Abstract:While colorimetric-based assays are very convenient to determine biomarkers in point-of-care testing (POCT), they often suffer from pretreatment procedures for separation of plasma or serum from whole blood samples. Here, we report a simple colorimetric paper-based analytical device (c-PAD) that is capable of performing sample-to-answer analysis by directly dropping the whole blood sample on paper. This is accomplished by utilizing sodium alginate hydrogel, which exhibits a nanometer-scale porous structure to effectively prevent the passage of large red blood cells and hemoglobin molecules, to encapsulate enzymes and chromogenic reagents. As the small targets in the blood sample enter the sensing region to trigger a chromogenic reaction, the resulting color signal is recorded by a smartphone. The interference from the red blood to the color signal can be completely avoided without the requirement of any separation process. The analytical performance of the method is evaluated by assaying glucose in real blood samples. The results show that rapid and accurate analysis can be achieved with the limit of detection as low as 0.12 mM. In addition, simultaneous detection of different targets (glucose, cholesterol, and triglycerides) in whole blood can be achieved by fabricating c-PAD with multiple sensing regions. Owing to its several essential advantages including an extremely simple procedure for fabrication, sample-to-answer analysis without tedious pretreatment, and capability to perform high-throughput analysis, the proposed c-PAD will be of great value in POCT applications of whole blood samples.

Abstract:Paper-based analytical devices (PADs) integrated with smartphones have shown great potential in various fields, but they also face challenges such as single signal reading, complex data processing and significant environmental impacting. In this study, a colorimetric PAD platform has been proposed using bimetallic nickel-cobalt selenides as highly active peroxidase mimic, smartphone with 3D-printing dark-cavity as a portable detector and an artificial neural network (ANN) model as multi-signal processing tool. Notably, the optimized nickel-cobalt selenides (Ni0.75Co0.25Se with Ni to Co ratio of 3/1) exhibit excellent peoxidase-mimetic activities and are capable of catalyzing the oxidation of four chromogenic reagents in the presence of H2O2. Using a smartphone with image capture function as a friendly signal readout tool, the Ni0.75Co0.25Se based four channel colorimetric sensing paper is used for multi-signal quantitative analysis of H2O2 by determining the Grey, red (R), green (G) and blue (B) channel values of the captured pictures. An intelligent on-site detection method for H2O2 has been constructed by combining an ANN model and a self-programmed easy-to-use smartphone APP with a dynamic range of 5 μM to 2 M. Noteworthy, machine learning-assisted smartphone sensing devices based on nanozyme and 3D printing technology provide new insights and universal strategies for visual ultrasensitive detection in a variety of fields, including environments monitoring, biomedical diagnosis and safety screening.

Abstract:One of the effective approaches to safeguard human health is the rapid and sensitive identification of food additives, environmental pollutants, and disease markers. This study developed a portable intelligent paper-based sensor for point-of-care detection of hydrogen peroxide (H 2 O 2 ) and sarcosine (SA) using smartphone colorimetric analysis, supplemented by UV-Vis verification. The paper-based sensor utilizes a chemical modification of conventional filter paper by immobilizing horseradish peroxidase (HRP) from the horseradish plant, achieved through boronate affinity and metal chelation techniques. For H 2 O 2 detection, the sensor exhibited a linear range of 2–100 μmol L −1 and a limit of detection (LOD) of 1.0 μmol L −1 . Similarly, a range of 2–400 μmol L −1 for SA detection and an LOD of 1.0 μmol L −1 . The sensors were successfully employed for determining the H 2 O 2 concentration in liquor samples, environmental water, and the amount of SA in urine. Significantly, the detection ranges of these sensors surpassed those of commercially available test strips. In summary, we first proposed enzyme-immobilized paper-based sensors for immobilizing HRP from plants to detect H 2 O 2 and SA, which are intelligent, portable, fast, green, low-cost, highly sensitive, and selective, with broad application prospects in food analysis, environmental monitoring, and medical diagnostics.

