Abstract:Revaprazan in overall yield of 42% was synthesized by condensation of p-fluoroaniline with cyanamide, and then condensation with methyl 2-ethylacetoacetate to give pyrimidine, followed by chlorination and then condensation with 1-methyl-1,2,3,4-tetrahydroisoquinoline.

Abstract:2‐Chloro‐3‐amino‐4‐methylpyridine (1), a key intermediate in the synthesis of nervirapine, was prepared from 2‐cyanoacetamide and 4,4‐dimethoxyl‐2‐butanone via condensation, cyclization, one‐pot reaction of chlorination and hydrolysis, and Hofmann reaction. Utilization of the quadratic orthogonal test resulted in a high yield (62.1%) of the whole process.

Abstract:An improved synthesis technology of 1-methyl-3-trifluoromethyl-4-pyrazole carboxylic acid which is an important intermediate of pesticides was studied.The title compound was synthesized by sodium cyanoacetate via condensation,acylation,cyclization and hydrolysis.The suitable reaction conditions were found as follows.The condensation reaction was carried out at 40~45℃ for 5 h,and in which the ethanol was used as solvent,the molar ratio of sodium cyanoacetate to N-(methoxymethylene)-N,N-dimethylaminium methanesulfonate is 1:2.5,and under above conditions the yield was improved from 67%(reported in the literatures) to 93.7%.During acylation,TEA was used as acid-capturer,trifluoroacetyl chloride was used to replace phosgene as acylating reagent and the yield of 78.6% was obtained when the molar ratio of 3-(dimethylamino)acrylonitrile,trifluoroacetyl chloride and TEA is 1:1.5:2.The cyclization reaction was carried out in a mixed solvent of methanol and water,and the product yield of 74.3% was obtained.The structures of the intermediates and product were confirmed by 1H-NMR.The proposed process has quite good industrial value because of its high yields,cheap and easily available raw materials,moderate reaction conditions and convenient operations.

Abstract:Pyridazine was prepared by hydrogenolysis of 3,6-dichloropyridazine obtained via chlorination of 3,6-pyridazine diol with phosphorus oxychloride.3,6-Pyridazine diol was prepared by cyclization of hydrazine dihydrochloride and maleic anhydride.The feasible reaction conditions were as follows: cyclization: the mole ratio of maleic anhydride to hydrazine dihydrochloride was 1∶1,100°C,3 h;chlorination: the mole ratio of 3,6-pyridazine diol to phosphorus oxychloride was 1∶12.5,100～105°C,5 h;catalytic hydrogenolysis: the mole ratio of 3,6-dichloropyridazine to triethylamine was 1∶2.6,50°C,H_2 0.5 MPa,6 h.The overall yield was 62.2%(mol).The product was identified by melting point test,()~1H NMR and elemental analysis.

Abstract:An improved synthetic approach to tenatoprazole 1 is described. It started from 2,3,5-trimethyl-4-nitropyridine-N-oxide 2 with acetic anhydride via rearrangement and hydrolysis to give 3, Chlorination with SOCl2 yielded 2-chloromethyl-3,5-dimethyl-4-nitropyridine hydrochloride 4, then 4 condensed with 2-mercapto-5-methoxy imidazole [4,5-b]pyridine 5 to give 5-methoxy-2-[(4-nitro-3,5-dimethyl-2-pyridinyl)methylthio]imidazole[4,5-b]pyridine 6. At last the title compound 1 was produced by two methods: 6 was oxidized with MCPBA and then methoxylated with CH3ONa to give 1 and 6 was first methoxylated with CH3ONa and then oxidized with MCPBA to give 1. The overall yield is around 26% for both five-step syntheses.

Abstract:3-Nitropyrazole was prepared via two steps, nitration and rearrangement, using pyrazole as a raw material. Pyrazole was nitrated in a mixed acid of nitric acid and sulfuric acid below 15 ℃for 3.5 hours with stirring, giving N-nitropyrazole with a yield of 86.6%, which was then refluxed in n-octanol at 185-190℃, giving 3-nitropyrazole with ayield of 87.8%. This nitration reaction was innovated by using sulfuric acid as the proton donor instead of acetic acid and acetic anhydride, and solvent n-octanol could be recycled without further treatment.

Abstract:The synthetic method on(R)-rolipram was described in detail.Using isoacetovanillone as materials,(R)-rolipram was synthesized by six steps of alkylation,Wittig-Horner,bromo reaction,phthalimide,enantioselective conjugate reduction and deprotection followed by cyclization reaction.The product yield was 34%.The structure of the product was determined by 1H NMR and MS.

