Abstract:Rabies, one of the most lethal global zoonoses, affects all mammals. It remains circulating worldwide in sylvatic cycles through terrestrial and airborne reservoirs, and in Brazil, bats are currently the main reservoirs and source of transmission. Wild boars, an important invasive alien species in Brazil, are a proven food source for hematophagous bats and may participate in the Brazilian sylvatic cycle of rabies. We evaluated the presence of this pathogen in hunted wild boars from the São Paulo state using histopathology, the direct fluorescent antibody test (DFA), viral isolation in cell culture (VICC), the rapid fluorescent focus inhibition test (RFFIT), and quantitative reverse transcription polymerase chain reaction (RT-qPCR). The results of histopathological, DFA, VICC, and RT-qPCR analysis were negative for all samples; seven serum samples tested positive in the RFFIT, and titers ranged from 0.13 IU/mL to 0.5 IU/mL. The presence of rabies virus-neutralizing antibodies in the studied wild boars suggests the circulation of the virus in these animals. Educative actions directed at hunters should include information on the prevention of this important zoonosis.

Abstract:Abstract The present study updates previously published biodiversity/faunistics of the flea beetles of Mexico published by the author after examination of 6132 specimens from 8 institutional collections. The following 9 genera were selected as indicators of the effects of known diversity only through examination of museum specimens (i.e., “indoor collecting”): Alagoasa Bechyné; Asphaera Chevrolat; Capraita Bechyné; Disonycha Chevrolat; Kuschelina Bechyné; Omophoita Chevrolat; Prasona Baly; Systena Chevrolat; and Walterianella Bechyné. From the specimens examined in these genera from the 8 collections, there were 394 new records for Mexican states of the 287 new species records representing 47% new records of the species recorded from those states. Total new state records 287 from 80 species. States with most new records: Chiapas (32); Nayarit (27); Sinaloa (24). 80 spp. (47%) with new state records. Systena oberthuri Baly is reported from Mexico for the first time. The current total of Alticinae in Mexico is 90 genera/626 species. The difficulties of the generic boundaries between Systena and Prasona, Alagoasa and Kuschelina; as well as the specific boundaries between A. jacobiana and A. decemguttatus and the specific level pattern variation in Disonycha glabrata and Alagoasa decemguttatus are discussed. Kuschelina semipurpurea, formerly placed in Alagoasa, is placed is considered as a new combination.
High risk of bat bites in an indigenous village in Brazil: Warning of the re-emergence of rabies among the Maxakali People

Abstract:Bat-mediated human rabies is a viral zoonotic disease that poses a serious threat to the public health of traditional peoples, especially indigenous populations that maintain primitive cultural and social habits, such as the Maxakali ethnic group, located in the southeastern region of Brazil. The sociocultural habit of this population led to the emergence between April and May 2022 of the viral spillover of rabies transmitted by bats, which decimated the lives of four children from this population who maintained contact with this animal as a recreational practice. Because the vampire bats Desmodus rotundus have exceptional ecology and social characteristics that can have important effects on the dynamics of viral dispersion in this indigenous population, I present the dynamics of contact between native children and the bat and the meaning of this relationship, which involves ritualistic and recreational significance. As important as knowing the reasons for this practice is discussing some intrinsic and extrinsic factors that imply risks that intensify the vulnerability of this population to the transmission of the rabies virus at any time. In view of this, I warn of the need to adopt efficient strategies to mitigate the risks of a new emergency in this region. Although emergency containment measures were carried out during the critical period of the outbreak, such animal and environmental control actions must become routine programmatic and structuring interventions. Essential for rabies surveillance in this population is to develop culturally adapted interethnic health education campaigns to guarantee the accessibility of the Maxakali indigenous people to the content taught, so that any attempt at domestication, captivity and recreational practices with bats of any species is discouraged, thus avoiding a possible re-emergence of this anthropozoonosis that has impacted not only the epidemiological scenario in this region, but throughout Brazil, and also throughout Latin America.

