Abstract:An improved synthesis technology of 1-methyl-3-trifluoromethyl-4-pyrazole carboxylic acid which is an important intermediate of pesticides was studied.The title compound was synthesized by sodium cyanoacetate via condensation,acylation,cyclization and hydrolysis.The suitable reaction conditions were found as follows.The condensation reaction was carried out at 40~45℃ for 5 h,and in which the ethanol was used as solvent,the molar ratio of sodium cyanoacetate to N-(methoxymethylene)-N,N-dimethylaminium methanesulfonate is 1:2.5,and under above conditions the yield was improved from 67%(reported in the literatures) to 93.7%.During acylation,TEA was used as acid-capturer,trifluoroacetyl chloride was used to replace phosgene as acylating reagent and the yield of 78.6% was obtained when the molar ratio of 3-(dimethylamino)acrylonitrile,trifluoroacetyl chloride and TEA is 1:1.5:2.The cyclization reaction was carried out in a mixed solvent of methanol and water,and the product yield of 74.3% was obtained.The structures of the intermediates and product were confirmed by 1H-NMR.The proposed process has quite good industrial value because of its high yields,cheap and easily available raw materials,moderate reaction conditions and convenient operations.
Synthesis of 3-Aminopyrrolidine and its N-alkylating Derivatives

Abstract:The target compounds——3-aminopyrrolidine dihydrochloride and its acetate are the key intermediates of tosufloxacin and other quinolone antibiotics. One of the synthetic routes of the target compounds is from benzylamine and ethylacrylate, which lead to 3-benzylaminopropionate by additional reaction, then affords 3-[N-(methoxycarbonylmethene)benzylamino]propionate through N-alkylation with methyl chloroacetate, further by Dieckmann cyclization, decarboxylation to obtain 1-benzyl-3-pyrrolidine, then oximate and hydrogenate it to gain the intermediate 3-amino-1-benzy1-pyrrolidone, which can be changed to target compounds easily. This route is much of industrial value because of the cheap and easily gained materials. But the original method presents many weakness such as low overall yield and troublesome operate process. We carried out our research work to improve the above method and got an obvious increase of N-alkylation yield from 62.4% to more than 95%, and also some improvement in some reaction process. Thus we succeed in getting an industrial route characterized by cheap and easily gained materials, convenient operate process and low cost.

Abstract:An effective synthetic method to achieve difluoromethylated oxazolidin-2-imine has been developed via visible-light promoted three-component tandem reaction of aryl allylamines, 2-BTSO2CF2H (BT = Benzothiazole) and isocyanates. This method features mild reaction conditions and good functional group tolerance, and the reaction mechanism was confirmed by experiments and interpreted by quantum chemical calculations.

Abstract:The three-component reaction between an isoquinoline, a dialkyl acetylenedicarboxylate and 2,2,2-trifluoro-1-phenylethanone involving a 1,4-dipolar cycloaddition cascade approach is developed using a continuous-flow microreactor. The reaction conditions (solvent, flow rate and temperature) are optimized leading to the synthesis 2-(trifluoromethyl)-2H-[1,3]oxazino[2,3-a]isoquinoline derivatives in 79-83% isolated yields, whilst eliminating the formation of 11bH-pyrido[2,1-a]isoquinoline-12,3,4-tetracarboxylate by-products. The continuous-flow method is also applied to similar reactions using aromatic aldehydes as the carbonyl component.

Abstract:As one kind of tetrahydrophthalimide herbicides with high efficiency to control the weed in corn and soybean fields,flumiclora-pentyl was firstly synthesized by Sumitomo Chemical.Based on the traditional synthesis process,using p-fluorophenol as a starting material,a new synthesis process of flumiclora-pentyl was developed with several steps such as chlorination,etherification,nitrification,reduction and condensation.The yield of the product was 33%,and the purity was higher than 95%.The structures of the product and the intermediate were confirmed by Elementary Analysis,1H-NMR,MS and IR methods.

Abstract:A novel and convenient one-step method for the synthesis of 1,3,4,-Triaryl-substituted-3,4,5,6-tetrahydropyrimidin-2(1H)-ones (3) was developed by cyclization of aromatic isocyanates (1) with 3-arylamino-1-phenylpropan-1-ones (2) under refluxing of toluene. Effects of reaction conditions on the converting ratio and yield were investigated. The method offered several advantages including simple experimental procedure, easily available starting materials and excellent yields (79%similar to 96%).

Abstract:The reactions of α-bromoacetophenones with methylhydrazine in refluxing acetic acid generated 2-methyl-4-aryl-2H-[1,2,3]triazoles in good yields. The method was developed by the reactions of α-bromoacetophenones with phenylhydrazines in the presence of cupric ion, leading to 2,4-diary-2H-[1,2,3]triazoles. The structures were established on the basis of corresponding IR, 1H NMR, and elemental analysis data.

