Abstract:An improved synthesis technology of 1-methyl-3-trifluoromethyl-4-pyrazole carboxylic acid which is an important intermediate of pesticides was studied.The title compound was synthesized by sodium cyanoacetate via condensation,acylation,cyclization and hydrolysis.The suitable reaction conditions were found as follows.The condensation reaction was carried out at 40~45℃ for 5 h,and in which the ethanol was used as solvent,the molar ratio of sodium cyanoacetate to N-(methoxymethylene)-N,N-dimethylaminium methanesulfonate is 1:2.5,and under above conditions the yield was improved from 67%(reported in the literatures) to 93.7%.During acylation,TEA was used as acid-capturer,trifluoroacetyl chloride was used to replace phosgene as acylating reagent and the yield of 78.6% was obtained when the molar ratio of 3-(dimethylamino)acrylonitrile,trifluoroacetyl chloride and TEA is 1:1.5:2.The cyclization reaction was carried out in a mixed solvent of methanol and water,and the product yield of 74.3% was obtained.The structures of the intermediates and product were confirmed by 1H-NMR.The proposed process has quite good industrial value because of its high yields,cheap and easily available raw materials,moderate reaction conditions and convenient operations.
Novel Synthesis Process of 5H-Dibenz[b,f]azepine and Removal of Chloroformyl Protection Group

Abstract:A novel synthesis process of 5H-dibenz[b,f]azepine was studied.It is an important intermediate of 5H-dibenz[b,f]azepine-5-carboxamide,used in genetic engineering,materials science and so on.With 10,11-dihydro-5H-dibenz[b,f]azepine as raw material,5-chloroformyl-5H-dibenz[b,f]azepine was prepared by reflux with triphosgene in toluene,brominated with bromine in chlorobenzene,and dehydrobrominated at 130℃,finally,the title compound was obtained by removing chloroformyl group with NaOH in isopropanol with 80.4% overall yield.The optimal reaction conditions and yields (temperature,time,molar yield) were as follows: acyl chlorination,110℃,10 h,93.0%; bromination,dehydrobromination,100℃,2 h,130℃,5 h,88.0%; removal of chloroformyl group,40℃,3 h,98.2%. The melting point of the title compound was the same as literature reported,and its structure was confirmed by 1H NMR.The optimal technology condition to remove chloroformyl group as protection group and its application of protection for some amine compounds were also studied.

Abstract:ZJ273,propyl 4-[2-[(4,6-dimethoxy-2-pyrimidinyl)oxy]benzylamino]benzoate,is a novel herbicide.The microsynthetic method of the compound was developed from 4,6-dimethoxy-2-pyrimidine,propyl(4-benzylamino) benzoate and phenol,and then purified by preparative-HPLC.The yield of end product was 53%.Its chemical structure was characterized by means of()~1H-NMR,MS,UV,and its chemical purity was determined to be exceed 98% by HPLC.The microsynthetic method of propyl 4[2-[(4,6-dimethoxy-2-pyrimidinyl)oxy] benzylamino]benzoate developed sets up a basis to synthesize the radioisotopic labeled target compound.

Abstract:As one kind of tetrahydrophthalimide herbicides with high efficiency to control the weed in corn and soybean fields,flumiclora-pentyl was firstly synthesized by Sumitomo Chemical.Based on the traditional synthesis process,using p-fluorophenol as a starting material,a new synthesis process of flumiclora-pentyl was developed with several steps such as chlorination,etherification,nitrification,reduction and condensation.The yield of the product was 33%,and the purity was higher than 95%.The structures of the product and the intermediate were confirmed by Elementary Analysis,1H-NMR,MS and IR methods.

Abstract:A new synthetic method for preparing two arylfluoroquinolone antimicrobial agents, difloxacin and temafloxacin, was reported. 2,4-Dichloro-5-fluoroacetophenone was used as a starting material to react with diethyl oxalate in place of diethyl carbonate giving the corresponding beta-keto ester in high yield. The beta-keto ester was allowed to ethyloxymethenylation, amination, cyclization and hydrolysis to afford an intermediate acid with one more carbonyl compared with that reported in the literature. After decarbonylation, it reacted with piperazine derivatives to yield the desired antibiotics. All new compounds were confirmed by H-1 NMR, IR, MS spectra and elemental analysis.

Abstract:A new and efficient method for the synthesis of 4-methoxyl-2,3,5-trimethylpyiidine with an overall yield of 76% from cheap and easily available 2,3,5-trimethylpyridine was developed. The structure of related compounds was identified by IR, H-1 NMR and C-13 NMR. The method could be carried out economically on a huge scale.

Abstract:p-trifluoromethylbenzonitrile was prepared from p-(trifluoromethyl)chlorobenzene and potussium cyanide catalyzed by tris(triphenylphosphine)nickle(0), which was prepared from anhydrous NiCl2 and triphenylphosphine in the presence of zinc.And then, the produced intermideate, p-trifluoromethylbenzonitrile was treated with hydrogen sulfide to give p-(trifluoromethyl)thiobenzamide and the yield of 90% with the purity of 99.7% was obtained for the objective product.The structure is correct through IR and GC-MS testing.The operation process is simple and reaction condition is moderate.

