Abstract:A method for the determination of 21 perfluorinated and 10 polyfluorinated alkyl substances (PFAS) was developed for application in different food matrices. Acetonitrile was used as the extraction solvent with solid phase extraction weak anion-exchange (SPE-WAX) clean up, with LC-MS/MS analysis using both surrogate and performance standards to correct for losses during sample preparation and matrix effects. The method has been evaluated in four different matrices (fish, pizza, chicken nuggets and spinach). Originally, the focus was to develop a method for foods commonly thought to be a source of PFASs (e.g. fish). It was expanded to include foods where PFAS exposure would be possible through their presence in grease-proof food packaging (e.g. pizza, chicken nuggets). Vegetables (lettuce) and fruit (tomato) have recently been considered as part of proficiency testing programmes, so the inclusion of some testing in a vegetable matrix (i.e. spinach) was also added to the testing. Limits of quantification ranged from 0.018 ng g-1 (L-PFDS) to 5.28 ng g-1 (FHEA), although method quantification limits for PFBA (12.4 ng g-1), 6:2 PAP (8.96 ng g-1) and 8:2 PAP (3.49 ng g-1) were elevated above instrumental limits owing to their consistent detection in reagent blank samples. PFAS analyses were strongly impacted by matrix, therefore the use of isotopically labelled internal standards was critical to the development of accurate results. The accuracy of the method using numerous proficiency testing schemes or interlaboratory comparison studies has shown the developed method to be successful with z-scores for all concerned analytes in all test matrices remaining within ±2.0, with the exception of PFBA in wheat flour which was -2.4.

Abstract:Per- and polyfluoroalkyl substances (PFAS), that are widely used in different industrial and commercial activities, have become emerging contaminants of concern. Thus, the evaluation of PFAS presence in diverse environmental matrices is of paramount importance, leading to the necessity in development of different types of analytical methods to analyse the trace amounts of PFAS with varying selectivity and sensitivity. Analytical methods that have been advanced and employed for PFAS identification are critically described with their strengths, limitations and future potentials in this review. This review includes, (i) instrument analytical methodologies, mainly focusing on sample preparation, extraction and recovery analysis, (ii) nanoparticle-based sensors, and an app-based smartphone portable sensor, for the monitoring and sensing of PFAS in different environmental matrices. Furthermore, (iii) total oxidisable precursor assay (TOPA), content of total organic fluorine (TOF) as well as total fluorine (TF) are also reviewed, which provide fundamental tools towards achieving precursor quantification and closing mass balance analysis. Abstract:To enable the monitoring of a wide scope of per- and polyfluoroalkyl substances (PFAS) in the ng/kg level in foodstuffs, an LC-MS/MS method comprising 57 analytes was developed and validated in seven different matrices (milk powder, milk-based infant formula, meat-based baby food puree, fish and fish oil, fresh egg, and soluble coffee). The analytical approach was based on an acetonitrile:water extraction followed by solid phase extraction clean-up with subsequent quantification of the extracted analytes either by isotope dilution (55 compounds) or by standard addition (2 compounds) mass spectrometry. The validation criteria followed the guidance document for the analysis of PFAS issued by the European Union Reference Laboratory for Halogenated Persistent Organic Pollutants. The lowest limits of quantification (LOQs) for the four recently regulated compounds (L-PFOS, PFOA, PFNA, L-PFHxS) were set at 0.010 μg/kg in baby and infant foods (as sold) but also in dairy ingredients. Exception was for PFOA in milk powder due to too large variability in the repeatability. Applicability of the method was further demonstrated in 37 commodity check matrices. Overall validation data demonstrated the robustness of the method for most of the compounds and the LOQs achieved were low enough to ensure compliance with Commission Regulation EU 2022/2388 but also to support future collection of occurrence data in ng/kg level in food.

