Abstract:Ascorbic acid, dopamine, and uric acid are important electroactive biomolecules for health monitoring and they coexist in serum or urine. Their quantitative determination by electrochemistry could provide the accurate reference for diseases diagnosis and treatment. However, the traditional electrochemical workstations are too large for on-field inspection. Hence, the design of handheld electrochemical system for the detection of biomolecules is significant for point-of-care testing (POCT). In this paper, a smartphone-based integrated voltammetry system using modified electrode was developed for simultaneous detection of biomolecules. The system contained a disposable sensor, a coin-size detector, and a smartphone equipped with application program. Screen-printed electrodes were used as sensors for the detection, on which reduced graphene oxide and gold nanoparticles were electrochemically deposited by the system. The detector was used with voltammetric methods, in which excitation voltage was applied on the sensors and subsequent current responses were detected. The smartphone is the core component to communicate with the detector, calculate data, and plot voltammograms in real-time. Then, the system was applied to detect standard solutions of the biomolecules and their mixtures as examples. The results showed that the peak currents of each substance increased with higher concentration and the method allowed the discrimination of the different potentials of the studied species. Finally, the practical applications of the system were tested through detections of the biomolecules in artificial urine. The results exhibited that the system could be used to detect electrochemical activity of biomolecules with linear, high sensitivity, and specific responses in point-of-care testing.

Abstract:Creatinine and albumin are crucial biomarkers for health monitoring and their ratio in urine is an effective approach for albuminuria assessment. Herein, to address the challenges of point-of-care and efficient analysis of the biomarkers simultaneously, we developed a fully integrated handheld smartphone-based photoelectrochemical biosensing system. A miniaturized printed circuit board included a potentiostat for photocurrent measurements and single-wavelength light-emitting diodes (LEDs) for photo-excitation, which was controlled with a Bluetooth-enabled smartphone. Graphitic carbon nitride (g-C3N4)/chitosan nanocomposites were modified on a transparent indium tin oxide (ITO) electrode as photoactive materials. Creatinine was detected through chelate formation with copper ion probes, while albumin was recognized specifically by an antigen-antibody reaction based on immunoassay. The biosensing system demonstrated good linearity and high sensitivity, with detection ranges of 100 μg mL-1 to 1500 μg mL-1 for creatinine, and 9.9 μg mL-1 to 500 μg mL-1 for albumin. Spiked artificial urine samples with different concentrations were tested to confirm the practical validity of the biosensing system, where an acceptable recovery rate ranged from 98.7% to 105.3%. This portable photoelectrochemical biosensing platform provides a convenient and cost-effective method for biofluid analysis, which has an extensive prospect in point-of-care testing (POCT) for mobile health.

Abstract:The paper-based analytical platforms have been applied in biochemical detection and environmental monitoring due to their low cost and easy operation. Despite their advantages, the complexity of fabrication has limited their further application. Here, a simple and innovative approach using parylene C, a material commonly used in semiconductors, is reported to fabricate paper-based chips and construct a point-of-care testing (POCT) platform through smartphones. Parylene C is deposited on the filter paper through a polydimethylsiloxane (PDMS) clamp and forms a hydrophobic barrier. A three-channel paper-based chip was utilized to detect aluminum (Al) ions on a smartphone platform by the chromogenic method. The color change after Al 3+ detection is represented with △E which was photographed and calculated by app on smartphone. The detection limit is 0.05 mg/L and the detection range is from 0.05 mg/L to 10 mg/L which indicates the application potential in the field of biochemical detection. This new paper-based sensing method and smartphone-based portable detection platform are simple, low-cost, fast, and highly sensitive, and provides a potential application for on-site, visual detection of ions and even other substances.

