Abstract:A method for the determination of 21 perfluorinated and 10 polyfluorinated alkyl substances (PFAS) was developed for application in different food matrices. Acetonitrile was used as the extraction solvent with solid phase extraction weak anion-exchange (SPE-WAX) clean up, with LC-MS/MS analysis using both surrogate and performance standards to correct for losses during sample preparation and matrix effects. The method has been evaluated in four different matrices (fish, pizza, chicken nuggets and spinach). Originally, the focus was to develop a method for foods commonly thought to be a source of PFASs (e.g. fish). It was expanded to include foods where PFAS exposure would be possible through their presence in grease-proof food packaging (e.g. pizza, chicken nuggets). Vegetables (lettuce) and fruit (tomato) have recently been considered as part of proficiency testing programmes, so the inclusion of some testing in a vegetable matrix (i.e. spinach) was also added to the testing. Limits of quantification ranged from 0.018 ng g-1 (L-PFDS) to 5.28 ng g-1 (FHEA), although method quantification limits for PFBA (12.4 ng g-1), 6:2 PAP (8.96 ng g-1) and 8:2 PAP (3.49 ng g-1) were elevated above instrumental limits owing to their consistent detection in reagent blank samples. PFAS analyses were strongly impacted by matrix, therefore the use of isotopically labelled internal standards was critical to the development of accurate results. The accuracy of the method using numerous proficiency testing schemes or interlaboratory comparison studies has shown the developed method to be successful with z-scores for all concerned analytes in all test matrices remaining within ±2.0, with the exception of PFBA in wheat flour which was -2.4.

Abstract:Per- and polyfluoroalkyl substances (PFAS), that are widely used in different industrial and commercial activities, have become emerging contaminants of concern. Thus, the evaluation of PFAS presence in diverse environmental matrices is of paramount importance, leading to the necessity in development of different types of analytical methods to analyse the trace amounts of PFAS with varying selectivity and sensitivity. Analytical methods that have been advanced and employed for PFAS identification are critically described with their strengths, limitations and future potentials in this review. This review includes, (i) instrument analytical methodologies, mainly focusing on sample preparation, extraction and recovery analysis, (ii) nanoparticle-based sensors, and an app-based smartphone portable sensor, for the monitoring and sensing of PFAS in different environmental matrices. Furthermore, (iii) total oxidisable precursor assay (TOPA), content of total organic fluorine (TOF) as well as total fluorine (TF) are also reviewed, which provide fundamental tools towards achieving precursor quantification and closing mass balance analysis. Abstract:To enable the monitoring of a wide scope of per- and polyfluoroalkyl substances (PFAS) in the ng/kg level in foodstuffs, an LC-MS/MS method comprising 57 analytes was developed and validated in seven different matrices (milk powder, milk-based infant formula, meat-based baby food puree, fish and fish oil, fresh egg, and soluble coffee). The analytical approach was based on an acetonitrile:water extraction followed by solid phase extraction clean-up with subsequent quantification of the extracted analytes either by isotope dilution (55 compounds) or by standard addition (2 compounds) mass spectrometry. The validation criteria followed the guidance document for the analysis of PFAS issued by the European Union Reference Laboratory for Halogenated Persistent Organic Pollutants. The lowest limits of quantification (LOQs) for the four recently regulated compounds (L-PFOS, PFOA, PFNA, L-PFHxS) were set at 0.010 μg/kg in baby and infant foods (as sold) but also in dairy ingredients. Exception was for PFOA in milk powder due to too large variability in the repeatability. Applicability of the method was further demonstrated in 37 commodity check matrices. Overall validation data demonstrated the robustness of the method for most of the compounds and the LOQs achieved were low enough to ensure compliance with Commission Regulation EU 2022/2388 but also to support future collection of occurrence data in ng/kg level in food.
Fast and Sensitive Analysis of Short- and Long-Chain Perfluoroalkyl Substances in Foods of Animal Origin