Abstract:Per- and polyfluorinated alkyl substances (PFAS) can be added to food contact materials (FCM) to increase their water and/or grease repellent properties. Some well-known PFAS are perfluoroalkyl carboxylic acids (PFCA), perfluoroalkyl sulfonic acids (PFSA), and polyfluorinated telomer alcohols (FTOH). Due to the strength of the carbon-fluorine bond, PFAS are chemically very stable and highly resistant to biological degradation, posing a risk to human health and the environment. To examine the presence of PFAS in paper-based FCM, various samples were collected, including popcorn bags, muffin cups, and pizza boxes with high total organic fluorine (TOF) content from the Danish and Spanish markets. The FCM composition was characterised by FTIR. Quantification of some well-known PFAS such as PFCA, PFSA, and FTOH was performed in food simulants using LC-MS/MS, and in addition a non-targeted screening approach was performed by LC-Orbitrap-HRMS. Among analysed samples, the highest concentrations of PFAS were found in a muffin cup made of cellulose (PFCA ∼ 1.41 μg kg −1 food, FTOH ∼ 11.5 μg kg −1 food), and the results were used to estimate dietary exposures to PFAS migrated from this FCM. Compared to measured TOF value in this sample, the fluorine from all quantified PFAS accounted for only 0.6%. Thus, a more powerful analytical approach was used to further investigate PFAS occurrence in this sample. Using non-targeted screening, an additional twenty compounds were identified, among them five with confidence level 1 and ten with confidence level 2. Many of them were either fluorotelomer carboxylic acids or sulfonic acids or ether-containing compounds.

Abstract:In response to the European Food Safety Authority's establishment of a tolerable weekly intake (TWI) for the sum of PFOA, PFNA, PFHxS, and PFOS, a method was developed to quantify and confirm 20 PFASs at the sub-parts-per-trillion level in fruit and vegetables. Improved sensitivity was achieved by (i) increasing the sample intake, (ii) decreasing the solvent volume in the final extract, and (iii) using a highly sensitive mass spectrometer. Except for PFTrDA, target PFASs could be quantitatively determined with an apparent recovery of 90-119%, limits of quantitation down to 0.5 ng/kg, and a relative standard deviation under within-laboratory reproducibility conditions of <28%. The method was successfully applied to 215 fruit and vegetable samples obtained from local grocery stores and markets. Leafy vegetables prove to be the main vegetable category responsible to PFAS exposure, mainly of PFOA, followed by PFHpA and PFHxA.

Abstract:The availability of sensitive analytical methods to detect per- and polyfluoroalkyl substances (PFASs) in food of animal origin is fundamental for monitoring programs to collect data useful for improving risk assessment strategies. The present study aimed to develop and validate a fast and sensitive method for determining short and long-chain PFASs in meat (bovine, fish, and swine muscle), bovine liver, hen eggs, and cow's milk to be easily applicable in routine analysis of food. A QuEChERS extraction and clean-up method in combination with liquid chromatography coupled to mass spectrometry (LC-MSMS) were used. The method resulted in good linearity (Pearson's R > 0.99), low limits of detection (7.78–16.35 ng/kg, 8.26–34.01 ng/kg, 6.70–33.65 ng/kg, and 5.92–19.07 ng/kg for milk, liver, egg, and muscle, respectively), and appropriate limits of quantification (50 ng/kg for all compounds except for GenX and C6O4, where the limits of quantification were 100 ng/kg). Trueness and precision for all the tested levels met the acceptability criteria of 80–120% and ≤20%, respectively, regardless of the analyzed matrix. As to measurement uncertainty, it was <50% for all compound/matrix combinations. These results demonstrate the selectivity and sensitivity of the method for simultaneous trace detection and quantification of 14 PFASs in foods of animal origin, verified through the analysis of 63 food samples.

