Abstract:Objective To establish a GC-MS method for the detection method of sedative-hypnotic drugs illegally added in health foods and traditional Chinese medicines.Methods The solvent ultrasonic extraction or the liquid-liquid extraction was adopted to extract and enrich the samples.The qualitative analysis was carried out by GC-MS and the Scan mode.The analytical capillary column was DP-5 ms(30 m×0.25 mm,0.25 μm).The initial temperature was set at 120 ℃(1 min),then was risen to 230 ℃with 10 ℃/min,to 270 ℃(20 min) by 15 ℃/min by temperature programming.The EI ion source was selected and the solvent was delayed by 2.5 min.The MS scanning was set at the range of 40-350 amu.Results Under the selected chromatographic conditions,14 kinds of sedative-hypnotic drugs could get a better separation anf the detection was rapidly completed within 30 min.Conclusion This method is rapid,simple,accurate and suitable for the fast qualitative detection in crackdown on counterfeit goods.

Abstract:A method of high performance liquid chromatography-mass spectrometry (HPLC-MS) for the simultaneous determination of 8 illegal synthetic drugs in antihypertensive dietary supplements was developed. The chromatography was performed using a Spherigel C18 column, with the mobile phase of 0.005 mol/L ammonium formate buffer (pH 3.0)-acetonitrile-methanol and cialis as internal standard. In ESI mode, the lowest concentration limit of detection (LOD) was 2.5 microg/L and the lowest concentration limit of quantification (LOQ) was 8.3 microg/L, while in ESI- mode, the lowest concentration limit of detection (LOD) was 50 microg/L. The average recoveries were between 63.3% - 107.4%. The clonidine, dihydrochlorothiazide, triamterene, metoprolol, phentolamine, chlorthalidone, furosemide and valsartan in the samples were separated well under the optimized conditions. The method was well validated by systematical searching for satisfactory conditions using the experimental designs including precision, repeatability and spiked recovery.

Abstract:An analytical method has been developed and validated for the simultaneous determination of 18 pharmaceuticals and personal care products (PPCPs), including antibiotics (trimethoprim, erythromycin x 2H2O, norfloxacin, ofloxacin, pencilline G, penicillin V potassium salt, cephalexin and sulfamethoxazole), beta-bloker (atenolol), anophelifuge (N, N-diethyl-3-methylbenzoylamide, DEET), antiepileptics (carbamazepine), central nervous system stimulant (caffeine), lipid modifying agent (clofibric acid), non-steroidal anti-inflammatory drugs (ibuprofen, naproxen and diclofenac sodium salt) and antimicrobial agents (triclosan and triclocarban). The detection and qualification of the target compounds were performed by ultra-high performance liquid chromatography-tandem mass spectrometry. The optimized mobile phases were methanol as organic phase, 0. 3% (v/v) formic acid-5 mmol/L ammonium acetate for positive electrospray ionization (ESI+) and 5 mmol/L ammonium acetate for ESI- as inorganic phase. Water samples were concentrated by solid phase extraction at 2 mL/min, and all the target PPCPs were efficiently extracted at pH 7. The extracted PPCPs were eluted by the mixture of methanol and acetonitrile (1 : 1, v/v). The average recoveries of the target compounds in the spiked pure water samples ranged from 53.9% - 112%. The average recoveries of the target compounds ranged from 45.1% - 156.6% with the relative standard deviations ranged from 2.4% - 15.7%, in the surface water samples spiked at 100 ng/L. The surface water samples collected from Yu Hangtang River in Hangzhou were detected. The results showed that nine PPCPs were detected including caffeine that reached a maximum concentration of 550.7 ng/L. It proved that this analytical method is reliable and acceptable.

Abstract:An analytical method was developed for the determination of 34 illegally adulterated weight loss compounds in foods and dietary supplements by ultra high performance liquid chromatography-triple quadrupole mass spectrometry (UHPLC-MS/MS). A Waters CORTECS T3 column (100 mm×2.1 mm, 2.7 μm) was used with 0.1% (v/v) formic acid aqueous solution-acetonitrile containing 0.1% (v/v) formic acid as mobile phases by gradient elution. The compounds were detected by electrospray ion source in positive or negative mode with multiple reaction monitoring (MRM) mode. The calibration curves showed good linearity in the range of 0.5-10 μg/L for 29 compounds such as sibutramine, and in the range of 2.5-50 μg/L for five compounds such as chlorothiazide. The correlation coefficients (r) of the standard calibration curves for the 34 analytes were all greater than 0.99. The recoveries of the 29 compounds at spiked levels of 5, 10 and 20 μg/kg were in the range of 49.2%-136.2%, and the RSDs were 0.7%-15.0% (n=6). The recoveries of the five compounds at spiked levels of 25, 50 and 100 μg/kg were in the range of 51.5%-130.9%, and the RSDs were 0.8%-14.0% (n=6). The limits of detection (LODs) and limits of quantification (LOQs) were 5 μg/kg and 10 μg/kg for the 29 compounds, 25 μg/kg and 50 μg/kg for the five compounds, respectively. The method was successfully applied to the analysis of actual samples, and 12 compounds were checked out, which combated the illegal adulteration behavior effectively.

