Abstract:A catalytic 1,2‐oxyhalogenation method that converts non‐conjugated internal alkynes into tetrasubstituted alkenes with high regio‐ and stereoselectivity is described. Mechanistically, the reaction involves a Pd II /Pd IV catalytic cycle that begins with a directed oxypalladation step. The origin of regioselectivity is the preference for formation of a six‐membered palladacycle intermediate, which is facilitated by an N,N‐bidentate 2‐(pyridin‐2‐yl)isopropyl (PIP) amide directing group. Selectivity for C(alkenyl)−X versus −N (X=halide) reductive elimination from the Pd IV center depends on the identity of the halide anion; bromide and iodide engage in C(alkenyl)−X formation, while intramolecular C(alkenyl)−N reductive elimination occurs with chloride to furnish a lactam product. DFT calculations shed light on the origins of this phenomenon.

Abstract:A novel strategy of visible‐light photoredox catalysis ipso‐annulation of activated alkynes with polychloroalkanes for the synthesis of 3‐polychloroalkyl spiro[4,5]trienones, which used 4‐methoxybenzenediazonium tetrafluoroborate salt as a hydrogen atom transfer reagent, is described. This polychloroalkylation/ spirocyclization of alkynes offers a widely range of 3‐polychloroalkyl‐spiro[4,5]trienone derivatives in high yields with a broad substrate scope via selective C(sp3)‐H homolytic cleavage. The results of controlled experiments showed that this method involves a radical process. The polyhaloalkyl radical species generated from intermolecular hydrogen atom transfer (HAT) between aryl radical and polychloroalkanes.

Abstract:A simple and efficient catalyst-free method for isothiocyanatoalkylthiation of styrenes has been developed. The protocol uses (alkylthio)pyrrolidine-2,5-diones and trimethylsilyl isothiocyanate as the isothiocyanatoalkylthiolating reagents, dimethylformamide (DMF) as the solvent, and the reactions were completed within one hour with tolerance of some functional groups. No catalyst and additive were required in the reaction system, which avoided any contamination of transition metal catalysts in the target products. Therefore, the present method provides a convenient, efficient and practical strategy for the synthesis of compounds by a combination of sulfides and isothiocyanates.

Abstract:A versatile method for radical reductive cross-coupling of iodides and activated olefins under mild conditions, facilitated by NHC-BH3 through photocatalysis, was developed. This method exhibits high efficiency and regioselectivity, achieving yields of up to 98% for aryl, heterocyclic, and alkyl iodides. The reaction employs a minimal amount (1.0 mol%) of photocatalyst and can be performed on a gram scale. Importantly, this approach avoids the use of transition metal co-catalysts, eliminating the need for traditional C-I bond cleavage steps such as oxidative addition, migration insertion, and halogen exchange. This work unveils a novel mechanism for the reductive coupling of iodides in a photocatalytic NHC-BH3 system, providing insights into iodide modification and the functional exploration of NHC-BH3.

Abstract:An electrochemically enabled C(sp3)-C(sp) cross-coupling reaction is herein described for the synthesis of internal alkynes. This method is suitable not only to a diverse of acetylenic sulfones, but also to a wide range of alkyl radical precursors, such as alkyl iodides, N-hydroxyphthalimide esters and Katritzky salts. This reaction can also be applied to vinylation, thioetherification, selenation and hydrogenation reactions. This reaction shows a lot of significant advantages such as mild conditions, broad functional group tolerance, simple operation and convenience of purification and enlargement.

Abstract:To synthesize meso-ethynylene-conjugated porphyrin arrays, the Sonogashira cross-coupling reaction is straightforward to construct the C(sp)-C(sp2) bonds, but the reaction is often accompanied by side reactions such as the Glaser homocoupling. The rate-determining oxidative addition step results in the unexpected kinetic competition with the Glaser homocoupling, which is desired to be circumvented. We here propose two sets of improved strategies for the synthesis of arylene-ethynylene-linked porphyrin arrays from the meso-brominated porphyrin and alkynes. First, we explored the solvent-modulated approach employing dichloromethane as the reaction solvent to minimize the formation of the copper acetylide dimer as the intermediate for the Glaser homocoupling, while the scope of the approach is limited. Subsequently, we have developed the trimethylsilanolate-promoted Hiyama-type approach that activates alkynyl trimethylsilanes (TMSs) by use of potassium trimethylsilanolate under amine-free conditions. The latter approach is advantageous not only in skipping the preprotodesilylation of the TMS group but also in achieving an excellent isolated yield.

Abstract:Enabled by merging halogen atom transfer and hydrogen atom transfer, a novel photocatalytic [3+2] cycloaddition of alkynes and alkyl/aryl iodides is accomplished, providing a highly regio‐ and diastereoselective approach for the direct assembly of ubiquitous and important cyclopentane derivatives from readily available chemicals. Comprehensive Summary A visible light photocatalytic [3+2] cycloaddition of alkynes with readily accessible organic iodides as the C3 synthon is developed herein. By merging halogen atom transfer (XAT) and hydrogen atom transfer (HAT), alkyl/aryl iodides serve as a formal diradical precursor and add across C‐C triple bonds to deliver a number of functionalized cyclopentanes in moderate to high yields with exceptional regio‐ and diastereoselectivity. A reductive radical‐polar crossover mechanism, involving the cascade XAT, radical addition, 1,5‐HAT, polar effect‐promoted 5‐endo annulation, single electron transfer (SET) reduction, and protonation, may account for this unprecedented dehalogenative [3+2] cycloaddition. This work not only expands the repertoire of the traditional RATC methodology, but also provides a robust platform for the expedient assembly of cyclopentanes, a valuable structural motif in the realms of medicinal chemistry and material sciences.

Abstract:A highly regio- and stereoselective halohydroxylation of 1,2-allenyl sulfoxides with X+ and water was developed. The reaction shows E-stereoselectivity. In the iodohydroxylation reaction, 12 was used to introduce the iodine atom. For bromohalohydroxylation, CuBr2, NBS, or Br-2 can be used. When using 12, NBS, or Br2, the addition of LiOAc.2H(2)O is necessary for high yields of the halohydroxylation products. The chlorohydroxylation reaction was preformed by milling 1,2-allenyl sulfoxides and CuCl2.2H(2)O with silica gel. Under the catalysis of a Pd(0) complex, the halohydroxylation products, that is, E-2-halo-1-phenylsulfinyl-1-alken-3-ols, can undergo Sonogashira, Suzuki, and Negishi cross-coupling reactions leading to Z-2-substituted-1-phenyisulfonyl-1-alken-3-ols. The C-S bond of the coupling product may undergo a further coupling reaction with organozincs under the catalysis of an Ni catalyst. Here, the presence of a hydroxyl group is important for a smooth coupling involving the C-S bond. Thus, optically active stereodefined multisubstituted allylic alcohols can be prepared by the reaction of the easily available optically active propargylic alcohols with sulfinyl chloride followed by E-halohydroxylation and a selective Pd- or Ni-coupling reaction.

Abstract:Summary of main observation and conclusionAn efficient strategy for the formation of alkenyl‐functionalized spirocarbocyclic scaffolds from alkyne‐containing phenol‐based biaryls via sequential iodine‐induced cyclization/dearomatization and Pd‐catalyzed coupling of N‐tosylhydrazones is developed. The approach provides various spirocarbocyclic compounds in moderate to excellent yields with good functional tolerance.
