Abstract:Three adult black howler monkeys (Alouatta pigra) were vasectomized for the purpose of population control. Two different surgical approaches (inguinal and pelvic) were used to vasectomize the animals under general anesthesia. One monkey was vasectomized with a pelvic approach, and the other two monkeys were vasectomized with an inguinal approach. The inguinal approach was minimally invasive and proved to be an effective field procedure, utilizing minimal surgical equipment with a mean operative time of 35 min. The inguinal approach allowed for better visualization of the spermatic cord, which prompted easier ligation and transection of the ductus deferens. Identification of the ductus deferens was successfully performed intraoperatively using a tuberculin syringe to aseptically aspirate cord contents, as histopathology was not available. All howler monkeys fully recovered without complication. The social hierarchy of the troop and mating behavior has not been affected. There have been no new pregnancies reported in the troop since the vasectomies were performed. The inguinal approach is preferred as it is minimally invasive and can be performed confidently in a field setting.

Abstract:The capybara ( Hydrochoerus hydrochaeris ) is currently the largest known representative of the Rodentia order. It is an important herbivorous animal from the zootechnical point of view, found in different countries in South America, mainly in Brazil, where it can be found in all its States. The study of the anatomy of the lumbosacral plexus and medullary cone of the species could contribute to the development of comparative anatomy, and to adoption of future clinical and surgical procedures in the species. The present study proposed to conduct an exploration of the nerves of the lumbosacral plexus in capybara to highlight their relationship with the adjacent muscle tissues. Four adult capybaras, two males and two females, were used for dissection of the muscles innervated by the nerves that make up the lumbosacral plexus. The results allowed to verify that the lumbosacral plexus is formed by ventral roots originating in the six lumbar vertebrae and the three sacral vertebrae. The medullary cone was found between L6 and S1 in the sixth lumbar vertebra (L6) and the first sacral vertebra (S1). The identification and the recognition of these nerves in capybaras are important to improve the assessment of possible injuries caused by inadequate management, accidental mechanical shocks, or peripheral nervous system disorders. Additionally, the study included the topographic analysis of the medullary cone, essential for improving the anesthetic techniques by epidural route. The expansion of this type of research with capybaras can be useful for developing management techniques that preserve the species health and well-being.

Abstract:Polar bears (Ursus maritimus) face a number of challenges that threaten the survival of the species. Captive breeding represents one essential facet of species conservation, but aspects of the polar bear's reproductive physiology, such as follicle maturation, coitus-induced ovulation, and pseudopregnancy, are poorly characterized and present challenges for enhancing natural reproductive success and the application of advanced reproductive techniques. Due to the absence of a reliable transrectal or transabdominal ultrasound method for ovarian examination in the species, the ovaries of two adult female polar bears were examined laparoscopically to evaluate the feasibility of surgical access to the ovaries, oviduct, and uterus. The minimally invasive procedure was easily and rapidly performed in both bears and all procedures. Direct visual assessment of the ovary was possible after dissection of a fatty bursal sac, which completely enclosed the ovaries. In the second bear, laparoscopic manipulation of the ovary to draw it closer to the body wall enabled transcutaneous ultrasound. Laparoscopy may be a valuable tool to aid in the application of advanced reproductive technologies in polar bears.

Abstract:To compose the experiment, 160 male Topgen pigs - 80 surgically castrated (treatment 1) and 80 immunocastrated (treatment 2) - were randomly selected from a commercial swine farm at the moment of slaughter. Surgical castration was performed when the animals were seven days of age and immunocastration was performed by administering two doses (2 mL each) of immunocastration vaccine (analogue of GnRF linked to a carrier protein, development of anti-GnRF antibodies, 200 mg of a GnRF-protein conjugate/mL) when they were 104 and 132 days of age. Animals from both treatments were maintained in masonry stalls, where they received water and ad libitum diet (the same feed for both groups). The animals were slaughtered at 160 days of age, and the length and width of the testicles of immunocastrated animals were evaluated, along with the degree and number of carcass lesions, carcass traits and meat quality of both treatments. The means of these measurements were calculated and compared by Student ́s t-test. For the immunocastrated treatment, Pearson ́s correlation coefficients were also calculated for testicle length and width with backfat thickness. Approximately 80% of the immunocastrated animals had testicle widths of 11 cm or less. Immunocastrated animals showed higher degrees of lesions, pH (initial and 8 hours), hue, muscle depth and loin eye area and lower brightness, redness, chroma and backfat thickness than the surgically castrated animals. The correlation between testicle length and width with backfat thickness was inverse. Immunocastration can be an alternative to improve the proportion of lean meat (longissimus thoracis) instead of fat (backfat thickness) leading to better carcass and meat quality, since fat has become undesirable from a nutritional point of view in swine.