Abstract:Herein, a simple microfluidic paper-based analytical device (μPAD) by using platinum nanoparticles (Pt NPs) as highly active peroxidase mimic for simultaneous determination of glucose and uric acid was fabricated. The μPAD consisted of one sample transportation layer, four paper-based detection chips, and two layers of hydrophobic polyethylene terephthalate (PET) films. The four detection chips were immobilized with various chromogenic reagents, Pt NPs, and specific oxidase (glucose oxidase or uricase). H2O2 generated by specific enzymatic reactions could oxidize co-immobilized chromogenic reagents to produce colored products by using Pt NPs as efficient catalyst. The multi-layered structure of μPAD could effectively improve the color uniformity and color intensity. Total color intensity from each two detection chips modified with distinct chromogenic reagents were used for quantitative analysis of glucose and uric acid, respectively, resulting in significantly improved sensitivity. The linear range for glucose and uric acid detection was 0.01–5.0 mM and 0.01–2.5 mM, respectively. Satisfied results were obtained for glucose and uric acid detection in real serum samples. An easy-to-use smartphone APP was developed for convenient and intelligent detection. The developed μPAD integrated with smartphone as detector holds great applicability for simple and portable on-site analysis.

Cardiac venous endothelial dysfunction after myocardial ischemia and reperfusion in dogs.

Abstract:Endothelial dysfunction is a prominent occurrence in coronary arteries after myocardial ischemia and reperfusion. However, this has not been studied in coronary veins. Endothelium-dependent vasorelaxation was studied in cardiac venous rings obtained from dogs subjected to 60 min of coronary artery occlusion followed by 270 min of reperfusion, as well as from dogs subjected to sham ischemia-reperfusion. Myocardial ischemia resulted in a 96 +/- 2% decrease in coronary flow to the ischemic area 60 min after occlusion of the left anterior descending (LAD) coronary artery, which led to a significant degree of cardiac necrosis (i.e., 32.9 +/- 3.9% of area at risk). Cardiac venous rings isolated from sham ischemia-reperfusion dogs relaxed 68 +/- 3% to 200 microM ADP, 65 +/- 3% to 2 microM A23187, and 76 +/- 4% to 200 microM sodium nitrite (NaNO2). Corresponding values for cardiac venous rings isolated from ischemic-reperfused dogs were 32 +/- 3% for 200 microM ADP (P less than 0.01 vs. sham), 31 +/- 3% for 2 microM A23187 (P less than 0.01 vs. sham), and 74 +/- 3% for 200 microM NaNO2 (NS from sham). In rings from control dogs, vasorelaxation to ADP and A23187 was markedly inhibited by 4 mM NG-methyl-L-arginine (L-NMMA) and 10 methylene blue and restored after NG-nitro-L-arginine by 3 mM L-arginine. These results demonstrate that a significant degree of endothelial dysfunction occurred in cardiac venous rings after ischemia and reperfusion.(ABSTRACT TRUNCATED AT 250 WORDS)

Abstract:Vitronectin, a multifunctional glycoprotein present in blood and extracellular matrix, is not only a member of the cell adhesion molecules, but also a regulator of proteolytic enzyme cascades, thereby providing a unique regulatory factor for proteolytic degradation of extracellular matrix and tissue remodeling. Vitronectin interacts with the cell surface through integrins of the alpha(v)-related system. Because vitronectin and its receptor may have a role in various renal physiological and pathological processes, we evaluated their expression in renal tissues of streptozotocin-induced short- and long-term hyperglycemic rats by applying quantitative immunoelectron microscopy and Western blot analysis. Vitronectin was shown over the glomerular basement membrane (GBM) and mesangial matrix (MM), whereas alpha(v) was located along the plasma membrane of endothelial, epithelial, and mesangial cells. Although distribution patterns of vitronectin and alpha(v) integrin labeling in renal tissues from short- and long-term hyperglycemic rats, as well age-matched normoglycemic rats, were similar, increases in their immunoreactive sites were detected in hyperglycemic conditions. Changes also were present in old compared with young normoglycemic animals. The diabetes-related increase in vitronectin was more significant in the GBM than MM, whereas the increase in alpha(v) integrin was as significant in podocytes as mesangial cells. Western blot analysis, performed on isolated glomerular material from normoglycemic and hyperglycemic animals, confirmed those changes. Our results suggest that vitronectin and its receptor, alpha(v) integrin, must have defined roles in molecular mechanisms involved in the pathogenesis of both diabetic and aging nephropathy.
Smartphone-Based Chemiluminescent Origami µPAD for the Rapid Assessment of Glucose Blood Levels