Abstract:Rabies transmitted by wildlife is the main source of human rabies mortality in Latin America and considered an emerging disease. The common marmoset Callithrix jacchus of Brazil is the only known primate reservoir of rabies worldwide. We tested whether alive free-ranging C. jacchus were exposed to rabies in four northeast states that have previously reported rabies-positive dead C. jacchus (Pernambuco and Bahia) or not (Paraíba and Rio Grande do Norte). Our results show no evidence of rabies antibodies or infection in the sampled C. jacchus , suggesting that apparently healthy marmosets are not widely exposed to rabies over their natural range.

Abstract:This study aimed to investigate the presence of ectoparasites and the occurrence of natural infection by Rickettsia spp. and Trypanosoma spp. in bats from Rio Grande do Sul (RS), Brazil. The evaluated animals were obtained from the Instituto de Pesquisas Veterinárias Desidério Finamor, sent by the Centro Estadual de Vigilância Sanitária, to carry out rabies diagnostic tests, during the period from 2016 to 2021. The bats came from 34 municipalities in RS. Of the 109 animals surveyed, 35.8% (39/109) had 385 ectoparasites, with an average of 9.9 parasites per animal. Of these bats, all had insectivorous feeding habits, with 35.9% (14/39) females and 64.1% (25/39) males. The co-parasitism of Chirnyssoides sp., Ewingana inaequalis, and Chiroptonyssus robustipes on Molossus currentium (Mammalia, Chiroptera) was recorded for the first time. All bats surveyed were negative for infection by the protozoan and bacteria. Thus, the expansion of the occurrence of these ectoparasites in insectivorous bats in RS was observed. Furthermore, this study corresponds to the first recorded interspecific associations for the species.

Abstract:The importance of the common vampire bat Desmodus rotundus for the transmission of the rabies virus does not lie solely in its ability to transmit this disease to other mammals, but also in its capacity to adapt to environmental and climatic changes, granting them a wide geographical distribution. Control of this disease is currently based on culling of the vampire bat and vaccination of the livestock. A transmission model incorporating geographic and behavioral determinants of the vampire bat was proposed to direct and optimize the epidemiological surveillance and control of livestock rabies. This model was built using a bimodal network connecting 260 vampire bat roosts among themselves (roost-roost-network) and with 1557 farms (roost-farm network) in eastern Sao Paulo State, Brazil. These roosts were grouped in 9 communities, some very interconnected, and some not and the farms were grouped in 14 communities. From 2013 to 2017, 44 livestock rabies outbreaks occurred in the area, circulating among the farm communities during the entire period, with possible introductions from neighboring areas. Based on the network and environment parameters, it was possible to reasonably predict both the roosts' occupation type (harem, bachelor, overnight and empty) and livestock rabies outbreak occurrence. The network approach brings light to the importance of phylogenetic studies of bats and rabies virus. Finally, the understanding of the interactions between bats and their feeding sources, influenced by the environment, allows to establish more precise surveillance and control measures and, ultimately, with a lower cost-benefit ratio of these actions.

Abstract:In 2018, an outbreak of human rabies caused by the hematophagous bat Desmodus rotundus hit the Brazilian Amazon Basin community of Melgaço, Brazil, resulting in the death of 10 people, 9 of them children. The incidence of rabies has been on the rise among populations in conditions of vulnerability in this ecosystem due to human expansion into sylvatic environments and limited access to public health services. To address this issue, in September 2019, a collaborative effort from national, local, and international institutions promoted and executed a pilot for pre-exposure prophylaxis of a population in high-risk areas for hematophagous bat-mediated rabies. This measure is usually only implemented in response to outbreaks. The pilot was conducted in Portel, in a nearby location to the previous outbreak, with the use of fluvial transportation, and 2987 individuals in 411 dwellings were successfully vaccinated. It established a methodology for pre-exposure prophylaxis for populations in conditions of vulnerability, identifying logistics and costs, as well as characterizing the target riverine population regarding risk factors associated with bites by hematophagous bats. This approach offers a proactive measure to prevent future outbreaks and provides valuable insights into how to address the issue of rabies in remote and difficult-to-reach areas.
Cross-border surveillance of sheep and goat rabies in Brazil