Abstract:This paper presents an application of ultrahigh-performance liquid chromatography and quadrupole Orbitrap high-resolution mass spectrometry (UHPLC-Q-Orbitrap HR MS) for the screening, confirmation and quantification of 11 antidiabetics in herbal medicines and dietary supplements. The mass spectrometer was operated in Full MS/dd-MS(2) (data-dependent MS(2)) mode. The full MS scan acquired data for identification and quantification, and dd-MS(2) scan obtained product ion spectra for confirmation. UHPLC-Q-Orbitrap MS quantification was achieved using matrix-matched standard calibration curves with phenacetin as internal standard. The method validation that included selectivity, sensitivity, calibration curve, accuracy and precision, recovery, matrix effect and stability was evaluated. The response showed good linear relationship with the concentrations of analytes over wide ranges (e.g., 0.0004-1 μg/g for metformin) with all the coefficients of correlation (r(2)) >0.9991. The detection limits (LODs) were in the range of 0.05-0.5 ng/g for different analytes. The recoveries yielded results higher than 74.3% for all compounds. The accuracy was in the range of -6.75 to 3.85%, while the intra- and inter-day precision ranged from 0.048 to 11.5%. Among 63 batches of herbal medicines and 34 batches of dietary supplements samples, 7 batches of dietary supplements were positive, while all the herbal medicines were negative. Overall, the novel UHPLC-Q-Orbitrap has demonstrated great performance for identification, confirmation and quantification of antidiabetics in herbal medicines and dietary supplements, ensuring food safety and public health.

Abstract:Objective:To establish a method for simultaneous determination of three consitituents in AEC Tablet containing as- pirin,ethenzamide and caffeine.Method:Inertsil-ODS column is used.The mobile phase is composed of methanol -0.1% di- ethylamine-acetic acid(40:56:4)with flow rate of 1.0mL/min.The detection wavelength is 280nm.Results:The linear re- sponse range were 120-480μg/mL for aspirin(r=0.9998),40-160μg/mL for ethenzamide(r=0.9998),20-80μg/mL for caf- feine(r=0.9997).The mean recoveries of aspirin,ethenzamide,caffeine were 100.3%,100.7%,99.7% respectively.Conclu- sion:The method is simple,quick and suitable for determination of these three constituents in compound preparation simulta- neously.

Abstract:A simple and sensitive method was developed for determination of illegal adulterants (yohimbine, sildenafil, vardenafil and tadalafil) in dietary supplements by HPLC-MS/MS. The separation was achieved on a C-18 column with the mobile phase consisting of acetonitrile and 0.1% acetic acid aqueous solution with a gradient elution at a flow rate of 0.5 mL/min. The analytes were quantified and identified by two characteristic transitions using the multiple-reaction monitoring mode. The recoveries of the analytes ranged from 77.5 to 109.3% with the RSD less than 8.1% (n = 6). The method has been successfully applied to screen illegal adulterations of natural dietary supplements.

