Abstract:Converting common biomass materials to high-performance biomedical products could not only reduce the environmental pressure associated with the large-scale use of synthetic materials, but also increase the economic value. Chitosan as a very promising candidate has drawn considerable attention owing to its abundant sources and remarkable bioactivities. However, pure chitosan materials usually exhibit insufficient mechanical properties and excessive swelling ratio, which seriously affected their in vivo stability and integrity when applied as tissue engineering scaffolds. Thus, simultaneously improving the mechanical strength and biological compatibility of pure chitosan (CS) scaffolds becomes very important. Here, inspired by the fiber-reinforced construction of natural extracellular matrix and the porous structure of cancellous bone, we built silk microfibers/chitosan composite scaffolds via ice-templating technique. This biomimetic strategy achieved 500% of mechanical improvement to pure chitosan, and meanwhile still maintaining high porosity (> 87%). In addition, the increased roughness of chitosan pore walls by embedded silk microfibers significantly promoted cell adhesion and proliferation. More importantly, after subcutaneous implantation in mice for four weeks, the composite scaffold showed greater structural integrity, as well as better collagenation, angiogenesis, and osteogenesis abilities, suggesting its great potential in biomedicine.
Investigation of Silk Fibroin Nanoparticle-Decorated Poly(l-Lactic Acid) Composite Scaffolds for Osteoblast Growth and Differentiation

Abstract:Attempts to reflect the physiology of organs is quite an intricacy during the tissue engineering process. An ideal scaffold and its surface topography can address and manipulate the cell behavior during the regeneration of targeted tissue, affecting the cell growth and differentiation significantly. Herein, silk fibroin (SF) nanoparticles were incorporated into poly(L-lactic acid) (PLLA) to prepare composite scaffolds via phase-inversion technique using supercritical carbon dioxide (SC-CO2). The SF nanoparticle core increased the surface roughness and hydrophilicity of the PLLA scaffolds, leading to a high affinity for albumin attachment. The in vitro cytotoxicity test of SF/PLLA scaffolds in L929 mouse fibroblast cells indicated good biocompatibility. Then, the in vitro interplay between mouse preosteoblast cell (MC3T3-E1) and various topological structures and biochemical cues were evaluated. The cell adhesion, proliferation, osteogenic differentiation and their relationship with the structures as well as SF content were explored. The SF/PLLA weight ratio (2:8) significantly affected the MC3T3-E1 cells by improving the expression of key players in the regulation of bone formation, ie, alkaline phosphatase (ALP), osteocalcin (OC) and collagen 1 (COL-1). These results suggest not only the importance of surface topography and biochemical cues but also the potential of applying SF/PLLA composite scaffolds as biomaterials in bone tissue engineering.
Fabrication and properties of PLA/nano-HA composite scaffolds with balanced mechanical properties and biological functions for bone tissue engineering application

Abstract:Abstract Repair of critical bone defects is a challenge in the orthopedic clinic. 3D printing is an advanced personalized manufacturing technology that can accurately shape internal structures and external contours. In this study, the composite scaffolds of polylactic acid (PLA) and nano-hydroxyapatite (n-HA) were manufactured by the fused deposition modeling (FDM) technique. Equal mass PLA and n-HA were uniformly mixed to simulate the organic and inorganic phases of natural bone. The suitability of the composite scaffolds was evaluated by material characterization, mechanical property, and in vitro biocompatibility, and the osteogenesis induction in vitro was further tested. Finally, the printed scaffold was implanted into the rabbit femoral defect model to evaluate the osteogenic ability in vivo . The results showed that the composite scaffold had sufficient mechanical strength, appropriate pore size, and biocompatibility. Most importantly, the osteogenic induction performance of the composite scaffold was significantly better than that of the pure PLA scaffold. In conclusion, the PLA/n-HA scaffold is a promising composite biomaterial for bone defect repair and has excellent clinical transformation potential.