Abstract:p-trifluoromethylbenzonitrile was prepared from p-(trifluoromethyl)chlorobenzene and potussium cyanide catalyzed by tris(triphenylphosphine)nickle(0), which was prepared from anhydrous NiCl2 and triphenylphosphine in the presence of zinc.And then, the produced intermideate, p-trifluoromethylbenzonitrile was treated with hydrogen sulfide to give p-(trifluoromethyl)thiobenzamide and the yield of 90% with the purity of 99.7% was obtained for the objective product.The structure is correct through IR and GC-MS testing.The operation process is simple and reaction condition is moderate.

Abstract:Telaprevir(1) was prepared by(2S)-2-cyclohexyl-N-(2-pyrazinylcarbonyl) glycyl-3-methyl-L-Valine(2) through one-pot condensation with(1S,3aR,6aS)-octahydro cyclopenta[c]pyrrole-1-carboxylic acid ethyl ester hydrochloride(3),hydrolysis with NaOH,another one-pot condensation with(3S)-3-amino-N-cyclopropyl-2-hydroxy-Hexanamide hydrochloride(4),Dess-Martin oxi-dation with overall yield of64%and the HPLC purity of 99. 9%.

Abstract:Aripiprazole in overall yield of 68% was synthesized from 2,3-dichloroaniline by a three-step nucleophilic substitution reaction in yield of 83%,91% and 90%,respectively.The structure was characterized by 1H NMR,IR and elemental analysis.

Abstract:OBJECTIVE:To modify the synthetic method of fluvoxamine. METHODS:Fluvoxamine was synthesized from 4-trifluoromethylbenzonitrile by the steps of Grignard reaction, hydrolysis and oximation. RESULT:The chemical structure of the synthesized product was confirmed by (1)HNMR, and the total yield reached to 36.16%. CONCLUSION:The results indicate that the synthetic route is practical.

Abstract:Nevirapine was synthesized from condensation of ethyl acetylacetate and cyanoacetamide followed bychlorination, hydrolysis, Hofmann reaction, hydrogenation, regioselective chlorination to give the key intermediate 2-chloro-3-amino-4-methylpyridine, which subjected to acylation with 2-chloronicotinoyl chloride and tandem reaction withcyclopropylamine and cyclization in 39% overall yield.

Abstract:Ethyl 4-(4-chlorophenyl)-3-methyl-2,4-dioxobutyrate(4)was prepared from 4-chloropropiophenone by protection with chlorotrimethylsilane,then condensation with ethyl oxalyl chloride.4 reacted with 2,4-dichlorophenylhydrazine hydrochloride,then performed an intramolecular cyclization in the presence of acetyl chloride and hydrolyzed to afford the key intermediate of rimonabant,5-(4-chloro phenyl)-1-(2,4-dichlorophenyl)-4-methylpyrazole-3-carboxylic acid.The overall yield was 30.3%.The structure was confirmed by 1H NMR and MS.

Abstract:(R)-N-(5-(2-methoxy-2-phenylacetyl)-1, 4, 5, 6-tetrahydropyrrolo [3, 4-c]pyrazol-3-yl)-4-(4-methylpiperazin-1-yl) benzamide was synthesized from glycine via Micheal reaction, esterification, protection of amino, cyclization to give tert-Butyl-3-Amino-4, 6-dihydro-1H-pyrrolo [3, 4-c] pyrazole-5(1H)-carboxylate, which followed by acylation, deprotection of boc, acylation with mandelic acid and deprotection with an overall yield of about 24.5%.

Abstract:Favipiravir, an antivirus drug, was synthesized from diethyl aminomalonate hydrochloride by ammonolysis and cyclization with glyoxal to give 3-oxo-3,4-dihydropyrazin-2-carboxamide(T1105), which was subjected to bromination with bromine, chlorination and dehydration with POCl3 and halogen exchange with KF to afford 3,6-difluoropyrazin-2-carbonitrile, followed by selective hydrolysis of 3-fluoro and cyano group with an overall yield of about 9%.

Abstract:(2S, 3R)-2-acetooxymethanyl-3-(P-nitrophenyl)-N-tosylaziridine 5 was synthesized from (1S, 2S)-2-amino-1-(4-nitrophenyl)-1, 3-propanediol 1 in four steps with a 24.8% overall yield. This reaction is stereospecific and occurs an expectable configuration inversion at α-carbon atom of benzyl group. The structure of substituted aziridine was determined on FT-IR, NMR, MS and so on.

Abstract:We developed a facile, efficient method for synthesizing highly substituted 2-aminopyridines from unstable vinyl carbodiimides generated in situ in a one-pot transformation. A series of novel highly substituted 3-functionalized 2-aminopyridines were produced in good yields. Reaction mechanism studies, which included control experiments and density-functional theory (DFT) calculations, demonstrated that Rh and potassium carbonate played key roles in the cyclization step.