Abstract:Microfluidic paper analytical devices (µPADs) represent one of the most appealing trends in the development of simple and inexpensive analytical systems for diagnostic applications at the point of care (POC). Herein, we describe a smartphone-based origami µPAD for the quantitative determination of glucose in blood samples based on the glucose oxidase-catalyzed oxidation of glucose leading to hydrogen peroxide, which is then detected by means of the luminol/hexacyanoferrate(III) chemiluminescent (CL) system. By exploiting the foldable µPAD format, a two-step analytical procedure has been implemented. First, the diluted blood sample was added, and hydrogen peroxide was accumulated, then the biosensor was folded, and a transport buffer was added to bring hydrogen peroxide in contact with CL reagents, thus promoting the CL reaction. To enable POC applicability, the reagents required for the assay were preloaded in the µPAD so that no chemicals handling was required, and a 3D-printed portable device was developed for measuring the CL emission using the smartphone's CMOS camera. The µPAD was stable for 30-day storage at room temperature and the assay, displaying a limit of detection of 10 µmol L-1, proved able to identify both hypoglycemic and hyperglycemic blood samples in less than 20 min.

Abstract:Microfluidic paper-based analytical devices (μPADs) for detection of hydrogen peroxide and glucose have been developed. The analytical performance of colorimetric detection using the conventional starch-iodine color reaction has been significantly improved by using gelatin as the surface modifier which retains the enzyme activity in the dry filter paper strip, improves antioxidability, as well as decreases the strong background signal. Under optimal conditions, the color intensities show a good linear relationship with glucose concentration ranging from 0.5 to 5 mM and hydrogen peroxide concentration from 0.5 to 6 mM, with the detection limit of 0.05 mM and 0.1 mM, respectively. In addition, the accuracy of colorimetric sensor has been successfully assessed in detecting glucose from real human serum samples and recovery value ranges from 95.7% to 97%, which are approaching to the glucose oxidase endpoint. The new colorimetric assay exhibits high sensitivity, good selectivity, acceptable stability and reproducibility. The present approach is promising for monitoring glucose for point of care diagnostic applications, especially in regions with resource-limited settings.

Abstract:With advances in point-of-care testing (POCT), lateral flow assays (LFAs) have been explored for nucleic acid detection. However, biological samples generally contain complex compositions and low amounts of target nucleic acids, and currently require laborious off-chip nucleic acid extraction and amplification processes (e.g., tube-based extraction and polymerase chain reaction (PCR)) prior to detection. To the best of our knowledge, even though the integration of DNA extraction and amplification into a paper-based biosensor has been reported, a combination of LFA with the aforementioned steps for simple colorimetric readout has not yet been demonstrated. Here, we demonstrate for the first time an integrated paper-based biosensor incorporating nucleic acid extraction, amplification and visual detection or quantification using a smartphone. A handheld battery-powered heating device was specially developed for nucleic acid amplification in POC settings, which is coupled with this simple assay for rapid target detection. The biosensor can successfully detect Escherichia coli (as a model analyte) in spiked drinking water, milk, blood, and spinach with a detection limit of as low as 10-1000 CFU mL(-1), and Streptococcus pneumonia in clinical blood samples, highlighting its potential use in medical diagnostics, food safety analysis and environmental monitoring. As compared to the lengthy conventional assay, which requires more than 5 hours for the entire sample-to-answer process, it takes about 1 hour for our integrated biosensor. The integrated biosensor holds great potential for detection of various target analytes for wide applications in the near future.

Abstract:The paper-based analytical platforms have been applied in biochemical detection and environmental monitoring due to their low cost and easy operation. Despite their advantages, the complexity of fabrication has limited their further application. Here, a simple and innovative approach using parylene C, a material commonly used in semiconductors, is reported to fabricate paper-based chips and construct a point-of-care testing (POCT) platform through smartphones. Parylene C is deposited on the filter paper through a polydimethylsiloxane (PDMS) clamp and forms a hydrophobic barrier. A three-channel paper-based chip was utilized to detect aluminum (Al) ions on a smartphone platform by the chromogenic method. The color change after Al 3+ detection is represented with △E which was photographed and calculated by app on smartphone. The detection limit is 0.05 mg/L and the detection range is from 0.05 mg/L to 10 mg/L which indicates the application potential in the field of biochemical detection. This new paper-based sensing method and smartphone-based portable detection platform are simple, low-cost, fast, and highly sensitive, and provides a potential application for on-site, visual detection of ions and even other substances.