Abstract:Preparation of biological scaffolds with complex shapes and controllable internal structures is a significant development direction for the tissue engineering field. Due to its simplicity, low-cost and cost-effectiveness, fused deposition molding (FDM) is one of the most popular 3D printing technologies. In this paper, polyvinyl alcohol/polylactic acid/hydroxyapatite composites were fabricated successfully. On this basis, composite scaffolds with different pore structures were designed and constructed by using FDM technology. The material structure was characterized by scanning electron microscopy (SEM) and dynamic thermomechanical analyzer. The results showed that the cross-sectional morphology of the system presented a "sea-island" structure. The introduction of PLA and HA significantly increased the modulus of the material, which was beneficial to achieve its FDM processing. The compressibility, mineralization behavior, and biocompatibility of the viscoelastic characteristics to natural cartilage subjected to the condition of filling angle of 45° was systemically invesitgated. Notably, the concentrations of Ca2+ and PO43− in simulated body fluid were reduced by 34.7% and 58.2% on 8th day, indicating a good bone formation and mineralization ability, which was further corroborated by SEM and fourier-transform infrared spectroscopy (FTIR) results. Biotoxicity test showed that the scaffold presented excellent biocompatibility.

Abstract:In bone tissue engineering, the fabrication of a scaffold with a hierarchical architecture, excellent mechanical properties, and good biocompatibility remains a challenge. Here, a solution of polylactic acid (PLA) and Tussah silk fibroin (TSF) was electrospun into nanofiber yarns and woven into multilayer fabrics. Then, composite scaffolds were obtained by mineralization in simulated body fluid (SBF) using the multilayer fabrics as a template. The structure and related properties of the composite scaffolds were characterized using different techniques. PLA/TSF (mass ratio, 9:1) nanofiber yarns with uniform diameters of 72±9μm were obtained by conjugated electrospinning; the presence of 10wt% TSF accelerated the nucleation and growth of hydroxyapatite on the surface of the composite scaffolds in SBF. Furthermore, the compressive mechanical properties of the PLA/TSF multilayer nanofiber fabrics were improved after mineralization; the compressive modulus and stress of the mineralized composite scaffolds were 32.8 and 3.0 times higher than that of the composite scaffolds without mineralization, respectively. Interestingly, these values were higher than those of scaffolds containing random nanofibers. Biological assay results showed that the mineralization and multilayer fabric structure of the composite nanofiber scaffolds significantly increased cell adhesion and proliferation and enhanced the mesenchymal stem cell differentiation toward osteoblasts. Our results indicated that the mineralized nanofiber scaffolds with multilayer fabrics possessed excellent cytocompatibility and good osteogenic activity, making them versatile biocompatible scaffolds for bone tissue engineering.

Abstract:Polyetheretherketone (PEEK) has been one of the most promising materials in bone tissue engineering in recent years, with characteristics such as biosafety, corrosion resistance, and wear resistance. However, the weak bioactivity of PEEK leads to its poor integration with bone tissues, restricting its application in biomedical fields. This research effectively fabricated composite porous scaffolds using a combination of PEEK, nano-hydroxyapatite (nHA), and carbon fiber (CF) by the process of fused deposition molding (FDM). The experimental study aimed to assess the impact of varying concentrations of nHA and CF on the biological performance of scaffolds. The incorporation of 10% CF has been shown to enhance the overall mechanical characteristics of composite PEEK scaffolds, including increased tensile strength and improved mechanical strength. Additionally, the addition of 20% nHA resulted in a significant increase in the surface roughness of the scaffolds. The high hydrophilicity of the PEEK composite scaffolds facilitated the in vitro inoculation of MC3T3-E1 cells. The findings of the study demonstrated that the inclusion of 20% nHA and 10% CF in the scaffolds resulted in improved cell attachment and proliferation compared to other scaffolds. This suggests that the incorporation of 20% nHA and 10% CF positively influenced the properties of the scaffolds, potentially facilitating bone regeneration. In vitro biocompatibility experiments showed that PEEK composite scaffolds have good biosafety. The investigation on osteoblast differentiation revealed that the intensity of calcium nodule staining intensified, along with an increase in the expression of osteoblast transcription factors and alkaline phosphatase activities. These findings suggest that scaffolds containing 20% nHA and 10% CF have favorable properties for bone induction. Hence, the integration of porous PEEK composite scaffolds with nHA and CF presents a promising avenue for the restoration of bone defects using materials in the field of bone tissue engineering.

