Abstract:The amino acid sequence and structure of a uterotonic polypeptide extracted from the African plant Oldenlandia affinis DC have been determined. The peptide, kalata B1, consists of 29 amino acid residues and is rich in cysteine (6), threonine (5), and glycine (5). Enzyme cleavage studies show that the polypeptide backbone is cyclic. The three-dimensional solution structure has been determined using two-dimensional nuclear magnetic resonance (NMR) spectroscopy and distance-restrained simulated annealing. Kalata B1 is composed mainly of beta-strands connected by tight turns, forming regions of beta-sheet, except in the case of one section which forms a longer, less structured loop. The tertiary fold, together with the disulfides that form a sulfur core, produces a striking and unusual surface in which the majority of the hydrophobic residues form a solvent-exposed patch. The hydrophobic side of kalata B1 is flanked by two diametrically opposed and opposite-charged residues. The structure calculations have been used to predict the previously unknown disulfide bond connectivities using two approaches. In the first, a family of structures was calculated on the basis of NOE constraints without the assumption of a specific disulfide connectivity. The resultant structures were examined to determine whether the calculated position of the sulfur atoms suggested that one set of disulfide connectivities was more likely than the other, theoretically possible, sets. In the second approach, a separate family of structures (50 per set) was calculated for each of the 15 possible disulfide-bonded molecules. The resultant families of structures were compared to see whether one was favored over the others. Both approaches led to the same global fold, and the most likely disulfide connectivity is predicted to be 5-22, 13-27, and 17-29. In the calculated structure the cyclic peptide backbone is folded back onto itself and braced with disulfide pairs across diagonally opposed beta-strands. This structure involves one of the disulfide bonds (5-22) threading through the eight amino acid loop formed by the other two disulfide bonds and the peptide fragments connecting them.

Abstract:Coastal water quality is facing increasing threats due to human activities. Their contamination by sewage discharges poses significant risks to the environment and public health. We aimed to investigate the presence of antibiotic-resistant Enterococcus in beach waters. Over a 10-month period, samples were collected from four beaches in the State of São Paulo (Brazil). Enterococcus isolates underwent matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF/MS) and molecular analysis for accurate genus and species identification. The antimicrobial susceptibility for 14 antibiotics was evaluated using the disc diffusion method followed by a multidrug-resistance (MDR) classification. PCR amplification method was used to detect antimicrobial resistance genes (ARGs). Our findings revealed the prevalence of Enterococcus faecalis, E. faecium and E. hirae. Out of 130 isolates, 118 were resistant to multiple antibiotics. The detection of resistance genes provided evidence of the potential transfer of antibiotic resistance within the environment. Our findings underscore the necessity for continuous research and surveillance to enhance understanding of the pathogenicity and antimicrobial resistance mechanisms of Enterococcus, which is crucial to implement effective measures to preserve the integrity of coastal ecosystems.

Abstract:Human population growth, especially in coastal urban cities, increases the potential for fecal pollution of adjacent waterways, requiring continued advances in pollution monitoring and management. Infections remain the largest health risk from contact with fecal- and sewage-polluted waters, and a small number of fecal indicator bacteria (FIB) are used as primary pollution assessment tools. While FIB continue to be useful tools, some of the assumptions about the behavior of FIB in the environment, and the associated pathways for pathogen exposure, have come into question. Research into the extra-enteric ecology of these indicators has identified management-relevant complexities including particle association, prolonged environmental persistence, and multidirectional microbial exchange among water, sediment, and air. These complexities provide opportunities for improving current monitoring and modeling strategies and to better understand exposure pathways for sewage-related infections.

