Abstract:Aripiprazole in overall yield of 68% was synthesized from 2,3-dichloroaniline by a three-step nucleophilic substitution reaction in yield of 83%,91% and 90%,respectively.The structure was characterized by 1H NMR,IR and elemental analysis.

Abstract:Objective To synthesize aripiprazole,a novel antipsychotic drug.Methods aripiprazole was prepared from 1-(2,3-dichlorophenyl)piperazine hydrochloride by condensation with 1,4-dibromobutane followed by reaction with 7-hydroxy-3,4-dihydrocarbostyril.Results The overall yield was 45% aripiprazole was achieved.The process is suitable for industrial producing.the intermediate's structure is 8-(2,3-dichloro-phenyl)-5,8-diaza-spiro[4,5]decane bromide.Conclusion The synthetic method is economical,convenient and suitable for industrial production.It's proved that the intermediate's structure is 8-(2,3-dichloro-phenyl)-5,8-diaza-spiro[4,5]decane bromide and not 1-(4-bromo-butyl)-(2,3-dichloro-phenyl)-piperazine reported in the literature.

Abstract:An improved and commercially valuable process is developed for the scalable synthesis of 2-chloro-3-amino-4-methylpyridine (CAPIC), a key intermediate of Nevirapine. The synthesis was accomplished in four steps, featuring condensation starting from 4,4-dimethoxyl-2-butanone and cyanoacetamide with ammonium acetate and acetic acid as catalysts. The total yield of the process is 62.1%. The pure CAPIC sample was confirmed with FTIR, 1H NMR, and 13C NMR spectra.

Abstract:An improved synthesis technology of 1-methyl-3-trifluoromethyl-4-pyrazole carboxylic acid which is an important intermediate of pesticides was studied.The title compound was synthesized by sodium cyanoacetate via condensation,acylation,cyclization and hydrolysis.The suitable reaction conditions were found as follows.The condensation reaction was carried out at 40~45℃ for 5 h,and in which the ethanol was used as solvent,the molar ratio of sodium cyanoacetate to N-(methoxymethylene)-N,N-dimethylaminium methanesulfonate is 1:2.5,and under above conditions the yield was improved from 67%(reported in the literatures) to 93.7%.During acylation,TEA was used as acid-capturer,trifluoroacetyl chloride was used to replace phosgene as acylating reagent and the yield of 78.6% was obtained when the molar ratio of 3-(dimethylamino)acrylonitrile,trifluoroacetyl chloride and TEA is 1:1.5:2.The cyclization reaction was carried out in a mixed solvent of methanol and water,and the product yield of 74.3% was obtained.The structures of the intermediates and product were confirmed by 1H-NMR.The proposed process has quite good industrial value because of its high yields,cheap and easily available raw materials,moderate reaction conditions and convenient operations.

Abstract:3-Nitropyrazole was prepared via two steps, nitration and rearrangement, using pyrazole as a raw material. Pyrazole was nitrated in a mixed acid of nitric acid and sulfuric acid below 15 ℃for 3.5 hours with stirring, giving N-nitropyrazole with a yield of 86.6%, which was then refluxed in n-octanol at 185-190℃, giving 3-nitropyrazole with ayield of 87.8%. This nitration reaction was innovated by using sulfuric acid as the proton donor instead of acetic acid and acetic anhydride, and solvent n-octanol could be recycled without further treatment.

Abstract:(S)-6-Nitroindoline-2-carboxylic acid, a substructure occurring in numerous biologically active natural products, was synthesized with moderate yield (53%) and high enantiomeric excess (> 99.5%) starting from the nitration of L-phenylalanine, which is a commercially available chiral pool compound, followed by successively bromination and intramolecular cyclization. The route was carried out in gram quantities and it is suitable for industrial application due to its convenient reaction conditions and low cost.

Abstract:Quetiapine hemifumarate wassynthesized in 63.4% overall yield from 2-aminodiphenyl sulfide and triphosgene to give 2-isocyanato diphenyl sulfide, which subjected to cyclization in polyphosphoric acid, chlorination and condensation with piperazine to afford 11-piperazin-1-yl-dibenzo[b,f][1,4]thiazepine followed by N4-alkylation of piperazine and then salification with fumaric acid.

