Abstract:A decarboxylative cross-coupling reaction of α-amino acids with nitrones via visible-light-induced photoredox catalysis has been established for easy access to β-amino hydroxylamines and vicinal diamines with structural diversity, which is featured with simple operation, mild conditions, readily available α-amino acids, and a broad scope of nitrone substrates. The application of this protocol can furnish efficient synthetic strategies for some valuable vicinal diamine-containing molecules.

Abstract:A novel and efficient decarboxylative alkynylation of N-(acetoxy)phthalimides of α-amino acids with terminal alkynes has been developed by merging photoredox with copper catalysis at room temperature, and the reaction provided the valuable propargylamines in good to excellent yields with assistance of [Ru(bpy)3]Cl2/CuI and visible light. The simple protocol, mild reaction conditions, and high efficiency of this method make it an important strategy for synthesis of diverse molecules containing amino acid fragments.

Abstract:The asymmetric cross-coupling reaction is developed as a straightforward strategy toward 1,1-diaryl alkanes, which are a key skeleton in a series of natural products and bioactive molecules in recent years. Here we report an enantioselective benzylic C(sp(3))-H bond arylation via photoredox/nickel dual catalysis. Sterically hindered chiral biimidazoline ligands are designed for this asymmetric cross-coupling reaction. Readily available alkyl benzenes and aryl bromides with various functional groups tolerance can be easily and directly transferred to useful chiral 1,1-diaryl alkanes including pharmaceutical intermediates and bioactive molecules. This reaction proceeds smoothly under mild conditions without the use of external redox reagents.

Abstract:Tetrahydropyridazines constitute an important structural motif found in numerous natural products and pharmaceutical compounds. Herein, we report an aminoacylation reaction of alkenes that enables the synthesis of 1,4,5,6-tetrahydropyridazines through cooperative N-heterocyclic carbene (NHC) and photoredox catalysis. This approach involves the 6-endo-trig cyclization of N-centered hydrazonyl radicals, generated via single-electron oxidation of hydrazones, followed by a radical-radical coupling step. The mild process tolerates a wide range of common functional groups and affords a variety of tetrahydropyridazines in moderate to high yields. Preliminary investigations using chiral NHC catalysts demonstrate the potential of this protocol for asymmetric radical reactions.

Abstract:Amines are a quintessential moiety in bioactive molecules, pharmaceuticals and organic materials. Transition-metal-catalysed C–N coupling of aryl electrophiles has been established as a powerful and reliable method for amine synthesis. However, the analogous C–N coupling of alkyl electrophiles is largely under-developed due to the decomposition of metal alkyl intermediates by β-hydrogen elimination and difficulty in C(sp3)–N reductive elimination. Here, we provide a general strategy for amination of alkyl electrophiles by merging photoredox and copper catalysis. Photoredox catalysis allows the use of alkyl redox-active esters, recently established as a superior class of alkyl electrophiles, whereas copper catalysis enables C(sp3)–N cross-coupling. Decarboxylative amination can be used for the synthesis of a diverse set of alkyl anilines with high chemoselectivity and functional-group compatibility. Rapid functionalization of amino acids, natural products and drugs is demonstrated.

Abstract:Radical-mediated Hydrogen Atom ion of Csp3-H bonds has become a powerful tool for the asymmetric functionalization of organic feedstocks. Here, we present an asymmetric synthesis of α-aryl amides via carbamoylation of alkylarenes with isocyanates as electrophiles. The synergistic combination of a photoredox and a chiral nickel-catalyst, enables the use of readily available and neutral reagents under mild reaction conditions and provides straightforward access to pharmacologically relevant motifs in enantiomerically pure form.

Abstract:Herein, a direct cross-dehydrogenative C-H amination of indoles has been successfully achieved, enabled by the merger of photocatalysis with nickel catalysis. This developed process does not require stoichiometric oxidants and prefunctionalization of amine partners, providing a concise platform for C-N bond formation. Moreover, the synthetic practicality of this transformation was well revealed by its high step- and atom-economy, high reaction efficiency, and broad functional group tolerance.

Abstract:Direct acylation of amides at C(sp3)–H sites from aldehydes is one of the most convenient methods to access highly valuable keto-amides, yet it still remains challenging and less development. Herein, we reported a photoredox cooperative NHC/Pd-catalyzed method that enables C(sp3)–H acylation for quick construction of keto-amide derivatives from the readily available amides and aldehydes. This work demonstrates the generation of ketyl radicals from aldehydes under NHC/Pd catalysis and alkyl radicals via 1,5-hydrogen atom transfer from aryl radicals under photoredox/Pd catalysis, thus providing two- and three-component reactions for the synthesis of α- and γ-keto-amides through radical−radical coupling reactions. This work features with good compatibility with a large variety of functional groups as demonstrated by more than 75 examples, including complex bioactive compounds. Mechanistic studies including several control reactions shed light on the mechanism of this photoredox cooperative NHC/Pd-catalyzed reaction.

Abstract:An iridium photoredox catalyst in combination with a phosphoric acid catalyzes the decarboxylative alpha-aminoalkylation of natural and unnatural alpha-amino acid-derived redox-active esters (N-hydroxyphthalimide esters) with a broad substrate scope of N-heteroarenes at room temperature under irradiation. Dipeptide- and tripeptide-derived redox-active esters are also amenable substrates to achieve decarboxylative insertion of a N-heterocycle at the C-terminal of peptides, yielding molecules that have potential medicinal applications. The key factors for the success of this reaction are the following: use of a photoredox catalyst of suitable redox potential to controllably generate alpha-aminoalkyl radicals, without overoxidation, and an acid cocatalyst to increase the electron deficiency of N-heteroarenes.