Abstract:Wastewater treatment is one of the major biotechnological processes used to treat municipal and industrial sewage. All the steps involved in the removal of contaminants from wastewaters to treat municipal and industrial sewage represent a reservoir of a dynamic microbial communities with specific key players in the different process types. Aerosolized biological particles, defined as bioaerosols, can be generated during different steps of the wastewater treatment process. The goal of this study is to offer a comprehensive indoor-air microbiota description of numerous wastewater treatments plants using an amplicon-based high-throughput sequencing approach. To fill up the gap of the wastewater-bioaerosol microbial diversity literature, a site-related and seasonal variation of bioaerosol emission in eight indoor wastewater treatments plants is described targeting the 16S rRNA gene for bacterial community analyses. No significant differences were observed between summer and winter in terms of microbial diversity and composition. However, indoor pre-treatment and secondary treatment steps suggest the presence of different bacterial taxa, some of them being pathogens or opportunistic pathogens. Gut-associated flora was most abundant in the air collected during the biodegradation of organic matter of the wastewater treatment step and suggests that fecal contamination can persist in aerosols until the last steps of the process. The results suggest that wastewater workers could be exposed to pathogenic and opportunistic pathogenic microorganisms in aerosols released at every treatment step with a peak during degritting and degreasing. This study offers a comprehensive indoor-air microbiota description of waste water treatment plants—concluding a significant potential occupational risk.

Bioaerosol emissions from wastewater treatment process at urban environment and potential health impacts

Abstract:The inlet of wastewater treatment plants (WWTPs) contains pathogenic microorganisms which during aeration and by mechanical mixing through wind typically aerosolized microbes into ambient air. Bioaerosol emission and its characterization (bacterial and fungal) was investigated considering low-flow and high-flow inlet of wastewater treatment plant. Generation of bioaerosols was found influenced by prevailing seasons while both during summer and winter, fungal concentration (winter: 1406 ± 517; summer: 1743 ± 271 CFU/m3) was higher compared to bacterial concentration (winter: 1077 ± 460; summer: 1415 ± 588 CFU/m3). Bioaerosols produced from WWTPs were predominately in the size range of 2.1-4.7 μm while fraction of fungal bioaerosols were also in ultra-fine range (0.65 μm). Bioaerosols reaching to the air from WWTPs varied seasonally and was calculated by aerosolization ratio. During summer, aerosolization of the bioaerosols was nearly 6 times higher than winter. To constitute potential health effects from the exposure to these bioaerosols, biological characterization, antibiotics resistance and the health survey of the nearby area were also performed. The biological characterization of the bioaerosols samples were done through metagenomic approach using 16s and ITS metagenomic sequencing. Presence of 167 genus of bacteria and 41 genus of fungi has been found. Out of this, bacillus (73%), curtobacterium (21%), pseudomonas, Exiguo bacterium, Acinetobacter bacillaceae, Enterobacteriaceae and Prevotella were the dominant genus (top 10) of bacteria. In case of fungi, xylariales (49%), Hypocreales (19%), Coperinopsis (9%), Alternaria (8%), Fusarium (6%), Biopolaris, Epicoccum, Pleosporaceae, Cladosporium and Nectriaceae were dominant. Antibiotics like, Azithromycin and cefixime were tested on the most dominant bacillus showed resistance on higher concentration of cefixime and lower concentration of azithromycin. Population-based health survey in WWTP nearby areas (50-150 m periphery) found several types of diseases/symptoms including respiratory problem, skin rash/irritation, change in smell and taste, eye irritation within the resident population and workers.

Abstract:Bioaerosols are particles of great importance for several fields of research, and spores produced by fungi can exist as bioaerosols when suspended in the air. Microbiological standards for environmental monitoring of outdoor air parameters can be achieved by analyzing the relationship between airborne microorganisms and the prevailing environmental conditions. The outdoor air of the Metropolitan Region of São Paulo and the rural area in a city of the state of São Paulo (Ibiúna/SP), both in Brazil, were evaluated for the presence of microorganisms using the MAS-100 ECO (Merck®, Fr.) and M Air T (Millipore®) air sample collectors. Dichloran Rose-Bengal Chloramphenicol and Tryptic Soy Agars were used for fungal and bacterial isolation, respectively. Bacterial colonies were counted, and the plates with fungal colonies were sent for phenotypic identification up to genus and species level, respectively. Data on pollutant concentrations were obtained from the Environmental Company of the State of São Paulo. The highest number of Colony-Forming Units/m 3 (CFU/m 3 ) of microorganisms was measured in the winter and summer seasons, respectively, but the greatest Spore-Forming Units (SFU) of fungi were found in the rural area, where pollutant concentrations were lower. Nitrogen dioxide (NO 2 ) had a slightly positive influence on the concentration of SFU of fungi in both areas studied. Sulfur dioxide (SO 2 ) pollutant concentrations had both positive and negative great relations showing influence on microbial counts in the air of the rural area. In the rural area, the low bacteria count was influenced negatively by the low concentration of carbon monoxide (CO). The microbial counts were related to each other, as well as to the concentrations of pollutants, shown by all the correlations seen, indicating microorganisms as biomarkers of pollution in outdoor areas. The influence of environmental factors on the population and outdoor air biome is also explicit.