Abstract:Objective: To report the treatment and outcome of a a captive chimpanzee (Pan troglodytes) undergoing 3-portal laparoscopic hysterectomy. Additionally, the technique used for successful urinary catheterization is described. Animals: A 29-year-old female intact chimpanzee with uterine bleeding. Study design: Clinical case report. Methods: Uterine changes consistent with adenomyosis and/or endometriosis were noted on abdominal ultrasonographic and computed tomographic examinations. A urinary catheter was placed before a 3-portal laparoscopic hysterectomy with a uterine manipulator (VCare) and a vessel sealer (Ligasure). The uterus was submitted for histopathology. Results: Preoperative urinary catheterization was achieved with several modifications and prevented bladder protrusion into the surgical field. Laparoscopy provided excellent visualization of the pelvic structures and VCare effectively maneuvered the uterus for a safe and efficient dissection. The use of the vessel sealer was effective, and bleeding was minimal. Anesthesia and surgery lasted 240 and 150 minutes, respectively. No complications were encountered. Histopathological changes of the uterus were consistent with adenomyosis and myometrial hyperplasia. The chimpanzee recovered uneventfully and returned to normal behavior with no recurrence of uterine bleeding 5 months after surgery. Conclusion: The 3-portal laparoscopic technique reported here allowed hysterectomy without complication in this chimpanzee. Urinary catheterization was technically challenging but successful.

Abstract:Introduction/Objective Vasectomy is a widely performed male sterilization surgery with rapid recovery. In Brazil, it is carried out following legal criteria for family planning. Vasectomy reversal is a procedure with a high success rate and is considered a cost-effective option compared to other fertility techniques. The microsurgical technique, developed in 1975, is widely adopted for reconstructing the vas deferens. Vasovasostomy is the gold standard technique, using non-absorbable sutures (9-0 or 10-0). The aim of this study is to correlate the literature's results with those achieved through the microsurgical technique employed at the Pedro Ernesto University Hospital. Method This study was a retrospective and descriptive analysis of medical records and a review of the literature on a surgical technique. We analyzed 56 cases performed between January 2012 and December 2022 at the Pedro Ernesto University Hospital. Additionally, scientific articles published between 1999 and 2022 were collected. Surgeries were performed under general or spinal anesthesia, using a single-layer technique with Prolene 7-0 sutures. All patients underwent sperm analysis at 1, 3, and 6 months postoperatively. Results A total of 56 vasectomy reversals were performed in patients with an average age of 42.91 years and an average interval of 7.65 years between vasectomy and reversal. The patency rate was 82.14%, and the pregnancy rate among those who were successfully reversed was 15.22%. Reversal was more successful (87.18% patency and 25% pregnancy) in patients with an interval of less than 10 years between surgeries. Conclusion The results of 56 vasectomy reversals by vasovasostomy using the modified microsurgical technique applied at the Pedro Ernesto University Hospital demonstrated satisfactory patency and pregnancy rates, closely matching those reported in the literature. The technique proved to be effective and safe, with no significant complications. Financing No conflict.

[Salmeterol and fluticason and mortality in COPD patients].

Abstract:In this study 6,112 COPD patients were treated with salmeterol 50 microg bid, fluticason 500 microg bid, salmeterol/fluticason combination 50/500 microg bid (SFK) or placebo for 3 years. The primary effect parameter was mortality and 875 patients had died after 3 years. Hazard ratio for death for SFK compared with placebo was 0.825, 95% confidence interval 0.681-1.002, p=0.052, or an absolute risk reduction of 2.6%. Active treatment reduced exacerbations and improved quality of life and lung function. The risk of pneumonia was higher in the fluticason and SFK treated groups than in the placebo group.