Abstract:The availability of sensitive analytical methods to detect per- and polyfluoroalkyl substances (PFASs) in food of animal origin is fundamental for monitoring programs to collect data useful for improving risk assessment strategies. The present study aimed to develop and validate a fast and sensitive method for determining short and long-chain PFASs in meat (bovine, fish, and swine muscle), bovine liver, hen eggs, and cow's milk to be easily applicable in routine analysis of food. A QuEChERS extraction and clean-up method in combination with liquid chromatography coupled to mass spectrometry (LC-MSMS) were used. The method resulted in good linearity (Pearson's R > 0.99), low limits of detection (7.78–16.35 ng/kg, 8.26–34.01 ng/kg, 6.70–33.65 ng/kg, and 5.92–19.07 ng/kg for milk, liver, egg, and muscle, respectively), and appropriate limits of quantification (50 ng/kg for all compounds except for GenX and C6O4, where the limits of quantification were 100 ng/kg). Trueness and precision for all the tested levels met the acceptability criteria of 80–120% and ≤20%, respectively, regardless of the analyzed matrix. As to measurement uncertainty, it was <50% for all compound/matrix combinations. These results demonstrate the selectivity and sensitivity of the method for simultaneous trace detection and quantification of 14 PFASs in foods of animal origin, verified through the analysis of 63 food samples.

Abstract:In response to the European Food Safety Authority's establishment of a tolerable weekly intake (TWI) for the sum of PFOA, PFNA, PFHxS, and PFOS, a method was developed to quantify and confirm 20 PFASs at the sub-parts-per-trillion level in fruit and vegetables. Improved sensitivity was achieved by (i) increasing the sample intake, (ii) decreasing the solvent volume in the final extract, and (iii) using a highly sensitive mass spectrometer. Except for PFTrDA, target PFASs could be quantitatively determined with an apparent recovery of 90-119%, limits of quantitation down to 0.5 ng/kg, and a relative standard deviation under within-laboratory reproducibility conditions of <28%. The method was successfully applied to 215 fruit and vegetable samples obtained from local grocery stores and markets. Leafy vegetables prove to be the main vegetable category responsible to PFAS exposure, mainly of PFOA, followed by PFHpA and PFHxA.

Abstract:The group of per- and polyfluoroalkyl substances (PFAS) comprises thousands of chemicals, which are used in various industrial applications and consumer products. In this study, a feeding experiment with laying hens and feed grown on a contamination site was conducted, and PFAS were analyzed in the feed and eggs to assess the transfer of PFAS into eggs. A targeted analysis of perfluoroalkyl acids (PFAAs) and different sulfonamides was performed. Additionally, the total oxidizable precursor (TOP) assay was modified by fully oxidizing small amounts of the samples instead of oxidizing their extracts in order to overcome potential losses during extraction. Targeted analysis showed the presence of known PFAAs and four sulfonamides in the feed and egg yolk samples. In the plant-based feed, short-chain PFAAs, methyl and ethyl perfluorooctane sulfonamidoacetic acid (Me- and EtFOSAA), and perfluorooctane sulfonic acid (PFOS) were the most abundant PFAS. In the eggs, PFOS, FOSAA, and its alkylated homologues showed the highest concentrations. The TOP assay revealed the presence of substantial amounts of precursors with different chain lengths from C4 to C8. The highest relative increase of PFOA after oxidation was observed in egg yolk from the end of the exposure period (828%). The results of this study demonstrate the transfer of PFAAs and their precursors into hens' eggs and emphasize the contribution of (known and unidentified) precursors to the overall PFAS burden in edible products. The modified TOP assay approach was shown to be a powerful tool to better assess the total burden of samples with PFAS.

Abstract:Poly- and perfluoroalkyl substances ( PFAS s) have attracted great scientific attention because of their environmental persistence, bioaccumulation potentials, and toxicity. This study validated a simple and efficient sample preparation method for determining extractable PFASs, including 12 perfluoroalkyl carboxylic acids and 4 perfluoroalkyl sulfonates. The samples underwent ultrasonic extraction, and the extracts were cleaned up by dispersive solid-phase extraction technique with graphitized carbon if necessary. PFAS quantification was performed by a liquid chromatography-tandem mass spectrometry system. The instrument provided analytical signals for all target compounds with good repeatability (RSD < 15%), reproducibility (RSD < 20%), linearity ( R 2 > 0.99), and low detection limits (0.10–1.0 ng/mL). The validated method exhibited adequate recovery (70–115%), precision (RSD < 15%), and method detection limits (0.020–0.70 ng/g). This validated method was applied to analyze PFASs in several food packaging samples collected in Vietnam, showing very low levels (range 0.090–2.9 ng/g; median 0.72 ng/g) derived from unintentional sources.