Abstract:Ballast water represents a significant vector for the transfer of aquatic organisms and chemical pollutants. Although various groups of transported microorganisms can have a negative impact on native species of aquatic fauna, the available literature usually focuses on larger organisms. This is important because microorganisms cause changes in the balanced aquatic environment, including a stable trophic pyramid. The objectives of this study were twofold: (i) to determine the seasonal changes in the microbiota of the ballast water of long- and short-range ships entering the southern Baltic port, with a focus on fish pathogenic microorganisms and (ii) to potentially assess the threat to the ichthyofauna caused by the introduction of these microorganisms into the aquatic environment. The analytical results demonstrated notable variability in microbial density across the samples, contingent on the distance traversed by the ships. The samples of ballast water collected in autumn exhibited the highest microbial density compared to those collected in spring and summer. The samples contained yeast (1.00–2.98 log cfu/mL), mold (1.30–3.26 log cfu/mL), and bacteria (2.18–4.61 log cfu/mL), including amylolytic bacteria (0.95–3.53 log cfu/mL), lipolytic bacteria (0.70–2.93 log cfu/mL), and proteolytic bacteria (0.70–2.39 log cfu/mL). The most prevalent were the Pseudomonas bacteria (0.48–4.40 log cfu/mL), including Pseudomonas fluorescens (0.20–2.60 log cfu/mL. The port waters in spring and summer were primarily characterized by the presence of bacteria belonging to the genus Bacillus. Additionally, the samples exhibited the presence of Intestinimonas, Oceanobacillus, and Virgibacillus bacteria. The short-range vessel samples were populated primarily by bacteria belonging to the genus Bordetella, accompanied by Oligella, Brackiella, and Basilea oraz Derxia, while the ballast water of long-range ships contained mainly Acholeplasma and Clostridium, accompanied by Bacillus, Peptosteptococcus, Intestinibacter, Terrisporobacter, Anaerobacillis, Anaerofustis, Oxobacter, and Listeria. A phylogenetic analysis of the bacteria recorded in the ballast water revealed the presence of species, including Bordetella and Acholeplasma, which can facilitate the colonization of aquatic organisms by pathogenic entities. The results of this study showed that despite the use of water treatment systems on ships, ballast waters carry microorganisms that can negatively impact new environments, including local fish populations (e.g., P. fluorescens). These observations point to the need for further research on the effectiveness of ballast water management systems used to date to minimize the environmental impact of organisms carried in ships' ballast water to preserve natural resources and environmental sustainability in port waters.

Abstract:The coastal atmosphere adjacent to large urban areas can be strongly affected by the emission of air pollutants, among them, major ions species. In this study, the chemical composition and sources of carboxylates and other water-soluble ions in fine and coarse aerosols as well as estimates of particle dry deposition fluxes were studied at a tropical coastal site affected by an urban environment. The mean concentrations of the total carboxylates were 78 ng m–3 in fine fraction and 81 ng m–3 in coarse fraction of particulate matter (PM). The corresponding values for the total inorganic ions were 2143 ng m–3 and 4880 ng m–3 respectively. Main sources for fine particles were: (i) photochemical formation of carboxylic acids in vapor phase and a posterior gas-to-particle conversion onto sea salt particles; (ii) emissions from anthropic sources with long range transportation processes; and (iii) the interchanging of volatile species among atmospheric phases. In turn, for coarse particles, the predominant sources were: (i) gas-phase species and ab/adsorbed onto pre-existing particles afterwards; (ii) primary emission of coarse particles from anthropogenic sources; and (iii) sea salt spray and/or soil resuspension. Finally, particle dry deposition was a very important mechanism representing air-to-sea fluxes of major species.
Assessment of anthropogenic pollution in Guanabara Bay (SE Brazil) through biogeochemical data and stable isotope mixing models