Abstract:Cardiovascular disorders are among the leading causes of death worldwide, especially hypertension, a silent killer syndrome requiring multiple drug therapy for appropriate management. Hydrochlorothiazide is an extensively utilized thiazide diuretic that combines with several antihypertensive drugs for effective treatment of hypertension. In this study, sustainable, innovative and accurate high performance liquid chromatographic methods with diode array and tandem mass detectors (HPLC–DAD and LC–MS/MS) were developed, optimized and validated for the concurrent determination of Hydrochlorothiazide (HCT) along with five antihypertensive drugs, namely; Valsartan (VAL), Amlodipine besylate (AML), Atenolol (ATN), Amiloride hydrochloride (AMI), and Candesartan cilextil (CAN) in their diverse pharmaceutical dosage forms and in the presence of Chlorothiazide (CT) and Salamide (DSA) as HCT officially identified impurities. The HPLC–DAD separation was achieved utilizing Inertsil ODS-3 C 18 column (250 × 4.6 mm, 5 μm) attached with photodiode array detection at 225.0 nm. Gradient elution was performed utilizing a mixture of solvent A (20.0 mM potassium dihydrogen phosphate, pH 3.0 ± 0.2, adjusted with phosphoric acid) and solvent B (acetonitrile) at ambient temperature. Linearity ranges were 0.1–100.0 μg/mL for HCT, VAL, AML and CAN, 0.05 –100.0 μg/mL for both ATN and AMI and 0.05–8.0 μg/mL for both CT and DSA. Additionally, this work describes the use of liquid chromatography–electrospray–tandem mass spectrometry for the accurate detection and quantification of the impurities; CT and DSA in the negative mode utilizing triple quadrupole mass spectrometry. The linearity ranges for those impurities were 1.0–200.0 ng/mL and 5.0–200.0 ng/mL for CT and DSA, respectively. Developed methods' validation was achieved in accordance with International Conference on Harmonization (ICH) guidelines. Upon applying liquid chromatographic techniques for the drug analysis, a green and sustainable assessment have to be handled due to the consumption of energy and many solvents. Through the use of the HEXAGON, Analytical Greenness (AGREE) and White Analytical Chemistry (WAC) tools, greenness and sustainability have been statistically assessed. The optimized HPLC–DAD and LC–MS/MS methods were fast, accurate, precise, and sensitive, and consequently could be applied for conventional analysis and quality control of the proposed drugs in their miscellaneous dosage forms for the purpose of reducing laboratory wastes, time of the analysis time, effort, and cost.

Solid Phase Extraction-Ultra High Performance Liquid Chromatography-Tradem Mass Spectrometry for Simultaneous Determination of 13 Restricted Food Additives in Beverages

Abstract:In this work,an ultra-high performance liquid chromatography-mass spectrometry/mass spectrometry(UPLC-MS/MS)method was established for the separation and determination of 13 restricted food additives,including 3sweeteners,5preservatives and 5pigments in beverage samples.The target compounds were separated in 10minutes on a Waters BEH C18column(50×2.1mm,1.7μm)in gradient elution mode using HPLC methanol and 10mmol/L ammonium acetate aqueous solution(with 0.1%acetic acid)as mobile phases with a flow-rate of 0.2mL/min at 35℃,and were then confirmed and quantified by mass spectrum via positive electrospray ionization(ESI+)for pigments and via negative electrospray ionization(ESI-)for sweeteners and preservatives.Under multiple reaction monitoring(MRM)mode,regression equations revealed acceptable linearity(correlation coefficients0.990)across the working-standard range(2-500μg/L)for the 13analytes and the detection limits of the analytes were in the range of 0.20-6.79μg/L.The solid phase extraction was performed on a Waters HLB SPE column with the optimized pretreatment method,the recoveries in spiked samples(10μg/L,50μg/L and 100μg/L)ranged from 85.6%to 110.4% with the relative standard deviations less than 10%(n=5).With the advantages of accessibility,high sensitivity and good reproducibility,this simple method can be used to separate and determine 13food additives in beverage samples simultaneously.

Abstract:The abuse of herbicides has emerged as a great threat to food security. Herein, a low-background interference detection method based on UPLC-MS was developed for the simultaneous determination of glufosinate, glyphosate, and its metabolite aminomethylphosphonic acid (AMPA) in foods. Initially, this study proposed a simple and rapid pretreatment method, utilizing water extraction and PRiME HLB purification to isolate glyphosate, glufosinate, and AMPA from food samples. After the optimization of pretreatment conditions, the processed samples are then analyzed directly by ultra-performance liquid chromatography–tandem mass spectrometry (UPLC-MS) without pre-column derivatization. The method can effectively reduce interference from by-products of pre-column derivatization and background substrates of food sample, showing low matrix effects (ME) ranging from − 24.83 to 32.10%. Subsequently, the method has been validated by 13 kinds of food samples. The recoveries of the three herbicides in the food samples range from 84.2 to 115.6%. The limit of detection (LOD) is lower to 0.073 mg/kg, 0.017 mg/kg, and 0.037 mg/kg, respectively. Moreover, the method shows an excellent reproducibility with relative standard deviations (RSD) within 16.9%. Thus, the method can provide high trueness, reproducibility, sensitivity, and interference-free detection to ensure human health safety.

