Abstract:On-site multiplex enzyme detection is crucial for diagnosis, therapeutics and prognostic. To date, it is still a daunting challenge to develop portable, low-cost, and efficient multi-enzyme detection methods. Herein, a novel sample-in-result-out platform integrating ratiometric fluorescent assays with 3D origami microfluidic paper-based device (μPAD) was developed for simultaneous visual point-of-care testing (POCT) of alkaline phosphatase (ALP) and butyrylcholinesterase (BChE). Cascade catalytic reaction with the same two fluorescent signal indicators was rationally designed to ratiometric fluorescent detection of ALP and BChE: substrate of ALP (pyrophosphate) and product of BChE (thiocholine) can strongly complex with Cu2+, Cu2+ oxidizes o-phenylenediamine to fluorescent 2,3-diaminophenazine (oxOPD) (emission, 565 nm), oxOPD quenches the fluorescence of carbon dots (CDs, emission at 445 nm) via inner filter effect, thus oxOPD/CDs values are relevant to ALP and BChE activities. Then 3D origami μPAD composing of four layers and two parallel channels was fabricated and simply prepared by one-step plotting with black oil-based marker and specific metal molds. After simple folding and unfolding neighboring layers to sequentially initiate reactions of pre-loaded reagents, fluorescent images on the detection zone can be captured by smartphone and analyzed by red-green-blue software for quantitative analysis. Under optimal conditions, the proposed platform was successfully performed to detect ALP and BChE with activity difference at 3 orders of magnitude in human serum samples without any pretreatment procedures. Excellent selectivity, good precision, favorable linear range, and high accuracy were exhibited. Importantly, the platform opens a promising horizon for high-throughput POCT of multiplex biomarkers.
New Thiocarbonyl Ylides Derived from 3,3-Dichloro-2,2,4,4-tetramethylcyclobutanethione; Generation and Reactions.

Abstract:The reaction of 3,3-dichloro-2,2,4,4-tetramethylcyclobutanethione (1) with diazo compounds yielded spirocyclic 2,5-dihydro-1,3,4-thiadiazoles 3, which decomposed at ca. 45°C to give the corresponding thiocarbonyl ylide of type 4. In the absence of trapping agents, these thiocarbonyl ylides underwent a 1,3-dipolar electrocyclization to yield spirocyclic thiiranes 5. On the other hand, the thiocarbonyl methanide 4a was efficiently intercepted with C C, C=C, C=O, C=S, and N=N dipolarophiles leading to the [2+3] cycloadducts. A non-stereoselective cycloaddition took place when 3a was decomposed in the presence of the very electron-deficient dicyanofumarate or maleate, indicating a two step mechanism via an intermediate zwitterion. Furthermore, the thiocarbonyl methanide 4a could be trapped by the imidazole-2-thione 7 to give the 1,3-adduct 8. Treatment of 3a with secondary amines led to amidrazones of type 25 via base-catalyzed ring opening and condensation reaction.
Ultrasensitive Enzyme-free Biosensor by Coupling Cyclodextrin Functionalized Au Nanoparticles and High Performance Au-Paper Electrode.

Abstract:Microfluidic paper-based analytical device (μPAD), originally developed for improving healthcare in developing countries, presents a simple yet powerful platform for performing low-cost and portable diagnostic devices. Here, we reported an enzyme-free μPAD for the detection of two tumor markers. Firstly, a porous structure of gold nanoparticles (AuNPs) modified paper working electrode (Au-PWE), with a feature of all-around conductivity and plenty of active sites favoring for the biological ligands attachment, was firstly fabricated as a sensor substrate. Next cyclodextrin functionalized AuNPs (CD@AuNPs) as dual mimicking enzyme was prepared to load secondary antibodies or peptide. One the one sample zone, in the presence of carcinoembryonic antigen (CEA), CD@AuNPs could be introduced into the Au-PWE through a sandwich immunoreaction, boosting the electrochemical signal of o-phenylenediamine (o-PD) via the trigger of a cascade catalysis reaction towards glucose & o-PD, eventually resulting in a sensitive detection of CEA. On another working zone, with the introduction of another target prostate specific antigen (PSA), the peptide cleavage took place, which further led to the CD@AuNPs released from Au-PWE, and then the variation of electrochemical signals were recorded for detection of PSA. We demonstrated, using the device, the detection of CEA and PSA clinically had high sensitivity, wide linear ranges and low detection limits. We believe that our work provides a promising platform for point-of-care testing, especially in resource-limited regions.