Abstract:This work intends to identify pollution sources along the margins of Guanabara Bay (GB; SE Brazil) through a multiproxy approach and Bayesian stable isotopic mixture model (BSIMM). For this purpose, 33 surface sediment samples were collected and analyzed for granulometry, geochemistry (heavy metals, total organic carbon-TOC, stable isotopes of carbon and nitrogen-δ13C and δ15N, Rock-Eval pyrolysis parameters-REPP), and physicochemical parameters. Metal concentrations (E) dissolved in water (EW), adsorbed by organic matter (EOM) and by Mn hydroxides (EMn), and total extracted concentrations (ET) were analyzed. Sampling was conducted in 2018 after an oil spill from Reduc Oil Refinery. Potential Ecological risk index (PERI), based on metals, classified 85% of the analyzed stations as having moderate to considerable ecological risk. The metals with the potential to cause the highest ecological risk were CdW, CdOM, PbOM, and HgOM. The combination of BSIMM and REPP data was an effective proxy for oil spill detection by indicating the presence of polycyclic aromatic hydrocarbons (PAHs). Relatively high TOC contents suggested that the analyzed stations are eutrophicated environments. BSIMM discriminated three groups of stations with different sources of organic matter (OM), endorsing the result previously shown by the cluster analysis: (A) Niterói region, Botafogo marina, Glória marina, Fiscal and Fundão islands with diffuse sources of OM, including marine phytoplankton and material of continental origin from highly polluted rivers and domestic sewage; (B) region near Fundão and Governador islands and Mangue Channel outlet with OM (≃70%) supplied by highly polluted streams and a small contribution of PAHs; (C) Duque de Caxias and Botafogo-Urca inlet with significant contributions of PAHs, materials from C-3 plants and rivers polluted by urban sewage. Results of linear regressions in conjunction with BSIMM indicate that HgMn and PbOM mainly affect Group A's stations. Although the eastern margin of GB (Niterói; Group A) showed greater oceanic interaction than the other groups, it presented substantial concentrations of metals, potentially harmful (i.e., Hg and Pb) to marine biota and human health.

Abstract:The hospital sewage contains many pathogenic bacteria, which may produce bioaerosol and spread diseases during the sewage treatment process. In this study, the characteristics, seasonal variation, and risk assessment of the bioaerosol in hospital sewage treatment stations (HSTS) were analyzed. The concentration of bacteria in bioaerosol generated from BRT (251 CFU/m 3 ) was higher than that in the air at SG (75 CFU/m 3 ). Summer (506 CFU/m 3 ) and autumn (32 CFU/m 3 ) showed the highest levels of bacterial and intestinal bacterial concentration in the air at BRT, respectively. The percentage of microorganisms attached to small particles (<2.1 μm) (SP) ranged from 22.64% to 66.66%, which was most prevalent in winter (66.66%). Brevundimonas (38.99%), Bacteroides (11.25%), Pseudomonas (49.48%), and Ochrobactrum (41.02%) were the dominant genera in spring, summer, autumn, and winter, respectively. The dominant bacterial genera of bioaerosol in BRT and SG showed obvious seasonal variations, Bacillus were present in each season. The hazard quotient (HQ) of males was higher than that of females (HQ males : 2.60 × 10 −3 , HQ females : 2.18 × 10 −3 ), and the inhalation risks of the bacteria were highest in summer (5.98 × 10 −2 ). Temperature and relative humidity positively influenced intestinal bacteria in bioaerosols. The potentially pathogenic were highest in spring. One hour after the sampling point, the result of the Hybrid Single-Particle Lagrangian Integrated Trajectory (HYSPLIT) mode showed that the diffusion area of bioaerosol in winter (182.03 km 2 ) was the largest. This study focused on the bioaerosol produced by hospital sewage. The results effectively revealed the seasonal variation and risk characteristics of bioaerosols, which could serve as a theoretical foundation for the future control and reduction of bioaerosols in the process of purifying municipal water.