Abstract:Porcine circovirus type 2 (PCV-2) is the agent of one of the most important diseases in the swine industry. Although it has been controlled through vaccination, viremic piglets at birth may represent a risk by reducing vaccination efficacy. Since there are few reports on the viremic status of pre-suckling piglets regarding PCV-2 infection, we assessed the PCV-2 frequency in sows housed in 18 breeding farms with no history of clinical PCVAD in Brazil, using placental umbilical cord serum (PUCS). The selection criteria were: breeding farms with more than 1,000 sows; sows not vaccinated for PCV-2 at least for 2 years prior to the study; farms with no history of PCV-2 clinical disease in the last 12 months; and production systems with a maximum of two sites. Blood from the umbilical cords in sow placenta or directly from piglet's immediately after birth was collected from 30 litters on each farm for PCR. In addition, blood from 538 sows was collected for PCV-2 antibody detection. A total of 17.29% of the PUCS tested positive. The PCV-2 DNA was detected in PUCS from 94.4% of all farms. A total of 94.8% of the sows was positive for PCV-2 antibodies. However, seronegative sows were detected in 44.4% of farms. All 18 farms had at least 46.9% seropositive dams. A higher percentage of seronegative sows was observed for farms with more than 10% of PCV-2-positive litters compared to those with ≤10% of PCV-2 positive litters (8.9 +/−1.7% vs. 1.5 +/− 0.7%, p < 0.01, respectively).

[Spergualin, a new antitumor antibiotic].

Abstract:Spergualin is a new antitumor antibiotic which has a very unique structure. It was shown to have chemotherapeutic effects on mice inoculated with various mouse leukemias such as L-1210 (IMC), L-1210, P388, P815, C1489, EL4 and RL male 1. Among these, spergualin was especially effective against L-1210 (IMC) tumors, where the mice were completely cured at an appropriate drug dosage. These cured mice rejected the second challenge of L-1210, but not other tumors, indicating that specific immunity had been induced by spergualin. Preliminary data suggested that killer T cells were acting as effector cells. There was some correlation between the cytocidal activity of spergualin and the concentrations of amine oxidases in the serum. These results suggest that the induction of tumor-immunity plays an important role in the antitumor activity of spergualin.
Moment of induction and duration of experimental varicocele in rats: effects on semen quality

Abstract:Purpose: Varicocele is a condition known to cause damage to seminal parameters and sperm function. Furthermore, it has been hypothesized that the varicocele effect on fertility is time-dependent; however, little is known about the consequences of its establishment time on reproductive organs and/or sperm function. This study aimed to evaluate the effect of the duration of experimental varicocele on reproductive organs, sperm parameters, and sperm function. Materials and methods: Varicocele induction surgeries were performed in Wistar rats aged 40 or 100 days old. At 160-day-old, analyses were performed, including biometry of reproductive organs (prostate, seminal vesicles, epididymis, and testis), sperm parameters (vitality, morphology, and motility), and sperm function tests (nuclear DNA integrity, acrosome integrity, and mitochondrial activity). Results: The analysis of the biometry of reproductive organs showed no differences between distinct ages in which varicocele was induced. The total abnormal sperm morphology was bigger in animals with varicocele induced to 100 days old than in animals with varicocele induced to 40 days old. Regarding nuclear DNA integrity, animals of varicocele induced to 100 days old showed worse results compared to animals of varicocele induced to 40 days old. Other parameters analyzed showed no differences between varicocele groups. Conclusion: In this study conducted on rats, we conclude that varicocele adversely affects sperm, particularly its function. However, we did not observe a negative progressive effect on sperm.

Abstract:Abstract Background The objective of the present study was to explore the benefits of Porcine circovirus 2 (PCV-2) blanket vaccination in a sow herd on productive parameters, PCV-2 infection and immune status in sows and their progeny. For this purpose, 288 sows were distributed among four balanced experimental groups. One group remained as negative control group and the other three received 1 mL of PCV-2 Ingelvac Circoflex® intramuscularly at different productive cycle moments: before mating, mid gestation (42–49 days post-insemination) or late gestation (86–93 days post-insemination); phosphate buffered saline (PBS) was used as negative control item. Reproductive parameters from sows during gestation and body weight of their progeny from birth to weaning were recorded. Additionally, blood was collected from sows at each vaccination time and piglets at 3 weeks of age. Moreover, up to 4 placental umbilical cords (PUC) per sow were taken at peri-partum. Sera from sows and piglets were analysed for PCV-2 antibody detection using an enzyme-linked immunosorbent assay (ELISA). Sera from sows and PUC were tested to quantify viraemia using a real time quantitative polymerase chain reaction (qPCR) assay. Results Globally, results indicated that vaccinated sows showed heavier piglets at birth and at weaning, less cross-fostered piglets, lower viral load at farrowing as well as in PUC, and higher antibody levels at farrowing, compared to non-vaccinated ones. When all groups were compared among them, sows vaccinated at mid or late gestation had heavier piglets at birth than non-vaccinated sows, and lower proportion of PCV-2 positive PUC. Also, cross-fostering was less frequently practiced in sows vaccinated at pre-mating or mid gestation compared to non-vaccinated ones. Conclusions In conclusion, the present study points out that PCV-2 sow vaccination at different time points of their physiological status (mimicking blanket vaccination) offers benefits at production and serological and virological levels.