Abstract:A photoredox catalytic asymmetric method has been established for the installation of both aliphatic and aromatic side chains and the introduction of deuterium into the chiral methyleneoxazolidinone simultaneously. Efficient coupling of readily available boronic acids with the chiral auxiliary delivers structurally diverse α-deuterated α-amino acid derivatives with a high level of diastereoselectivity and deuteration.

Abstract:In this work, we have established a photoredox/Ti dual‐catalyzed dehydroxylation of cyclobutanone oximes. The resulting γ‐cyanoalkyl radicals undergo a cascade radical addition/cyclization with N‐acrylamides to afford cyanoalkyl substituted oxoindolines under mild conditions. This reaction also exhibits broad substrate scope and good functional group tolerance. Furthermore, it is applicable to late‐stage functionalization of complex molecules and gram‐scale synthesis, which illustrates its potential value in organic synthesis.

Abstract:In view of the widespread significance of amide functional groups in organic synthesis and pharmaceutical studies, an efficient and practical synthetic protocol that avoids the use of stoichiometric activating reagents or metallic reductants is highly desirable. A straight-forward pathway to access amides from abundant chemical feedstock would offer a strategic advantage in the synthesis of complex amides. We herein disclose a direct reductive amidation reaction using readily available aldehydes and nitroarenes enabled by photo-mediated hydrogen atom transfer catalysis. It avoids the use of metallic reductants and production of toxic chemical waste. While aldehydes represent a classic class of electrophilic synthons, the corresponding nucleophilic acyl radicals could be directly accessed by photo hydrogen atom transfer catalysis, enabling polarity inversion. Our method provides an orthogonal strategy to conventional amide couplings, tolerating nucleophilic substituents such as free alcohols and sensitive functional groups to amines such as carbonyl or formyl groups. The synthetic utilization of this reductive amidation is demonstrated by the late-stage modification of complex biologically active molecules and direct access of drug molecules leflunomide and lidocaine.

Abstract:A novel Pd/Cu-catalyzed chemoselective aerobic oxidative N-dealkylation/carbonylation reaction has been developed. Tertiary amines are utilized as a "reservoir" of "active" secondary amines in this transformation, which inhibits the formation of undesired by-products and the deactivation of the catalysts. This protocol allows for an efficient and straightforward construction of synthetically useful and bioactive (E)-α,β-unsaturated amide derivatives from easily available tertiary amines, olefins and CO.

Abstract:The functionalization of α-C(sp 3 )–H bonds in amines has become a focal point of contemporary research. Here, we report a new approach utilizing photocatalysis α-C(sp 3 )–H bond functionalization in alicyclic and aliphatic secondary amines facilitated by intramolecular 1,5-hydrogen atom transfer (HAT). This finding unlocks a sustainable method for rapidly constructing complex heterocyclics via cross-dehydrogenative C–C coupling of protected amines and nitrogen-containing heterocycles. This protocol boasts broad applicability to various substrates, exhibits tolerance to numerous functional groups, and supports the late-stage modification of drug molecules.

Abstract:Conjugation of amino acids to bioactive molecules has emerged as a promising strategy for optimizing pharmacological profiles of lead candidates in drug discovery. This study describes a photocatalytic Minisci‐type transfer reaction of N‐acyl amino acids into various N‐heterocycles. Notably, this protocol enables direct conjugation of amino acids into heterocyclic C–H bonds, eliminating the need for prefunctionalized substrates. A diverse array of N‐heterocycles, amino acids, oligopeptides, and drugs were used to demonstrate the potential of the proposed approach. In addition, the importance of this approach is demonstrated through its application in the DNA‐encoded library chemistry. Various synthetic transformations and preliminary mechanistic investigations were also explored.

Abstract:Organic thiocyanates are commonly used as an essential organic synthesis intermediate and widely present in various drug molecules and bioactive molecules. Herein, a copper and photoredox catalyzed thiocyanoalkylation reaction of terminal alkenes with NHPI esters and NH4SCN to access organicthiocyanates is described. This protocol employs more readily accessible NHPI ester as a radical precursor under redox-neutral conditions. Moreover, the reaction exhibits high compatibility with diverse primary, secondary and tertiary NHPI esters. And the latestage modification of some drugs and structurally complex natural products enables the potential application in the organic synthesis.

Abstract:A unique decarbonylation of an amino acid derivative catalytic system has been established via palladium-catalyzed C-C bond and C-N bond dual activations. By employing 8-aminoquinoline as the directing group, this transformation has been found to facilitate the high chemoselectivity to decarbonylation of amino acid derivatives rather than intramolecular deamination or cross-dehydrogenative coupling reactions. This method provides a straightforward avenue for constructing diverse functionalized amide compounds in good to excellent yields. We proposed a possible reaction pathway that may go through the C-C bond and C-N bond dual activations on the basis of the mechanistic studies.

Abstract:Herein, we describe a two‐component methodology developed for the synthesis of pyrimidine derivatives using tertiary alkylamines and amidines. Tertiary alkylamines serve as dual C2 synthons through copper‐catalyzed aerobic difunctionalization of the α,β‐C(sp3)‐H bonds. The process operates under mild conditions and uses atmospheric oxygen as the oxidant. Notably, this methodology yields up to 85% and shows a broad substrate scope. Mechanistic studies indicate that the annulation proceeds via a radical‐mediated oxidation and C‒N bond coupling process. This approach provides a pathway for synthesizing various heterocycles by employing tertiary alkylamines as dual C2 synthons.