Reviewing the effects of contamination on the biota of Brazilian coastal ecosystems: Scientific challenges for a developing country in a changing world

Abstract:Pollution is a major worldwide problem that is increasing with urban growth, mainly along coastal areas. Pollution is often worse, governance is poorer and managerial strategies to improve environmental quality are less advanced in developing than developed countries. Here, we present an overview of the current scientific knowledge of the impacts of contamination on the biota of coastal ecosystems of Brazil and evaluate the scientific challenges to provide baseline information for local managerial purposes. We compiled data from 323 peer-reviewed published papers from the extensive Brazilian coast. We critically evaluated the produced knowledge (target contaminants, sources, ecosystems, taxa, response variables) and the science behind it (rigour and setting) within its socioenvironmental context (land occupation, use of the coast, sanitation status, contamination history). Research was driven largely by environmental outcomes of industrial development with a focus on the single effects of metals on the biota. The current knowledge derives mainly from laboratory manipulative experiments or from correlative field studies of changes in the biota with varying levels of contamination. Of these, 70% had problems in their experimental design. Environmental impacts have mainly been assessed using standard indicators of populations, mostly in ecotoxicological studies. Benthic assemblages have mostly been studied using structural indicators in field studies. Future assessments of impacts should expand research to more taxonomic groups and ecosystem compartments, adding combined functional and structural responses. Furthermore, further investigations need to consider the interactive effects of contaminants and other environmental stressors. By doing so, researchers would deliver more robust and effective results to solve problems of pollution.

Abstract:Objective We assessed the influence of changes in steroid hormones across the menstrual cycle on the spinal nociceptive reflex. Method We studied in 14 healthy women during the follicular and luteal phase the nociceptive flexion reflex (RIII reflex), an objective neurophysiological method that allows exploring possible abnormal functioning of the pain-control system. The basal body temperature (BBT) was used to evaluate the different phases of the ovarian cycle. The menstrual distress questionnaire (MDQ) was also applied for monitoring somatic and psychological symptoms during the cycle. Results During the luteal phase, the threshold of the RIII reflex (Tr) and the psychophysical threshold for pain (Tp) were both significantly reduced compared with the follicular phase. Moreover, the reflex threshold in the luteal phase was negatively correlated to the total MDQ score of the recording day. Conclusions A higher sensitivity to pain stimuli was observed during the luteal phase of the menstrual cycle, which probably results from a reduction in the inhibitory descending control on spinal nociceptive flexion reflex. Complex neuromodulatory interactions of ovarian steroids with other systems of neurotransmission (especially serotonergic) may account for these observations.

Abstract:Environmental contamination by plastics poses a significant threat to both fauna and flora, manifesting in lethal and sub-lethal effects. Plastics can enter coastal and marine environments through wind and rain, with microplastics (< 5 mm; MPs) arising from the degradation of larger plastics or being manufactured for commercial use. Despite Brazil's extensive coastline, data on microplastic contamination is scarce. This study aimed to assess microplastic pollution on a beach in Rio Grande do Sul, Brazil. Samples were collected from 30 points along the high tide line, with beach sand processed through density separation and filtration. Microplastics were quantified using Nile Red stain under fluorescence microscopy and chemically identified via the μRaman technique. Results showed an average concentration of 650 MPs/kg of sediment, with higher concentrations near the Mampituba River, indicating a greater transport of plastic contaminants by the river to the coastline. The smallest particles (50–100 μm) were most abundant, and the predominant types of plastics identified were polyethylene (PE), polypropylene (PP), and polyamide (PA). This study provides the first quantification of microplastic pollution in this region, indicating that the concentration and types of microplastics are consistent with findings elsewhere in Brazil and globally. These results highlight the widespread nature of microplastic pollution and underscore the need for coordinated environmental remediation efforts.

Abstract:Marine heatwaves (MHWs) are a major concern worldwide due to their increasing impacts in recent years, and these extreme events may trigger deoxygenation of coastal waters affected by sewage and eutrophication. Here we investigate the combined effects of MHWs and nutrient enrichment on the water quality and biodiversity of the Bay of Santa Catarina Island (Brazil). We used historical (1994–2020) sea surface temperature data from satellites and in situ physical, chemical and biological parameters to assess temporal trends. Oxygen levels have been decreasing whilst phosphorus levels have been increasing in the bay. During the austral summer of 2020 a regional sea surface heatwave was detected by satellite, lasting for 9 days and coinciding with our research cruise. During this period, seawater temperatures reached 29.8°C and anoxia was detected for the first time in the bay. A decrease in macrobenthic and phytoplankton community richness correlated with decreases in oxygen both through time and towards more urbanized areas. Overall, poor wastewater treatment is a key stressor that combined with MHWs to degrade coastal waters. Mitigation strategies are needed to minimize the impact of MHWs, including improved sewage treatment, restoration and conservation of wetlands and the use of nature-based technologies to promote coastal ecosystem recovery.

