Abstract:Tea polyphenols are considered as an important indicator of tea quality. Rapid detection of tea polyphenol content plays a valuable role for tea breeding and quality inspection during tea production. In this work, a portable rapid non-destructive detection device of tea polyphenols in fresh tea leaves was developed, which integrated the fusion technology of visible/short-wave (400–1050 nm) and long-wave (1000–1650 nm) near-infrared spectroscopy (Vis/NIR). Experimental results indicated that the spectra within the overlapping region (1000–1050 nm) were assembled by applying the spectral data fusing method. Followed by spectral data preprocessing with the Savitzky–Golay smoothing (SG) method, least squares support vector regression (LS–SVR) models were established for detecting the tea polyphenol content of fresh tea leaves. Based on the fused Vis/NIR spectra (dual-band), the correlation coefficient of calibration (RC), root mean square error of calibration (RMSEC), correlation coefficient of prediction (RP), root mean square error of prediction (RMSEP), and residual predictive deviation (RPD) reached 0.976, 0.679%, 0.893, 0.897%, and 2.230, respectively, which were better than the visible/short-wave or long-wave near infrared spectral data (single-band). The sensitive spectral wavebands of tea polyphenols extracted using the random frog (RF) algorithm were distributed in 402–448 nm, 555–600 nm, 810–1042 nm, 1056–1103 nm, 1219–1323 nm, 1406–1416 nm, and 1499–1511 nm. This demonstrated that the prediction of tea polyphenol content using fused spectral data combined with the LS–SVR model depended on various functional groups such as auxochromes, chromogenic groups, and hydrogen-containing groups. The proposed device is capable of non-destructive detection of tea polyphenol content in fresh tea leaves, which can provide effective technical support for tea breeding and tea leaf quality control.

Abstract:The non-judicious use of pesticides in agro-food poses a severe threat to food safety and human health. As an emerging chromatographic fingerprint provider, surface-enhanced Raman spectroscopy analysis (SERS) sheds bright light on sensitive and nondestructive detection of pesticide residues. This research proposed a novel strategy to detect three-pesticide residues (thiabendazole, carbendazim, and chlorpyrifos) on tomato peel based on the flexible and sticky SERS substrate. After selecting the best commercial adhesive tape (3M9080), the SERS substrate was constructed by optimizing the parameters in the preparation process of AuNPs. Therefore, a new simple “tape-wrapped SERS” way for pesticide residue analysis was established with a simple procedure of “absorption, separation, and drop addition.” Based on chemometrics method, the limit of semiquantitative detection was 20, 36, and 80 ng/cm2 for thiabendazole, carbendazim, and chlorpyrifos, respectively, on tomato surface, which indicated that the proposed method could meet the requirement of actual application with a large prospect in agro-food safety detection.
On-site rapid detection of multiple pesticide residues in tea leaves by lateral flow immunoassay