Abstract:To improve the bone regeneration ability of pure polymer, varieties of bioactive components were incorporated to a biomolecular scaffold with different structures. In this study, polysilsesquioxane (POSS), pearl powder and dexamethasone loaded porous carbon nanofibers (DEX@PCNFs) were incorporated into polylactic (PLA) nanofibrous scaffold via electrospinning for the application of bone tissue regeneration. The morphology observation showed that the nanofibers were well formed through electrospinning process. The mineralization test of incubation in simulated body fluid (SBF) revealed that POSS incorporated scaffold obtained faster hydroxyapatite depositing ability than pristine PLA nanofibers. Importantly, benefitting from the bioactive components of pearl powder like bone morphogenetic protein (BMP), bone mesenchymal stem cells (BMSCs) cultured on the composite scaffold presented higher proliferation rate. In addition, by further incorporating with DEX@PCNFs, the alkaline phosphatase (ALP) level and calcium deposition were a little higher based on pearl powder. Consequently, the novel POSS, pearl powder and DEX@PCNFs multi-incorporated PLA nanofibrous scaffold can provide better ability to enhance the biocompatibility and accelerate osteogenic differentiation of BMSCs, which has potential applications in bone tissue regeneration.

Abstract:Bone defect caused by trauma, tumor, infection, and other reasons is a thorny problem that needs to be solved in orthopedic clinic. Customized bone repair biomaterials and their fabrication still need to be explored. Three-dimensional (3D) printing is a high-speed fabrication process for bone tissue biomaterials, which paves the way of solving clinical bone defect problems in a new way. In this study, the fused deposition modeling (FDM) technology was used to prepare the composite scaffolds of polylactic acid (PLA) and nano-hydroxyapatite (n-HA). The composite scaffold was optimized by material characterization, mechanical property test, and in vitro bone marrow mesenchymal stem cells biocompatibility test. Finally, a rabbit model was established to evaluate the osteogenic ability of PLA/n-HA scaffolds in vivo. The results showed that the PLA/n-HA composites proposed in this study were highly printable, and the printed scaffold showed tunable mechanical strength accompanied by the proportion of n-HA components. The biocompatibility and osteogenic induction properties were proved better than that of the pure PLA scaffold. This composite scaffold of PLA and n-HA provides a promising strategy for the repair of large bone defects.

Abstract:In this study, a novel porous 3D composite scaffold based on the biodegradable Poly(ε-caprolactone) (PCL), Polylactide Acid (PLA) and Calcium Citrate (CC) was developed via polymer blends and thermal-induced phase separation. The chemical structure, crystalline structure and micromorphology as well as mechanical strength of the scaffolds were characterized by Fourier Transform Infrared Spectroscopy (FTIR), X-ray Diffraction (XRD), Scanning Electron Microscope (SEM) and tensile tests. The results show that the obtained composite scaffold present a suitable bone-like porous structure and sufficient mechanical strength. Furthermore, the release of calcium ions in Simulated Body Fluid (SBF) indicates that the composite material can provide a stable calcium-ion environment and maintain a constant pH value during the soaking process. The cell proliferation results from CCK-8 and light microscopy show that MG63 cells exhibit excellent adhesion and proliferation on the stent. At the same time, animal implantation histology confirms that the composite scaffolds have good biocompatibility in vivo. The scaffold material has greatly potential application value in the field of bone tissue engineering.

