Abstract:The elementary and phase analysis of discarded copper ores from Yongping of China has been performed. The experiments of extracting copper from the discarded copper ores were done with the mixed bacteria obtained through a series of enrichment, separation, domestication and combination tests. The results show that in the process of bioleaching, the pH value rises at first and drops gradually. The E-h value keeps rising along with the time and the appropriate E-h value varying between 750 and 800 mV will benefit the bioleaching copper. The high concentration of ferric ions is detrimental to the bioleaching copper. The results of bioleaching copper are good. That is, the copper recovery is 31.8% after 27 days.

Abstract:The column bioleaching of copper flotation tailings was comparatively investigated using layered heap construction method (LM), agglomerate heap construction method (AM), and pellets-sintering heap construction method (PM). The bacterial communities of free, attached, weakly-attached, and strongly-attached microbes in the later bioleaching stage were investigated. In AM group, the addition of lump sulphide ore resulted in the low leachate pH, high ferric iron concentration, and rapid microbial adsorption, which obtained the maximum copper extraction (60.1%) compared with LM (54.6%) and PM (43.9%) groups. The relative abundance of dominant genera and microbial communities of different microbiota underwent changes in three heap construction methods. The alpha-diversity indexes of attached, weakly-attached, and strongly-attached microbes were different, while no significant change was observed in free bacteria. The variation of whole bacterial community was significantly associated with solution pH, total iron, and ferric iron concentrations. Pearson correlation analysis and partial least square path model both indicated that attached bacteria made larger contribution to the copper extraction of tailings.

Abstract:The effect of process parameters (pH, pulp density, inoculum volume, ferrous content, particle size) on the rate of copper solubilization was analyzed respectively through bioleaching tests with mixed bacteria in shake flasks. These results show that the optimal process parameters are: pH, 1.50–1.80; pulp density, 5wt%; inoculum volume, 10vol%; ferrous content, 2g-L−1; particle size, <0.076 mm. Furthermore, when the pH value is below 1.50 in leaching solution, the iron precipitated can be reduced to a great extent, but it shows low bacterial activity at this pH. Finally, it is emphasized that the optimal parameters are also determined by economical benefit.

Abstract:Against the backdrop of the increasing copper demand in a low-carbon economy, this work statistically forecasted the distribution of China's copper tailings for the first time, and then characterized them as finely crushed and low-grade mining solid wastes containing copper mainly in the form of chalcopyrite, bornite, covelline, enargite and chalcocite based on available research data. China is the globally leading refined copper producer and consumer, where the typical commercial-scale bioleaching of copper tailings is conducted in the Dexing, Zijinshan and Jinchuan mining regions. And these leaching processes were compared in this study. Widely used chemolithoautotrophic and mesophilic bacteria are Acidithiobacillus, Leptospirillum, Acidiphilium, Alicyclobacillus and Thiobacillus with varied metal resistance. They can be used to treat copper sulfide tailings such as pyrite, chalcopyrite, enargite, chalcocite, bornite and covellite under sufficient dissolved oxygen from 1.5 to 4.1 mg/L and pH values ranging from 0.5 to 7.2. Moderate thermophiles (Acidithiobacillus caldus, Acidimicrobium, Acidiplasma, Ferroplasma and Sulfobacillus) and extreme thermophilic archaea (Acidianus, Metallosphaera, Sulfurococcus and Sulfolobus) are dominant in leaching systems with operating temperatures higher than 40 °C. However, these species are vulnerable to high pulp density and heavy metals. Heterotrophic Acidiphilium multivorum, Ferrimicrobium, Thermoplasma and fungi use organic carbon as energy to treat copper oxides (malachite, chrysocolla and azurite) and weathered sulfides (bornite, chalcocite, digenite and covellite) under a wide pH range and high pulp density. We also compared autotrophs in a planktonic state or biofilm to treat different metal sulfides using various sulfur-cycling enzymes involved in the polysulfide or thiosulfate pathways against fungi that produce various organic acids to chelate copper from oxides. Finally, we recommended a bioinformatic analysis of functional genes involved in Fe/S oxidization and C/N metabolism, as well as advanced representation that can create new possibilities for the development of high-efficiency leaching microorganisms and insight into the mechanisms of bioleaching desired metals from complex and low-grade copper tailings.