Abstract:Herein, a simple microfluidic paper-based analytical device (μPAD) by using platinum nanoparticles (Pt NPs) as highly active peroxidase mimic for simultaneous determination of glucose and uric acid was fabricated. The μPAD consisted of one sample transportation layer, four paper-based detection chips, and two layers of hydrophobic polyethylene terephthalate (PET) films. The four detection chips were immobilized with various chromogenic reagents, Pt NPs, and specific oxidase (glucose oxidase or uricase). H2O2 generated by specific enzymatic reactions could oxidize co-immobilized chromogenic reagents to produce colored products by using Pt NPs as efficient catalyst. The multi-layered structure of μPAD could effectively improve the color uniformity and color intensity. Total color intensity from each two detection chips modified with distinct chromogenic reagents were used for quantitative analysis of glucose and uric acid, respectively, resulting in significantly improved sensitivity. The linear range for glucose and uric acid detection was 0.01–5.0 mM and 0.01–2.5 mM, respectively. Satisfied results were obtained for glucose and uric acid detection in real serum samples. An easy-to-use smartphone APP was developed for convenient and intelligent detection. The developed μPAD integrated with smartphone as detector holds great applicability for simple and portable on-site analysis.

Abstract:A low-cost, simple and sensitive colorimetric detection method for alkaline phosphatase (ALP) was developed on microfluidic paper-based analysis devices. In a typical colorimetric detection, 5-bromo-4-chloro-3-indolyl phosphate disodium salt (BCIP) and nitro blue tetrazolium (NBT) were firstly added onto the circle array zones of paper devices. Then ALP solutions were spotted to the array zones to perform the colorimetric reaction. The color reaction results were recorded by both Gel Documentation systems and a common digital camera, and analyzed with Quantity One software. All the reaction conditions were optimized. Under the optimal conditions, the colorimetric intensity showed a linear correlation to the concentration of ALP in the range of 1.5–20 U L−1 with a limit of detection (LOD) of 0.78 U L−1 (3σ), which was about two orders of magnitude lower than that of the reported method on µPADs. Besides, it was applied in the analysis of spiked real sample with a satisfactory result. In addition, the dual-color array-based paper strip was fabricated for the semiquantitative detection of ALP. The approximate activity of ALP could be simply distinguished by the color change with naked eyes.