Abstract:This research introduces a novel approach for detecting sartan antihypertensive drug adulteration in herbal oral liquids using cotton fiber-supported liquid extraction (CF-SLE) combined with high-performance liquid chromatography-fluorescence detection (HPLC-FLD). Optimal extraction parameters were determined through systematic method development, establishing a sample solution with a pH of 3.0, using 200 mg of cotton fiber, ethyl acetate as the extraction solvent, and a solvent volume of 4 mL. These conditions demonstrated robust extraction efficiency and were further validated for precision and accuracy, with intra- and inter-day relative standard deviations consistently below 7.5 % and relative recoveries ranging from 88.5 % to 106.1 %. The method exhibited excellent linearity for sartans, with R² values greater than 0.993 across a concentration range of 10-2000 ng/mL. Detection limits were effectively established in the range of 2.6-3.1 ng/mL, indicating that the method's sensitivity is adequate for the intended screening purposes. This validated method was then applied to real sample analysis, confirming its potential for routine use in detecting illegal additives within complex herbal matrices, thereby ensuring consumer safety and supporting regulatory compliance.

Abstract:Adulteration of herbal supplements with synthetic drugs is illegal. A rapid and reliable method which utilizes direct analysis in real time mass spectrometry (DART-MS) was developed for the identification of seven synthetic antidiabetic drugs used as adulterants in herbal dietary supplements. The supplement sample was simply extracted with methanol/water by manually shaking several times and directly analyzed using DART-MS. The presence of synthetic drug adulterants was confirmed through the accurate m/z values and MS/MS data obtained via quadruple time of flight mass spectrometry (QTOF MS). Parameters for the DART source were systematically optimized, and the limits of detection (LODs) in herbal supplement matrices were measured. This method was successfully applied to examine five commercial herbal dietary supplements, and two of them proved to be adulterated with metformin without labeling.
Rapid Determination of Pharmaceuticals from Multiple Therapeutic Classes in Wastewater by Solid-Phase Extraction and Ultra-Performance Liquid Chromatography Tandem Mass Spectrometry

Abstract:A new analytical method utilizing ultra-performance liquid chromatography (UPLC) tandem mass spectrometry (MS/MS) has been developed to determine 16 pharmaceuticals from 8 therapeutic classes in wastewater: bezafibrate, clofibric acid, carbamazepine, caffeine, chloramphenicol, diclofenac, gemfibrozil, indomethacin, ketoprofen, mefenamic acid, metoprolol, nalidixic acid, N,N-diethyl-meta- toluamide, propranolol, sulpiride and trimethoprim. Key parameters of MS/MS, UPLC and solid phase extraction (SPE) were optimized. In general, recovery of target pharmaceuticals was over 70% for the wastewater effluent samples and 50% for the influent samples. The effects of matrix suppression, loss during the pretreatment as well as instrument variability were successfully corrected by two internal standards, and acceptable relative recovery was obtained. Target compounds were quantitatively analyzed using multiple reaction monitoring (MRM) mode, and the detection limits ranged from 0.3 to 20 ng/L. A detailed study, matrix effect in effluent wastewater was also present. The method was applied to detecting pharmaceuticals in the wastewater from three wastewater treatment plants (WWTPs) in Beijing, China and the results demonstrated that most target compounds were detectable in both the influent and effluent, with the mean concentrations ranging from 20.5 to 5775.6 ng/L and 4.6 to 418.6 ng/L, respectively.
Simultaneous Determination of Adulterants in Dietary Food Supplements Using Multivariate Data Analysis after Preconcentration with Novel Nanosorbents and Chromatographic Measurement

