Abstract:Objective To establish a GC-MS method for the detection method of sedative-hypnotic drugs illegally added in health foods and traditional Chinese medicines.Methods The solvent ultrasonic extraction or the liquid-liquid extraction was adopted to extract and enrich the samples.The qualitative analysis was carried out by GC-MS and the Scan mode.The analytical capillary column was DP-5 ms(30 m×0.25 mm,0.25 μm).The initial temperature was set at 120 ℃(1 min),then was risen to 230 ℃with 10 ℃/min,to 270 ℃(20 min) by 15 ℃/min by temperature programming.The EI ion source was selected and the solvent was delayed by 2.5 min.The MS scanning was set at the range of 40-350 amu.Results Under the selected chromatographic conditions,14 kinds of sedative-hypnotic drugs could get a better separation anf the detection was rapidly completed within 30 min.Conclusion This method is rapid,simple,accurate and suitable for the fast qualitative detection in crackdown on counterfeit goods.

Abstract:With the rapid developments of food industry and the improvement of living condi-tions,the phenomenon of economically motivated adulteration( EMA)is an extremely issue of concern due to people’s increasing requirements for food safety. Nowadays,the major EMA incidents include substitution of varieties,mislabeling geographical origin,interfusion of poor quality,incorporation of inferiority, etc. Therefore, an efficient authentication technology needs to be established. Metabolomics is defined as the quantitative and qualitative analyses of large quantity of metabolites as a whole in a given sample,which has huge advantages in non-targeted analysis. Metabolomics has been adopted extensively by both domestic and abroad researchers to study the food safety problems,which provided an alternative research tool to identify the EMA incidents. In this paper,the progress and applications of metabolomics in identifying food varieties,geographical origin,quality and counterfeits have been reviewed, which provide technical support to ensure food quality and safety and protect the interests of consumers.

Abstract:Adrenergic receptor agonists are a class of adrenergic transmitters analogues that can bind and stimulate adrenergic receptors, which always have been illegally used in the animal husbandry since they can promote growth, reduce fat deposition, and improve lean meat rate. Thus, detecting these agonist residues is of great significance to ensure food safety and human health. Here, we proposed a comprehensive and critical review, firstly introducing the typical adrenergic receptor agonists that occurred in animal-derived foods. Then we summarized and discussed the recent residues detection methods including immunoassays, receptor-based assays, liquid chromatography-tandem mass spectrometry, gas chromatography-tandem mass spectrometry, and electrochemical methods. We highlighted the immunoassays due to their merits of simple operation, low cost, and high sensitivity for screening purpose. Lastly, we prospected the future development of adrenergic receptor agonist residues detection method.

Abstract:The increased availability and use of botanical dietary supplements and herbal remedies among consumers has been accompanied by an increased frequency of adulteration of these products with synthetic pharmaceuticals. Unscrupulous producers may add drugs and analogues of various classes, such as phosphodiesterase type 5 (PDE-5) inhibitors, weight loss, hypoglycemic, antihypertensive and anti-inflammatory agents, or anabolic steroids, to develop or intensify biological effects of dietary supplements or herbal remedies. The presence of such adulterated products in the marketplace is a worldwide problem and their consumption poses health risks to consumers. Analytical methods that allow rapid and reliable testing of dietary supplements for the presence of synthetic drugs are needed to address such fraudulent practices. Mass spectrometry (MS) and hyphenated techniques such as liquid chromatography–mass spectrometry (LC–MS) and gas chromatography–mass spectrometry (GC–MS) have become primary tools in this endeavor. The present review critically assesses the role and summarizes the applications of MS in the analysis of pharmaceutical adulterants in botanical dietary supplements and herbal remedies. The uses of MS techniques in detection, confirmation, and quantification of known pharmaceutical adulterants as well as in screening for and structure elucidation of unexpected adulterants and novel designer drugs are discussed.