Abstract:Biodegradable scaffolds based on biomedical polymeric materials have attracted wide interest in bone transplantation for clinical treatment for bone defects without a second operation. The composite materials of poly(trimethylene carbonate), poly(L-lactic acid), and hydroxyapatite (PTMC/PLA/HA and PTMC/HA) were prepared by the modification and blending of PTMC with PLA and HA, respectively. The PTMC/PLA/HA and PTMC/HA scaffolds were further prepared by additive manufacturing using the biological 3D printing method using the PTMC/PLA/HA and PTMC/HA composite materials, respectively. These scaffolds were also characterized by Fourier transform infrared spectroscopy (FT-IR), gel permeation chromatography (GPC), automatic contact-angle, scanning electronic micrographs (SEM), diffraction of X-rays (XRD), differential scanning calorimetry (DSC), and thermogravimetry (TG). Subsequently, their properties, such as mechanical, biodegradation, cell cytotoxicity, cell compatibility in vitro, and proliferation/differentiation assay in vivo, were also investigated. Experiment results indicated that PTMC/PLA/HA and PTMC/HA scaffolds possessed low toxicity, good biodegradability, and good biocompatibility and then enhanced the cell multiplication ability of osteoblast cells (MC3T3-E1). Moreover, PTMC/PLA/HA and PTMC/HA scaffolds enhanced the adhesion and proliferation of MC3T3-E1 cells and enabled the bone cell proliferation and induction of bone tissue formation. Therefore, these composite materials can be used as potential biomaterials for bone repatriation and tissue engineering.

Abstract:Scaffold‐based tissue engineering is a promising strategy to address the rapidly growing demand for bone implants, but developing scaffolds with bone extracellular matrix like structures, suitable mechanical properties and multiple biological activities remains a huge challenge. Here, we aimed to develop a wood‐derived composite scaffold with anisotropic porous structure, high elasticity, and good antibacterial, osteogenic and angiogenic activities. First, natural wood was treated with alkaline solution to obtain a wood‐derived scaffold with oriented cellulose skeleton and high elasticity, which can not only simulate collagen fiber skeleton in bone tissue, but also greatly improve the convenience of clinical implantation. Subsequently, chitosan quaternary ammonium salt (CQS) and dimethyloxalylglycine (DMOG) were further modified on the wood‐derived elastic scaffold through a polydopamine layer. Among them, CQS endows the scaffold with good antibacterial activity, while DMOG significantly improves the scaffold's osteogenic and angiogenic activities. Moreover, the anisotropic and elastic characteristics of the scaffold can induce cell polarization and osteogenic activity through YAP/TAZ signaling pathway. Interestingly, DMOG can also activate the YAP/TAZ by upregulating the expression of Runx‐2. That is to say, the mechanical characteristics of the scaffolds and the modified DMOG can synergistically enhance the expression of YAP/TAZ, thereby effectively promoting osteogenic differentiation. A Multimaterial Scaffold With Tunable Properties: Toward Bone Tissue Repair

Abstract:Polyetheretherketone (PEEK)/β-tricalcium phosphate (β-TCP) scaffolds are expected to be able to combine the excellent mechanical strength of PEEK and the good bioactivity and biodegradability of β-TCP. While PEEK acts as a closed membrane in which β-TCP is completely wrapped after the melting/solidifying processing, the PEEK membrane degrades very little, hence the scaffolds cannot display bioactivity and biodegradability. The strategy reported here is to blend a biodegradable polymer with PEEK and β-TCP to fabricate multi-material scaffolds via selective laser sintering (SLS). The biodegradable polymer first degrades and leaves caverns on the closed membrane, and then the wrapped β-TCP is exposed to body fluid. In this study, poly(l-lactide) (PLLA) is adopted as the biodegradable polymer. The results show that large numbers of caverns form on the membrane with the degradation of PLLA, enabling direct contact between β-TCP and body fluid, and allowing for their ion-exchange. As a consequence, the scaffolds display the bioactivity, biodegradability and cytocompatibility. Moreover, bone defect repair studies reveal that new bone tissues grow from the margin towards the center of the scaffolds from the histological analysis. The bone defect region is completely connected to the host bone end after 8 weeks of implantation.

Abstract:The repair of bone defect, especially large bone defect remains as a significant clinical challenge. In the present study, we fabricated a novel scaffold OCP/SA/SF which was a composite of octacalcium phosphate (OCP), sodium alginate (SA) and silk fibroin (SF). The SEM images showed that it was porous sponge-like and the pore sizes were suitable for the ingrowth of cells and facilitated nutritions and fluids exchange. FTIR spectra and XRD showed characteristic absorption peaks of the OCP and SF of the scaffolds. In vitro swelling and degradation showed good water-binding and biodegradable ability of the scaffolds. Compressive stress test showed that the compressive strength of the scaffolds increased gradually as the OCP contents increased. HA deposition onto the surface of the scaffolds was seen by SEM after 14 days of soaking the scaffolds in SBF. The cultivation of mBMSCs on the scaffolds for 1 day and 7 days showed good biocompatibility through CCK8 assay, SEM and CLSM images. Osteogenic differentiation genes detection showed osteogenic genes ALP, OC, COL1 and OPN were all upregulated when mBMSCs and the scaffolds were cocultured for 7 days and 14 days. In conclusion, this kind of scaffolds showed good mechanical, biocompatible and biodegradable properties, and were nontoxic to mBMSCs. It can promote proliferation and osteogenic differentiation of mBMSCs. Accordingly, it might be a promising biomaterial for the clinical application of bone defect.