Determination of per- and polyfluoroalkyl substances (PFAS) in six different fish species from Swiss lakes

Abstract:Per- and polyfluoroalkyl substances (PFAS) are persistent environmental contaminants with bioaccumulation potential, particularly affecting aquatic ecosystems and human health also via fish consumption. There is therefore a need for reliable extraction methods and studies to accurately assess PFAS levels in fish, crucial for understanding bioaccumulation and potential toxicological effects on both fish and humans through consumption. This study investigated PFAS levels in freshwater fish from Swiss lakes, focusing on six common species: Coregonus wartmanni, Cyprinus carpio, Oncorhynchus mykiss, Perca fluviatilis, Salmo trutta, and Squalius cephalus. Utilizing an optimized QuEChERS extraction method, 15 PFAS were analyzed in 218 fish fillet samples using liquid chromatography-mass spectrometry (LC-MS/MS). The results were compared to EU regulations and EFSA guidelines for tolerable weekly intake (TWI), with a specific focus on correlations between fish size and PFAS concentration. Our findings reveal significant PFAS contamination, particularly in Perca fluviatilis with perfluorooctane sulfonic acid (PFOS) and perfluorohexane sulfonic acid (PFHxS) levels often exceeding EU safety limits. TWI, calculated for a person of 70 kg body weight and an intake of 200 g of fish fillet, is exceeded in 95% of Coregonus wartmanni, 100% of Squalius cephalus, and in 55%, 50%, and 36% of the specimens Oncorhynchus mykiss, Salmo trutta, and Perca fluviatilis respectively. Correlation analysis between PFAS concentration and fish size in 121 Salmo trutta specimens revealed significant positive correlations for perfluorobutane sulfonic acid (PFBS), perfluorodecanoic acid (PFDA), and perfluorohexane sulfonic acid (PFHxS), and a negative correlation for perfluoropentanoic acid (PFPeA). These results underscore the critical need for continuous monitoring and regulatory efforts to mitigate PFAS exposure risks to both ecosystems and human health.
Dietary exposure to selected perfluoroalkyl acids (PFAAs) in four European regions

Abstract:The dietary exposure to selected PFAAs was estimated in four selected European states (Belgium, the Czech Republic, Italy and Norway) representing Western, Southern, Eastern and Northern Europe. The harmonised sampling programme designed in the European Union project PERFOOD was targeted at identifying seven selected PFAAs, including perfluorooctane sulfonic acid (PFOS) and perfluorooctanoic acid (PFOA), in food items that are most important both in terms of consumption and based on known high contamination patterns. The estimated average dietary exposure for adults (18-64 years) and children (3-9 years) is generally below or close to 1 ng kg⁻¹ BW day⁻¹ for all seven PFAAs. Considering the high consumption of food groups that contribute most to the exposure does not result in estimates exceeding 4 ng kg⁻¹ BW day⁻¹. Thus, based on the TDIs proposed by EFSA for PFOS (150 ng kg⁻¹ BW day⁻¹) and PFOA (1500 ng kg⁻¹ BW day⁻¹), no concern can be identified. There are distinct dietary exposure patterns from region to region as a result of different food consumption and contamination patterns. Foods of plant origin (e.g. fruit and vegetables) are most important for the dietary exposure to PFHxA, PFOA and PFHxS, while the consumption of foods of animal origin (particularly fish and seafood) mostly contributes to the dietary exposure to PFDA and PFUnDA. For the dietary exposure to PFNA and PFOS, food of animal and plant origin contributes with equal importance. In conclusion, region-to-region differences as well as the relative importance of food of different origin for each PFAA should be paid more attention in further research.

Abstract:Poly- and perfluoroalkyl substances ( PFAS s) have attracted great scientific attention because of their environmental persistence, bioaccumulation potentials, and toxicity. This study validated a simple and efficient sample preparation method for determining extractable PFASs, including 12 perfluoroalkyl carboxylic acids and 4 perfluoroalkyl sulfonates. The samples underwent ultrasonic extraction, and the extracts were cleaned up by dispersive solid-phase extraction technique with graphitized carbon if necessary. PFAS quantification was performed by a liquid chromatography-tandem mass spectrometry system. The instrument provided analytical signals for all target compounds with good repeatability (RSD < 15%), reproducibility (RSD < 20%), linearity ( R 2 > 0.99), and low detection limits (0.10–1.0 ng/mL). The validated method exhibited adequate recovery (70–115%), precision (RSD < 15%), and method detection limits (0.020–0.70 ng/g). This validated method was applied to analyze PFASs in several food packaging samples collected in Vietnam, showing very low levels (range 0.090–2.9 ng/g; median 0.72 ng/g) derived from unintentional sources.