Abstract:Since Brazil has the largest territory in South America and borders 10 other countries, rabies control is strategic to prevent cross-border spread. However, prevention and control of rabies in small ruminants is neglected. The present study evaluated the spatiotemporal distribution and temporal trends of rabies in small ruminants in Brazil between 2005 and 2023. Official data on rabies case notifications and the population density of goats and sheep from the Brazilian states were used. Descriptive epidemiology and temporal analyses of high-risk clusters and trends were conducted. Rabies cases were reported in all states, except for the Federal District, Roraima, Amapá, Amazonas, and Rondônia. In sheep, 174 cases were reported, with an emphasis on Paraná (25.29 %), being highest. There were 64 cases in goats, with a highlight on Bahia (37.50 %) and Maranhão (18.75 %). However, Espírito Santo presented the highest incidence risk (IR) for goats and sheep. The highest peaks in IR occurred in 2005, 2006 and 2013. A temporal trend of decreasing goat cases was observed in northeastern Brazil from 2005 to 2023. In sheep, the Northeast region showed a downward trend in rabies cases. Three high-risk clusters were identified: the primary cluster for goats occurred in 2006, and for sheep, between 2005 and 2013. Rabies in small ruminants occurs across all Brazilian regions, with high-risk areas in the Northeast, Southeast, and South, as well as a risk of cross-border transmission. These findings support animal health authorities in strengthening rabies control for small ruminants and reducing the risk of transboundary spread.

Abstract:Rabies, caused by the Lyssavirus genus, is a highly lethal zoonotic disease transmitted by animals such as bats and domestic and wild carnivores to humans, claiming nearly 100% of lives. In Brazil, recent evidence suggests an increasing role of bats in human deaths from rabies, particularly in the Amazon region. This neglected tropical disease disproportionately affects impoverished and vulnerable populations in rural areas, where approximately 80% of human cases are concentrated. This article presents research conducted in riverine communities of the Tapajós/Arapiuns Extractive Reserve in Brazil to combat rabies in September 2022. The study adopted a participatory and collaborative approach, involving community members, healthcare professionals, and educators. Prioritizing proactive interventions, the health team administered prophylactic vaccinations to 30 individuals residing in communities exposed to the Lyssavirus. Educational activities focused on dispelling myths and raising awareness about preventive measures, with 100% of individuals reporting prior doubts about the disease, emphasizing the essential nature of the clarification, especially regarding preventive aspects. This study underscores the importance of community involvement, personalized interventions, and ongoing education to effectively combat rabies. By reinforcing public health policies and promoting health education, we can empower communities to take proactive measures in rabies prevention, leading to a reduction in incidence and an improvement in quality of life.

Abstract:Rabies is a neglected disease that affects all mammals. To determine the appropriate sanitary measures, the schedule of preventive medicine campaigns requires the proper identification of the variants of the virus circulating in the outbreaks, the species involved, and the interspecific and intraspecific virus movements. Urban rabies has been eradicated in developed countries and is being eradicated in some developing countries. In Europe and North America, oral vaccination programs for wildlife have been successful, whereas in Latin America, Asia, and Africa, rabies remains a public health problem due to the habitation of a wide variety of wild animal species that can act as rabies virus reservoirs in their environment. After obtaining recognition from the WHO/PAHO as the first country to eliminate human rabies transmitted by dogs, Mexico faces a new challenge: the control of rabies transmitted by wildlife to humans and domestic animals. In recent years, rabies outbreaks in the white-nosed coati ( Nasua narica ) have been detected, and it is suspected that the species plays a significant role in maintaining the wild cycle of rabies in the southeast of Mexico. In this study, we discussed cases of rabies in white-nosed coatis that were diagnosed at InDRE (in English: Institute of Epidemiological Diagnosis and Reference; in Spanish: Instituto de Diagnostico y Referencia Epidemiologicos) from 1993 to 2022. This study aimed to determine whether white-nosed coatis might be an emergent rabies reservoir in the country. A total of 13 samples were registered in the database from the Rabies laboratories of Estado de Mexico ( n = 1), Jalisco ( n = 1), Quintana Roo ( n = 5), Sonora ( n = 1), and Yucatan ( n = 5). Samples from 1993 to 2002 from Estado de Mexico, Jalisco, and Sonora were not characterized because we no longer had any samples available. Nine samples were antigenically and genetically characterized. To date, coatis have not been considered important vectors of the rabies virus. The results from our research indicate that the surveillance of the rabies virus in coatis should be relevant to prevent human cases transmitted by this species.