Abstract:Protein sulfation in baby hamster kidney cells (BHK) and their polyoma virus transformants (PY-BHK) was studied comparatively. On in vivo labeling, [35S]-sulfate was incorporated into the 50K protein and proteins in the 100-180K range, represented by the 155K protein. The incorporation into both the 50K and 155K protein was elevated 2-3 fold in PY-BHK cells compared to in BHK cells. Tyrosine-O-sulfate was the only identifiable sulfated amino acid in both proteins. On in vitro labeling with [35S]3'-phosphoadenosine 5'-phosphosulfate (PAPS), at least 6 radioactive protein bands were discernible on gel electrophoresis. Of these, sulfation of the 57K and 60K proteins was elevated in PY-BHK cells compared to in BHK cells, whereas sulfation of the 39K protein was depressed in PY-BHK cells. Tyrosine-O-sulfate was the only identifiable sulfated amino acid in these proteins.

Abstract:A robust method was developed for simultaneous determination of nine trace perfluoroalkyl carboxylic acids (PFCAs) in various edible crop matrices including cereal (grain), root vegetable (carrot), leafy vegetable (lettuce), and melon vegetable (pumpkin) using ultrasonic extraction followed by solid-phase extraction cleanup and high liquid chromatography-tandem mass spectrometry (HPLC-MS/MS). The varieties of extractants and cleanup cartridges, the usage of Supelclean graphitized carbon, and the matrix effect and its potential influencing factors were estimated to gain an optimal extraction procedure. The developed method presented high sensitivity and accuracy with the method detection limits and the recoveries at four fortification levels in various matrices ranging from 0.017 to 0.180 ng/g (dry weight) and from 70% to 114%, respectively. The successful application of the developed method to determine PFCAs in various crops sampled from several farms demonstrated its practicability for regular monitoring of PFCAs in real crops.

Robust trace analysis of polar (C2-C8) perfluorinated carboxylic acids by liquid chromatography-tandem mass spectrometry: method development and application to surface water, groundwater and drinking water

Abstract:A simple and robust analytical method for the determination of perfluorinated carboxylic acids (PFCAs) with C2 to C8 chains, based on solid-phase extraction (SPE) and liquid chromatography-tandem mass spectrometry (LC-MS/MS), was developed, validated and applied to tap water, groundwater and surface water. Two stationary phases for LC (Obelisc N and Kinetex C18) and two materials with weak anion-exchange properties for SPE (Strata X-AW and Oasis WAX) were evaluated. Robust separation and retention was achieved with the reversed phase column and an acidic eluent. Quantitative extraction recoveries were generally achieved for PFCAs with C > 3, but extraction efficiencies were different for the two shortest chained analytes: 36 to 114% of perfluoropropanoate (PFPrA) and 14 to 99% of trifluoroacetate (TFA) were recovered with Strata X-AW, while 93 to 103% of PFPrA and 40 to 103% of TFA were recovered with Oasis WAX. The sample pH was identified as a key parameter in the extraction process. One-step elution-filtration was introduced in the workflow, in order to remove sorbent particles and minimise sample preparation steps. Validation resulted in limits of quantification for all PFCAs between 0.6 and 26 ng/L. Precision was between 0.7 and 15% and mean recoveries ranged from 83 to 107%. In groundwater samples from sites impacted by per- and polyfluoroalkyl substances (PFASs), PFCA concentrations ranged from 0.056 to 2.2 μg/L. TFA and perfluorooctanoate were the predominant analytes. TFA, however, revealed a more ubiquitous occurrence and was found in concentrations between 0.045 and 17 μg/L in drinking water, groundwater and surface water, which were not impacted by PFASs.