Abstract:Abstract Background Despite specific restrictions on their production and use, per- and polyfluoralkyl substances (PFAS) are still omnipresent in the environment, including aquatic ecosystems. Most biomonitoring studies have investigated the PFAS concentrations in indigenous organisms, whereas active biomonitoring has only been used sporadically. In the present study, accumulated PFAS concentrations were measured in indigenous fish, European perch ( Perca fluviatilis ) and European eel ( Anguilla anguilla ), and in translocated freshwater mussels ( Dreissena bugensis and Corbicula fluminea) at 44 sampling locations within the main water basins of Flanders, the northern part of Belgium. Finally, both human health risk and ecological risk were assessed based on accumulated concentrations in fish muscle. Results Among locations, ΣPFAS concentrations ranged from 8.56–157 ng/g ww (median: 22.4 ng/g ww) in mussels, 5.22–67.8 ng/g ww (median: 20.8 ng/g ww) in perch, and 5.73–68.8 ng/g ww (median: 22.1 ng/g ww) in eel. Concentrations of PFOA and PFTeDA were higher in mussels compared to fish, whereas for PFDA and PFUnDA the opposite was true. A comparison of concentrations on a wet weight basis between both fish species showed significantly higher PFDoDA, PFTrDA, PFTeDA and PFOA concentrations in eel compared to perch and significantly higher concentrations of PFDA and PFOS in perch. In mussels, PFAS profiles were dominated by PFOA and showed a higher relative contribution of short-chained PFAS, while PFAS profiles in fish were dominated by PFOS. Furthermore, all mussel species clearly occupied a lower trophic level than both fish species, based on a stable isotope analysis. Conclusions Biomagnification of PFDA, PFUnDA and PFOS and biodilution of PFOA and PFTeDA were observed. Translocated mussels have been proven suitable to determine which PFAS are present in indigenous fish, since similar PFAS profiles were measured in all biota. Finally, mean PFAS concentrations in fish did pose a human health risk for eel, although tolerable daily intake values for perch were close to the reported daily consumption rates in Belgium and exceeded them in highly contaminated locations. Based on the ecological risk of PFOS, the standard was exceeded at about half of the sampling locations (44% for perch and 58% for eel).

Acta physiol. scand. 1965. 64. 383–386. –Either I-C14-lauric acid or l-C14-myristic acid together with 9–10-H3-palmitic acid were i. v. injected into fasted and refed male rats and the disappearance of label from the blood and the tissue distribution of label studied. Both lauric and myristic acid disappeared more rapidly than palmitic acid from the blood. In refed rats lauric acid was extracted even more rapidly than myristic acid. Lauric acid was oxidized at a higher rate than myristic acid and myristic acid was more rapidly oxidized than palmitic acid. More lauric acid label and myristic acid label was found in the liver“Neutral lipid plus FFA” than in the liver phospholipids. This skewed distribution was most marked for lauric acid in the fasted rats. The uneven distribution is discussed in relation to the fatty acid composition of the liver lipids, to certain feeding experiments and to the interconversion with other fatty acids.

Abstract:Public health is increasingly emphasizing policy, systems, and environmental (PSE) change as a key strategy for population-level health promotion and disease prevention. When applied to childhood obesity, this strategy typically involves school systems, since children spend large portions of their days in school and are heavily influenced by this environment. While most school systems have implemented nutrition education and physical activity programs for some time, their understanding and use of PSE approaches to obesity prevention is accelerating based on several large federally funded initiatives. As part of one initiative's evaluation, key informant interviews reveal the specific obesity prevention PSE strategies schools are attempting and the corresponding barriers and facilitators to their implementation. These evaluation findings raise several fundamental issues regarding school-based obesity prevention, including the potential role of school personnel, the influence of grant funding on school health initiatives, and the fit between public health and educational priorities.