A novel standard‐free detection of adulteration method for sildenafil derivatives in dietary supplements

Abstract:The rapid and accurate detection of illegal adulteration of chemical drugs into dietary supplements is a big challenge in the food chemistry field. Detection of compounds without a standard reference is even more difficult; however, this is a common situation. Here in this study, a novel "standard‐free detection of adulteration" (SFDA) method was proposed and phosphodiesterase‐5 inhibitor derivatives were used as an example to figure out the possibility and reliability of this SFDA method. After analysis by quadrupole coupled time of flight–tandem mass spectrometry detection and multivariable statistics, six common fragment ions were chosen to indicate whether adulteration was present or not, while 20 characteristic fragment ions indicated whether adulteration was by nitrogen‐containing heterocycles or by anilines. Furthermore, the quantitative methods were conducted by high‐performance liquid chromatography–tandem mass spectrometry. In a word, this strategy allows for a quick determination of dietary supplement adulteration without any need for standard materials, improving the efficacy of food safety testing.

Abstract:Food adulteration and illegal supplementations have always been one of the major problems in the world. The threat of food adulteration to the health of consumers cannot be ignored. Food of questionable origin causes economic losses to consumers, but the potential health risks cannot be ignored. However, the traditional detection methods are time-consuming and complex. This review mainly discusses the types of adulteration and technologies used to detect adulteration. Matrix-assisted laser desorption ionization-time-of-flight mass spectrometry (MALDI-TOF MS) is also emphasized in the detection of adulteration and authenticity of origin analysis of various types of food (milk, meat, edible oil, etc.), and the future application direction and feasibility of this technology are analyzed. On this basis, MALDI-TOF MS was compared with other detection methods, highlighting the advantages of this technology in the detection of food adulteration. The future development prospect and direction of this technology are also emphasized.

Abstract:Background: The misuse of animal-derived stimulants in food is becoming increasingly common, and mass spectrometry (MS) is used extensively for their detection and analysis. There is a growing demand for abused-substances detection, highlighting the need for systematic studies on the advantages of MS-based methods in detecting animal-derived stimulants. Objective: We reviewed the application of chromatography-mass spectrometry to the screening and detection of food stimulants of animal origin. Specifically, we analyzed four common animal sources of synthetic steroids, β-receptor agonists, zearalenol (ZAL), and glucocorticoids. We also explored the potential of using chromatography-mass spectrometry to detect and analyze animal-derived foods. Methods: We searched and screened the Web of Science and Google Scholar databases until April 2023. Our inclusion criteria included a publication year within the last 5 years, publication language of English, and the research fields of food analysis, environmental chemistry, and polymer science. Our keywords were "mass spectrometry," "anabolic androgenic steroids," "β-2agonists," "glucocorticoids," "zearalenone," and "doping." Results: Although traditional techniques such as thin-layer chromatography and enzyme-linked immunoassays are simple, fast, and suitable for the initial screening of bulk products, they are limited by their relatively high detection limits. Among the methods based on MS, gas chromatography-mass spectrometry and liquid chromatography-tandem mass spectrometry are the most widely used for detecting food doping agents of animal origin. However, a sensitive method with high repeatability and a short analysis time for a large number of samples is still required. Advances in MS have enabled the detection of extremely low concentrations of these substances. Combining different techniques, such as high-resolution mass spectrometry, ultra-high performance liquid chromatography-tandem mass spectrometry, gas chromatography-combustion-isotope ratio mass spectrometry, ultra-high performance liquid chromatography-high resolution mass spectrometry, and two-dimensional chromatography, offers significant advantages for detecting trace illicit drugs in animal-derived foods. Due to advances in assay technology and sample preparation methods, sample collection and storage methods such as dried blood spots, dried urine spots, and volumetric absorptive microsampling are increasingly accepted because of their increased stability and cost-effectiveness. Significance: MS significantly improves the efficiency of detecting doping agents of animal origin. With the continuous development of MS technology, its application in the fields of doping detection and the analysis of doping agents of animal origin is expected to become more extensive.