Integration of target responsive hydrogel with cascaded enzymatic reactions and microfluidic paper-based analytic devices (µPADs) for point-of-care testing (POCT)

Abstract:Paper based microfluidics (µPADs) with advantages of portability, low cost, and ease of use have attracted extensive attention. Here we describe a novel method that integrates glucoamylase-trapped aptamer-crosslinked hydrogel for molecular recognition with cascaded enzymatic reactions for signal amplification and a µPAD for portable readout. Upon target introduction, the hydrogel decomposes to release glucoamylase, which catalyzes the hydrolysis of amylose to produce a large amount of glucose. With a simple folding of the µPAD, the sample solution containing glucose product wicks and diffuses in parallel to each test-zone to carry out homogeneous assays, where glucose is used to produce I2 for brown color visualization through multiple enzymatic and chemical cascade reactions. Through color gradient changes based on different concentrations of the target, a semiquantitative assay is achieved by the naked eye, and quantitation can be obtained by handheld devices. Detection of cocaine in buffer and urine was performed to demonstrate the utility of the hydrogel-µPAD system. More importantly, the hydrogel-µPAD system can be extended to the detection of various targets by incorporating the corresponding aptamer into the hydrogel. The hydrogel-µPAD system reported here provides a new platform for portable, disposable and visual detection of a wide range of targets.
Fabrication of a paper-based facile and low-cost microfluidic device and digital imaging technique for point-of-need monitoring of hypochlorite

Abstract:Lab-on-a-paper-based devices are promising alternatives to the existing arduous techniques for point-of-need monitoring. The present work reports an instant and facile method to produce a microfluidic paper-based analytical device (μPAD). The fabricated μPAD has been used to detect hypochlorite (OCl-) by incorporating newly synthesized chromo-fluorogenic ratiometric probes 1 and 2 into the sample reception zone. The probes showed high selectivity and fast response (<10 s) toward OCl- with an excellent linear relationship in the concentration range of 0-100 μM. The concentration-dependent fluorometric change driven by the reaction of 1@μPAD with OCl- has been monitored using gel-doc imaging systems, which is unprecedented. Digitizing the intensity of the colour solution with different mathematical models of colour has developed a straightforward method for monitoring OCl- without any interference from its competitors. 1@μPAD can detect OCl- at ∼10 times lower than the WHO recommended limit. The detection limit of 1@μPAD via a digital camera-based fluorescence technique was found to be better over digital camera-based cuvette assays. Therefore, 1@μPAD has been successfully utilized to monitor OCl- in actual environmental water samples with portability, ease of use, and sensitivity. The analytical RSD was found to be ≤3% based on fluorimetric detection using 1@μPAD. The chemodosimetric reaction between OCl- and the probe was evidenced by UV-vis and fluorescence spectroscopy, 1H NMR, and ESI-MS. The rapid response time, biocompatibility, low cytotoxicity, 100% aqueous solubility, ratiometric feature, and exclusive OCl- selectivity over other competitive ROS/RNS successfully lead to the application of the probes for bioimaging of exogenous as well as endogenous OCl- in normal cells (HEK293) and cancerous cells (HeLa).
Engineering the nanozyme hydrogel beads by polyvinyl alcohol/chitosan encapsulation with recyclable and sustainable catalytic activity for visual analysis of hydrogen peroxide

Abstract:Background: Hydrogen peroxide (H2O2) plays a vital role in human health and have been regarded as a crucial analyte in metabolic processes, redox transformations, foods research and medical fields. Especially, the long-time and excessive digestion of H2O2 may even cause severe diseases. Although conventional instrumental methods and nanozymes-based colorimetric methods have been developed to accomplish the quantitative analysis of H2O2, the drawbacks of instrument dependence, cost-effectiveness, short lifespan, non-portable and unsustainable detection efficacies will limit their applications in different detection scenarios. Results: Herein, to address these challenges, we have proposed a novel strategy for nanozyme (RuO2) hydrogel preparation by the solid support from cross-linked polyvinyl alcohol (PVA) and chitosan (CS) to both inherit the dominant peroxidase-like (POD) activity and protect the RuO2 from losing efficacies. Taking advantages from the hydrogel, the encapsulated RuO2 were further prepared as the regularly spherical beads (PCRO) to exhibit the sustainable, recyclable, and robust catalysis. Moreover, the intrinsic color interferences which originated from RuO2 can be avoided by the encapsulation strategy to promote the detection accuracy. Meanwhile, the high mechanical strength of PCRO shows the high stability, reproducibility, and cyclic catalysis to achieve the recyclable detection performance and long lifetime storage (40 days), which enables the sensitively detection of H2O2 with the detection limit as lower to 15 μM and the wide detection linear range from 0.025 to 1.0 mM. Significance: On the basis of the unique properties, PCRO has been further adopted to construct a smartphone detection platform to realize the instrument-free and visual analysis of H2O2 in multi-types of milk and real water samples through capturing, processing, and analyzing the RGB values from the colorimetric photographs. Therefore, PCRO with the advanced detection efficacies holds the great potential in achieving the portable and on-site analysis of targets-of-interest.
Smartphone-assisted paper-based multifunctional assay for on-site tandem detection of inorganic phosphate, neutral red and heparin