Abstract:Nutrition(TN,TP,TK and organic materials) loss is one of problems in bioleaching process of heavy metals from sewage sludge,because of the acid environment.The removal of Zn and Cu from sludge from the Sibao wastewater treatment plant in Hangzhou and losses of nutrition after bioleaching were studied to supply reference to application of the bioleaching for heavy metal control.Two 10%(V/V) different inoculants,iron-oxidizing bacteria Thiobacillus ferrooxidans(T.fe) and sulfur-oxidizing bacteria Thiobacillus thiooxidans(T.th.),were used in the batch experiment.The results demonstrated that T.fe could faster start bioleaching.The removal rate of Zn reached 97.8% after 2 d and kept the high removal rate until the end of the experiment.The efficiency of Cu removing reached 37% at the 4th day and increased persistently.Both of bioleaching rates of Zn and Cu by T.th had high level after 6 d.The bioleaching of Zn and Cu in sludge were affected by pH.For Zn,high removal rates occurred at pH 3.0~4.0,while better Cu bioleaching rate occurred at pH<3.0.When a large number of heavy metals were removed from sludge,the losses of nutrition(TN,TP,TK and organic materials) concurred.Especially,the loss of TP reached 70%.The losses of nutrition in the system with T.fe were more than these in the system with T.th.

Abstract:Low-temperature bioleaching is relevant to the recovery of metals in alpine mines, but its development has been constrained by low bioleaching rates at high pulp concentrations. To this end, the bioleaching effect of the microbial community after the domestication of pulp concentration at 6 °C was studied. Domestication improved the bioleaching rate of copper. Environmental scanning electron microscopy (ESEM), X-ray diffraction (XRD), and electrochemical measurements revealed that the domestication process aggravated the corrosion of the chalcopyrite surface by accelerating its dissolution reaction. High-throughput sequencing technology indicated that Acidithiobacillus spp., Leptospirillum spp., and Acidiphilium spp. were the major lineages of the domesticated microbial community. The analysis of the microbial community revealed that domestication changed the microbial structure, enhancing the adaptability of the microbial community to pulp concentrations and acidic conditions. This study uncovered the mechanism by which domestication enhanced the bioleaching efficiency of the microbial community at low temperatures.

Abstract:Over the past several decades, extensive research has focused on improving bioleaching of different ores, and increasing research on applying bioleaching for the processing of tailings produced from mining operations around the world. As part of the research project Near-zero-waste recycling of low-grade sulfide mining waste for critical-metal, mineral and construction raw-material production in a circular economy (NEMO), we have studied the effect of nutrient availability, the effect of high concentration of dissolved salts (Al and Mg sulfates) on metal (Cu, Co, Mn, Ni, Zn) extraction rates by bioleaching in laboratory column tests at high temperature (48 °C and 60 °C), using secondary ore from the Sotkamo mine (Terrafame operation, Finland). Metal dissolution rates showed two stages: a rapid first stage, where acid addition and initial irrigation released acid-soluble metal species, and a second stage, with slower rates of metal dissolution by microbial oxidation of residual metal sulfides with Fe(III) as the oxidant. Experiments showed that adequate Fe(II) oxidation rates depended on the addition of salts but not so much as the addition of nutrients (P as phosphate, N as ammonium and K). High dissolved salt concentrations resulted in low redox potentials (745–760 mV vs standard hydrogen electrode), limiting Co dissolution rates. For example, Co extraction yield at 48 °C when added salts plus nutrients was 57 %, and without salts (with or without nutrients) ∼75 %. Other metal dissolution rates were not affected by the composition of the irrigation solution or temperature, e.g. Ni extraction rates were between 88 % and 92 % in all the experiments. This was mostly due to the high metal content as readily soluble phases formed during the first heap leaching as metal dissolution in mine waste (e.g. tailings) highly depends on the metal speciation. Recycling of the leaching solution for irrigation, a common practice in heap bioleaching, affects negatively metal extraction rates, the precise reason for this is still to be determined in further experiments, possible effects including changes in solution chemistry affecting mineral oxidation, precipitation of insoluble salts, or toxicity or inhibition of microbial growth rates limiting microbial activity and mineral leaching.