Abstract:A general synthetic route of aminopyridines which are useful organic intermediates was investigated.The target compounds were synthesized starting from corresponding cyanopyridines,via incomplete hydrolysis in the presence of hydrogen peroxide using mixture of acetone-water and dilute base as solvent,and Hoffman degradation reaction with freshly made NaBrO.The feasible conditions of the above synthetic process were discussed taking the preparation of 4-aminopyridine as example.The optimum concentration of sodium hydroxide solution in the transformation of cyanopyridine to pyridinecarboxamide was 1.0 %.And the Hoffman degradation would be efficiently carried out at 75 ℃ in 12.5 % sodium hydroxide solution.

Abstract:The synthetic method on(R)-rolipram was described in detail.Using isoacetovanillone as materials,(R)-rolipram was synthesized by six steps of alkylation,Wittig-Horner,bromo reaction,phthalimide,enantioselective conjugate reduction and deprotection followed by cyclization reaction.The product yield was 34%.The structure of the product was determined by 1H NMR and MS.

Abstract:A new and efficient method for the synthesis of 4-methoxyl-2,3,5-trimethylpyiidine with an overall yield of 76% from cheap and easily available 2,3,5-trimethylpyridine was developed. The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:A practical process for the resolution of racemic 6-fluoro-3, 4-dihydro-2H-1-benzopyran-2-carboxylic acid ((±)-A) was studied. (+)-Dehydroabietylamine was separated as the chiral resolving reagent from commercially available rosin amine with a purity of 98.8% and the optical purity 100%. Dehydroabietylamine and (±)-A with an amount of substance ratio of 1:1 were used to form the diastereomeric amides; the pure diastereomers were obtained separately from ethanol/methanol by fractional crystallization in a total yield of 59% (30%, 29% respectively); the optical acids were obtained by the hydrolysis of the corresponding diastereomers under a 40 h refluxing with a mass ratio of 0.15:2.00:1.00 of amide, acetic acid and concentrated HCl, totally yielding 32%. The spectra and physical properties of the diastereomers were given. The diastereomeric amides were hydrolyzed to give (±)-A in boiling glycol with KOH, and the mother liquor in the resolution was recovered 52% in (±)-A.

Abstract:OBJECTIVE To study the synthetic method of 2-aminoxy-1- aminoethane dihydrochloride. METHOD The objective compound was synthesized by steps of ring-opening, condensation, hydrazinolysis and hydrolysis, starting from phthalic anhydride.RESULTS The chemical structure of synthetic product was confirmed by IR and 1HNMR, the total yield was amount to 39.4%. CONCLUSION This synthetic route is practical.

Combined ultrasound-microwave assisted synthesis of aripiprazole: Process optimization using RSM-ANN

Abstract:The current work demonstrates combined microwave and ultrasonic assisted synthesis of aripiprazole (ARZ) drug from carbostyril intermediate with piperazine moiety in the presence of a mixture of a green medium. The traditional cooking (TC) technique is time-consuming (≥ 16.45 h), accounts for a significant number of contaminants (≥ 2.63%), and has the drawback of additional repetitive purification stages. The intensification tool simultaneous microwave-ultrasonic irradiation has been employed to overcome these challenges. Response surface methodology (RSM), along with central composite design (CCD), was used to assess the impact of different process parameters on process yield (Υ c ) and total impurity content (ὶ). A consistent process yield (≥ 91%) with less total impurity content (≤ 1.45%) was obtained at the optimum condition of reactant mole ratio (1:1), base mole ratio (2.4:1), reaction temperature (85°C) and process time (188 min). Among the exercised numerical tools, a neural network was found to be more precise than RSM for the current system. The synthesis of ARZ followed a second-order kinetic model. The activation energy was found to be 79.61 kJ/mol. Eventually, the qualitative analysis shows a good resemblance between the synthesized products obtained by these two techniques.

Abstract:OBJECTIVE To study and improve the synthesis of donepezil.METHODS 3, 4-dimethoxy-benzaldehyde was condensed with malomc acid in the presence of pyridine, then hydrogenated with Raney Ni to get3-(3,4-dimethoxy-phenyl)-propanic acid, which was cy-clized with poly-phosphoric acid (PPA) to yield 5,6-dimetrioxy-l-indenone (intermediate 1). N-benzyl piperidone was epoxidated with (CH3)2 SOCH2- to get epoxide, which was rearranged with magnesium bromide-etherate to get N-benzyl-4-formyl-piperidine (intermediate 2) . The two intermediates were condensed with NaOH, followed by selective hydrogenation with 5% Pd-C and hydrochlorination to yield donepezil. RESULTS The structures of intermediates and donepezil were identified by IR,1H-NMR and MS.CONCLUSION This method avoided using expensive reagents such as n-BuLi, IDA, and there was no need to use column chromatograph in all steps.