Abstract:OBJECTIVE:To modify the synthetic method of fluvoxamine. METHODS:Fluvoxamine was synthesized from 4-trifluoromethylbenzonitrile by the steps of Grignard reaction, hydrolysis and oximation. RESULT:The chemical structure of the synthesized product was confirmed by (1)HNMR, and the total yield reached to 36.16%. CONCLUSION:The results indicate that the synthetic route is practical.

Abstract:Three efficient methods to synthesize mono-and di-fluorinated benzimidazoles are reported. These methods provide 5-amino-6-fluoro-benzimidazoles (5), 5,7-difluoro-benzimidazoles (10), and 6-fluoro-benzimidazoles (13) starting from commercially available 1,5-difluoro-2,4-dinitrobenzene (DFDNB), 2,3,4,5-tetrafluoro-6-nitrobenzoic acid (TFNBA), and 2,4-difluoro-1-nitrobenzene (DFNB), respectively.

Abstract:sulfomethyltetrazole 5 thiol disodium salt, a side chain of cefonicid,was synthesized from sodium bisulfite,ammonium hydroxide and formaldehyde through five steps of reactions. The yield of product was 10.8% in the optimum condition. The process was more economic in which the iodomethane is substituted to the bromoethane in the second reaction.

Abstract:The research present an improved synthetic method of 7-chloro-6-fluoro-1-(2,4-difluorophenyl) -1,4-dihydro-4-oxo-quinoline-3-carboxylic acid, the important intermediate of temafloxacin. The reaction was started with 2,4-dichloro-5-fluorobenzoyl acetoformic ethyl ester, via amination, cyclization, hydrolysis and decarbonylation. The reaction temperature was moderate and the operation was easily controlled. Moreover, three new intermediates 4,5 and 6 were synthesized by the suggested method.

Abstract:A series of novel insecticides were designed and synthesized by introducing 1,2,4‐triazole into meta‐diamide compounds and the insecticidal activity of the target compounds were evaluated. In an effort to discover a new insecticide, we designed and synthesized a series of novel meta‐diamide compounds containing 1,2,4‐triazole with cyproflanilide as a lead compound. All the compounds were characterized by 1H NMR, 13C NMR, and HR‐MS. Both Plutella xylostella and Mythimna separata were tested for their insecticidal activity at 200 mg/L (P. xylostella: 0%–100%; M. separata: 0%–100%), 20 mg/L (P. xylostella: 0%–97%; M. separata: 0%–100%), and 2 mg/L (P. xylostella: 0%–97%; M. separata: 0%–100%), while Aphis craccivora was tested for its insecticidal activity at 400 mg/L (A. craccivora: 0%–36%). Further studies are needed to investigate the insecticidal activity of A. craccivora. Preliminary bioactivity results showed that most of the compounds had good insecticidal activity at 200 mg/L against P. xylostella and M. separata. Especially, the compound 7p, N‐(cyclopropylmethyl)‐N‐(5‐((2,6‐dibromo‐4‐(perfluoropropan‐2‐yl)phenyl) carbamoyl)‐2‐(1H‐1,2,4‐triazol‐1‐yl)phenyl)‐6‐(trifluoromethyl)nicotinamide (7p), showed good insecticidal activity at even lower doses of 2 mg/L (P. xylostella: 97%; M. separata: 100%), which was equivalent to that of the lead compound cyproflanilide (P. xylostella: 100%; M. separata: 100%), as well as significantly better than the two known compounds Ia (P. xylostella: 97%; M. separata: 0%) and Ib (P. xylostella: 60%; M. separata: 0%). Preliminary structure–activity relationship was also discussed based on insecticidal tests. The results indicate that meta‐diamide compounds containing 1,2,4‐triazole can be developed as novel insecticides.

Abstract:In searching for novel insecticide lead, 20 new meta‐diamide compounds containing triazole were designed and synthesized regarding Cyproflanilide as lead compound. All the compounds were characterized by 1H NMR, 13C NMR and HR MS. In preliminary bioassay, we found that one of the compounds: N‐(cyclopropylmethyl)‐N‐(5‐((2,6‐dibromo‐4‐(1,2,2,2‐tetrafluoro‐1‐(trifluoromethyl)ethyl)phenyl)carbamoyl)‐2‐(1H‐1,2,4‐triazol‐1‐yl)phenyl)‐6‐(trifluoromethyl) nicotinamide (16a) had high activity against the target organism Plutella xylostella at 1 mg/L and against the target organism Mythimna separata at 2 mg/L. It is obviously superior to the known compounds Ia and Ib, and it is equivalent to cyproflanilide, the LC50 values of the compound against P. xylostella and M. separata were 0.90 mg/L and 0.64 mg/L respectively. It is providing important clues for subsequent structure‐activity relationship studies. From this, we explored the structure‐activity relationship of these compounds. Through comprehensive analysis of their structural characteristics and insecticidal activity data, we found that different halogen substituents on benzene ring and different aromatic substituents on amino group in the compound had significant effects on insecticidal activity. This has certain guiding significance for further structural optimization and activity enhancement.