Abstract:Highlights • Rapid test for simultaneous detection of 9 pesticide residues in tea leaves. • An extract-purify-dilute method was optimized for sample rapid pretreatment. • The entire test is easy-to-use, cost-effective, and can be finished within 30 min. The application of pesticides (mostly insecticides and fungicides) during the tea-planting process will undoubtedly increase the dietary risk associated with drinking tea. Thus, it is necessary to ascertain whether pesticide residues in tea products exceed the maximum residue limits. However, the complex matrices present in tea samples comprise a major challenge in the analytical detection of pesticide residues. In this study, 9 types of lateral flow immunochromatographic strips (LFICSs) were developed to detect the pesticides of interest (fenpropathrin, chlorpyrifos, imidacloprid, thiamethoxam, acetamiprid, carbendazim, chlorothalonil, pyraclostrobin, and iprodione). To reduce the interference of tea substrates on the assay sensitivity, the pretreatment conditions for tea samples, including the extraction solvent, extraction time, and purification agent, were optimized for the simultaneous detection of these pesticides. The entire testing procedure (including pretreatment and detection) could be completed within 30 min. The detected results of authentic tea samples were confirmed by ultra-performance liquid chromatography-tandem mass spectrometry, which suggest that the LFICS coupled with sample rapid pretreatment can be used for on-site rapid screening of the target pesticide in tea products prior to their market release. Abstract:A 48-year-old woman presented with severe headache radiating to her neck and chest, followed by a brief period of loss of consciousness in the emergency department. After she regained consciousness, the patient described a 2-year history of right-sided pulsatile tinnitus and hearing loss. She also had a history of poorly controlled hypertension and of noncompliance with her medication. On examination her blood pressure was 239/90 mm Hg. Results of a neurologic examination were unremarkable. Brain CT showed a subarachnoid hemorrhage (figure 1, A and B). However, a large mass was present in the right jugular foramen with erosion of temporal bone and encroachment of the right internal acoustic canal. MRI confirmed a hypervascular mass that contained numerous flow voids. The mass was located lateral to the medulla and extended superiorly into the cerebellopontine angle cistern. It extended inferiorly to the superior aspect of the right parapharyngeal space and displaced the right internal carotid artery (figure 2C). Abstract:Silver nanoparticles (AgNPs) have been regarded as a highly promising substrate for surface-enhanced Raman scattering (SERS) sensors. In this study, we focused on the electrochemical synthesis method by developing three kinds of AgNPs using three different electrolytes: citrate (e-Ag-C), oleic acid (e-Ag-O) and fish mint ( Houttuynia cordata Thunb.) extract (e-Ag-bio). The as-prepared AgNPs were characterized and then employed as SERS substrates to detect the pesticide thiram. The obtained results show that e-Ag-O exhibits the best SERS performance. The effect of the coating agent was explained by chemical and electromagnetic enhancements (CM and EM). Although thiram could absorb onto e-Ag-C at the highest level, allowing its Raman signal to be best enhanced via the CM, the smallest interparticle distance of e-Ag-O could have resulted in the largest improvement in the EM. Using e-Ag-O to develop SERS-based sensors for thiram, we obtain the impressive detection limit of 1.04 × 10 −10 M in standard samples and 10 −9 M in tea leaves. The linear ranges are from 10 −4 M to 10 −7 M and from 10 −7 M to 10 −9 M, covering the maximum residue levels for plant commodities established by the United States Environment Protection Agency and European Food Safety Authority (2–13 ppm ∼7.7 × 10 −6 M to 5 × 10 −5 M).

Abstract:Acetamiprid is an organic sever toxic compound with molecular formula. Despite being widely used as a pesticide agent on a large scale, acetamiprid poses numerous health risks to living organisms, particularly humans. Herein, a strategy for detection acetamiprid in tea employing surface-enhanced Raman scattering (SERS) technology incorporated with a microfluidic chip was developed. Significantly, a seed-mediated growth approach was utilized to engineer Ag-coated tetrapod gold nanostars (core-shell Au@AgNSs) with four sharp tips. The synthesized Au@AgNSs showed an enhancement factor of 7.2×106. Solidworks software was used to figure out the two-channel microfluidic chip featuring four circular split hybrid structures, COMSOL (Software for Multiphysics Simulation) utilized to model the fusion effect between the substrate (Au@AgNSs) and the sample (acetamiprid). For the first time, the core-shell Au@AgNSs and acetamiprid were fused in the microfluidic channel to facilitate SERS detection of acetamiprid. The outcomes pointed out that the standard curve correlation coefficient between SERS intensity (876 cm-1) and concentration of acetamiprid in tea specimens was calculated as 0.991, while the limit of detection (LOD) was 0.048 ng/mL, which is well below the minimum limit set by the European Union (10 ng/mL). Thus, the developed technique combining SERS and microfluidics showed high potential in rapid and efficient detection of acetamiprid in tea.

The extraction efficiency is optimized through single-factor adjustments of key parameters, including adsorbent quantity (5-20 mg), adsorption duration (5-20 min), desorbent identity (methanol, acetonitrile), volume (100-600 μL), sodium chloride concentration (0-10%, m/v), and sample solution pH (3-8). The optimized method demonstrates linearity in the range of 2-500 μg L-1, with limits of detection (LODs) ranging from 0.6-1.5 μg L-1 and limits of quantification (LOQs) from 2.0 5.0 μg L-1. Recovery rates of 81.2-96.0% are achieved, with intra-day relative standard deviations (RSDs) of 1.4-2.8% and inter-day relative standard deviations of 2.0-3.9%, indicating high accuracy and repeatability. The successful application of detecting residues in commercial tea samples underscores the strong potential of the technique to determine benzoylurea analyte levels.