Biocompatibility and Physico-Chemical Properties of Highly Porous PLA/HA Scaffolds for Bone Reconstruction

Abstract:The major problem in bone tissue engineering is the development of scaffolds which can simultaneously meet the requirements of porous structure, as well as have the ability to guide the regeneration of damaged tissue by biological fixation. Composites containing biodegradable matrix and bioactive filler are the new hope in this research field. Herein we employed a simple and facile solvent casting particulate-leaching method for producing polylactide acid/hydroxyapatite (PLA/HA) composites at room temperature. FT-IR analysis confirmed the existence of necessary functional groups associated with the PLA/HA composite, whereas energy-dispersive X-ray (EDX) spectra indicated the uniform distribution of hydroxyapatite particles in the polymer matrix. The beehive-like surface morphology of the composites revealed the presence of macropores, ranged from 300 to 400 μm, whereas the thickness of the pores was noticed to be 1–2 μm. The total porosity of the scaffolds, calculated by hydrostatic weighing, was found to be 79%. The water contact angle of pure PLA was decreased from 83.6 ± 1.91° to 62.4 ± 4.17° due to the addition of hydroxyapatite in the polymer matrix. Thus, the wettability of the polymeric biomaterial could be increased by preparing their composites with hydroxyapatite. The adhesion of multipotent mesenchymal stromal cells over the surface of PLA/HA scaffolds was 3.2 times (p = 0.03) higher than the pure PLA sample. Subcutaneous implantation in mice demonstrated a good tolerance of all tested porous scaffolds and widespread ingrowth of tissue into the implant pores. HA-containing scaffolds showed a less pronounced inflammatory response after two weeks of implantation compared to pure PLA. These observations suggest that PLA/HA composites have enormous potential for hard tissue engineering and restoring maxillofacial defects.

Abstract:In this paper, a new polylactide (PLA)-based scaffold composite by biomimetic synthesis was designed. The novel composite mainly consists of nano-hydroxyapatite (n-HA), which is the main inorganic content in natural bone tissue for the PLA. The crystal degree of the n-HA in the composite is low and the crystal size is very small, which is similar to that of natural bone. The compressive strength of the composite is higher than that of the PLA scaffold. Using the osteoblast culture technique, we detected cell behaviors on the biomaterial in vitro by SEM, and the cell affinity of the composite was found to be higher than that of the PLA scaffold. The biomimetic three-dimensional porous composite can serve as a kind of excellent scaffold material for bone tissue engineering because of its microstructure and properties.