Abstract:Spurious/Falsely-labeled/Falsified/Counterfeit (SFFC)drugs have become a major threat to public health, especially in rural areas of developing countries.The goal of this review is to provide an overview of rapid detection technologies for counterfeits recently reported, such as Near Infrared Spectroscopy, Near Infrared Chemical Imaging, Raman Spectroscopy, X-Ray Fluorescence, X-RayPowder Diffraction, Ion Mobility Spectrometry, Ion MobilityMass Spectrometry,Isotope Ratio Mass Spectrometry and visual analytical methods The advantages of each of these detection methods are introduced. Examples of characterization of SFFC drugs using the detection technology mentioned are presented. In addition, new characteristics and trends of SFFC drugs are listed and the solution is discussed.

Simultaneous Determination of Adulterants in Dietary Food Supplements Using Multivariate Data Analysis after Preconcentration with Novel Nanosorbents and Chromatographic Measurement

Abstract:Background: The increasing popularity of dietary supplements and, consequently, related adulteration emphasizes the rising need to examine the association of food supplements with fraud. Intentional or unintentional fraud in food supplements by hazardous chemicals compounds is a problem that many countries are struggling with. Much effort have been made to effectively and reliably control the quality of food supplements. Objective: Due to the importance of the subject, an analytical method for the simultaneous and reliable detection and quantitative determination of three key adulterants in dietary food supplements was developed. The proposed method benefits from analytical methods and multivariate calibration methods to progress the determination of adulterants in a complex matrix. Methods: HPLC assisted by multivariate curve resolution-alternating least square (MCR-ALS) analysis was used to detect adulterants in real samples after separation and preconcentration using novel mesoporous carbon nanoparticles. Solid-phase extraction (SPE) optimization was accomplished by central composite design (CCD). In order to obtain the best results, the MCR-ALS model was compared with the parallel factor analysis 2 (PARAFAC2) model and validated by estimation of linearity, detection limits, and recovery. Results: The detection limits and linear dynamics were calculated as 1.5, 4.27, and 4.77 µg/mL, and 1-50, 5-20, and 5-20 µg/mL for caffeine, ephedrine, and fluoxetine, respectively. Mean recovery for determination of caffeine, ephedrine, and fluoxetine using the developed method was reported as 101.75, 91.7, and 92.36, respectively. Conclusion: The results showed that to avoid negative health outcomes associated with the excessive consumption of adulterated food supplements releasing such products should be carefully regulated. The developed method was validated using statistical factors and showed acceptable and reliable results. Highlights: (1) The application of MCR-ALS coupled with HPLC-Diode-Array Detection data sets allowed the simultaneous identification and quantification of three key adulterants (caffeine, ephedrine, and fluoxetine) in dietary food supplements. (2) A small amount of the novel adsorbent was successfully used to preconcentrate the trace amounts of adulterants in samples. (3) This method benefits from the chemometrics tools and experimental design to significantly reduce the use of toxic solvents and complicated instruments to propose a less time-consuming method for quantification of multicomponents in the presence of uncalibrated interferents.

Abstract:In recent years, people pay more and more attention to food quality and safety, which are significantly relating to human health. Food adulteration is a world-wide concerned issue relating to food quality and safety, and it is difficult to be detected. Modern detection techniques (high performance liquid chromatography, gas chromatography-mass spectrometer, etc.) can accurately identify the types and concentrations of adulterants in different food types. However, the characteristics as expensive, low efficient and complex sample preparation and operation limit the use of these techniques. The rapid, nondestructive and accurate detection techniques of food adulteration is of great and urgent demand. This paper introduced the principles, advantages and disadvantages of the nondestructive analysis techniques and reviewed the applications of these techniques in food adulteration screen in recent years. Differences among these techniques, differences on data interpretation and future prospects were also discussed.