Abstract:Deltamethrin pesticides can cause inflammation, nephrotoxicity and hepatotoxicity as well as affect the activity of antioxidant enzymes in tissues. As a result of this concern, there is a rising focus on the development of fast and reliable pesticide residue testing to minimise potential risks to humans. The goal of this study is to use Au-Ag colloid nanoparticles as liquid surface-enhanced Raman spectroscopy (SERS) to improve the Raman signal in the detection of deltamethrin pesticide in a brewed tea. The liquid SERS system is fascinating to study due to its ease of use and its unlikeliness to cause several phenomena, such as photo-bleaching, combustion, sublimation and even photo-catalysis, which can interfere with the Raman signal, as shown in the SERS substrate. Our liquid SERS system is simpler than previous liquid SERS systems that have been reported. We performed the detection of pesticide analyte directly on brewed tea, without diluting it with ethanol or centrifuging it. Femtosecond laser-induced photo-reduction was employed to synthesise the liquid SERS of Au, Au-Ag, and Ag colloidal nanoparticles. The SERS was utilised to detect deltamethrin pesticide in brewed tea. The result showed that liquid SERS-based Ag NPs significantly enhance the Raman signal of pesticides compared with liquid SERS-based Au NPs and Au-Ag Nanoalloys. The maximum residue limits (MRLs) in tea in Indonesia are set at 10 ppm. Therefore, this method was also utilised to detect and improve, to 0.01 ppm, the deltamethrin pesticide Limit of Detection (LOD).

Abstract:Chlorpyrifos is one of the most widely used broad-spectrum insecticides in agriculture. Given its potential toxicity and residue in food (e.g., tea), establishing a rapid and reliable method for the determination of chlorpyrifos residue is crucial. In this study, a strategy combining surface-enhanced Raman spectroscopy (SERS) and intelligent variable selection models for detecting chlorpyrifos residue in tea was established. First, gold nanostars were fabricated as a SERS sensor for measuring the SERS spectra. Second, the raw SERS spectra were preprocessed to facilitate the quantitative analysis. Third, a partial least squares model and four outstanding intelligent variable selection models, Monte Carlo-based uninformative variable elimination, competitive adaptive reweighted sampling, iteratively retaining informative variables, and variable iterative space shrinkage approach, were developed for detecting chlorpyrifos residue in a comparative study. The repeatability and reproducibility tests demonstrated the excellent stability of the proposed strategy. Furthermore, the sensitivity of the proposed strategy was assessed by estimating limit of detection values of the various models. Finally, two-tailed paired t-tests confirmed that the accuracy of the proposed strategy was equivalent to that of gas chromatography-mass spectrometry. Hence, the proposed method provides a promising strategy for detecting chlorpyrifos residue in tea.
Novel electrochemiluminescence sensing platform for ultrasensitive detection of malathion residue in tea based on SiO2NSs doped Luminol/AgNPs as a signal amplification strategy

Abstract:To address the potential hazards of organophosphorus pesticides (OPs) residues in tea, an electrochemiluminescence (ECL) aptasensor based on functionalized nanomaterials was constructed in this work. Firstly, gold nanoparticles (AuNPs) were attached on the surface of multi-walled carbon nanotubes (MWCNTs) by the constant potential electrodeposition to form a compound, and it was utilized to provide excellent immobilization sites for complementary DNA (cDNA). Subsequently, composite nanomaterials were synthesized by a one-pot method with aminated Luminol/silver nanoparticles@silica nanospheres (NH2-Luminol/Ag@SiO2NSs). Finally, NH2-Luminol/Ag@SiO2NSs was combined with a malathion aptamer (Apt) to obtain signal probes (SPs) for the construction of an aptasensor. The aptasensor had a wide linear range (1×10-3-1×103 ng/mL) and a low limit of detection (LOD) (0.3×10-3 ng/mL). It had the virtues of high sensitivity, wonderful stability and excellent specificity, which could be used for the detection of malathion residue in tea. The work provides a proven way for the construction of a rapid and ultrasensitive aptasensor with low-cost.
Development and application of gold immunostrip for the detection of acetamiprid residue in tea samples

Abstract:The development of a gold immunostrip for the rapid detection of acetamiprid residue in tea samples was described. It was based on the principle of direct competitive immunochromatography. The artificial antigen of acetamiprid was prepared for immunization, and a highly-sensitive monoclonal antibody was obtained with the titer of 1:10 000. Subsequently, the gold immunostrip was developed and the visual detection limit of acetamiprid was 10 ng/mL. The detection time was just 10 min. No cross reaction with other neonicotinoid pesticides, such as imidacloprid, thiamethoxam, and nitenpyram, was detected. Moreover, this method has been used to determine the acetamiprid residue in tea (green tea, black tea and extra-strong tea) by spiked recovery tests. And results indicated that this method could meet the detection requirement of Chinese MRL of acetamiprid (0.5 mg/kg) in tea. Also, it possessed some advantages such as high efficiency, convenience, and low cost. Overall, the newly-developed immunostrip can be used in field tests for rapid screening of fresh tea before tea harvest and the quality control of tea products.