Abstract:Glaucoma is the second leading cause of blindness in the world. The ultimate cause of vision loss due to glaucoma is thought to be retinal ganglion cell (RGC) apoptosis. Neuroprotection of RGC is becoming an important approach of glaucoma therapy. Several lines of evidence suggest that estrogen has neurotrophic and neuroprotective properties. In this study, we examine the role of estrogen in preventing RGC loss in DBA/2J mouse, an in vivo model of an inherited (pigmentary) glaucoma. Two‐month‐old female DBA/2J mice were anesthetized and ovariectomized with or without subcutaneous 17β‐estradiol (βE2) pellet implantation. RGC survival was evaluated from flat‐mounted whole retinas by counting retrograde‐labeled cells. The loss of nerve fibers and RGC were also evaluated in paraffin‐fixed retinal cross sections. Biochemical alterations in the retinas of DBA/2J mice in response to systemic injection of βE2 were also examined. We have made several important observations showing that: (1) βE2 treatment reduced the loss of RGC and neurofibers through inhibition of ganglion cell apoptosis, (2) βE2 activated Akt and cAMP‐responsive‐element‐binding‐protein (CREB), (3) βE2 up‐regulated thioredoxin‐1 (Trx‐1) expression, (4) βE2 reduced the increased activations of mitogen‐activated protein kinases (MAPK) and NF‐κB, (5) βE2 inhibited the increased interleukin‐18 (IL‐18) expression, and (6) treatment with tamoxifen, an estrogen receptor antagonist, blocked βE2‐mediated activation of Akt and inhibition of MAPK phosphorylation in the retinas of DBA/2J mice. These findings suggest the possible involvement of multiple biochemical events, including estrogen receptor/Akt/CREB/thioredoxin‐1, and estrogen receptor/MAPK/NF‐κB, in estrogen‐mediated retinal ganglion cell protection. © 2007 Wiley Periodicals, Inc. Nanoplasmonic biosensors for multicolor visual analysis of acetylcholinesterase activity and drug inhibitor screening in point-of-care testing

Abstract:The monitoring of acetylcholinesterase (AChE) activity and the screening of its inhibitors are significance of the diagnosis and drug therapy of nervous diseases. A metal ions-mediated signal amplification strategy was developed for the highly sensitive and multicolor assay of AChE activity and visually screening its drug inhibitors. After the specific reaction between AChE and acetylthiocholine (ATCh), the hydrolysis product thiocholine (TCh) can directly and decompose the α-FeOOH nanorods (NRs) to release amounts of Fe2+, which was regarded as Fenton reagent to efficiently catalyze H2O2 to produce ·OH. Then, the as-formed ·OH can further largely shorten the gold nanobipyramids (Au NBPs), generating a series of palpable color variations. The linear range for AChE activity was 0.01-500.0 U/L with the limit of detection as low as 0.0074 U/L. The vivid visual effects could be easily distinguished for the multicolor assay of AChE activity by naked eye in visible light. To achieve the point-of-care testing, Au NBPs were further assembled on polymeric electrospun nanofibrous films (ENFs) surface as test strips for the easy-to-use test of AChE activity by RGB values with a smartphone. Fascinatingly, this proposed strategy can be used for the visual screening AChE inhibitors or non-inhibitors. Comparing with the clinical drugs (rivastigmine tartrate, and donepezil), some natural alkaloids such as evodiamine, caffeine, camptothecin, and berberine hydrochloride were selected as inhibitor modes to confirm the drug screening capability of this method. This proposed strategy may have great potential in the other disease-related enzymatic biomarkers assay and the rapid screening of drug therapy.

Abstract:The development of on-site, portable and visual assays for trace elements detection in tea is essential to ensure the health of the population in consideration of its widespread availability and most familiar consumption. Herein, a composite nano-enzyme (FeNPs@g-C3N4) based smartphone platform was proposed for sensing F- and Al3+. The FeNPs@g-C3N4 nano-enzyme with excellent peroxidase activity was synthesized through Fe nanoparticles (FeNPs) in situ growth on graphite-phase carbon nitride nanosheets (g-C3N4), which could catalyze H2O2 to oxidize TMB to blue oxide (oxTMB). The peroxidase activity of FeNPs@g-C3N4 nano-enzyme could be inhibited by F-, and then restored by Al3+ because of the coordination reaction between Fe3+/Al3+ and F-. Furthermore, based on the fact that the solution color could be dynamic manipulated by the concentrations of F- and Al3+,a smartphone was employed in visual detection by capturing and identifying the Red-Green-Blue (RGB) value of the solution images with detection limits of 2.43 nM and 7.66 μM. The cooperation of smartphone camera and color recognition application has greatly shortened the detection time and reduced the cost of detection, which provided a new strategy for the on-site and convenient detection of F- and Al3+ in tea, and showed significant application potential in food safety evaluation.