Abstract:Here we synthesize current understanding of the magnitudes and methods for assessing human and wildlife exposures to poly‐ and perfluoroalkyl substances (PFAS). Most human exposure assessments have focused on two to five legacy PFAS and wildlife assessments are typically limited to targeted PFAS (up to ~30 substances). However, shifts in chemical production are occurring rapidly and targeted methods for detecting PFAS have not kept pace with these changes. Total fluorine (TF) measurements complemented by suspect screening using high resolution mass spectrometry are thus emerging as essential tools for PFAS exposure assessment. Such methods enable researchers to better understand contributions from precursor compounds that degrade into terminal perfluoroalkyl acids (PFAA). Available data suggest that diet is the major human exposure pathway for some PFAS but there is large variability across populations and PFAS compounds. Additional data on TF in exposure media and the fraction of unidentified organofluorine are needed. Drinking water has been established as the major exposure source in contaminated communities. As water supplies are remediated, and for the general population, exposures from dust, personal care products, indoor environments and other sources may be more important. A major challenge for exposure assessments is the lack of statistically representative population surveys. For wildlife, bioaccumulation processes differ substantially between PFAS and neutral lipophilic organic compounds, prompting a revaluation of traditional bioaccumulation metrics. There is evidence that both phospholipids and proteins are important for the tissue partitioning and accumulation of PFAS. New mechanistic models for PFAS bioaccumulation are being developed that will assist in wildlife risk evaluations.This article is protected by copyright. All rights reserved.

Abstract:The presence of per- and polyfluoroalkyl substances (PFASs) in commercial baby food products from various European countries was investigated in this study. A total of 96 samples were collected and analyzed to assess PFASs levels, composition profiles, and potential dietary intake among infants. The results indicated detectable levels of PFASs in the sampled baby food products, with carboxylic acid prevalence over sulfonic acids. Among the various baby food groups studied, dry cereals exhibited the highest PFASs concentrations. This finding emphasizes the need for further monitoring and investigation of PFASs contamination in this specific food category. While the concentrations detected were generally low, they indicated the widespread presence of PFASs in various types of baby food. Furthermore, a preliminary exposure assessment was conducted on the basis of the measured PFASs concentrations, providing an initial insight into the potential exposure levels among infants from three months to three years old. Calculations based on two scenario types revealed the best-case scenario likely underestimating actual exposure, while the worst-case scenario occasionally exceeded the limits set by the governmental institutions. Further research is needed to understand the sources, pathways, and potential health effects of PFASs exposure in this vulnerable population.
Consciousness in congenitally decorticate children: developmental vegetative state as self‐fulfilling prophecy

Abstract:According to traditional neurophysiological theory, consciousness requires neocortical functioning, and children born without cerebral hemispheres necessarily remain indefinitely in a developmental vegetative state. Four children between 5 and 17 years old are reported with congenital brain malformations involving total or near‐total absence of cerebral cortex but who, nevertheless, possessed discriminative awareness: for example, distinguishing familiar from unfamiliar people and environments, social interaction, functional vision, orienting, musical preferences, appropriate affective responses, and associative learning. These abilities may reflect‘vertical’plasticity of brainstem and diencephalic structures. The relative rarity of manifest consciousness in congenitally decorticate children could be due largely to an inherent tendency of the label‘developmental vegetative state’to become a self‐fulfilling prophecy.

Abstract:Per- and polyfluoroalkyl substances (PFAS) form a vast family comprising more than 4700 synthetic compounds. Their molecules contain a terminal functional group and a hydrophobic carbon tail (alkyl group) at which the hydrogen atoms are totally (in the case of perfluorinated compounds) or partially (in the case of polyfluorinated compounds) replaced by fluorine atoms. Due to the very specific properties of their structure, they have been used in a vast range of applications over the last 70 years. These substances are considered to be of concern for the environment. Their effects on human health are still poorly understood because studies are still too rare, but the cutaneous route could be a significant pathway of penetration. In this context, we made a qualitative study to assess the presence of PFAS in various cosmetics such as hygiene products, skin care products, make-up and perfumes. Among the 765 products studied, we found 11 different PFAS. Polytetrafluoroethylene (PTFE) and perfluorodecalin, present in 25.9% and 22.2% of products containing it, respectively, were the most frequent. Although the presence of this type of ingredient seems to be limited in Europe, make-up appears to be the type of product most likely to contain PFAS.