Abstract:Tea infusion, as one of the most popular beverages, is favored by people around the world. However, tea plants accumulate heavy metals from the soil during growth, which are gradually transferred to the leaves and subsequently decomposed into the tea infusion after soaking in hot water. Long-term consumption of tea infusion containing heavy metals may lead to severe health issues, such as liver dysfunction and cancer. Therefore, rapid, precise, and highly sensitive detection of heavy metals in tea infusion becomes crucial for human health. Traditional methods, such as atomic absorption spectroscopy (AAS), suffer from long detection cycles, high costs, and the risk of secondary pollution from chemical extraction reagents. Thus, this paper presents a method, using laser-induced breakdown spectroscopy combined with electrospinning technology (LIBS-ES), to detect trace amounts of Cr and Cu in tea infusion. The electrospun nanofiber membranes (ENM) were modified with gold nanoparticles (AuNPs) and silver nanoparticles (AgNPs), achieving ultra-sensitive detection of Cr and Cu. The experiment results show that the limits of detection for Cr and Cu are 5 μg/L and 10 μg/L, respectively. Moreover, this method was successfully applied to the detection of Cr and Cu in seven different types of tea infusion, achieving recovery rates of 99 %-106 % and 99 %-108 %, respectively. The method demonstrated good performance even under the interference of other ions at various concentrations, providing a reference for the rapid and highly sensitive detection of heavy metals in tea infusion.

A highly selective and sensitive europium-organic framework sensor for the fluorescence detection of fipronil in tea

Abstract:A europium-based metal organic framework (Eu-TFPA-MOF) was used for the fluorescence detection of fipronil in green tea and oolong tea for the first time. The red fluorescence of Eu-TFPA-MOF could be quenched significantly by low concentration (0.24 mM) of fipronil, and the "turn off" process exhibited quick response time (2 min), high sensitivity and selectivity, low detection limits (4.4 nM) and wide linear range (0-0.15 mM). The mechanism of fluorescence quenching was mainly attributed to static quenching process and the competitive absorption of excitation energy. Besides, the spiked and recovery test indicated that Eu-TFPA-MOF could be used in the fluorescence detection of fipronil in real green tea and oolong tea sample and the process had the advantages of simple pretreatment and satisfactory recoveries (98.33-106.17 %). More importantly, a simple, portable and low-cost smartphone-assisted test strip were designed for the visual detection of fipronil in real tea samples. The detection platform will be beneficial for tea quality safety and human heath, and is expected to be applied in other agricultural product safety field.

Abstract:Tea plants absorb chromium-contaminated soil and water and accumulate in tea leaves. Hexavalent chromium (Cr6+) is a very toxic heavy metal; excessive intake of tea containing Cr6+ can cause serious harm to human health. A reliable and sensitive surface-enhanced Raman spectroscopy (SERS) method was developed using Au@Ag nanoparticles as an enhanced substrate for the determination of Cr6+ in tea. The Au@AgNPs coated with carbimazole showed a highly selective reaction to Cr6+ in tea samples through a redox reaction between Cr6+ and carbimazole. The Cr6+ in the contaminated tea sample reacted with methimazole—the hydrolysate of carbimazole—to form disulfide, which led to the decrease in the Raman intensity of the peak at 595 cm−1. The logarithm of the concentration of Cr6+ has a linear relationship with the Raman intensity at the characteristic peak and showed a limit of detection of 0.945 mg/kg for the tea sample. The carbimazole functionalized Au@AgNPs showed high selectivity in analyzing Cr6+ in tea samples, even in the presence of other metal ions. The SERS detection technique established in this study also showed comparable results with the standard ICP-MS method, indicating the applicability of the established technique in practical applications.