Abstract:Today, easy availability and efficient detection assays in low-resource settings are mandatory. Therefore, a smartphone-assisted paper-based detection strategy using eosin Y (EY)-embed ZIF-8 nanocomposite for phosphate (Pi), neutral red (NR) and heparin (HP) on site testing was fabricated. After adding the targets in a specific order, the prepared strips produced brilliant color transitions (astonishing colors, ranging from pistachio to bright yellow for Pi, from bright yellow to jacinth for NR and from jacinth to light orange for HP). These variation on tonality can be evaluated by the smartphone-based readout device (Color Assist App) through the ratio change of B/(R + G + B), which exhibits superior performance of on-site detection in wastewater, including high sensitivity (LOD is 0.015, 0.031 and 0.014 mM for Pi, NR and HP, respectively), ultra-fast response time and excellent selectivity. This strips and smartphone dual-mode monitoring strategy may offer a convenient idea to identify Pi, NR and HP, with great prospects for environmental wastewater applications. Additionally, the probe was enabled for bioimaging of Pi, NR and HP in living cells. Finally, the related anti-counterfeiting fluorescent ink of probe was proposed on schedule.

Abstract:A novel and facile method by sewing low-cost fiber electrodes on paper is proposed to fabricate electrochemical paper-based analytical devices (ePADs). Prussian Blue (PB) and Ag/AgCl were deposited on carbon fibers to prepare the working and reference electrodes. The hydrophilic carbon fiber was employed as the counter electrode. All electrodes were sewed on the paper substrate. The linear voltammetry scanning (LSV) and physical characterization results showed that the physico-chemical characteristics of the prepared electrode were maintained after sewing. As a proof of concept, the ePAD fabricating by sewing was used to detect hydrogen peroxide (H2O2). The limit of detection (LOD) achieved 0.9 µM and the ePAD possessed high anti-interference ability to a variety of common ions and biological molecules. Additionally, the robustness of the ePADs against bending (error 6.2 %) manifested that the ePAD had excellent practical applicability. The good reproducibility (3.65 %) displayed by the current responses confirmed that this strategy can be profitably adopted to easily assemble ePADs in a highly flexible manner. In addition, a user-friendly reagent-free ePAD was proposed to determine the content of H2O2 in real water sample without treatment. The good recovery percentage (95.7–99.7 %) demonstrated the high feasibility of the reagent-free ePAD with facile carbon fiber-sewed electrodes in the detection of H2O2 from complex water samples. These results confirm that the sewing method is feasible and promising. This work provides a new insight into fabrication of highly sensitive and highly selective ePADs with fiber electrodes.

Abstract:On-site multiplex enzyme detection is crucial for diagnosis, therapeutics and prognostic. To date, it is still a daunting challenge to develop portable, low-cost, and efficient multi-enzyme detection methods. Herein, a novel sample-in-result-out platform integrating ratiometric fluorescent assays with 3D origami microfluidic paper-based device (μPAD) was developed for simultaneous visual point-of-care testing (POCT) of alkaline phosphatase (ALP) and butyrylcholinesterase (BChE). Cascade catalytic reaction with the same two fluorescent signal indicators was rationally designed to ratiometric fluorescent detection of ALP and BChE: substrate of ALP (pyrophosphate) and product of BChE (thiocholine) can strongly complex with Cu2+, Cu2+ oxidizes o-phenylenediamine to fluorescent 2,3-diaminophenazine (oxOPD) (emission, 565 nm), oxOPD quenches the fluorescence of carbon dots (CDs, emission at 445 nm) via inner filter effect, thus oxOPD/CDs values are relevant to ALP and BChE activities. Then 3D origami μPAD composing of four layers and two parallel channels was fabricated and simply prepared by one-step plotting with black oil-based marker and specific metal molds. After simple folding and unfolding neighboring layers to sequentially initiate reactions of pre-loaded reagents, fluorescent images on the detection zone can be captured by smartphone and analyzed by red-green-blue software for quantitative analysis. Under optimal conditions, the proposed platform was successfully performed to detect ALP and BChE with activity difference at 3 orders of magnitude in human serum samples without any pretreatment procedures. Excellent selectivity, good precision, favorable linear range, and high accuracy were exhibited. Importantly, the platform opens a promising horizon for high-throughput POCT of multiplex biomarkers.