Induction of hypersensitivity of brain by hydralazine treatment in rats.

Abstract:Pretreatment with hydralazine caused prolongation of hypnosis induced by hexobarbital (80 mg/kg, i.p.) or intracerebroventricular injection of phenobarbital (1 mg/rat). Under the same condition, brain concentration of hexobarbital was lower than that of no hydralazine treatment. In addition, no appreciable change in liver microsomal drug metabolizing enzyme activities of rats after hydralazine treatment was observed. These findings strongly suggested that hydralazine increases the sensitivity of brain to barbiturates, which leads to prolongation of sleeping time without inhibition of drug metabolizing enzymes.

Abstract:We have realized a 46F/sup 2/ SRAM cell size of 0.294 /spl mu/m/sup 2/ with 80 nm technology and single stack S/sup 3/ cell technology. SSTFTs and vertical node contacts are major keys in the S/sup 3/ cell technology. The stacked single crystal silicon thin film is developed for the load pMOS SSTFT of the S/sup 3/ SRAM cell. The load pMOS SSTFT is stacked on ILD to reduce the SRAM cell size. Fully working 64 Mbit SRAM is achieved by this S/sup 3/ cell technology. The basic reliability of SSTFT, with 80 nm length, is also investigated in this study.

Abstract:Introduction and Objective Varicocele is the most common identifiable cause of male infertility, and varicocelectomy is the most frequently performed surgery for its treatment. This condition impacts infertility cases and sperm quality. Microsurgical varicocelectomy is considered the gold standard treatment for varicocele. This retrospective cohort aims to evaluate the effects of microsurgical varicocelectomy on seminal parameters, serum testosterone, and pregnancy rate in the context of male infertility in 39 patients who underwent the procedure at a university hospital in Rio de Janeiro. Methods This study is a retrospective cohort conducted between January 2016 and February 2023, involving 39 men who underwent subinguinal microsurgical varicocele repair in the context of marital infertility. Pre- and post-operative analyses of seminal parameters were based on the criteria of the World Health Organization from 2010. Post-operative semen analysis and serum testosterone levels were assessed 3 to 6 months after varicocelectomy. The pregnancy rate was observed through interviews until February 28, 2023. Results Subinguinal microsurgical varicocelectomy is associated with a significant increase in testosterone levels and an improvement in seminal parameters, all with statistical significance. Testosterone levels increased by an average of 84.2 (±14.7) ng/dl. The pregnancy rate also increased significantly, with 51.3% of couples achieving pregnancy as of the time of this study. Conclusion In this patient population, infertile men with clinical varicocele who underwent subinguinal microsurgical varicocelectomy showed a significant increase in testosterone levels, improvement in seminal parameters, and a significant reduction in the proportion of azoospermia. Furthermore, there was an increase in the occurrence of pregnancy in couples with marital infertility. Statistical ResultsFinancing No conflict.