Abstract:Orthopedic tumor resection, trauma, or degenerative disease surgeries can result in large bone defects and often require bone grafting. However, standard autologous bone grafting has been associated with donor site morbidity and/or limited quantity. As an alternate, allografts with or without metallic or polyether-etherketone have been used as grafting substitutes. However, these may have drawbacks as well, including stress shielding, pseudarthrosis, disease-transmission, and infection. There is therefore a need for alternative bone substitutes, such as the use of mechanically compliant three-dimensional (3D)-printed scaffolds. Several off-the-shelf materials are available for low-cost fused deposition 3D printing such as polylactic acid (PLA) and polycaprolactone (PCL). We have previously described the feasibility of 3D-printed PLA scaffolds to support cell activity and extracellular matrix deposition. In this study, we investigate two medical-grade filaments consistent with specifications found in American Society for Testing and Materials (ASTM) standard for semi-crystalline polylactide polymers for surgical implants, a pure polymer (100M) and a copolymeric material (7415) for their cytocompatibility and suitability in bone tissue engineering. Moreover, we assessed the impact on osteo-inductive properties with the addition of beta-tricalcium phosphate (β-TCP) minerals and assessed their mechanical properties. 100M and 7415 scaffolds with the additive β-TCP demonstrated superior mesenchymal stem cells (MSCs) differentiation detected via increased alkaline phosphatase activity (6-fold and 1.5-fold, respectively) and mineralized matrix deposition (14-fold and 5-fold, respectively) in vitro . Furthermore, we evaluated in vivo compatibility, biosafety and bone repair potential in a rat femur window defect model. 100M +β - TCP implants displayed a positive biosafety profile and showed significantly enhanced new bone formation compared to 100M implants evidenced by μCT (39 versus 25% bone volume/tissue volume ratio) and histological analysis 6 weeks post-implantation. These scaffolds are encouraging composite biomaterials for repairing bone applications with a great potential for clinical translation. Further analyses are required with appropriate evaluation in a larger critical-sized defect animal model with long-term follow-up.

Abstract:<p>Polylactic acid/Hydroxyapatite (PLA/HA) composite was widely studied and applied in the field of biomaterials for its good processability, bioactivity, and mechanical properties. In addition to traditional preparation methods, additive manufacturing has also been adopted to prepare PLA/HA composites with customized geometries. This work combined the comprehensive optimized PLLA (L-polylactic acid)/nano-HA (nHA) composite with the low-cost and stable Fused deposition modeling (FDM) technology to successfully prepare PLLA/nHA porous bone repair scaffolds. The results showed that PLLA/nHA composite ink satisfied the smoothness of printing, and the accuracy also met the requirements of personalized bone repair application. The high loaded nHA scaffold had suitable compressive strength was significantly higher than those of pure HA ceramic scaffold and cancellous bone. Besides, in vitro bone-like apatite formation on the surface in the degradation process and in vivo evaluations further verified its good osteogenic property. Compared with other complex and cutting-edge 3D printing technologies, this study provides a low-cost, stable, simple and fast way to realize personalized printing of bone repair scaffolds, which is undoubtedly conductive to the improvement and rapid deployment of personalized biomaterials in clinical applications.</p>

Abstract:Bioceramics have been extensively used to improve osteogenesis of polymers because of their excellent bone-forming capabilities. However, the inadequate interfacial bonding between ceramics and polymers compromises their mechanical properties. In this study, zeolitic imidazolate framework-8 (ZIF-8) was grown in situ on strontium carbonate (SrCO3) to construct a core-shell SrCO3@ZIF-8, which was then added to poly-l-lactic acid (PLLA) to print a SrCO3@ZIF-8/PLLA composite scaffold using selective sintering technology. First, ZIF-8 characterized by its multiple organic ligands, forms a robust interface with PLLA. Second, SrCO3 characterized by its negative zeta potential in solution, exhibits the ability to adsorb positively charged zinc ions. This, in turn, promotes the in situ growth of ZIF-8 on SrCO3, eventually achieving perfect bonding between the second phase and the PLLA matrix. Our findings indicated that the composite scaffold exhibited the highest compressive strength (21.93 MPa) and significantly promoted the osteogenic differentiation of mouse mesenchymal stem cells. Moreover, the in vivo results established that the SrCO3@ZIF-8/PLLA scaffold significantly accelerated bone regeneration efficiency in rat femur defects. The prepared scaffold, with its favorable mechanical properties and osteogenic activity, shows considerable promise for applications in bone repair.

Abstract:Mechanical performance is crucial for biomedical applications of scaffolds. In this study, the stress distribution of six lattice-inspired structures was investigated using finite element simulations, and scaffolds with pre-designed structures were prepared using selective laser sintering (SLS) technology. The results showed that scaffolds with face-centered cubic (FCC) structures exhibited the highest compressive strength. Moreover, scaffolds composed of polylactic acid/anhydrous calcium hydrogen phosphate (PLA/DCPA) showed good mechanical properties and bioactivity. An in vitro study showed that these scaffolds promoted cell proliferation significantly and showed excellent osteogenic performance. Composite scaffolds with FCC structures are promising for bone tissue engineering.