Abstract:I WOULD like to begin by expressing appreciation both for myself and on behalf of the American Society for Clinical Investigation for the opportunity to participate in this symposium. It cannot help but further cement relationships between our sister societies. Many of our problems are essentially the same and stem primarily from the somewhat unique and difficult position of the clinical investigator. He attempts to play the role of a two-faced Janus with one face directed toward clinical medicine and the other toward the basic sciences. Added to this are his interests in teaching and his desires to be a humanistic scholar. It is no easy task for him to be a scientist. The puristic view that he is a second-class citizen in the science community is prevalent in certain circles, and it is not readily dispelled by the concept that we are engaged in the salutary avocation of basic research in human biology. However, my primary topic concerns the more mundane question of the support of clinical investigation in the U.S.A. Following World War II, widespread popular recognition of the achievements of science and research developed in the United States. The war had revealed the enormous potential of economic capability in support of science and technology. The conviction grew that such enterprise could be successfully directed toward the conquest of disease. The anomalous situation arose where there actually was political pressure for action. Private fortunes and the resources of public foundations were inadequate to finance the large-scale medical research effort that many envisioned. As a result we have witnessed continued increases in the appropriations of funds by the Congress. Support available to the National Institutes of Health (NIH) for research grants alone rose from 3.5 million in 1947 to 97 million in 1957, nearly a 30-fold increase. During the fiscal year 1962, the NIH will expend over 300 million in grants for research projects. This seems like a gigantic increase in appropriations for medical research. However, it should be remembered that all research and development has increased rapidly, so that the medical component has remained a relatively constant, quite small, proportion of the nation's total research and development effort. It remains less than 5%o, hardly a figure to excite undue alarm concerning perspective. Just how much of this appropriation for medical research goes to clinical investigation is difficult to evaluate. Certainly, a considerable portion goes to the more basic sciences, particularly biochemistry and physiology, and it frequently is not disease-oriented. Abstract:Hydrogen peroxide (H2O2) and hydrogen sulfide (H2S) play a key role in many cellular signaling pathways. The efficient and accurate in situ detection of H2O2 and H2S released from living cells has attracted extensive research interests. Herein, we presented a portable colorimetric and electrochemical dual-mode sensor for detection of cellular secreted H2O2 and H2S based on MOF-818 nanozyme. MOF-818 containing trinuclear copper centers not only possesses excellent peroxidase-like activity under 37 °C and acidic conditions, but also exhibits outstanding electrocatalytic reduction activity to H2O2 (reduction peak potential at −0.08 V vs Ag/AgCl). Meanwhile, the copper sites of MOF-818 can be coordinated with sulfide, reducing the catalytic activity of MOF-818. As a result, the colorimetric and electrochemical sensing of H2O2 and H2S were established based on “signal-on” and “signal-off” strategies, respectively. Additionally, the combination of a smartphone and a miniature electrochemical analyzer enables dual-mode colorimetric and electrochemical sensing platform for in situ detection of H2O2 and H2S released from live cells. This portable dual-mode sensor based on MOF nanozyme not only opens a new path for point-of-care testing, but also has potential applications in the field of cell biology and clinical diagnosis.

Abstract:Hydrogen peroxide (H2O2) is closely involved in the fields of food industry. Excessive residual H2O2 in food samples will exert adverse effects on the nutritional value of foods and pose great threat to the health of consumer. Development of a reliable analytical method for on-spot quantitative detection of H2O2 levels in food is of urgent necessity. Here, we presented a colorimetric and near-infrared (NIR) ratiometric fluorescent sensor for on-spot quantitative detection of H2O2 in various food samples. The sensor displayed colorimetric and NIR ratiometric fluorescence dual-signal response to H2O2 with high selectivity and sensitivity. The limits of detection (LOD) was measured to be 23.08 nM (S / N = 3). The sensor has been applied for detecting exogenous and endogenous H2O2 in living cells by fluorescence imaging. Also, the changes of H2O2 level in potato tissue infected by phytophthora infestans has been monitored. Particularly, to achieve on-spot quantitative detection of H2O2 in food samples, a portable and field deployable detection device was rationally developed. The sensor, together with portable detection devices and smartphones, performed on-spot quantitative detection of H2O2 in chicken wings and milk through dual signal outputs of colorimetry and fluorescence. We anticipate that the analytical method developed here could be served as convenient tool for on-spot quantitative detection of H2O2 in different foods in near future.