Abstract:Guanabara Bay is the second largest bay in the coast of Brazil, with an area of 384 km(2). In its surroundings live circa 16 million inhabitants, out of which 6 million live in Rio de Janeiro city, one of the largest cities of the country, and the host of the 2016 Olympic Games. Anthropogenic interference in Guanabara Bay area started early in the XVI century, but environmental impacts escalated from 1930, when this region underwent an industrialization process. Herein we present an overview of the current environmental and sanitary conditions of Guanabara Bay, a consequence of all these decades of impacts. We will focus on microbial communities, how they may affect higher trophic levels of the aquatic community and also human health. The anthropogenic impacts in the bay are flagged by heavy eutrophication and by the emergence of pathogenic microorganisms that are either carried by domestic and/or hospital waste (e.g., virus, KPC-producing bacteria, and fecal coliforms), or that proliferate in such conditions (e.g., vibrios). Antibiotic resistance genes are commonly found in metagenomes of Guanabara Bay planktonic microorganisms. Furthermore, eutrophication results in recurrent algal blooms, with signs of a shift toward flagellated, mixotrophic groups, including several potentially harmful species. A recent large-scale fish kill episode, and a long trend decrease in fish stocks also reflects the bay's degraded water quality. Although pollution of Guanabara Bay is not a recent problem, the hosting of the 2016 Olympic Games propelled the government to launch a series of plans to restore the bay's water quality. If all plans are fully implemented, the restoration of Guanabara Bay and its shores may be one of the best legacies of the Olympic Games in Rio de Janeiro.

Abstract:This study made use of four asynchronously coupled numerical simulations (four Eulerian and four Lagrangian) to investigate an event of a sudden arrival of more than 50 tons of marine litter on the northeastern Brazilian coast. This material mysteriously landed on several beaches, compromising water quality and impacting tourism, therefore raising serious concerns about its origin from local authorities. A total of 87,030 virtual particles were released and had their trajectories investigated, as well as their relationship with major physical forcings, including tides, winds and the North Brazil Under Current (NBUC). The virtual particles followed predominantly towards the north/northwest, mainly due to the presence of the NBUC and southeast trade winds. This pattern indicated that the flow of marine surface litter followed the continental shelf northwards, and the role of winds was key in providing the conditions for oceanic originated material to being deposit along the coastline. This study provided important insights about the regional circulation and main forcings that act in the transport and deposition of floating materials that reach the shallow shelf of the Brazilian northeast. Further investigation on the variability of these forcings along the seasonal cycle seems to be extremely important and should be undertaken soon.

Abstract:Monitoring microbial water quality is essential for recreational beaches in order to protect human health. To evaluate the relative importance and impacts of various types of non-point microbial sources at a subtropical beach (Hobie Beach, Miami, USA), we utilized a coastal ocean circulation model (Delft3D) with a microbe transport-fate model. Those non-point sources include beach sediment, dog feces, bather shedding, and rainfall runoff. The hydrodynamic model results agreed well with tidal elevations recorded by a nearby NOAA tidal station and also field data collected by pressure sensors, acoustic Doppler current profiler (ADCP). We modeled enterococci levels from four different types of non-point sources on the beach and Staphylococcus aureus levels from only the bather shedding. Model results suggest that dog feces are spotty sources of enterococci and can result in transient spikes of enterococci levels for hours. Beach sands are pervasive sources of enterococci and may explain observed persistent elevations of enterococci levels at this site. Runoff may also significantly increase enterococci levels during rainfall events while bather shedding contribution of enterococci is almost negligible. Bather is the only Staphylococcus aureus source considered in the study and simulated levels are in the same order as prior field measurements.