Abstract:Background: The increasing popularity of dietary supplements and, consequently, related adulteration emphasizes the rising need to examine the association of food supplements with fraud. Intentional or unintentional fraud in food supplements by hazardous chemicals compounds is a problem that many countries are struggling with. Much effort have been made to effectively and reliably control the quality of food supplements. Objective: Due to the importance of the subject, an analytical method for the simultaneous and reliable detection and quantitative determination of three key adulterants in dietary food supplements was developed. The proposed method benefits from analytical methods and multivariate calibration methods to progress the determination of adulterants in a complex matrix. Methods: HPLC assisted by multivariate curve resolution-alternating least square (MCR-ALS) analysis was used to detect adulterants in real samples after separation and preconcentration using novel mesoporous carbon nanoparticles. Solid-phase extraction (SPE) optimization was accomplished by central composite design (CCD). In order to obtain the best results, the MCR-ALS model was compared with the parallel factor analysis 2 (PARAFAC2) model and validated by estimation of linearity, detection limits, and recovery. Results: The detection limits and linear dynamics were calculated as 1.5, 4.27, and 4.77 µg/mL, and 1-50, 5-20, and 5-20 µg/mL for caffeine, ephedrine, and fluoxetine, respectively. Mean recovery for determination of caffeine, ephedrine, and fluoxetine using the developed method was reported as 101.75, 91.7, and 92.36, respectively. Conclusion: The results showed that to avoid negative health outcomes associated with the excessive consumption of adulterated food supplements releasing such products should be carefully regulated. The developed method was validated using statistical factors and showed acceptable and reliable results. Highlights: (1) The application of MCR-ALS coupled with HPLC-Diode-Array Detection data sets allowed the simultaneous identification and quantification of three key adulterants (caffeine, ephedrine, and fluoxetine) in dietary food supplements. (2) A small amount of the novel adsorbent was successfully used to preconcentrate the trace amounts of adulterants in samples. Abstract:In recent years, dietary supplements and herbal medicines are increasing in popularity all over the world. However, it is problematic that some manufacturers illegally included synthetic drugs in their products. Due to the extremely complex matrices of those products, most existing methods for screening illegal adulterations are time-consuming and liable to false positive. In this paper, a robust LC/MS/MS method for the high-throughput, sensitive and reliable determination of illegal adulterations from herbal medicines and dietary supplements was established. Minimal LC separation was employed and MRM was used to simultaneously monitor the three transitions under their respective optimal collision energy for each compound. Positive results were determined only if well-defined peaks appeared at all of the three transitions and the ratios among the peak areas were within given threshold. In this study, the method had been applied for the screening of nine most commonly adulterated therapeutic substances, such as sildenafil (Viagra) and famotidine, and the lower limits of detection of these compounds ranged from 0.05 to 1.5 ng/ml. Little sample preparation was needed for this method and the analysis time was less than 5 min/sample. The reliability has been demonstrated by the test with blank matrix. Over 200 products that were under suspicion by SDA of China had been assayed and till now no false negative or positive result was found. This method is rapid, simple, reliable and capable of screening multiple adulterants in one run.

Abstract:Various types of milk powder purportedly providing diverse health functions have emerged with the growth of the country's elderly population. Some manufacturers illegally add chemical drugs to their products to achieve their reported benefits, which poses a threat to consumer health. The existing standard methods are inapplicable to such complex sample matrices and require testing based on functional claims and classification. These limitations not only consume manpower and resources but also seriously impede daily regulatory efforts to detect unknown risk substances. In this study, a high-throughput method for the screening and quantitative analysis of 300 illegally added chemical drugs in functional milk powder and an identification strategy for unknown structural analogues were established using Zeno SWATH® data-independent acquisition (DIA) ultra-high performance liquid chromatography-high resolution mass spectrometry (UHPLC-HRMS) technology combined with a QuEChERS sample purification method. The QuEChERS purification process was developed according to the characteristics of milk powder matrix. The supernatant was separated on a Kinetex F5 column (100 mm×3.0 mm, 2.6 μm) by gradient elution using 5 mmol/L ammonium formate aqueous solution (0.1% (v/v) formic acid, ) and methanol-acetonitrile (1∶1, v/v) as mobile phases. The method was validated in terms of selectivity, linearity, limits of detection and quantification (LODs and LOQs, respectively), matrix effect, accuracy, and precision. Based on a screening database for the 300 target substances, electron-activated dissociation (EAD) fragmentation was applied to obtain rich secondary MS fragmentation information, and unknown structural analogues were identified and confirmed through fragment attribution analysis. The results indicated that all compounds had good linear relationships in certain ranges with correlation coefficients >0.99. The LODs and LOQs were 0.04-2.7 and 0.2-8.0 μg/kg, respectively. The average recoveries at three spiked levels were in the range of 73.1%-125.2%, and the relative standard deviations were ≤14.8% (n=6). When the developed method was applied to detect illegally added chemicals in 60 functional milk powder samples, it detected benzoguanidine and sildenafil and successfully identified ethylphenidate, which is the structural analogue of an amphetamine. The proposed method is simple, sensitive, and accurate; thus, it may have practical application value for the daily supervision and law enforcement of milk powders with reported health functions.

Abstract:A sensitive method for the determination of three new pesticides (trifludimoxazin, pyraziflumid and picarbutrazox) in cereals, vegetables and fruits samples has been developed using ultra-performance liquid chromatography/tandem mass spectrometry. The analytes were extracted using acetonitrile containing 1% formic acid, and then the extracts were purified using primary secondary amine and octadecylsilane. The compounds were eluted in < 3.5 min using an electrospray ionization source in positive mode. Good linearities (R2 > 0.9916) were acquired for the analytes in all matrices. The limit of detection and quantification were 0.05–5 μg kg−1 and 10 μg kg−1 for all the three target compounds, respectively. The recoveries were 74.4–105.1% and the relative standard deviations values were lower than 12.4% at three spiked levels (10, 100 and 500 μg kg−1). The results indicated that the method is reliable for monitoring the three pesticides in cereals, vegetables and fruits.