Abstract:Adulteration of herbal supplements with synthetic drugs is illegal. A rapid and reliable method which utilizes direct analysis in real time mass spectrometry (DART-MS) was developed for the identification of seven synthetic antidiabetic drugs used as adulterants in herbal dietary supplements. The supplement sample was simply extracted with methanol/water by manually shaking several times and directly analyzed using DART-MS. The presence of synthetic drug adulterants was confirmed through the accurate m/z values and MS/MS data obtained via quadruple time of flight mass spectrometry (QTOF MS). Parameters for the DART source were systematically optimized, and the limits of detection (LODs) in herbal supplement matrices were measured. This method was successfully applied to examine five commercial herbal dietary supplements, and two of them proved to be adulterated with metformin without labeling.

Abstract:In recent years, dietary supplements and herbal medicines are increasing in popularity all over the world. However, it is problematic that some manufacturers illegally included synthetic drugs in their products. Due to the extremely complex matrices of those products, most existing methods for screening illegal adulterations are time-consuming and liable to false positive. In this paper, a robust LC/MS/MS method for the high-throughput, sensitive and reliable determination of illegal adulterations from herbal medicines and dietary supplements was established. Minimal LC separation was employed and MRM was used to simultaneously monitor the three transitions under their respective optimal collision energy for each compound. Positive results were determined only if well-defined peaks appeared at all of the three transitions and the ratios among the peak areas were within given threshold. In this study, the method had been applied for the screening of nine most commonly adulterated therapeutic substances, such as sildenafil (Viagra) and famotidine, and the lower limits of detection of these compounds ranged from 0.05 to 1.5 ng/ml. Little sample preparation was needed for this method and the analysis time was less than 5 min/sample. The reliability has been demonstrated by the test with blank matrix. Over 200 products that were under suspicion by SDA of China had been assayed and till now no false negative or positive result was found. This method is rapid, simple, reliable and capable of screening multiple adulterants in one run.

Abstract:Obesity is a chronic disease associated with serious health problem such as metabolic syndrome, diabetes, hypertension, and cardiovascular disease. Recently, anti-obesity drugs, designer analogues and natural weight-loss ingredients have been found in dietary supplements for rapid weight loss or maintenance. In addition, structurally modified designer analogues are being continuously synthesized to avoid governmental inspections. 25 illegal compounds were assayed using a liquid chromatography coupled with photodiode array detector (LC-PDA) and tandem mass spectrometry (UPLC-MS/MS) analyses. A simultaneous determination method was established and fully validated. In 76 of 370 samples, nine different adulterants were detected including yohimbine, followed by β-phenylethylamine (β-PEA), sibutramine, sennoside A and B, β-methyl phenylethylamine (BMPEA), fluoxetine, ephedrine, phenolphthalein, and icariin. And 12 adulterated products contained a mixture of two or more adulterants. Adequate use of the existing regulation and the development of alternative methods for detecting adulterants are necessary consumers from consuming dietary supplements adulterated with illegal compounds.

Abstract:Main detection techniques of illegal chemical additives in food were reviewed in this paper. The latest developments in detection methods were briefly introduced. Some sample pretreatment tech- niques were also introduced. According to the chemical properties of different kinds of illegal chemical additives, some modern analysis method were developed to detect them, such as spectrometry, gas chro- matography/mass, liquid chromatography, liquid chromatography/mass spectrometry, ion chromatogra- phy, capillary electrophoresis and so on. The enzyme-linked immunosorbent assay,raman spectrum and biosensors were studied by many researchers and are potential for replacing complex analysis methods.

Abstract:In the last few years, the consumption of dietary supplements, especially those having plants as ingredients, has been increasing due to the common idea that they are natural products posing no risks to human health. In the European Union and the United States, dietary supplements are legally considered as foods/special category of foods, thus are not being submitted to any safety assessment prior to their commercialization. Among the issues that can affect safety, adulteration by the illegal addition of pharmaceutical substances or their analogs is of major concern since unscrupulous producers can falsify these products to provide for quick effects and to increase sales. This review discusses the various classes of synthetic drugs most frequently described as being illegally added to dietary supplements marketed for weight loss, muscle building/sport performance and sexual performance enhancement. Information regarding regulation and consumption is also presented. Finally, several conventional and advanced analytical techniques used to detect and identify different adulterants in dietary supplements and therefore also in foods, with particular emphasis on plant food supplements, are critically described. This review demonstrates that dietary supplement adulteration is an emerging food safety problem and that an effective control by food regulatory authorities is needed to safeguard consumers.