Abstract:On-site multiplex enzyme detection is crucial for diagnosis, therapeutics and prognostic. To date, it is still a daunting challenge to develop portable, low-cost, and efficient multi-enzyme detection methods. Herein, a novel sample-in-result-out platform integrating ratiometric fluorescent assays with 3D origami microfluidic paper-based device (μPAD) was developed for simultaneous visual point-of-care testing (POCT) of alkaline phosphatase (ALP) and butyrylcholinesterase (BChE). Cascade catalytic reaction with the same two fluorescent signal indicators was rationally designed to ratiometric fluorescent detection of ALP and BChE: substrate of ALP (pyrophosphate) and product of BChE (thiocholine) can strongly complex with Cu2+, Cu2+ oxidizes o-phenylenediamine to fluorescent 2,3-diaminophenazine (oxOPD) (emission, 565 nm), oxOPD quenches the fluorescence of carbon dots (CDs, emission at 445 nm) via inner filter effect, thus oxOPD/CDs values are relevant to ALP and BChE activities. Then 3D origami μPAD composing of four layers and two parallel channels was fabricated and simply prepared by one-step plotting with black oil-based marker and specific metal molds. After simple folding and unfolding neighboring layers to sequentially initiate reactions of pre-loaded reagents, fluorescent images on the detection zone can be captured by smartphone and analyzed by red-green-blue software for quantitative analysis. Under optimal conditions, the proposed platform was successfully performed to detect ALP and BChE with activity difference at 3 orders of magnitude in human serum samples without any pretreatment procedures. Excellent selectivity, good precision, favorable linear range, and high accuracy were exhibited. Importantly, the platform opens a promising horizon for high-throughput POCT of multiplex biomarkers.

Abstract:Achieving rapid, cost effective, and intelligent identification and quantification of flavonoids is challenging. For fast and uncomplicated flavonoid determination, a sensing platform of smartphone-coupled colorimetric sensor arrays (electronic noses) was developed, relying on the differential competitive inhibition of hesperidin, nobiletin, and tangeretin on the oxidation reactions of nanozymes with a 3,3',5,5'-tetramethylbenzidine substrate. First, density functional theory calculations predicted the enhanced peroxidase-like activities of CeO2 nanozymes after doping with Mn, Co, and Fe, which was then confirmed by experiments. The self-designed mobile application, Quick Viewer, enabled a rapid evaluation of the red, green, and blue values of colorimetric images using a multi-hole parallel acquisition strategy. The sensor array based on three channels of CeMn, CeFe, and CeCo was able to discriminate between different flavonoids from various categories, concentrations, mixtures, and the various storage durations of flavonoid-rich Citri Reticulatae Pericarpium through a linear discriminant analysis. Furthermore, the integration of a "segmentation-extraction-regression" deep learning algorithm enabled single-hole images to be obtained by segmenting from a 3 × 4 sensing array to augment the featured information of array images. The MobileNetV3-small neural network was trained on 37,488 single-well images and achieved an excellent predictive capability for flavonoid concentrations (R2 = 0.97). Finally, MobileNetV3-small was integrated into a smartphone as an application (Intelligent Analysis Master), to achieve the one-click output of three concentrations. This study developed an innovative approach for the qualitative and simultaneous multi-ingredient quantitative analysis of flavonoids.

Abstract:Multimodal biosensors with independent signaling pathways can self-calibrate and improve the reliability of disease biomarker detection. Herein, a colorimetric-fluorescent dual-mode paper-based biosensor with PAN/Fe(III)-CNOs (FPCs) as core components has been developed, which information is recognized by smartphone and naked eye. Using 1-(2-pyridylazo)-2-naphthol (PAN) as a mediator, Fe(III) is enriched on the surface of carbon nano-onions (CNOs), endowing FPCs with excellent mimetic enzyme activity and photothermal conversion ability, which allows it to output amplified colorimetric signals under laser irradiation. In addition, the complexation of PAN with Fe(III) broadens its absorption spectrum, which makes FPCs more suitable to be energy acceptors to quench fluorescence of polymer dots (Pdots), resulting in the changes of output fluorescent signal. Based on the above design, a portable colorimetric-fluorescent dual-mode biosensor is proposed for trypsin detection with Pdots as fluorescence sources and FPCs as fluorescence quenchers and nanoenzymes. This work provides a convenient way for constructing portable visual multimodal biosensors, which is expected to applied in various disease diagnosis.