Abstract:Per- and polyfluorinated alkyl substances (PFAS) can be added to food contact materials (FCM) to increase their water and/or grease repellent properties. Some well-known PFAS are perfluoroalkyl carboxylic acids (PFCA), perfluoroalkyl sulfonic acids (PFSA), and polyfluorinated telomer alcohols (FTOH). Due to the strength of the carbon-fluorine bond, PFAS are chemically very stable and highly resistant to biological degradation, posing a risk to human health and the environment. To examine the presence of PFAS in paper-based FCM, various samples were collected, including popcorn bags, muffin cups, and pizza boxes with high total organic fluorine (TOF) content from the Danish and Spanish markets. The FCM composition was characterised by FTIR. Quantification of some well-known PFAS such as PFCA, PFSA, and FTOH was performed in food simulants using LC-MS/MS, and in addition a non-targeted screening approach was performed by LC-Orbitrap-HRMS. Among analysed samples, the highest concentrations of PFAS were found in a muffin cup made of cellulose (PFCA ∼ 1.41 μg kg −1 food, FTOH ∼ 11.5 μg kg −1 food), and the results were used to estimate dietary exposures to PFAS migrated from this FCM. Compared to measured TOF value in this sample, the fluorine from all quantified PFAS accounted for only 0.6%. Thus, a more powerful analytical approach was used to further investigate PFAS occurrence in this sample. Using non-targeted screening, an additional twenty compounds were identified, among them five with confidence level 1 and ten with confidence level 2. Many of them were either fluorotelomer carboxylic acids or sulfonic acids or ether-containing compounds.

Abstract:Per- and polyfluoroalkyl substances (PFAS) analysis has become crucial due to their presence in the environment, their persistence and potential health risks. These compounds are commonly used in food contact materials (FCM) as a coating to provide water and grease-repellent properties. One of the pathways for PFAS to enter the human body is either through direct consumption of contaminated food or indirectly through migration from FCM into food. The purpose of this study was to investigate where the initial contamination of paper FCM occurs. We analysed paper material consisting of fresh fibre and secondary materials, intended to produce food packaging for the presence of PFAS. The samples were extracted and analysed for 23 different PFAS substances using the targeted approach with LC tandem mass spectrometry (LC-MS/MS). This analytical technique detects specific, easily ionisable PFAS with high sensitivity. However, one drawback of this approach is that it allows the identification of less than 1% of the PFAS known today. For this reason, we used combustion ion chromatography (CIC) to determine the content of extractable organic fluorine compounds (EOF) and compare it to the total fluorine content. The targeted analysis using LC-MS/MS measured an average sum concentration of PFAS of 0.17 ng g −1 sample. Our research shows that the primary PFAS contamination happens during the recycling process since all of the samples in which the targeted PFAS were measured belonged to the secondary material. The most frequently detected analytes were PFOA and PFOS, detected in 90% and 62% of the samples, respectively, followed by PFBS (in 29% of the samples). CIC showed that measured PFAS via LC-MS/MS amount to an average of 2.7 × 10 −4 % of total fluorine content, whereas the EOF was under the LOD in all of the measured samples. This result highlights the complexity of the accurate determination of PFAS compounds, displaying what kind of information the chosen methods provide.