Abstract:The recovery of precious metals from solid waste through bioleaching has become a research hotspot in recent years. Thus, in this study, different strategies, such as chemical sulfuric acid leaching and mixed consortium bioleaching, were adopted to extract copper from Copper-Containing Electroplating Sludge. The results showed that, compared to chemical leaching, bioleaching showed a much better performance. Indeed, copper bioleaching efficiency reached 94.3% on day 7 (21.1% higher than that of chemical leaching). The results also indicated that the process of bioleaching involved more mechanisms and reactions than that of chemical leaching. The SEM and EDX tests showed that the surface morphology of the sludge changed significantly after bioleaching, and that an insignificant amount of copper remained in the leached residues. Furthermore, the leached residues passed the characteristic leaching toxic test and thus can be considered as non-hazardous raw materials for the construction industry. Hence, adopting a mixed consortium leaching process to extract copper from Copper-Containing Electroplating Sludge will not only significantly reduce environmental pollution, but will also use metal resources more efficiently.
Continuous Bioleaching of Arsenic-Containing Copper-Zinc Concentrate and Shift of Microbial Population under Various Conditions

Abstract:The goal of this work was to study the bioleaching of arsenic-containing polymetallic concentrate that contained 6.2% Cu, 7.3% Zn and 1.7% As, depending on different temperatures and in the presence of CO2 and molasses in the medium, as well as the difference in the composition of microbial population formed under various conditions. A mixed population of moderately thermophilic and thermotolerant acidophilic microorganisms formed during the continuous bioleaching of copper concentrate was used as an inoculum. The experiments were carried out in a continuous mode in laboratory scale reactors, with a temperature range of 40 °C to 60 °C. To assess the effect of CO2 and molasses on metal leaching and microbial population composition, the experiments were carried out in three reactors: CO2 (~0.01 L/min) was supplied into the first reactor; 0.02% molasses were added to the pulp of the second reactor; and no additional carbon sources were supplied into the control reactor. The highest copper recovery (27%) was achieved at 50°C in the experiment with molasses, while the highest zinc recovery (82.1%) was reached at 45°C in the control experiment. Additional carbon sources affected the extraction of non-ferrous metals only at 60 °C and increased the extraction of copper and zinc by 12.6% and 24.2%, respectively. Both the temperature and carbon source used affected the microbial population composition. The main microbial genera revealed in the populations by next generation sequencing (NGS) were bacteria of the genera Sulfobacillus and Acidithiobacillus, as well as archaea of the genera Ferroplasma, Acidiplasma, and Cuniculiplasma. At low temperatures (40 and 45 °C), Acidithiobacillus, Sulfobacillus, and Ferroplasma predominated, while at temperatures 50–55 °C, the decrease in relative abundance of these genera occurred, and the predominance of Acidiplasma archaea was observed. The usage of both CO2 and molasses led to the increase in Sulfobacillus and Acidiplasma in relative abundance.

Abstract:Bioleaching is a promising technology to remediate sediments contaminated by heavy metals. However, the complex heterogeneities of the sediments can reduce the acidification efficiency and the heavy metal removal rate, thus hindering the practical application of sediment bioleaching. This experiment conducted comparative bioleaching experiments between the inoculated group (average leaching percentages: Cu 67.64%; Zn 54.44%; Ni 29.59%) and the non-inoculated control group (Cu 37.10%; Zn 41.04%; Ni 19.89%) on 28 sediments characterized by different physicochemical properties to explore the key factors influencing bioleaching. The results indicated that the bioleaching process was predominated by the indigenous bioleaching bacteria and the bioleaching inoculum, respectively. The ACCpH=4 (acid-consuming capacity), TOC (total organic carbon), and TN (total nitrogen) of the sediments played an essential role in influencing the microbial community structure and bioleaching performance: the ACCpH=4, as the inhibitive factor, could influence the succession growth of the indigenous bioleaching bacteria and the inoculum during the bioleaching process, while the TOC and TN, as the contributing factor, could influence the metabolism of the indigenous bioleaching bacteria. Based on these results, the bioleaching process was improved with the classification and pretreatments of sediment to realize successful bioleaching of all types of the sediments examined in this research.

Abstract:The distribution of the metal-bearing mineral grains within a particulate ore prepared for leaching, and the impact of this spatial heterogeneity on overall extraction efficiency is of key importance to a mining industry that must continuously target ever-reducing grades and more complex ore bodies. If accessibility and recovery of the target minerals is to be improved, a more detailed understanding of the behaviour of the system must be developed. We present an in situ analysis using X-ray computed tomography to quantify the rates of volume reduction of sulfide mineral grains in low grade agglomerated copper bearing ores during a miniature laboratory scale column leaching experiment. The data shows the scale of the heterogeneity in the leaching behaviour, with an overall reduction of sulphide mineral grains of 50%, but that this value masks significant mm(3) to cm(3) scale variability in reduction. On the scale of individual ore fragments, leaching efficiency ranged from 22% to 99%. We use novel quantitative methods to determine the volume fraction of the sulfide that is accessible to the leachate solution.