Abstract:A ratiometric fluorescence sensing platform with smartphone was designed for highly sensitive detection of glutathione (GSH), ascorbic acid (AA), and alkaline phosphatase (ALP) activity by modulation the inner filter effect (IFE) between persistent luminescent nanoparticles (PLNPs) and 2, 3-diaminophenazine (DAP). As an oxidase-mimetic nanomaterial, MnO2 nanosheets (MnO2 NS) can oxidize o-phenylenediamine (OPD) to DAP, which exhibited a yellow fluorescence emission signal at 558 nm, whereas the fluorescence of PLNPs at 475 nm was quenched through IFE. The presence of GSH and AA reduced MnO2 NS to Mn2+, which inhibited the oxidation of OPD, and effectively preserved the blue fluorescence of PLNPs at 475 nm. Further, ALP can catalytically hydrolyze L-ascorbic acid 2-phosphate trisodium salt (AAP) to yield AA. Therefore, a simple, high sensitivity and reproducibility, and good selectivity ratiometric fluorescence sensing platform was fabricated to detect GSH, AA and ALP activity by measuring the ratio of the fluorescence intensity of DAP and PLNPs. The linear range for GSH, AA and ALP activity were 1-100 μM, and 1 to 5 mU/mL, and the limit of detection were 0.2 μM, 0.33 μM and 0.3 mU/mL, respectively. Finally, the proposed sensor was utilized in spiked human serum and urine samples detection with satisfactory recoveries. Moreover, the red-green-blue (RGB) intensities of a color image were acquired via scanning measurement analysis with the help of the color-scanning application on smartphones and portable UV lamps, enabling real-time and in-situ detection of ALP activity, GSH, and AA.

Smartphone-assisted hydrogel platform based on BSA-CeO2 nanoclusters for dual-mode determination of acetylcholinesterase and organophosphorus pesticides

Abstract:A dual-mode sensor was developed for detecting acetylcholinesterase (AChE) and organophosphorus pesticides (OPs) via bifunctional BSA-CeO2 nanoclusters (NCs) with oxidase-mimetic activity and fluorescence property. The dual-mode sensor has the characteristics of self-calibration and self-verification, meeting the needs of different detection conditions and provide more accurate results. The colorimetric sensor and fluorescence sensor have been successfully used for detecting AChE with limit of detection (LOD) of 0.081 mU/mL and 0.056 mU/mL, respectively, while the LOD for OPs were 0.9 ng/mL and 0.78 ng/mL, respectively. The recovery of AChE was 93.9-107.2% and of OPs was 95.8-105.0% in actual samples. A novel strategy was developed to monitor pesticide residues and detect AChE level, which will motivate future work to explore the potential applications of multifunctional nanozymes.

Abstract:Amantadine (AMD) is an antiviral drug used to treat animal-based infectious diseases such as avian influenza. However, its illegal use in animal-derived food industries endangers human health. Therefore, an efficient, portable, and visual sensor to detect AMD in food is crucial for ensuring food safety. A multifunctional nanocomposite NH2-UiO-66@PtNPs was exploited to design a smartphone-assisted fluorescence-colorimetric immunosensing microarray for multidimensional detection of AMD in animal-derived foods. Platinum nanoparticles (NPs) were immobilized on a Zr-based metal–organic framework (NH2-UiO-66). Pt NPs exhibited peroxidase-like activity, whereas NH2-UiO-66 stabilized Pt NPs and improved the colorimetric signal of the sensor. Moreover, the fluorescence ligand NH2-BDC in NH2-UiO-66 was released via alkaline hydrolysis, amplifying the fluorescence signal and improving detection sensitivity. The smartphone-assisted color recognition system accurately identified the color changes of the microarray. The immunocompetitive reaction of AMD and the antibody (Ab) in the microarray NH2-UiO-66@PtNPs@Ab2 allowed the quantitative and visual detection of AMD in a wide concentration range (0.1–1000 ng/mL) with high sensitivity [limit of detection = 0.0022 ng/mL (fluorescence), 0.069 ng/mL (colorimetric), and 0.68 ng/mL (smartphone)]. This detection strategy was verified in AMD-spiked food samples with satisfactory recoveries (82.0–103.0%) and stability (RSD: ≤7.1%, n = 3), showing its great potential for visual screening and portable detection of other food contaminants.