Abstract:Glaucoma is the second leading cause of blindness in the world. The ultimate cause of vision loss due to glaucoma is thought to be retinal ganglion cell (RGC) apoptosis. Neuroprotection of RGC is becoming an important approach of glaucoma therapy. Several lines of evidence suggest that estrogen has neurotrophic and neuroprotective properties. In this study, we examine the role of estrogen in preventing RGC loss in DBA/2J mouse, an in vivo model of an inherited (pigmentary) glaucoma. Two‐month‐old female DBA/2J mice were anesthetized and ovariectomized with or without subcutaneous 17β‐estradiol (βE2) pellet implantation. RGC survival was evaluated from flat‐mounted whole retinas by counting retrograde‐labeled cells. The loss of nerve fibers and RGC were also evaluated in paraffin‐fixed retinal cross sections. Biochemical alterations in the retinas of DBA/2J mice in response to systemic injection of βE2 were also examined. We have made several important observations showing that: (1) βE2 treatment reduced the loss of RGC and neurofibers through inhibition of ganglion cell apoptosis, (2) βE2 activated Akt and cAMP‐responsive‐element‐binding‐protein (CREB), (3) βE2 up‐regulated thioredoxin‐1 (Trx‐1) expression, (4) βE2 reduced the increased activations of mitogen‐activated protein kinases (MAPK) and NF‐κB, (5) βE2 inhibited the increased interleukin‐18 (IL‐18) expression, and (6) treatment with tamoxifen, an estrogen receptor antagonist, blocked βE2‐mediated activation of Akt and inhibition of MAPK phosphorylation in the retinas of DBA/2J mice. These findings suggest the possible involvement of multiple biochemical events, including estrogen receptor/Akt/CREB/thioredoxin‐1, and estrogen receptor/MAPK/NF‐κB, in estrogen‐mediated retinal ganglion cell protection. © 2007 Wiley Periodicals, Inc. Abstract:Multimodal biosensors with independent signaling pathways can self-calibrate and improve the reliability of disease biomarker detection. Herein, a colorimetric-fluorescent dual-mode paper-based biosensor with PAN/Fe(III)-CNOs (FPCs) as core components has been developed, which information is recognized by smartphone and naked eye. Using 1-(2-pyridylazo)-2-naphthol (PAN) as a mediator, Fe(III) is enriched on the surface of carbon nano-onions (CNOs), endowing FPCs with excellent mimetic enzyme activity and photothermal conversion ability, which allows it to output amplified colorimetric signals under laser irradiation. In addition, the complexation of PAN with Fe(III) broadens its absorption spectrum, which makes FPCs more suitable to be energy acceptors to quench fluorescence of polymer dots (Pdots), resulting in the changes of output fluorescent signal. Based on the above design, a portable colorimetric-fluorescent dual-mode biosensor is proposed for trypsin detection with Pdots as fluorescence sources and FPCs as fluorescence quenchers and nanoenzymes. This work provides a convenient way for constructing portable visual multimodal biosensors, which is expected to applied in various disease diagnosis.

Abstract:Paper-based analytical devices (PADs) integrated with smartphones have shown great potential in various fields, but they also face challenges such as single signal reading, complex data processing and significant environmental impacting. In this study, a colorimetric PAD platform has been proposed using bimetallic nickel-cobalt selenides as highly active peroxidase mimic, smartphone with 3D-printing dark-cavity as a portable detector and an artificial neural network (ANN) model as multi-signal processing tool. Notably, the optimized nickel-cobalt selenides (Ni0.75Co0.25Se with Ni to Co ratio of 3/1) exhibit excellent peoxidase-mimetic activities and are capable of catalyzing the oxidation of four chromogenic reagents in the presence of H2O2. Using a smartphone with image capture function as a friendly signal readout tool, the Ni0.75Co0.25Se based four channel colorimetric sensing paper is used for multi-signal quantitative analysis of H2O2 by determining the Grey, red (R), green (G) and blue (B) channel values of the captured pictures. An intelligent on-site detection method for H2O2 has been constructed by combining an ANN model and a self-programmed easy-to-use smartphone APP with a dynamic range of 5 μM to 2 M. Noteworthy, machine learning-assisted smartphone sensing devices based on nanozyme and 3D printing technology provide new insights and universal strategies for visual ultrasensitive detection in a variety of fields, including environments monitoring, biomedical diagnosis and safety screening.
Novel "turn on-off" paper sensor based on nonionic conjugated polythiophene-coated CdTe QDs for efficient visual detection of cholinesterase activity