Immunoscintigraphy of primary and metastatic colorectal cancers with radiolabelled monoclonal antibodies anti-CEA.

Abstract:Thirteen patients with a diagnosis of primary colorectal tumour and 68 patients previously operated for colon cancer underwent an immunoscintigraphy carried out with monoclonal antibodies anti-CEA F(ab')2, labelled with 131I or 111In. These studies led to the detection of all primary tumours and of most of their associated lesions (7/8). In this group 15 neoplastic deposits previously undetected were demonstrated, allowing an improvement in the patient staging before the operation. In the group of the 68 operated patients, immunoscintigraphy succeeded in imaging 96 out of 121 known lesions: the best outcomes were obtained for abdominal and pelvic recurrences as well as for lymph nodes lesions. The lowest levels of sensitivity were observed for liver metastases. An enhancement of the immunoscintigraphy sensitivity was obtained when the radiopharmaceutical was injected intraperitoneally. In 50 patients, this alternative administration route was successful in detecting 91/107 cancer deposits and, in particular, liver metastases were found out in 36/42 cases.

Abstract:Rabies virus (RABV), a fatal zoonotic pathogen, remains a significant public health concern, with bat-maintained lineages accounting for all currently documented cases in Brazil. Despite the availability of pharmacological prophylaxis for humans and animals, the high genetic diversity of RABV in diverse natural bat hosts and continued circulation in multiple animals pose challenges for effective surveillance. Here, we developed and validated a novel, rapidly deployable amplicon-based sequencing approach for rabies virus (RABV) genomic surveillance. This "all-in-one" protocol integrates whole RABV genome sequencing with host species identification through COI gene amplification and sequencing, addressing the challenges posed by RABV's high genetic diversity and complex transmission dynamics. We assessed the protocol's effectiveness by sequencing 25 near-complete RABV genomes from host species across four distinct families (Bovidae, Equidae, Felidae, and Microchiroptera) obtained from the Rabies Control and Surveillance Program in Rio Grande do Sul State, Southern Brazil. The method achieved an average genome coverage of 91.4% at a minimum 5x read depth, with a mean depth coverage of 816x across sequenced genomes. The results demonstrated significant Bat-Clade sublineage diversity, which was classified using the MADDOG RABV lineage system. The protocol successfully identified three bat species (Tadarida brasiliensis, Desmodus rotundus, and Myotis nigricans) among the samples, highlighting its capability for precise host identification. This study presents a powerful tool for high-resolution evaluation of RABV genomic features and host identification, enabling more targeted public health interventions. This new approach has the potential to enhance RABV surveillance capabilities, contributing to more effective rabies control strategies within a One Health framework.

Abstract:Agile methods depend on active communication and effective knowledge sharing among team members for producing high quality working software systems in short releases and iterations. However, effective communication in Distributed Agile Development (DAD) can be challenging due to a number of different factors, such as physical locations, multi-cultures and time-zones. The agile body of knowledge mainly discusses some technology and non-technology solutions and strategies to mitigate the DAD communication challenges from a project management perspective. Nevertheless, it has recently been argued that there is a need to understand and analyze DAD communication from other related but different perspectives, such as enterprise strategy, enterprise architecture and service management. Due to the fact that agile EA provides a holistic view and blueprint of the whole environment in which a number of projects are developed and managed, we attempt in this study to explore the effect of agile Enterprise Architecture (EA) on DAD communication. Particularly, we propose the development of an agile EA driven approach from the architecture body of knowledge for handling the DAD communication challenges that have not been thoroughly investigated before.

Abstract:A standardized interview administered at intake to a cohort of 611 applicants to a community mental health center and to 433 of their family members showed that patients who received hospitalization as their initial assignment were assessed as significantly more ill by their families than those who received other initial outpatient assignments. Patients who were hospitalized did not see themselves as being more ill, however. Patients assigned to Medicine Clinic saw themselves as significantly less psychologically ill.