Single injection of eCG/hCG leads to successful estrous synchronization in the collared peccary (Pecari tajacu Linnaeus, 1758)

Abstract:The establishment of protocols for the control of the ovarian function of collared peccaries is recommended for the development of assisted reproductive techniques. The goals were to (1) compare a gonadotropin combination with prostaglandin analogue to synchronize timing of onset of estrus among animals, and (2) elucidate the effects of the most desirable protocol for performing an artificial insemination study and macroscopic evaluation of the ovaries. Three of five females treated with a double administration of 120 μg prostaglandin (cloprostenol) at a 9-day interval expressed symptoms of estrus 9 days after the second injection. One female presented estrus after 6 days, whereas other did not respond to the treatment. All females (5/5) treated with a single dose containing 400 IU eCG and 200 IU hCG manifested estrus 6 days after the hormone injection. In a second experiment, ten females that were estrous synchronized using eCG/hCG, were artificially inseminated with fresh semen and monitored for pregnancy every 30 days. Although there was no detection of fetuses by ultrasonic examination, seven females (7/10) had greater than basal progesterone values for 60 days after the treatments were imposed. Ovaries from two females treated with eCG/hCG were collected 6 days post-injection. There was confirmation of an ovarian stimulation as a result of the presence of 88 and 25 antral follicles, as well as three and eight hemorrhagic structures in ovaries of each female, respectively. It, therefore, is proposed that eCG/hCG can be used as an effective treatment for estrous synchronization in collared peccaries.

Thus, this study evaluated the ovulation rate, presence of functional corpora lutea and fecal glucocorticoid levels following treatments promoting superovulation in captive brown brocket deer.MethodsThe crossover design used six hinds, allocated to two groups (n = 6): eCG Treatment, CIDR for 8 days, followed by 0.25 mg of EB on day 0, 700 IU of eCG on day 4 following device insertion and 265 mug of PGF2alfa on day 8; and FSH Treatment, CIDR for 7.5 days, followed by 0.25 mg of EB on day 0, 130 mg of FSH in 8 equal doses and 265 mug of PGF2alfa on day 7.5. Induced adrenal activity and treatment efficacy were evaluated by corpora lutea (CL) counts and fecal glucocorticoid and progestin concentration (ng/g feces) analyses for five different phases: Pre, two days before treatment; Early, first four days of treatment; Late, last four days of treatment; Total, entire treatment period; and Post, five days posttreatment.ResultseCG Treatment resulted in the highest number of CL (P lower than 0.05). There was no significant difference for fecal glucocorticoid concentrations in five different time periods between the treatments; however Pre fecal glucocorticoid concentrations (90.06+/−19.64) were significantly different from Late (200.76+/−26.39) within FSH Treatment. The mean fecal progestin concentration and mean ovulation rate were higher in eCG Treatment (4293.69+/−769.47, 7.0+/−1.8) than in FSH Treatment (1571.26+/−240.28, 2.6+/−0.8) (P lower than or equal to 0.05).ConclusionsAlthough the eCG Treatment induced a good superovulatory response, with the formation of functional corpora lutea, we cannot yet affirm that we have established a suitable protocol for induction of SOV in the species M. gouazoubira because approximately 65% of the deer showed premature regression of the corpora lutea. Moreover, multiple FSH applications in FSH Treatment resulted in a low ovulation rate and induced an increase in fecal glucocorticoid levels.

Abstract:The effects of antibiotics on sperm longevity in collared peccary (Pecari tajacu) fresh diluted semen was evaluated. Semen samples from six adult males were collected by electroejaculation and diluted in Tris-citrate-fructose alone (control) and plus streptomycin-penicillin (2 mg/ml-2000 IU/ml) or gentamicin (70 µg/ml). Membrane integrity and functionality, mitochondrial activity and sperm morphology were assessed subjectively. Sperm motility and other kinetic parameters were objectively assessed using CASA (computer-assisted semen analysis). The semen diluted according to the treatments were submitted to the thermoresistance test, incubated at 37 ° C, and the sperm parameters analyzed at 0, 30, 60, 120 and 180 min. The average values of the treatments were compared with each other and between the times. There were no differences (P > 0.05) between treatments until the end of the test. Control and streptomycin-penicillin samples maintained sperm function for up to 180 min (with total motility of 24.3 ± 7.1% and 28 ± 8.7%, respectively). Gentamicin aliquots retained most parameters until the end of the incubation, except for membrane integrity and mitochondrial activity that declined (P < 0.05) at 180 min (53.1 ± 7.1% and 50.7 ± 6.2%, respectively) compared to 0 min (80.5 ± 4.7% and 86.3 ± 3.4%, respectively). In conclusion, a multiparametric thermoresistance test proved that Tris-based extenders used for collared peccary semen can be effectively supplemented by streptomycin-penicillin (2 mg/ml-2000 IU/ml) or gentamicin (70 µg/ml), especially during 180-min incubation at 37 °C.