Abstract:Herein, a copper-catalyzed 2,2,2-trifluoroethylthiolation reaction of aryl bromides and iodides with elemental sulfur, and 1,1,1-trifluoro-2-iodoethane is described. The reaction showed excellent functional group tolerance and allowed the synthesis of various substituted aryl 2,2,2-trifluoroethyl thioethers with good to excellent yields. This transformation constitutes a one-pot synthesis of 2,2,2-trifluoroethylthiolated compounds from inexpensive, readily available starting materials. Utility of the protocol was further demonstrated in the late-stage synthesis of the pirfenidone derivative. The copper thiolate species were prepared and proposed as key intermediates in the catalytic cycle.

Abstract:A combinatorial nickel‐catalyzed direct trifluoroethylation of aryl iodides with an industrial raw material CF3CH2I has been developed, demonstrating high efficiency, excellent functional‐group compatibility, especially with large sterically hindered groups. The key to success is the combination of nickel with readily available nitrogen and phosphine ligands. The powerful potential of this strategy is further demonstrated by the late‐stage modification of several derived bioactive molecules.

Abstract:In this work, a series of 4-aryl-3-(trifluoromethyl)-1 H -1,2,4-triazol-5(4 H )-one derivatives is synthesized through nucleophilic intramolecular cyclization reactions of ethyl 2-(2,2,2-trifluoro-1-(arylimino)ethyl)hydrazine-1-carboxylate intermediates, which are themselves prepared in high yields via reactions of ethyl chloroformate and N -aryl-2,2,2-trifluoroacetimidoyl chloride derivatives. Some of the merits of the reported procedure are an operationally simple and concise method, the ready availability of starting materials, excellent product yields, no formation of harmful by-products and easy purification of the products. Abstract:A novel method has been developed for the synthesis of 1,3-thiazin-4-one compounds containing trifluoromethyl groups utilizing 2-trifluoromethyl acrylic acid and thioamides as key starting materials. This protocol is characterized by its simplicity, practicality, and tolerance toward various functional groups. Given the straightforward nature of the procedure, the ready availability of both starting materials, and the significance of drugs containing trifluoromethyl, it is anticipated that this reaction will have wide-ranging applications.

Abstract:Ibuprofen is an essential nonsteroidal anti-inflammatory drug (NSAID) which is still inaccessible to more than 100 million patients worldwide. Although three elegant continuous-flow approaches toward this NSAID have been developed, the major defect of requiring the use of a stoichiometric amount of highly toxic, corrosive, and expensive oxidizing reagents (such as di(acetoxy)phenyl iodide (PhI(OAc)2) and iodine monochloride (ICl)) prevents their application in industrial manufacturing. Herein, a novel and catalytic synthesis of ibuprofen from commercially available isobutylbenzene is performed by a linear continuous-flow sequence involving four chemical transformations without conducting solvent exchanges or intermediate purifications. This newly developed proof-of-principle regime provided access to ibuprofen in a global yield of 80.6% with an average of 94.7% for each step in a total reaction time of 32.5 min, corresponding to a throughput of 15.22 g/h.

Abstract:An improved and practical route is reported here for accessing 3,5-dichloro-4-(1,1,2,2-tetrafluoroethoxy)aniline ( 1 ), a key intermediate of hexaflumuron. The overall yield for the preparation of 1 is 74.0%, with a purity of 99.7%, after a one-pot process with catalytic hydrogenation and Bamberger rearrangement and a greener addition reaction in a continuous flow reactor. Compared with the original process route, the new synthetic route has the advantages of fewer reaction steps, higher overall yield, less process safety hazard and environmental impact.

Abstract:Using 5-fluorouracil as starting material,8-fluoro-5-methoxy-2H-[1,2,4]triazolo[4,3-c]pyrimidine-3-thione was synthesized by chlorination,alcoholysis,hydrazinolysis,cyclization,in a total yield of 70.1 %,which is a key intermediate of florasulam.The structure of 5-fluorouracil was confirmed by NMR analysis.The influence of the ratio of reactants,temperature and time factor on the yield was investigated.

Abstract:Readily available 2-unsubstituted imidazole N-oxides were examined as starting materials for the preparation of fully substituted 1,4,5-aryl/alkyl 2-trifluoromethylsulfanyl-imidazoles. Whereas activation of the N-oxide function followed by attempted nucleophilic addition of the -SCF3 was in vain, the alternative approach involving "sulfur transfer reaction" and subsequent electrophilic trifluoromethylation with Togni reagent provided target products in high yield via a one-pot procedure. The structure of representative enantiomerically pure imidazol-2-yl trifluoromethyl sulfide was confirmed by X-ray analysis.

Abstract:A new synthetic route has been developed on a large scale for the synthesis of fluphenazine hydrochloride. We started the synthesis with the cheap and readily available tert-butyl piperazine-1-carboxylate and 1- bromo-3-chloropropane.This approach is scalable nucleophilic substitution reaction between tert-butyl piperazine-1-carboxylate and 1-bromo-3- chloropropane to afford tert-butyl 4-(3-chloropropyl) piperazine-1-carboxylate, the resulting amino chloro compound was used for N-alkylation with 2- (trifluoromethyl)-10H-phenothiazine and the obtained N- alkylated product was deprotected and then Nalkylated with chloro ethanol to afford fluphenazine hydrochloride. During the synthectic approach, we identified unknown impurity which was properly isolated and analysed by spectral data. Our synthetic strategy is very effective for large scale synthesis of fluphenazine hydrochloride.