Abstract:A new rapid modified one-step QuEChERS (quick, easy, cheap, effective, rugged, and safe) approach was developed for the simultaneous determination of 20 perfluoroalkyl substances (PFASs) from samples of cucumber, lettuce, eggplant, tomato, and leek. Several parameters were optimized. Similar pretreatment approaches were employed for comparison; the results verified the satisfactory performance of this new method. The detection limits of this method for the selected matrices ranged from 0.003 to 0.034 mu g kg(-1), and the method was verified to be satisfactory in terms of precision, accuracy, and matrix effects. 35 greenhouse vegetable samples were determined. PFASs were detected in 26 samples, with perfluorooctanoic acid (PFOA) being detected most frequently and much higher levels of PFBA and PFPeA being found in some samples. The total PFAS concentrations ranged from not detectable to 0.683 mu g kg(-1). This method could be applied for large scale determination of vegetables to research the migration and accumulation trends of PFASs from the environment to crops in the future. (C) 2019 Elsevier Ltd. Abstract:Analysis of neutral and ionic per- and polyfluoroalkyl substances (PFAS) required two discrete techniques- with methods based on gas chromatography–mass spectrometry (GC-MS) for the former and ultrahigh performance liquid chromatography-tandem mass spectrometry (UPLC-MS/MS) for the latter. In this study, we developed a method for simultaneous analysis of 58 neutral and ionic PFAS in environmental samples, obviating the need for distinct extraction and instrumental methods. The UPLC-MS/MS method was optimized and validated for sensitivity, selectivity, linearity, precision, and accuracy. The instrumental detection limits for fluorotelomer alcohols (FTOHs) ranged from 0.08 to 0.50 pg on the column. Select environmental samples, including food packaging, indoor dust, roadside soil, polyurethane foam, carpet, and consumer products, were analyzed. The highest concentrations of polyfluoroalkyl phosphates (Σ6PAPs, 650 ± 910 ng/g) and fluorotelomer sulfonates (Σ4FTSs, 36 ± 51 ng/g) were found in indoor dust, whereas Σ5FTOHs (81 ± 69 ng/g) were found in carpet. Emerging PFAS, including F-53B, ADONA, and Gen-X, were found at sub ng/g concentrations. Hydrogen-substituted polyfluorocarboxylates (H-PFCAs), such as 7H-PFHpA, were found at concentrations of up to 4 ng/g in indoor dust. This method can expedite the analysis of neutral PFAS precursors, including FTOHs, PAPs, and FTSs along with legacy iPFAS in environmental samples.

Abstract:Per-and polyfluoroalkyl substances (PFASs) have garnered significant attention owing to their prevalence and adverse effects on humans. The direct dietary intake of perfluoroalkyl acids (PFAAs) and PFAAs precursors (pre-PFAAs) biotransformation are considered major contributors to human exposure to PFASs. However, little information is available on analytical methods for the simultaneous detection of PFAAs and pre-PFAAs. In the present study, a single-step sample-treatment-based supramolecular solvents-dispersed liquid-liquid microextraction (SUPRASs-DLLME) technique was established for the analysis of 16 PFAAs and 7 pre-PFAAs in aquatic food. SUPRASs were synthesized using 1-heptanol (3 mL) and tetrahydrofuran (4 mL), which were self-assembled in water. The parameters for microextraction, such as extraction method and enrichment capacity, were optimized. Under the optimum conditions, the limit of detection (LOD) and limit of quantification (LOQ) were 0.03-0.15 ng·g-1 and 0.1-0.5 ng·g-1, respectively. Good linearities (R2 > 0.996) were obtained for all the target compounds, and the recoveries ranged 81.1-120 % with relative standard deviations (RSDs) lower than 20 %. This method was applied to the analysis of 16 PFAAs and 7 pre-PFAAs in aquatic food samples (crabs, prawns, and fish). This study provides a new idea for analyzing other pollutants in biological samples.

Food Additives and Contaminants - Part A Chemistry, Analysis, Control, Exposure and Risk Assessment

Abstract:Food contact materials (FCMs) from three countries were analysed for all extractable organofluorines (EOFs) from the materials and subsequently by target and non-target analysis for per- and polyfluoroalkyl substances (PFAS). The EOF varied by two orders of magnitude for FCM from UK and Saudi Arabia ranging between 2.14 and 483 ng cm −2 (0.2–48 ng g −1 ) showing that one quarter of all samples were above the Danish regulation for PFAS in FCM. Target PFAS showed high variability in composition and accounted for less than 1% of the EOF. Non-target PFAS screening using HPLC–ICP-MS and coupled simultaneously to HRMS showed the occurrence of organofluorines which were identified by neither LC–MS/MS nor LC–HRMS. This illustrates that the current target PFAS approaches fail to identify EOFs from FCM, which would be a problem with the new EU proposal to ban all PFAS.