Abstract:An increasing number of patients are living with Alzheimer's disease (AD); thus, the need for a method to detect AD early and sensitively has become urgent, and the demand for an intelligent analytical platform is growing year by year. Abnormal levels of acetylcholinesterase (AChE) and butyrylcholinesterase (BChE) are known to be indicative of AD. In this work, a novel conjugated polythiophene (CP) compound was successfully combined with CdTe quantum dots (QDs) to improve their selectivity and sensitivity. The QDs successfully enabled the detection of low concentrations of AChE by turning on the fluorescence of the CdTe/CP via the interaction between CP and thiocholine produced by ATCh hydrolysis and aggregation induced emission enhancement (AIEE). Under optimal conditions, we reached a low detection limit of 0.14 U L-1, which is 7.9 times lower than that of pristine QDs. More importantly, an efficient, inexpensive, and disposable paper-based platform, which allows the efficient visual detection of AChE activity via the color variation of CdTe/CP, was designed. Moreover, the accuracy of the method was demonstrated by conducting a recovery test in human serum, in which the recoveries reached 107% and 110%, proving that CdTe/CP has considerable potential to be used for analyzing real biological samples. The advantages of this method are its simplicity, fast detection capability, affordability, and the fact that it can be used for on-site detection of AChE activity. Furthermore, it has certain guiding significance for detecting AD.
Integration of target responsive hydrogel with cascaded enzymatic reactions and microfluidic paper-based analytic devices (µPADs) for point-of-care testing (POCT)

Abstract:Paper based microfluidics (µPADs) with advantages of portability, low cost, and ease of use have attracted extensive attention. Here we describe a novel method that integrates glucoamylase-trapped aptamer-crosslinked hydrogel for molecular recognition with cascaded enzymatic reactions for signal amplification and a µPAD for portable readout. Upon target introduction, the hydrogel decomposes to release glucoamylase, which catalyzes the hydrolysis of amylose to produce a large amount of glucose. With a simple folding of the µPAD, the sample solution containing glucose product wicks and diffuses in parallel to each test-zone to carry out homogeneous assays, where glucose is used to produce I2 for brown color visualization through multiple enzymatic and chemical cascade reactions. Through color gradient changes based on different concentrations of the target, a semiquantitative assay is achieved by the naked eye, and quantitation can be obtained by handheld devices. Detection of cocaine in buffer and urine was performed to demonstrate the utility of the hydrogel-µPAD system. More importantly, the hydrogel-µPAD system can be extended to the detection of various targets by incorporating the corresponding aptamer into the hydrogel. The hydrogel-µPAD system reported here provides a new platform for portable, disposable and visual detection of a wide range of targets.

Abstract:The development of paper-based sensors with the distance readout is particularly attractive for the point-of-care testing (POCT) of biomarkers. Herein, we report a paper-based sensor that can convert the viscosity change caused by the hydrogel degradation into the variance of water flow distance on the paper. The detection of alkaline phosphatase (ALP) and alpha-fetoprotein (AFP) is demonstrated as examples. Copper alginate (Cu-Alg) hydrogel can be formed when mixing the aqueous solutions of Cu 2+ and alginate. The introduction of pyrophosphate ion (PPi) induces the gel-sol transition due to the high affinity between Cu 2+ and PPi. However, in the presence of ALP, the gel-sol transition is prevented due to the hydrolysis of PPi by ALP. In these processes, the viscosity changes of the hydrogels are translated into the differences of the flow distances on the pH test papers. In addition, the tilt of the papers can increase the accuracy of detection. The detection of AFP is also fulfilled via the construction of the paper-based immunosensor using the ALP-linked AFP detection antibody. The developed paper-based sensors show excellent performance for the quantitative detection of biomarkers in real samples with remarkable selectivity. Overall, this convenient method has great potential in POCT applications.