Abstract:Tea is an important economic crop and health beverage in China. The presence of heavy metal ions in tea poses a significant threat to public health. Here, we prepared an aptamer biosensor labelled with AIEgen nanospheres to detect Pb2+ in tea. The dsDNA modified by amino and phosphoric acid was combined with the carboxylated AIEgen NPs to form AIEgen-DNA with a fluorescence group, which was then fixed to the surface of Zr-MOFs to quench the fluorescence of AIEgen NPs. At the same time, PEG was added to remove nonspecific adsorption. Then Pb2+ was added to cut the DNA sequences containing the cutting sites, and AIEgen NPs and part of the DNA sequences were separated from the Zr-MOFs surface to recover the fluorescence. By comparing the fluorescence changes before and after adding Pb2+, the detection limit of Pb2+ can reach 1.70 nM. The fluorescence sensor was applied to detect Pb2+ in tea, and the detection results showed that the tea purchased on the market did not contain the concentration of Pb2+ within the detection range. This study provides new insights into monitoring food and agriculture-related pollutants based on fluorescent biosensors.
Detection of systemic pesticide residues in tea products at trace level based on SERS and verified by GC–MS

Abstract:Surface-enhanced Raman spectroscopy (SERS) has the potential to detect pesticide residues in agricultural products. However, some systemic pesticides, such as chlorpyrifos, can enter the plant tissue, and not just stay on the surface. Consequently, many SERS studies halted at practical application because of its complexity. In this work, SERS technology was used to detect chlorpyrifos residues in tea products at the semiquantitative level. A simple pretreatment method effectively avoided interference of other fluorescent substances, and all major peaks could be distinguished on the basis of a novel substrate. A principal component analysis algorithm was applied to form a regression model, and a nanogram detection limit was obtained. Furthermore, chlorpyrifos residues in the same tea products were also measured by gas chromatography–mass spectrometry, and the results show a small range of errors. From the comparative study of the two detection methods, the results suggest the great promise of SERS technology for rapid inspection of agricultural products.
Screening and Evaluation of Thiamethoxam Aptamer Based on Pressurized GO-SELEX and Its Sensor Application

Abstract:Thiamethoxam, a nicotinic pesticide used worldwide, can cause great harm to the environment and even to human health, and aptamers, known as chemical antibodies, have high affinity and specificity for the target, as well as great potential in detecting small molecules such as pesticides. In this paper, we report a highly sensitive biosensor system for thiamethoxam residue detection based on aptamer technology. After 15 rounds of screening with the pressurized GO-SELEX technology, we found that the aptamer libraries of the 5th and 9th rounds showed high affinity by the capture method. Four candidate aptamers were obtained by high-throughput sequencing and secondary structure prediction. Among them, the aptamer named Thi-5R-18 from the 5th round was demonstrated to possess the highest affinity by isothermal titration calorimetry, with a dissociation constant (Kd) of 4.935 × 10−5 M. The results of molecular docking showed that thiamethoxam and Thi-5R-18 were combined with bases G-15, A-19, and T-71 through hydrogen bonding and π–π interaction.Thi-5R-18 was used as a recognition element to construct a AuNPs colorimetric aptasensor, achieving an ultralow detection limit of 0.37 nM. More importantly, this colorimetric aptasensor can be used for quantitative detection of thiamethoxam on tea leaves, with a recovery of 96.94%~105.86%. This study provides a highly sensitive biosensor for detection of thiamethoxam residue.

Rapid Limit Test of Seven Pesticide Residues in Tea Based on the Combination of TLC and Raman Imaging Microscopy

Abstract:Numerous studies have shown that pesticide residues in tea exceeding the maximum residue limits (MRL) can cause harmful effects on the human body. There are many limitations in the existing analytical methods for pesticide residues in tea, so new analytical methods need to be developed. We developed a limit test method that combines thin-layer chromatography with Raman imaging microscopy (TLC-RIM). Seven residual pesticide components in tea (Avermectin, Methomyl, Carbendazim, Imidacloprid, Chlorothalonil, Azoxystrobin, and Acetamiprid) could be preliminarily separated by TLC and then irradiated by a 532 nm laser. Raman spectra of seven pesticides obtained by Raman imaging microscopy could be used to test whether the pesticide residues in tea exceed the MRL. The limits of detection of the seven pesticides were 0.04, 0.10, 0.24, 0.20, 0.12, 0.12, and 1.0 mg/mL, respectively. The simulated positive test showed that the matrix in tea did not interfere with the test of the seven pesticides. When the pesticides were tested within 8 h, the RSD of the peak heights of the seven pesticides were 1.2%~9.6%; the test results of three batches of tea showed that the imidacloprid in one batch of tea exceeded its MRL, and the results were consistent with that by UPLC-MS/MS. The TLC-RIM is fast, sensitive, stable, specific, and reliable.