Novel "turn on-off" paper sensor based on nonionic conjugated polythiophene-coated CdTe QDs for efficient visual detection of cholinesterase activity

Abstract:An increasing number of patients are living with Alzheimer's disease (AD); thus, the need for a method to detect AD early and sensitively has become urgent, and the demand for an intelligent analytical platform is growing year by year. Abnormal levels of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are known to be indicative of AD. In this work, a novel conjugated polythiophene (CP) compound was successfully combined with CdTe quantum dots (QDs) to improve their selectivity and sensitivity. The QDs successfully enabled the detection of low concentrations of AChE by turning on the fluorescence of the CdTe/CP via the interaction between CP and thiocholine produced by ATCh hydrolysis and aggregation induced emission enhancement (AIEE). Under optimal conditions, we reached a low detection limit of 0.14 U L-1, which is 7.9 times lower than that of pristine QDs. More importantly, an efficient, inexpensive, and disposable paper-based platform, which allows the efficient visual detection of AChE activity via the color variation of CdTe/CP, was designed. Moreover, the accuracy of the method was demonstrated by conducting a recovery test in human serum, in which the recoveries reached 107% and 110%, proving that CdTe/CP has considerable potential to be used for analyzing real biological samples. The advantages of this method are its simplicity, fast detection capability, affordability, and the fact that it can be used for on-site detection of AChE activity. Furthermore, it has certain guiding significance for detecting AD.

Abstract:When a semipreparative capillary electrochromatography (CEC) capillary is packed with silica particles and exposed to solvent, its mechanical strength is markedly reduced. In our studies, a fused‐silica capillary (internal diameter > 200 μm and wall thickness < 150 μm) was packed under pressure (approximately 200 psi) with spherical silica particles (1.5—5 μm) suspended in water or various common organic solvents. After one hour of exposure, the capillary can be readily deformed, and it keeps its deformed shape upon release of the force causing deformation. It is suggested that capillary softening is promoted through the propagation of internal microcracks that have been caused by action of the particles during packing in the presence of solvent. Application of a protective coating to the inside of the capillary is found to reduce or eliminate capillary softening.
Paper-based colorimetric sensor using bimetallic Nickel-Cobalt selenides nanozyme with artificial neural network-assisted for detection of H2O2 on smartphone

Abstract:Paper-based analytical devices (PADs) integrated with smartphones have shown great potential in various fields, but they also face challenges such as single signal reading, complex data processing and significant environmental impacting. In this study, a colorimetric PAD platform has been proposed using bimetallic nickel-cobalt selenides as highly active peroxidase mimic, smartphone with 3D-printing dark-cavity as a portable detector and an artificial neural network (ANN) model as multi-signal processing tool. Notably, the optimized nickel-cobalt selenides (Ni0.75Co0.25Se with Ni to Co ratio of 3/1) exhibit excellent peoxidase-mimetic activities and are capable of catalyzing the oxidation of four chromogenic reagents in the presence of H2O2. Using a smartphone with image capture function as a friendly signal readout tool, the Ni0.75Co0.25Se based four channel colorimetric sensing paper is used for multi-signal quantitative analysis of H2O2 by determining the Grey, red (R), green (G) and blue (B) channel values of the captured pictures. An intelligent on-site detection method for H2O2 has been constructed by combining an ANN model and a self-programmed easy-to-use smartphone APP with a dynamic range of 5 μM to 2 M. Noteworthy, machine learning-assisted smartphone sensing devices based on nanozyme and 3D printing technology provide new insights and universal strategies for visual ultrasensitive detection in a variety of fields, including environments monitoring, biomedical diagnosis and safety screening.
Accelerated and precise identification of antioxidants and pesticides using a smartphone-based colorimetric sensor array

Abstract:The integration of smartphones with conventional analytical approaches plays a crucial role in enhancing on-site detection platforms for point-of-care testing. Here, we developed a simple, rapid, and efficient three-channel colorimetric sensor array, leveraging the peroxidase (POD)-like activity of polydopamine-decorated FeNi foam (PDFeNi foam), to identify antioxidants using both microplate readers and smartphones for signal readouts. The exceptional catalytic capacity of PDFeNi foam enabled the quick catalytic oxidation of three typical peroxidase substrates (TMB, OPD and 4-AT) within 3 min. Consequently, we constructed a colorimetric sensor array with cross-reactive responses, which was successfully applied to differentiate five antioxidants (i.e., glycine (GLY), glutathione (GSH), citric acid (CA), ascorbic acid (AA), and tannic acid (TAN)) within the concentration range of 0.1–10 μM, quantitatively analyze individual antioxidants (with AA and CA as model analytes), and assess binary mixtures of AA and GSH. The practical application was further validated by discriminating antioxidants in serum samples with a smartphone for signal readout. In addition, since pesticides could be absorbed on the surface of PDFeNi foam through π-π stacking and hydrogen bonding, the active sites were differentially masked, leading to featured modulation on POD-like activity of PDFeNi foam, thereby forming the basis for pesticides discrimination on the sensor array. The nanozyme-based sensor array provides a simple, rapid, visual and high-throughput strategy for precise identification of various analytes with a versatile platform, highlighting its potential application in point-care-of diagnostic, food safety and environmental surveillance.