Abstract:Fabrication of a multi-signal readout assay with high sensitivity and selectivity is highly desirable for clinical and biochemical analysis, but remains a challenge due to laborious procedures, large-scale instruments, and inadequate accuracy. Herein, a straightforward, rapid, and portable detection platform based on palladium(II) methylene blue (MB) coordination polymer nanosheets (PdMBCP NSs) was unveiled for the ratiometric dual-mode detection of alkaline phosphatase (ALP) with temperature and colorimetric signal readout properties. The sensing mechanism is the ALP-catalyzed generation of ascorbic acid for competitive binding and etching PdMBCP NSs to release free MB in a quantitive means for detection. Specifically, ALP addition led to the decrease of temperature signal readout from the decomposed PdMBCP NSs under 808 nm laser excitation, and simultaneous increase of the temperature from the generated MB with a 660 nm laser, together with the corresponding absorbance changes at both wavelengths. Notably, this ratiometric nanosensor exhibited a detection limit of 0.013 U/L (colorimetric) and 0.095 U/L (photothermal) within 10 min, respectively. The reliability and satisfactory sensing performance of the developed method were further confirmed by clinic serum samples. Therefore, this study provides a new insight for the development of dual-signal sensing platforms for convenient, universal, and accurate detection of ALP.

Abstract:Alkaline phosphatase (ALP) is a phosphomonoester hydrolase and serves as a biomarker in various diseases. However, current detection methods for ALP rely on bulky instruments, extended time, and complex operations, which are particularly challenging in resource-limited regions. Herein, we synthesized a MOF-derived Fe-N-C nanozyme to create biosensors for the coulometric and visual detection of ALP. Specifically, we found the Fe-N-C nanozyme can efficiently oxidize 3,3′,5,5′-tetramethylbenzidine (TMB) to generate blue-colored tetramethyl benzidine (TMBox) without the need for H2O2. To construct the biosensor, we incorporated the ALP enzymatic catalytic reaction to inhibit the oxidation of TMB by Fe-N-C oxidase nanozyme. This biosensor showed rapid and highly sensitive detection of ALP in both buffer and clinical samples. The limit of detection (LOD) of our approach could be achieved at 3.38 U L−1, and the linear range was from 5 to 60 U L−1. Moreover, we also developed a visual detection for ALP by using a smartphone-based assay and facilitated practical and accessible point-and-care testing (POCT) in resource-limited areas. The visual detection method also achieved a similar LOD of 2.12 U L−1 and a linear range of 5–60 U L−1. Our approach presents potential applications for other biomarker detections by using ALP-based ELISA methods.

Hydrogel Paper-Based Analytical Devices: Separation-Free In Situ Assay of Small-Molecule Targets in Whole Blood.

Abstract:While colorimetric-based assays are very convenient to determine biomarkers in point-of-care testing (POCT), they often suffer from pretreatment procedures for separation of plasma or serum from whole blood samples. Here, we report a simple colorimetric paper-based analytical device (c-PAD) that is capable of performing sample-to-answer analysis by directly dropping the whole blood sample on paper. This is accomplished by utilizing sodium alginate hydrogel, which exhibits a nanometer-scale porous structure to effectively prevent the passage of large red blood cells and hemoglobin molecules, to encapsulate enzymes and chromogenic reagents. As the small targets in the blood sample enter the sensing region to trigger a chromogenic reaction, the resulting color signal is recorded by a smartphone. The interference from the red blood to the color signal can be completely avoided without the requirement of any separation process. The analytical performance of the method is evaluated by assaying glucose in real blood samples. The results show that rapid and accurate analysis can be achieved with the limit of detection as low as 0.12 mM. In addition, simultaneous detection of different targets (glucose, cholesterol, and triglycerides) in whole blood can be achieved by fabricating c-PAD with multiple sensing regions. Owing to its several essential advantages including an extremely simple procedure for fabrication, sample-to-answer analysis without tedious pretreatment, and capability to perform high-throughput analysis, the proposed c-PAD will be of great value in POCT applications of whole blood samples.