Abstract:Various types of milk powder purportedly providing diverse health functions have emerged with the growth of the country's elderly population. Some manufacturers illegally add chemical drugs to their products to achieve their reported benefits, which poses a threat to consumer health. The existing standard methods are inapplicable to such complex sample matrices and require testing based on functional claims and classification. These limitations not only consume manpower and resources but also seriously impede daily regulatory efforts to detect unknown risk substances. In this study, a high-throughput method for the screening and quantitative analysis of 300 illegally added chemical drugs in functional milk powder and an identification strategy for unknown structural analogues were established using Zeno SWATH® data-independent acquisition (DIA) ultra-high performance liquid chromatography-high resolution mass spectrometry (UHPLC-HRMS) technology combined with a QuEChERS sample purification method. The QuEChERS purification process was developed according to the characteristics of milk powder matrix. The supernatant was separated on a Kinetex F5 column (100 mm×3.0 mm, 2.6 μm) by gradient elution using 5 mmol/L ammonium formate aqueous solution (0.1% (v/v) formic acid, ) and methanol-acetonitrile (1∶1, v/v) as mobile phases. The method was validated in terms of selectivity, linearity, limits of detection and quantification (LODs and LOQs, respectively), matrix effect, accuracy, and precision. Based on a screening database for the 300 target substances, electron-activated dissociation (EAD) fragmentation was applied to obtain rich secondary MS fragmentation information, and unknown structural analogues were identified and confirmed through fragment attribution analysis. The results indicated that all compounds had good linear relationships in certain ranges with correlation coefficients >0.99. The LODs and LOQs were 0.04-2.7 and 0.2-8.0 μg/kg, respectively. The average recoveries at three spiked levels were in the range of 73.1%-125.2%, and the relative standard deviations were ≤14.8% (n=6). When the developed method was applied to detect illegally added chemicals in 60 functional milk powder samples, it detected benzoguanidine and sildenafil and successfully identified ethylphenidate, which is the structural analogue of an amphetamine. The proposed method is simple, sensitive, and accurate; thus, it may have practical application value for the daily supervision and law enforcement of milk powders with reported health functions.

Mycotoxin and marine biotoxins which are two kinds of natural toxic substances associated with food safety, have the characteristics of strong toxicity and pollute widely and a great harm to human health. Among the detection means of mycotoxin and marine biotoxin, the technique of liquid chromatography coupled with mass spectrometry (LC-MS) is widely used for its sensitive and efficient characteristics. Abstract:BackgroundHerbal products have been commonly used all over the world for centuries. Its products have gained remarkable acceptance as therapeutic agents for a variety of disorders. However, following recent research disclosing discrepancies between labeling and actual components of herbal products, there is growing concern about the efficacy, quality and safety of the products. The admixture and adulteration of herbal medicinal products pose a risk of serious health compromise and the well-being of the consumers. To prevent adulteration in raw ingredients and final herbal products, it is necessary to use approaches to assess both genomes as well as metabolomics of the products; this offers quality assurance in terms of product identification and purity. The combinations of molecular and analytical methods are inevitable for thorough verification and quality control of herbal medicine.Methods and ResultsThis review discusses the combination of DNA barcoding, DNA metabarcoding, mass spectroscopy as well as HPLC for the authentication of herbal medicine and determination of the level of adulteration. It also discusses the roles of PCR and real-time PCR techniques in validating and ensuring the quality, purity and identity of the herbal products.ConclusionsIn conclusion, each technique has its own pros and cons, but the cumulative of both the chemical and molecular methods is proven to be the best strategy for adulteration detection. Moreover, CRISPR diagnosis tools equipped with multiplexing techniques may be implemented for screening adulteration from herbal drugs, this will play a crucial role in herbal product authentication in the future.Graphical abstract