Abstract:Immunoassays such as the enzyme-linked immunosorbent assay (ELISA) are utilized extensively for detecting protein biomarkers and small molecules in healthcare, environmental monitoring, and food analysis. Unfortunately, the current strategies for immunoassays often require sophisticated apparatus such as a microplate reader, which might not be available in resource-limited areas. To mitigate this problem, we designed a compact smartphone based-device and a multicolor response immunosensor. First, we designed a compact and cost-effective 3D-printed attachment, where a light-emitting diode was used as a light excitation source and a smartphone captured the fluorescent emission signals. Second, by combining quantum dots hybrid and chemical redox reaction, multiple color responses were displayed in the presence of the analyte at different concentrations. Third, solutions with distinct tonality could be readily distinguished by the naked eye and they were suitable for quantitative analysis using the hue-saturation-lightness color space based on a smartphone application. The versatility of the proposed sensing system was demonstrated by implementing an indirect competitive ELISA for analyzing trace drug residues in foodstuffs. The multicolor response of this sensing strategy allows us to visually quantify drug residues in foodstuffs. Moreover, the smartphone-based immunosensor can assess the exact concentration of the analyte by using a self-designed mobile application. The proposed assay provides a highly sensitive performance that the limit of detection was 0.37 ng/mL by visual detection and 0.057 ng/mL using the compact device. Due to its advantages in terms of portability, straightforward visual detection, high sensitivity, and cost effectiveness, the proposed immunosensor has great potential for applications in areas without access to laboratories or expensive infrastructure.
Smartphone-assisted paper-based multifunctional assay for on-site tandem detection of inorganic phosphate, neutral red and heparin

Abstract:Today, easy availability and efficient detection assays in low-resource settings are mandatory. Therefore, a smartphone-assisted paper-based detection strategy using eosin Y (EY)-embed ZIF-8 nanocomposite for phosphate (Pi), neutral red (NR) and heparin (HP) on site testing was fabricated. After adding the targets in a specific order, the prepared strips produced brilliant color transitions (astonishing colors, ranging from pistachio to bright yellow for Pi, from bright yellow to jacinth for NR and from jacinth to light orange for HP). These variation on tonality can be evaluated by the smartphone-based readout device (Color Assist App) through the ratio change of B/(R + G + B), which exhibits superior performance of on-site detection in wastewater, including high sensitivity (LOD is 0.015, 0.031 and 0.014 mM for Pi, NR and HP, respectively), ultra-fast response time and excellent selectivity. This strips and smartphone dual-mode monitoring strategy may offer a convenient idea to identify Pi, NR and HP, with great prospects for environmental wastewater applications. Additionally, the probe was enabled for bioimaging of Pi, NR and HP in living cells. Finally, the related anti-counterfeiting fluorescent ink of probe was proposed on schedule.

Abstract:The colorimetric sensor usually depends on enzyme-mediated signal amplification to achieve trace analysis of ochratoxin A (OTA) residues in food samples. However, the enzyme labeling and manual addition of reagents steps increased assay time and operation complexity, restricting their application in point-of-care testing (POCT). Herein, we report a label-free colorimetric device integrating a 3D paper-based analytical device and a smartphone as handheld readout for rapid and sensitive detection of OTA. Using vertical-flow design, the paper-based analytical device enables the specific recognition of target and self-assembly of G-quadruplex (G4)/hemin DNAzyme to be performed, then employs DNAzyme for transducing the OTA binding event signal into a colorimetric signal. The design of independent functional units, including biorecognition unit, self-assembly unit and colorimetric units, which can address crowding and disorder of biosensing interfaces and improve the recognition efficiency of aptamer (apta). In addition, we eliminated signal losses and nonuniform coloring by introducing carboxymethyl chitosan (CMCS) to obtain perfectly focused signals on colorimetric unit. On the basis of parameter optimization, the device exhibited a detection range of 0.1–500 ng/mL and a detection limit of 41.9 pg/mL for OTA. Importantly, good results were obtained in spiked real samples, indicating applicability and reliability of developed device.
