1) and 2a-oxa-2-oxo-5alpha-hydroxy-3,4-dinor-24-ethylcholesta-24(28)-ene (2), together with five known steroids, fucosterol (3), 24-ethylcholesta-4,24(28)-dien-3,6-dione (4), 24xi-hydroperoxy-24-vinylcholesterol (5), 24-ketocholesterol (6), 24R,28R- and 24S, 28S-epoxy-24-ethylcholesterol (7), were isolated from the brown alga Sargassum carpophyllum as active compounds causing morphological abnormality of Pyricularia oryzae mycelia. Compounds 1, 3, 4, 5 and 7 also exhibited cytotoxic activity against various cancer cell lines. The IC50 values for 1 and 5 against HL-60 were 7.8 and 8.5 microg/ml, 3 and 4 against P-388 were 0.7 and 0.8 microg/ml, whereas 7 against MCF-7, HCT-8, 1A9, HOS and PC-3 were 4.0, 8.8, 10.0, 10.0 and 7.2 microg/ml, respectively.

Abstract:A new 19-oxygenated steroid, 25-acetate-nebrosteroid K (1), and five known steroids (2–6), along with one known 19-hydroxy steroidal glycoside (7) were obtained from the gorgonian Dichotella gemmacea collected from the South China Sea. The structure and relative configuration of 1 were elucidated by using comprehensive spectroscopic data including NOESY spectra. Compound 1 represents the first example of 19-oxygenated steroid with a 19-oic acid methyl ester group isolated from gorgonians. All of the isolated compounds were evaluated for lethal activity to brine shrimp Artemia salina and cytotoxicity against A549 and HL-60 cell lines. Among them, 1 showed strong lethality towards A. salina.

Abstract:Seven undescribed C27 steroidal glycosides, Seladelicatulasine A–G, including six cholestanol glycosides and one spirostanol glycoside, were isolated from Selaginella delicatula. Their structures were elucidated by 1D/2D NMR spectra and HRESIMS analyses. The absolute configurations of the sugars were determined by enzymatic hydrolysis and GC/MS analyses. These cholestanol glycosides were isolated from the family Selaginellaceae for the first time. Seladelicatulasine F is characterized as a rare B-5,6-secosteroid. In addition, all the compounds were evaluated for their inhibitory activities against cholinesterase (AChE/BChE) and monoamine oxidase (MAO-A/MAO-B). These steroidal glycosides displayed selective inhibition activities on cholinesterase. Seladelicatulasine A, B and E inhibited the AChE activity with IC50 values of 0.31, 0.09, and 0.04 μM, respectively. Seladelicatulasine A and F showed the strongest inhibition activity on BChE with IC50 values of 0.37 and 0.65 μM, respectively.

Abstract:Six selaginellin derivatives, including three new analogues selaginellins D-F (1-3), were isolated from the EtOAc extract of the whole plant of Selaginella pulvinata (Hook. et Grev.) Maxim. Their structures were determined on the basis of extensive physical and chemical evidence. Compounds 1 and 4 demonstrated antifungal activities against Candida albicans; compounds 4-6 exhibited significant antibacterial activity against Staphylococcus aureus.

Abstract:A new steroid, 24 xi, 25-dimethyl-3 alpha-hydroxyl-cholest-5-ene-2 beta-ol sodium sulfate (1), together with a known steroid, 24 xi, 25-dimethyl-cholest-5-ene-2 beta, 3 alpha-diol disodium sulfate (2), was isolated from the ethanol extract of marine sponge Halichondria rugosa. Their structures were elucidated on the base of spectroscopic analysis. Both compounds showed cytotoxicity to four human cancer cell lines (BEL-7402, HT-29, SPC-A1 and U-251) with IC50 values between 6.5 and 23.1 mu M.