In this system, cobalt hydroxide (CoOOH) nanoflakes can oxidize O-phenylenediamine (OPD) to produce 2,3-diaminophenazine (OxOPD), resulting in a strong fluorescence at 565 nm and an absorbance at 420 nm, respectively. The ascorbic acid 2-phosphate (AAP) can be hydrolyzed by alkaline phosphatase (ALP) to yield ascorbic acid (AA). Then, AA reduces the CoOOH nanoflakes to produce Co2+, and AA is oxidized to form dehydroascorbic acid (DHAA), thereby inhibiting the formation of OxOPD. The reaction product DHAA further combines with OPD to yield 3-(1,2-dihydroxyethyl)furo[3,4-b]quinoxalin-1(3H)-one (DFQ) accompanied by a strong fluorescence at 430 nm. Based on this, the fluorometric assay for ALP has a wide linear range from 0.8 to 190 U/L with a low detection limit of 0.16 U/L, and the colorimetric assay from 3 to 130 U/L with a detection limit of 1.94 U/L. Moreover, a portable smartphone sensing platform integrated with fluorescent and colorimetric signals was established for rapid determination of ALP without spectrometers. Recoveries of 97-104% for spiked samples and relative standard deviations (RSD) of less than 2% (n = 3) confirmed the feasibility of the developed platform in complicated samples, opening up new horizons for on-site evaluation in the biomedical field.

Abstract:A novel colorimetric and fluorescent dual-mode sensing system based on molybdophosphoric heteropoly acid (PMA) and fluorescent microspheres (FMs) was established for monitoring the activity of alkaline phosphatase (ALP). In the presence of ALP, L-ascorbic acid-2-phosphate (AAP) could be hydrolyzed catalytically to ascorbic acid (AA), which could reduce PMA to phosphorus molybdenum blue (PMB), accompanied by the generation of colorimetric signals depending on the level of ALP. Meanwhile, the fluorescence of FMs was quenched markedly by the PMB produced due to the inner-filter effect, which constituted the response mechanism for the dual-mode sensing systems of ALP. On this basis, a PMA-FMs based dual-mode sensing system was used for the detection of ALP, which not only possessed remarkable sensitivity, with a limit of detection of 0.27 U L-1 and 0.11 U L-1, but also exhibited good analytical performance in biological samples with satisfactory results. Moreover, a simple and portable test kit for the visual detection of ALP in real serum samples was fabricated utilizing a smartphone with image-recognition and data-processing capabilities as a visual-detection platform.

Abstract:Alkaline phosphatase (ALP), as an immunological label, is widely used in biochemical assays. Here, a simple yet effective strategy for ALP activity detection was proposed on the basis of in situ formation of Prussian blue nanoparticles and polychromatic superposition effect. Firstly, ascorbic acid, a product from ALP-catalyzed hydrolysis of 2-phospho-L-ascorbic acid (AAP), converted yellow ferricyanide into ferrocyanide. Then, the specific reaction between ferrocyanide and ferric ions (Fe3+) initiated the generation of Prussian blue nanoparticles in situ. Meanwhile, the residual AAP chelated with Fe3+, and a stable Fe3+-AAP complex was quickly formed. When Prussian blue nanoparticles mixed with brown Fe3+-AAP complex and yellow ferricyanide at different ratios, a distinct color variation was presented. Therefore, a sensitive multicolor assay of ALP activity with a detection limit of 1.0 U/L was realized by simply blending commercially available reagents. Furthermore, magnetic sandwich and competitive sensing platforms for multiple biomarkers detection were constructed by combining the ALP-regulated multicolor system with the well-developed aptasensor. The feasibility of the sensors was convincingly demonstrated by using thrombin and prostate specific antigen as model targets. In addition, the proposed multicolor strategy was employed for evaluating inhibition efficiency, and shows potential in visual screening of enzyme inhibitors. Such a facile, sensitive and low-cost sensing strategy provides a new perspective to develop universal platforms of point-of-care testing.

Abstract:Sir, It is now well known that aplastic anemia (AA) may represent a phase preceding ALL (pre-ALL). Despite numerous reports on this subject including more than 40 reported cases1-9 the etiology and pathogenesis of pre-ALL has not been adequately explained. Out of 552 children treated in our department for ALL during 1969-1992, pre-ALL has only been detected in eight patients (1.5%). The table summarizes our material.