Abstract:The aim of this investigation was to determine the efficacy of chalcopyrite bioleaching using pure cultures of Acidithiobacillus ferrooxidans or Acidithiobacillus thiooxidans and a mixed culture composed of Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans. The experiments were carried out in shake flasks with [Fe2+] 4gL−1 and S 1gL−1 at pH 1.80, 130rpm and 30°C. Inocula of both iron-oxidizing bacteria were obtained from cultures with Fe2+ as the sole energy source, whereas Acidithiobacillus thiooxidans inoculum was taken from a culture growing on sulphur. Tests showed that the copper extraction in a mixed culture of Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans was higher than in pure cultures. On the other hand, an important potential of Acidithiobacillus thiooxidans to leaching chalcopyrite was indicated. It can prevent jarosites accumulation on the substrate and allow further copper solubilization through the action of ferric ion. The selection of the suitable pH during the bioleaching processes would be important.

Abstract:Study on the bioleaching of heavy metals in lead⁃zinc mine tailings using Acidithiobacillus ferrooxidans and the transformation in the speciation of heavy metals during the bioleaching is helpful to understand the bioleaching process. In this study, recovery of heavy metals from lead-zinc mine tailings in different solid concentration by bioleaching was investigated. The results show that the bioleaching strongly affected pH and the recovery efficiencies of heavy metals. With increasing solid concentration in the bioleaching, the pH decrease and the leaching rate of heavy metals increased first and then decreased. About 85.45% of Fe and 97.85% of Zn were recovered from mine tailings under the optimum condition with 50 g·L-1 of solid. The analysis results indicate that the bioleaching had a significant impact on the speciation of heavy metals. In the bioleaching process, the transformation in the speciation of heavy metals was a gradual process. The mobile fractions of the heavy metals were first bioleached, followed by stable fractions.

Abstract:Low-temperature biohydrometallurgy is implicated in metal recovery in alpine mining areas, but bioleaching using microbial consortia at temperatures <10°C was scarcely discussed. To this end, a mixed culture was used for chalcopyrite bioleaching at 6°C. The mixed culture resulted in a higher copper leaching rate than the pure culture of Acidithiobacillus ferrivorans strain YL15. High-throughput sequencing technology showed that Acidithiobacillus spp. and Sulfobacillus spp. were the mixed culture’s major lineages. Cyclic voltammograms, potentiodynamic polarization and electrochemical impedance spectroscopy unveiled that the mixed culture enhanced the dissolution reactions, decreased the corrosion potential and increased the corrosion current, and lowered the charge transfer resistance and passivation layer impedance of the chalcopyrite electrode compared with the pure culture. This study revealed the mechanisms via which the mixed culture promoted the chalcopyrite bioleaching.

Abstract:In this study, the contact effects between mesophile bacteria and minerals were observed to enhance the bioleaching kinetics, which demonstrated that the adsorption of bacteria to mineral surfaces could improve the dissolution of arsenic and copper. The microstructure results also demonstrated that after the contact bioleaching, residue surfaces were rougher and with more corrosion bars and pits. Compared to contact oxidation approach, ferric ions, acting as the only oxidants in the non-contact bioleaching process, play a more important role in the copper extraction and arsenic dissolution, which is consistent with the previous works about indirect leaching mechanism. Due to the presence of various mesophiles in the contact oxidation, their activities and corresponding preferences for energy sources influenced by temperature were observed, causing differences in the sulfides' dissolution: ferrous ions were inclined to be oxidized biologically at 30 °C, whereas sulfide minerals, including enargite, were preferentially oxidized at 45 °C.

Abstract:This study aims to realize copper recovery from tailings and make further application of bioleached tailings residues as microbial-cemented tailings backfilling materials (MCTB). Results show that copper contained in tailings can be recovered by bioleaching. Behaviors of microbial-cemented tailings backfilling materials made from bioleached tailings residues are evaluated. Better performances with high compressive strength, small water-holding capacity and shrinkage ratio are obtained through using bioleached tailings residues. A mercury injection test is conducted, which reveals the effect of bioleached tailings residues on microstructure evolution in microbial-cemented tailings backfilling materials. Small pore volume along with uniform pore distribution and small specific surface area is obtained after using bioleached tailings. Simultaneously, concentrations of hazardous ions including Zn 2+ , Pb 2+ , As 3+ , Cd 2+ and Hg 2+ , can be reduced during spraying experiment by using bioleached tailings residues. Effect of bacterial species, especially Acidithiobacillus ferrooxidans , Leptospirillum ferriphilum , Acidibacillus ferrooxidans and Escherichia coli , on reducing hazardous ions concentration is analyzed. Possible reasons suggesting the role of bioleached tailings residues contributing to low concentration of hazardous ions and good performances of microbial-cemented tailings backfilling materials are put forward.