Abstract:A novel tandem reductive amination/intermolecular nucleophilic aromatic substitution (SNAr) sequence has been established for the synthesis of amine containing pyrimidine in formation of one carbon-oxygen and one carbon-nitrogen bonds in a one-pot fashion. Treatment of aldehyde with arylamine, 2-methanesulfonyl-4,6-dimethoxypyrimidine and sodium borohydride provides good overall yield. The p-toluenesulfonic acid (PTSA) can be used as activator and is generally needed in the reaction. Dioxane is the preferred reaction solvent, but reactions can also be carried out in tetrahydrofuran (THF), MeCN, toluene and dichloromethane. The procedure is carried out effectively in the presence of K2CO3. The reaction proceeds smoothly with aromatic aldehydes and arylamines possessing electron-donating or -withdrawing groups. This method can be applied to the synthesis of the oilseed rape herbicide and is superior to the classical one in several aspects: cutting out several purification steps, minimizing solvent use and chemical waste, and saving time. Its advantages such as operational convenience, high-efficient synthesis, and starting material availability make it a desirable method for preparing amines with molecular diversity and biological activity.

Abstract:The synthesis of 3-(N-ethyl-N-methoxycarbonylmethyl) amino acetanilide via two-step reactions was reported,with 3-aminoacetanilide as starting material.The designed product was yielded by the ethylated compound(from 3-aminoacetanilide reacting with acetaldehyde) reacting with methyl chloroacetate.The optimized reaction conditions were as follows: in the first step,the yield of 3-(Nethyl) amino acetanilide was 90.7% and its purity was up to 97.5%,when the molar ratio of acetaldehyde to 3-aminoacetanilide was 1.0∶1.2 and when 5% Pd/C was used as catalyst under 3.5 MPa of hydrogen atmosphere at 70 ℃.In the second step,the designed product was obtained in 94.6% yield and 96.8% purity with 0.6 equiv.of sodium carbonate as deacid reagent under 70 ℃.

Abstract:Pyridazine was prepared by hydrogenolysis of 3,6-dichloropyridazine obtained via chlorination of 3,6-pyridazine diol with phosphorus oxychloride.3,6-Pyridazine diol was prepared by cyclization of hydrazine dihydrochloride and maleic anhydride.The feasible reaction conditions were as follows: cyclization: the mole ratio of maleic anhydride to hydrazine dihydrochloride was 1∶1,100°C,3 h;chlorination: the mole ratio of 3,6-pyridazine diol to phosphorus oxychloride was 1∶12.5,100～105°C,5 h;catalytic hydrogenolysis: the mole ratio of 3,6-dichloropyridazine to triethylamine was 1∶2.6,50°C,H_2 0.5 MPa,6 h.The overall yield was 62.2%(mol).The product was identified by melting point test,()~1H NMR and elemental analysis.

Abstract:A series of new 1-(4-(3,4-dihydroquinolin-2(1H)-one-7-yloxy)butyl)-3-methyl-2,7-diphenyl-1,4-diazepan-5-one analogs of aripiprazole was synthesized by the replacement of 2,3-dichlorophenyl-4-piperazine/diazepane moiety with 3-methyl-2,7-diphenyl-1,4-diazepan-5-one to explore the influence of structural features. All the synthesized compounds are characterized by elemental analysis, Fourier Transform Infrared (FTIR), Proton Nuclear Magnetic Resonance (1H-NMR), Carbon-13 Nuclear Magnetic Resonance (13C-NMR), Heteronuclear Single Quantum Coherence (HSQC) (2D NMR) and mass spectrometry of compound 4a. All the multi target ligands have been docked against, human A2A adenosine receptor and human β2-adrenergic G-protein-coupled Receptors (GPCRs), both receptor and ligand interaction shows an excellent dock score. Absorption Distribution, Metabolism and Excretion (ADME), properties were also evaluated in the desirable range, finally these compounds have orally drug-likeness property. In this event was done to screening the neuroleptic activity of the synthesized compounds with different anti-psychotic animal models.