Abstract:Brazil has been promoting essential improvements in health indicators by implementing free-access health programs, which successfully reduced the prevalence of neglected zoonosis in urban areas, such as rabies. Despite constant efforts from the authorities to monitor and control the disease, sylvatic rabies is a current issue in Amazon's communities. The inequalities among Amazon areas challenge the expansion of high-tech services and limit the implementation of active laboratory surveillance to effectively avoid outbreaks in human and non-human hosts, which also reproduces a panorama of vulnerability in risk communities. Because rabies is a preventable disease, the prevalence in the particular context of the Amazon area highlights the failure of surveillance strategies to predict spillovers and indicates the need to adapt the public policies to a “One Health” approach. Therefore, this work assesses the distribution of free care resources and facilities among Pará's regions in the oriental Amazon; and discusses the challenges of implanting One Health in the particular context of the territory. We indicate a much-needed strengthening of the sylvatic and urban surveillance networks to achieve the “Zero by 30” goal, which is inextricable from multilateral efforts to combat the progressive biome's degradation.

Molecular detection and characterization of Anaplasmataceae agents, Bartonella spp. and hemoplasmas in armadillos and anteaters from Brazil

Abstract:Armadillos and anteaters exhibit a wide range of interactions with various pathogens and ectoparasites, which, along with their physiological and ecological characteristics, contribute to their potential as hosts for a broad variety of pathogens with zoonotic potential. However, there has been limited research into the occurrence and diversity of vector-borne agents in this group of mammals. The present study aimed to investigate the occurrence and molecular identity of Ehrlichia spp., Anaplasma spp., Bartonella spp. and hemotropic mycoplasmas (hemoplasmas) in free-ranging armadillos and anteaters sampled in the southeast and central-west regions of Brazil. To this purpose, 167 biological samples (139 blood and 28 spleen) from Xenarthra mammals sampled in the states of São Paulo and Mato Grosso do Sul were analyzed: 48 from six-banded armadillos (Euphractus sexcinctus), 29 from giant armadillos ( Priodontes maximus ), 4 from nine-banded armadillos ( Dasypus novemcinctus ), 1 from naked-tailed armadillo ( Cabassous unicinctus ), 79 from giant anteaters ( Myrmecophaga tridactyla ) and 6 from southern tamanduas ( Tamandua tetradactyla ). Screening PCR assays were performed for Ehrlichia spp. based on the dsb gene, nested PCR for Anaplasma spp. and PCR for hemotropic mycoplasmas based on the 16S rRNA gene, and qPCR for Bartonella spp. based on the 16-23S rRNA intergenic region (ITS). The positive samples were additionally subjected to PCR assays targeting different molecular markers for molecular characterization. As a result, 1/48 (0.59%) blood sample from E. sexcintus was positive for Anaplasma spp., and 1/79 (0.59%) blood sample from M. tridactyla was positive for Ehrlichia spp. The 16S rRNA sequence of Anaplasma sp. detected in E. sexcintus clustered within the same clade as 'Candidatus Anaplasma brasiliensis ', previously detected in T. tetradactyla . The dsb sequence of Ehrlichia sp. detected in M. tridactyla clustered within the same clade as Ehrlichia minasensis . In the PCR tests for hemoplasmas, 31/46 (64.5%) E. sexcinctus , 9/29 (65.5%) P. maximus , 2/4 (50%) D. novemcinctus , 33/79 (41.7%) M. tridactyla , and 2/6 (33.3%) T. tetradactyla tested positive. The 16S rRNA and 23S rRNA sequences of hemoplasmas found in E. sexcinctus clustered within the same clade as 'Candidatus Mycoplasma haematomaximus ', which was previously detected in P. maximus in Brazil. High positivity rates were also observed for Bartonella spp., with 23/48 (47.9%) E. sexcinctus , 1/4 (25%) D. novemcinctus , 9/29 (31%) P. maximus , 21/79 (26.5%) M. tridactyla , and 3/6 (50%) T. tetradactyla showing positive results. The ftsZ sequences of Bartonella sp. detected in E. sexcinctus clustered with ' Candidatus Bartonella washoensis subsp. brasiliensis', previously identified in six-banded armadillos in Brazil. These findings reinforce the presence of ' Candidatus Anaplasma brasiliensis' and ' Candidatus Bartonella washoensis subsp. brasiliensis' in armadillos, and Ehrlichia minasensis in anteaters. This is the first report of ' Candidatus Mycoplasma haematomaximus', a hemoplasma previously described in giant armadillos, in six-banded armadillos. The zoonotic potential and real significance of infection by these agents in xenarthrans' health and conservation efforts are yet to be investigated.