Abstract:To overcome the problems of long cycle and low efficiency of bioleaching the lead⁃zinc sulfide tailings, this study applied Leptospirillum ferriphilum and Acidithiobacillus caldus in equal proportion to constructed a moderate thermophilic consortia to leach lead⁃zinc sulfide. The influence of introducing a facultative autotrophic iron / sulfur⁃oxidizer Sulfobacillus benefaciens on the process of bioleaching tailings was investigated. The results showed that Zn leaching rate of by L.ferriphilum and A.caldus consortia reached 93.94% only in 8 days. With the introduction of S.benefaciens, the leaching system maintained a higher redox potential and microbial activity, and the ferric iron and total iron concentration increased during the leaching process. The relative leaching rate of Zn increased by 13.63% within 4 days, and the Zn leaching rate reached 96.08% within 8 days. Therefore, the addition of S.benefaciens enhanced the leaching efficiency of the moderate thermophilic consortia. Correlation analysis indicated that polysaccharides, protein, and dissolved organic carbon (DOC) showed a significant correlation with the changes in solution media parameters during the leaching process. Moreover, the result of surface functional groups and extracellular polymeric substances (EPS) confirmed that the introduction of S.benefaciens in the leaching process promoted the consortia to produce more proteins and carbohydrates, which could improve the leaching efficiency of moderate thermophilic bacteria, accelerated the leaching of valuable metals in lead⁃zinc sulfide tailings, and reduced its environmental toxicity.

Abstract:To determine the effect of organics (yeast extract) on microbial community during chalcopyrite bioleaching at different temperature, real-time polymerase chain reaction (PCR) was employed to analyze community dynamics of major bacteria applied in bioleaching. The results showed that yeast extract exerted great impact on microbial community, and therefore influencing bioleaching rate. To be specific, yeast extract was adverse to this bioleaching process at 30°C due to decreased proportion of important chemolithotrophs such as Acidithiobacillus ferrooxidans and Acidithiobacillus thiooxidans. However, yeast extract could promote bioleaching rate at 40°C on account of the increased number and enhanced work of Ferroplasma thermophilum, a kind of facultative bacteria. Similarly, bioleaching rate was enhanced under the effect of yeast extract at 50°C owing to the work of Acidianus brierleyi. At 60°C, bioleaching rate was close to 100% and temperature was the dominant factor determining bioleaching rate. Interestingly, the existence of yeast extract greatly enhanced the relative competitiveness of Ferroplasma thermophilum in this complex bioleaching microbial community.

Abstract:This study investigated the effect of designed microbial consortia on biomining of low grade base-metal sulfide tailings. The results show the amount of recycled metals were equal if the tailings were leached by mixed cultures of Leptospirillum ferriphilum and Acidithiobacillus thiooxidans at three different ratios or by pure culture of L. ferriphilum, which was better than the pure culture of Acidithiobacillus ferrooxidans. qPCR (quantitative polymerase chain reaction) demonstrated only L. ferriphilum functioned in the mixtures at initial stage. The results of extracellular polymeric substances (EPS) via three-dimensional excitation-emission matrix combined with parallel factor analysis (3DEEM-PARAFAC) collected from mixed or pure cultures indicated there were no interactions between two strains. Secondary minerals were formed, but did not influence the leaching process. A new strategy for tailings biomining was proposed: only ferrous oxidizers should be added during the initial and middle biomining stage, while sulfur oxidizers should be added at the end.

Abstract:In this work, bioleaching of chalcopyrite-pyrite and chalcopyrite-bornite mixed ores in the presence of moderately thermophilic microorganisms (A.caldus, L.ferriphilum and mixed culture) were carried out. Bioleaching results showed that bioleaching behaviors of chalcopyrite-pyrite and chalcopyrite-bornite mixed ores in the presence of these moderately thermophilic microorganisms were all in accordance with the proposed optimum redox potential theory. Results showed that high copper extraction was obtained in different bioleaching systems if the redox potential was between EL to EH. Real-time PCR technique was used to analyze the change of percentages of A.caldus and L.ferriphilum during bioleaching by mixed culture consisting of A.caldus and L.ferriphilum. Results showed that the change rules were similar between bioleaching system of sole chalcopyrite and two kinds mixed ores, indicating that the chemical factor instead of biological factor should be the main cause for the high copper extraction of chalcopyrite-pyrite and chalcopyrite-bornite mixed ores.
